4Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. 5基于脉冲激励的地铁运营引起邻近建筑物内振动预测方法显示文摘根据振动系统传递特性,提出一种新的环境振动预测方法——实测脉冲传递函数预测方法。通过脉冲试验测得振源与邻近建筑物内测点的振动加速度,计算得到振动系统的振动响应传递函数;将实测的地铁运营振源的振动加速度代入传递函数公式,实现对建筑物内测点的振动加速度时程、频谱、1/3倍频程和z振级的预测。应用该方法预测隧道中心楼内测点振动加速度结果表明:隧道结构与隧道中心楼内测点振动加速度传递函数峰值频段为30~50Hz,隧道中心楼内振动加速度峰值频段为40~60Hz;土层和隧道中心楼基础结构对60Hz以上频段的振动具有很强的衰减作用,楼房基础结构有效地隔断了70Hz以上的高频振动。该方法可用于预测地铁运营引起的建筑物内振动。 Wang, Wenbin Liu, Weining Sun, Ning Li, Kefei Liu, Li 2012中国铁道科学2012,,4:12
6NIR-II ?uorescence in vivo confocal microscopy with aggregation-induced emission dots显示文摘Significantly reduced tissue scattering of fluorescence signals in the second near-infrared(NIR-Ⅱ,1,000–1,700 nm)spectral region offers opportunities for large-depth in vivo bioimaging.Nowadays,most reported works concerning NIR-II fluorescence in vivo bioimaging are realized by wide-field illumination and 2D-arrayed detection(e.g.,via InGaAs camera),which has high temporal resolution but limited spatial resolution due to out-of-focus signals.Combining NIR-II fluorescence imaging with confocal microscopy is a good approach to achieve high-spatial resolution visualization of biosamples even at deep tissues.In this presented work,a NIR-II fluorescence confocal microscopic system was setup.By using a kind of aggregation-induced emission(AIE)dots as NIR-II fluorescent probes,800 lm-deep 3D in vivo cerebrovascular imaging of a mouse was obtained,and the spatial resolution at 700 lm depth could reach 8.78 lm.Moreover,the time-correlated single photon counting(TCSPC)technique and femtosecond laser excitation were introduced into NIR-II fluorescence confocal microscopy,and in vivo confocal NIR-II fluorescence lifetime microscopic imaging(FLIM)of mouse cerebral vasculature was successfully realized.Wenbin Yu Bing Guo Hequn Zhang Jing Zhou Xiaoming Yu Liang Zhu Dingwei Xue Wen Liu Xianhe Sun Jun Qian 2019Science Bulletin2019,64,6:10
7The Assessment of Global Surface Temperature Change from 1850s:The C-LSAT2.0 Ensemble and the CMST-Interim Datasets显示文摘Based on C-LSAT2.0,using high-and low-frequency components reconstruction methods,combined with observation constraint masking,a reconstructed C-LSAT2.0 with 756 ensemble members from the 1850s to 2018 has been developed.These ensemble versions have been merged with the ERSSTv5 ensemble dataset,and an upgraded version of the CMSTInterim dataset with 5°×5°resolution has been developed.The CMST-Interim dataset has significantly improved the coverage rate of global surface temperature data.After reconstruction,the data coverage before 1950 increased from 78%−81%of the original CMST to 81%−89%.The total coverage after 1955 reached about 93%,including more than 98%in the Northern Hemisphere and 81%−89%in the Southern Hemisphere.Through the reconstruction ensemble experiments with different parameters,a good basis is provided for more systematic uncertainty assessment of C-LSAT2.0 and CMSTInterim.In comparison with the original CMST,the global mean surface temperatures are estimated to be cooler in the second half of 19th century and warmer during the 21st century,which shows that the global warming trend is further amplified.The global warming trends are updated from 0.085±0.004℃(10 yr)^(–1)and 0.128±0.006℃(10 yr)^(–1)to 0.089±0.004℃(10 yr)^(–1)and 0.137±0.007℃(10 yr)^(–1),respectively,since the start and the second half of 20th century.Wenbin SUN Qingxiang LI Boyin HUANG Jiayi CHENG Zhaoyang SONG Haiyan LI Wenjie DONG Panmao ZHAI Phil JONES 2021Advances in Atmospheric Sciences2021,38,5:8
8Studies of Primary Angle Qosure Glaucoma in China显示文摘The resent researches on primary angle closure glaucoma (PACG) in China were reviewed. The prevalence of PACG (0.41%) in chinese is between the Caucasian and Eskimos, and chronic type is the dominant type in PACG. However, with the economic development and medical care improvements, the prevalence condition of PACG may change; several researches discovered that the mechanism of PACG, especially the chronic type, is not only because of pupil-blocking but also closely related to the mechanism of crowding of peripheral iris and the anterior location of the ciliary body. We supposed that PACG can be divided into several subtypes based on this machnism. Also, we believe that the use of ultrasound biomicroscopy (UBM) and ophthalmic laser in China has promoted the research of PACG and improved the level of prevention and treatment of PACG. 9Earth's Temporal Principal Moments of Inertia and Variable Rotation显示文摘Based on the gravity field models EGM96 and EIGEN-GL04C, the Earth's time-dependent principal moments of inertia A, B, C are obtained, and the variable rotation of the Earth is determined. Numerical results show that A, B, and C have increasing tendencies; the tilt of the rotation axis increases 2.1×10-8 mas/yr; the third component of the rotational angular velocity, ω 3, has a decrease of 1.0×10-22 rad/s2, which is around 23% of the present observed value. 10Quantitative Real Time Measurement of Iris Configuration in Human Eyes显示文摘Purpose: To evaluate intraobserver and interobserver reproducibility of real time measurement of iris morphology in living human eyes.Methods: Base on the platform of software (Autocad, version 12), we developed an ultrasound biomicroscopy (UBM) image asisstant measuring system. By using the system, we can perform the iris configuration quantitative measurement in living eyes. The measuring parameters including: iris rest length,radius of iris curvature,and the thickness in different parts of iris. Ten anterior segment images of one normal individual were obtained by a single operator to evaluate the intraobserver reproducibility of image capture, and ten times measurement of one image were performed by a single operator to assess the reproducibilityof image measurement. The measurement of three independent observers were compared to investigate interobserver reproducibility in quantitative measurement. 11Environmentally Tough and Stretchable MXene Organohydrogel with Exceptionally Enhanced Electromagnetic Interference Shielding Performances显示文摘Conductive hydrogels have potential applications in shielding electromagnetic(EM)radiation interference in deformable and wearable electronic devices,but usually suffer from poor environmental stability and stretching-induced shielding performance degradation.Although organohydrogels can improve the environmental stability of materials,their development is at the expense of reducing electrical conductivity and thus weakening EM interference shielding ability.Here,a MXene organohydrogel is prepared which is composed of MXene network for electron conduction,binary solvent channels for ion conduction,and abundant solvent-polymer-MXene interfaces for EM wave scattering.This organohydrogel possesses excellent anti-drying ability,low-temperature tolerance,stretchability,shape adaptability,adhesion and rapid self-healing ability.Two effective strategies have been proposed to solve the problems of current organohydrogel shielding materials.By reasonably controlling the MXene content and the glycerol-water ratio in the gel,MXene organohydrogel can exhibit exceptionally enhanced EM interference shielding performances compared to MXene hydrogel due to the increased physical cross-linking density of the gel.Moreover,MXene organohydrogel shows attractive stretching-enhanced interference effectiveness,caused by the connection and parallel arrangement of MXene nanosheets.This well-designed MXene organohydrogel has potential applications in shielding EM interference in deformable and wearable electronic devices.Yuanhang Yu Peng Yi Wenbin Xu Xin Sun Gao Deng Xiaofang Liu Jianglan Shui Ronghai Yu 2022Nano-Micro Letters2022,14,5:6

16Expression of rabbit defensin NP-1 gene in transgenic tobacco plants and its activity against bacterial wilt显示文摘A cDNAderived 200 bp fragment encoding a rabbit defensin NP1 mature peptide was ligated into the GUSless pBI121 to yield pBIC35NP1. 19Using nerve segment insert grafting to reconstruct neural pathways of brain-derived paralysis显示文摘Since 1992,task groups have used free nerve auto grafts to bridge partially transected nerves from the dominant area of the normal cerebral cortex to nerves that innervate spastic muscles from a diseased cerebral cortex,using transplanting sutures to alleviate the muscle spasm of cerebral palsy caused by different diseases. This has facilitated rebuilding of some of its neurological function. In this study,80 such patients were followed up,including 20 patients with traumatic brain injury,32 patients with stroke,and 28 pediatric patients with cerebral palsy. 20Stable perovskite solar cells with efficiency of 22.6%via quinoxaline-based polymeric hole transport material显示文摘Poor stability of spiro-OMe TAD hole transport materials(HTMs)with dopant is a major obstacle for the commercialization of perovskite solar cells(pero-SCs).Herein,we demonstrate a series of quinoxaline-based D-A copolymers PBQ5,PBQ6 and PBQ10 as the dopant-free polymer HTMs for high performance pero-SCs.The D-A copolymers are composed of fluorothienyl benzodithiophene(BDTT)as D-unit,difluoroquinoxaline(DFQ)with different side chains as A-unit,and thiophene asπ-bridge,where the side chains on the DFQ unit are bi-alkyl for PBQ5,bi-alkyl-fluorothienyl for PBQ6,and alkoxyl for PBQ10.All the three copolymers are adopted as the dopant-free HTM in the pero-SCs.The planar n-i-p structured pero-SCs based on(FAPb I_(3))_(0.98)(MAPb Br_(3))_(0.02)with PBQ6 HTM demonstrated the high power conversion efficiency(PCE)of 22.6%with Vocof1.13 V and FF of 80.8%,which is benefitted from the suitable energy level and high hole mobility of PBQ6.The PCE of 22.6%is the highest efficiency reported in the n-i-p structured pero-SCs based on dopant-free D-A copolymer HTM.In addition,the peroSCs show significantly enhanced ambient,thermal and light-soaking stability compared with the devices with traditional spiroOMe TAD HTM.Chenxing Lu Can Zhu Lei Meng Chenkai Sun Wenbin Lai Shucheng Qin Jinyuan Zhang Wenchao Huang Jiaqi Du Yiyang Wang Yongfang Li 2021Science China Chemistry2021,64,11:2
