1Andrographolide sulfonate ameliorates lipopolysaccharide-induced acute lung injury in mice by down-regulating MAPK and NF-κB pathways显示文摘Acute lung injury(ALI) or acute respiratory distress syndrome(ARDS) is a severe, lifethreatening medical condition characterized by widespread inflammation in the lungs, and is a significant source of morbidity and mortality in the patient population. New therapies for the treatment of ALI are desperately needed. In the present study, we examined the effect of andrographolide sulfonate, a water-soluble form of andrographolide(trade name: Xi-Yan-Ping Injection), on lipopolysaccharide(LPS)-induced ALI and inflammation. Andrographolide sulfonate was administered by intraperitoneal injection to mice with LPS-induced ALI. LPS-induced airway inflammatory cell recruitment and lung histological alterations were significantly ameliorated by andrographolide sulfonate. Protein levels of pro-inflammatory cytokines in bronchoalveolar lavage fluid(BALF) and serum were reduced by andrographolide sulfonate administration. m RNA levels of pro-inflammatory cytokines in lung tissue were also suppressed. Moreover, andrographolide sulfonate markedly suppressed the activation of mitogen-activated protein kinase(MAPK) as well as p65 subunit of nuclear factor-κB(NF-κB). In summary, these results suggest that andrographolide sulfonate ameliorated LPS-induced ALI in mice by inhibiting NF-κB and MAPK-mediated inflammatory responses. 2The structural and accessory proteins M,ORF 4a,ORF 4b,and ORF 5 of Middle East respiratory syndrome coronavirus(MERS-CoV)are potent interferon antagonists显示文摘The newly emerged Middle East respiratory syndrome coronavirus(MERS-CoV)is a highly pathogenic respira-tory virus with pathogenic mechanisms that may be driven by innate immune pathways.The goal of this study is to characterize the expression of the structural(S,E,M,N)and accessory(ORF 3,ORF 4a,ORF 4b,ORF 5)proteins of MERS-CoV and to determine whether any of these pro-teins acts as an interferon antagonist.Individual structural and accessory protein-coding plasmids with an N-terminal HA tag were constructed and transiently transfected into cells,and their native expression and subcellular localiza-tion were assessed using Wes tern blotting and indirect immunofl uorescence.While ORF 4b demonstrated majorly nuclear localization,all of the other proteins demonstrated cytoplasmic localization.In addition,for the fi rst time,our experiments revealed that the M,ORF 4a,ORF 4b,and ORF 5 proteins are potent interferon antagonists.Further exami-nation revealed that the ORF 4a protein of MERS-CoV has the most potential to counteract the antiviral effects of IFN via the inhibition of both the interferon production(IFN-βpromoter activity,IRF-3/7 and NF-κB activation)and ISRE promoter element signaling pathways.Together,our re-sults provide new insights into the function and pathogenic role of the structural and accessory proteins of MERS-CoV.Yang Yang Ling Zhang Heyuan Geng Yao Deng Baoying Huang Yin Guo Zhengdong Zhao Wenjie Tan 2013Protein & Cell2013,4,12:25
3Future Changes and Uncertainties in Temperature and Precipitation over China Based on CMIP5 Models显示文摘Climate changes in future 21 st century China and their uncertainties are evaluated based on 22 climate models from the Coupled Model Intercomparison Project Phase 5(CMIP5). By 2081–2100, the annual mean surface air temperature(SAT) is predicted to increase by 1.3℃± 0.7℃, 2.6℃± 0.8℃ and 5.2℃± 1.2℃ under the Representative Concentration Pathway(RCP) scenarios RCP2.6, RCP4.5 and RCP8.5, relative to 1986–2005, respectively. The future change in SAT averaged over China increases the most in autumn/winter and the least in spring, while the uncertainty shows little seasonal variation.Spatially, the annual and seasonal mean SAT both show a homogeneous warming pattern across China, with a warming rate increasing from southeastern China to the Tibetan Plateau and northern China, invariant with time and emissions scenario.The associated uncertainty in SAT decreases from northern to southern China. Meanwhile, by 2081–2100, the annual mean precipitation increases by 5% ± 5%, 8% ± 6% and 12% ± 8% under RCP2.6, RCP4.5 and RCP8.5, respectively. The national average precipitation anomaly percentage, largest in spring and smallest in winter, and its uncertainty, largest in winter and smallest in autumn, show visible seasonal variations. Although at a low confidence level, a homogeneous wetting pattern is projected across China on the annual mean scale, with a larger increasing percentage in northern China and a weak drying in southern China in the early 21 st century. The associated uncertainty is also generally larger in northern China and smaller in southwestern China. 4Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20

6Asiatic acid inhibits lung cancer cell growth in vitro and in vivo by destroying mitochondria显示文摘Asiatic acid(AA), a pentacyclic triterpene found in Centella asiatica, displays significant antiproliferative effects on cancer cells in vitro although the underlying mechanism of this effect remains unknown. This study investigated the efficacy and mechanism of action of AA against lung cancer both in vivo and in vitro. Using the MTT assay, AA was found to induce apoptosis in a dose- and timedependent manner, an effect enhanced by pretreatment with an autophagy inhibitor. It also elevated expression of microtubule-associated protein 1 light chain 3(LC3) and decreased the expression of p62.Furthermore, exposure to AA resulted in collapse of the mitochondrial membrane potential and generation of reactive oxygen species(ROS), suggesting mitochondria are the target of AA. In the mouse lung cancer xenograft model, oral administration of AA significantly inhibited tumor volume and weight accompanied by significant apoptosis of lung cancer cells. In addition, it led to a significant decrease in the expression of proliferating cell nuclear antigen(PCNA). 7SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 8Therapeutic effect in patients with coronary heart disease based on information analysis from Traditional Chinese Medicine four diagnostic methods显示文摘OBJECTIVE:To study information gained by Traditional Chinese Medicine(TCM)four diagnostic methods and characteristics of syndrome development before and after treatment in patients with coronary heart disease,and to probe into assessment indexes of therapeutic effects with distinctive TCM features.METHODS:Information from the four diagnostic methods before and after treatment in 100 patients with coronary heart disease was collected using the TCM interrogation scale,a pulse condition instrument,a tongue-face diagnosis instrument,anda voice diagnosis information collection system.Changes in the four diagnostic method results from before and after treatment were analyzed with frequency analysis and t-test methods.RESULTS:Before treatment,deficiency syndrome complicated with hyperactivity of pathogenic factors was most common.After treatment,deficiency syndrome was most common.This change from complex syndromes to single syndromes indicates disease and syndrome alleviation.Frequencies of symptoms gained by interrogation after treatment for syndrome of deficiency of heart-Qi,syndrome of deficiency of heart-Yin,and turbid phlegm syndrome were all less severe than those before treatment.Parameters of face color,color on all sub-regions of tongue,color of tongue fur,septic and greasy fur,and lip color after treatment all had significant changes.After treatment,part of the voice diagnosis parameters in the deficiency of heart-Yin and turbid phlegm syndromes had significant changes,but no significant changes were found in the deficiency of heart-Qi syndrome.CONCLUSION:Use of the TCM four diagnostic methods can provide an effective basis forTCM syndrome diagnosis,observation of development of state of illness,and evaluation of clinical therapeutic effects.Yiqin Wang Jin Xu Rui Guo Chaoxia Xu Yiming Hao Chunfeng Chen Yijian Hong Xiangqun Xiao Wenjie Xu Jing Hong Zhao Lei 2014Journal of Traditional Chinese Medicine2014,34,1:12
9Future changes in coverage of 1.5℃ and 2℃warming thresholds显示文摘The areas covered by 1.5 ℃ and 2.0 ℃ warming thresholds under RCP2.6, RCP4.5, and RCP8.5 were analyzed based on 22 CMIP5 models. More than 90% of the model runs are in agreement that by the end of the 21 st century, near-surface air temperature changes over ~5%(~2%), ~40%(~18%), and ~92%(~86%)of the globe will cross the 1.5 ℃(2.0 ℃) threshold under RCP2.6, RCP4.5, and RCP8.5, respectively. Under RCP8.5, nearly the whole of North America, Europe + Russia, Africa, and Asia–Russia will cross the 1.5 ℃(2.0 ℃) threshold in ~2050(~2060), while the coverage rates over South America and Oceania are ~80%(~75%) and ~50%(~30%), respectively. The threshold-onset time(TOT) for 2 ℃ warming is earliest over Europe + Russia and North America, followed by Africa, Asia–Russia, South America, and finally Oceania under the RCP4.5 and RCP8.5 scenarios. 11Effect of lithium or aluminum substitution on the characteristics of graphite for anode of lithium ion batteries显示文摘Modification of graphite for anode of lithium ion batteries is investigated. Results of X-ray diffraction shows lithium and aluminum exists as Li compound (CH3COOLi@2H2O) and Al compound (AlD3) in the graphite, respectiovely.The Bmnauer-Emmer-Teller (BET) surface area of the modified graphite increases. According to the electrochemical measurements of Li/C cell and prototype Li-ion batteries, the Li-doped graphite has large reversible capacity of 312.2mA@h/g, low irreversible capacity of 52.9 mA@h/g, and high initial coulombic efficiency of 85.51%. The 063448 size prototype battery with Li-doped graphite anode has large discharge capacity of 845 mA@h and good cycling performance. 12A Brief Introduction to BNU-HESM1.0 and Its Earth Surface Temperature Simulations显示文摘Integrated assessment models and coupled earth system models both have their limitations in understanding the interactions between human activity and the physical earth system. In this paper,a new human–earth system model,BNUHESM1.0,constructed by combining the economic and climate damage components of the Dynamic Integrated Model of Climate Change and Economy to the BNU-ESM model,is introduced. The ability of BNU-HESM1.0 in simulating the global CO2 concentration and surface temperature is also evaluated. We find that,compared to observation,BNU-HESM1.0underestimates the global CO2 concentration and its rising trend during 1965–2005,due to the uncertainty in the economic components. However,the surface temperature simulated by BNU-HESM1.0 is much closer to observation,resulting from the overestimates of surface temperature by the original BNU-ESM model. The uncertainty of BNU-ESM falls within the range of present earth system uncertainty,so it is the economic and climate damage component of BNU-HESM1.0 that needs to be improved through further study. However,the main purpose of this paper is to introduce a new approach to investigate the complex relationship between human activity and the earth system. 13In situ plasmonic optical fiber detection of the state of charge of supercapacitors for renewable energy storage显示文摘In situ and continuous monitoring of electrochemical activity is key to understanding and evaluating the operation mechanism and efficiency of energy storage devices.However,this task remains challenging.For example,the present methods are not capable of providing the real-time information about the state of charge(SOC)of the energy storage devices while in operation.To address this,a novel approach based on an electrochemical surface plasmon resonance(SPR)optical fiber sensor is proposed here.This approach offers the capability of in situ comprehensive monitoring of the electrochemical activity(the electrode potential and the SOC)of supercapacitors(used as an example).The sensor adopted is a tilted fiber Bragg grating imprinted in a commercial single-mode fiber and coated with a nanoscale gold film for high-efficiency SPR excitation.Unlike conventional“bulk”detection methods for electrode activity,our approach targets the“localized”(sub-μm-scale)charge state of the ions adjacent to the electrode interface of supercapacitors by monitoring the properties of the SPR wave on the fiber sensor surface located adjacent to the electrode.A stable and reproducible correlation between the real-time charge–discharge cycles of the supercapacitors and the optical transmission of the optical fiber has been found.Moreover,the method proposed is inherently immune to temperature cross-talk because of the presence of environmentally insensitive reference features in the optical transmission spectrum of the devices.Finally,this particular application is ideally suited to the fundamental qualities of optical fiber sensors,such as their compact size,flexible shape,and remote operation capability,thereby opening the way for other opportunities for electrochemical monitoring in various hard-to-reach spaces and remote environments.Jiajie Lao Peng Sun Fu Liu Xuejun Zhang Chuanxi Zhao Wenjie Mai Tuan Guo Gaozhi Xiao Jacques Albert 2018Light(Science & Applications)2018,7,1:8
14Performance and capacity fading reason of LiMn_2O_4/graphite batteries after storing at high temperature显示文摘Spinel LiMn2O4 was synthesized by a solid-state method. A 204468-size battery was fabricated and stored at 55°C. The structure and morphology of the LiMn2O4 cathode were analyzed by X-ray diffraction (XRD) and scanning electron microscopy (SEM) technique. Energy dispersive spectroscopy (EDS) was used to analyze the surface component of the carbon anode. The discharge capacities of LiMn2O4 stored for 0, 24, 48, and 96 h are 106, 98, 96, and 92 mAh?g?1, respectively. The cyclic performance is improved after storage. The capacity retentions of LiMn2O4 stored for 0, 24, 48, and 96 h are 83.8%, 85.8%, 86.9%, and 88.6% after 180 cycles. The intensity of all the LiMn2O4 diffraction peaks is weakened. Mn is detected from the carbon electrode when the battery is stored for 96 h. Cyclic voltammograms and electrochemical impedance spectroscopy (EIS) were used to examine the surface state of the electrode after storage. 15Global warming projections using the human–earth system model BNU-HESM1.0显示文摘Future climate change is usually projected by coupled earth system models under specific emission scenarios designed by integrated assessment models(IAMs),and this offline approach means there is no interaction between the coupled earth system models and the IAMs.This paper introduces a new method to design possible future emission scenarios and corresponding climate change, in which a simple economic and climate damage component is added to the coupled earth system model of Beijing Normal University(BNU-ESM). With the growth of population and technological expertise and the declining emission-to-output ratio described in the Dynamic Integrated Climate-Economy model, the projected carbon emission is 13.7 Gt C, resulting in a 2.4 °C warming by the end of the twenty-first century(2080–2099) compared with1980–1999. This paper also suggests the importance of the land and ocean carbon cycle in determining the CO2 concentration in the atmosphere. 16Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 17Reduction and characterization of bioaerosols in a wastewater treatment station via ventilation显示文摘Bioaerosols from wastewater treatment processes are a significant subgroup of atmospheric aerosols. In the present study,airborne microorganisms generated from a wastewater treatment station(WWTS) that uses an oxidation ditch process were diminished by ventilation.Conventional sampling and detection methods combined with cloning/sequencing techniques were applied to determine the groups,concentrations,size distributions,and species diversity of airborne microorganisms before and after ventilation. There were 3021 ± 537 CFU/m3 of airborne bacteria and 926 ± 132 CFU/m3 of airborne fungi present in the WWTS bioaerosol.Results showed that the ventilation reduced airborne microorganisms significantly compared to the air in the WWTS. Over 60% of airborne bacteria and airborne fungi could be reduced after4 hr of air exchange. The highest removal(92.1% for airborne bacteria and 89.1% for fungi) was achieved for 0.65–1.1 μm sized particles. The bioaerosol particles over 4.7 μm were also reduced effectively. Large particles tended to be lost by gravitational settling and small particles were generally carried away,which led to the relatively easy reduction of bioaerosol particles0.65–1.1 μm and over 4.7 μm in size. 18Influence of the carbon cycle on the attribution of responsibility for climate change显示文摘The carbon cycle is one of the fundamental climate change issues.Its long-term evolution largely affects the amplitude and trend of human-induced climate change,as well as the formulation and implementation of emission reduction policy and technology for stabilizing the atmospheric CO2concentration.Two earth system models incorporating the global carbon cycle,the Community Earth System Model and the Beijing Normal University-Earth System Model,were used to investigate the effect of the carbon cycle on the attribution of the historical responsibility for climate change.The simulations show that when compared with the criterion based on cumulative emissions,the developed(developing)countries’responsibility is reduced(increased)by 6%–10%using atmospheric CO2concentration as the criterion.This discrepancy is attributed to the fact that the developed world contributed approximately61%–68%(61%–64%)to the change in global oceanic(terrestrial)carbon sequestration for the period from 1850 to2005,whereas the developing world contributed approximately 32%–49%(36%–39%).Under a developed world emissions scenario,the relatively larger uptake of global carbon sinks reduced the developed countries’responsibility for carbon emissions but increased their responsibility for global ocean acidification(68%).In addition,the large emissions from the developed world reduced the efficiency of the global carbon sinks,which may affect the long-term carbon sequestration and exacerbate global warming in the future.Therefore,it is necessary to further consider the interaction between carbon emissions and the carbon cycle when formulating emission reduction policy.Ting Wei Wenjie Dong Wenping Yuan Xiaodong Yan Yan Guo 2014Chinese Science Bulletin2014,59,19:6
19Stiffeners layout design of thin-walled structures with constraints on multi-fastener joint loads显示文摘The purpose of this paper is to present an extended topology optimization method for the stiffeners layout design of aircraft assembled structures. Multi-fastener joint loads and manufacturing constraints are considered simultaneously. On one hand, the joint loads are calculated and constrained within a limited value to avoid the failure of fasteners. On the other hand, the manufacturing constraints of the material distribution in the machining directions of stiffeners are implemented by an improved piecewise interpolation based on a beveled cut-surface. It is proven that the objective function is strictly continuous and differentiable with respect to the piecewise interpolation. The effects of the extended method with two different constraints are highlighted by typical numerical examples. 20In situ construction of Z-scheme FeS_(2)/Fe_(2)O_(3) photocatalyst via structural transformation of pyrite for photocatalytic degradation of carbamazepine and the synergistic reduction of Cr(Ⅵ)显示文摘Pyrite is the most abundant sulfide semiconductor mineral with excellent optical properties.However,few reports have investigated its photocatalytic activity because of the low photogenerated carrier separation efficiency.In this work,a Z-scheme FeS_(2)/Fe_(2)O_(3)composite photocatalyst was fabricated in situ via structural transformation of pyrite through heat treatment.A remarkably enhanced photocatalytic performance was observed over the FeS_(2)/Fe_(2)O_(3)composite photocatalyst.Compared with the pristine pyrite,the degradation efficiency of carbamazepine(CBZ)reached 65%at the added hexavalent chromium(Cr(Ⅵ))concentration of 20 mg/L and the Cr(Ⅵ)was nearly completely reduced in the mixed system using FeS_(2)/Fe_(2)O_(3)within 30 min under simulated solar light irradiation.The enhanced photocatalytic activity can be attributed to the efficient separation and transfer of photogenerated carriers in the FeS_(2)/Fe_(2)O_(3)composite photocatalyst.This facilitated the generation of·OH,hole(h^(+))and·O_(2)^(-)species,which participated in the photocatalytic reaction with CBZ.Based on the measurement of the active species and electric properties,a Z-scheme electron transfer pathway was proposed for the FeS_(2)/Fe_(2)O_(3)composite photocatalyst.This work broadens the application potential of pyrite in environmental remediation.Qian Guo Guangbei Tang Wenjie Zhu Yongming Luo Xiaoya Gao 2021Journal of Environmental Sciences2021,33,3:4
