| 2 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 3 | Determination of metastable zone and induction time of analgin for cooling crystallization显示文摘The solubility, metastable zone width, and induction time of analgin for unseeded batch cooling crystallization in ethanol–aqueous system were experimentally determined. The solubility data could be well described by the van't Hoff equation model. The metastable zone width at various cooling rates was measured, and some parameters of nucleation kinetic were calculated using the Ny'vlt theory. Furthermore, the induction period of various temperatures and supersaturation ratios was also measured. According to classical nucleation theory, some nucleation parameters and interfacial energy was calculated through the induction time(t_(ind)) data. | 4 | Changes in T lymphocyte subsets after severe traumatic brain injury显示文摘BACKGROUND: Besides local changes of cranial parenchymal cells, hemorrhage, etc., severe traumatic brain injuries also cause the changes of total body fluid and various functions, and the changes of lymphocytes and T lymphocyte subsets should be paid more attention to. OBJECTIVE: To reveal the changing laws of T lymphocyte subsets after severe traumatic brain injury, and compare with mild to moderate brain injury. DESIGN: A comparative observation. SETTINGS: Department of Neurosurgery, Longgang District Buji People's Hospital of Shenzhen City; Central Laboratory of Shenzhen Hospital of Prevention and Cure for Chronic Disease. PARTICIPANTS: All the subjects were selected from the Department of Neurosurgery, Longgang District Buji People's Hospital of Shenzhen City from August 2002 to August 2005. Thirty patients with severe brain injury, whose Glasgow coma score (GCS) was ≤ 8 points, were taken as the experimental group, including 21 males and 9 females, aging 16–62 years. Meanwhile, 30 patients with mild traumatic brain injury were taken as the control group (GCS ranged 14–15 points), including 18 males and 12 females, aging 15–58 years. All the subjects were in admission at 6 hours after injury, without disease of major organs before injury. Informed consents were obtained from all the patients or their relatives. METHODS: ① The T lymphocytes and the subsets in peripheral blood were detected with immunofluorescent tricolor flow cytometry at 1, 3, 7 and 14 days after injury in both groups. ② The conditions of pulmonary infections were observed at 4 days after injury. The differences of measurement data were compared with the t test. MAIN OUTCOME MEASURES: Changes of T lymphocytes subsets at 1–14 days after severe and mild or moderate traumatic injury. RESULTS: Finally, 28 and 25 patients with mild to moderate traumatic brain injury, whereas 25 and 21 patients with severe traumatic brain injury were analyzed at 7 and 14 days respectively, and the missed ones died due to the development of disease. ① Changes of T lymphocyte subsets: At 1 and 3 days after injury, CD3, CD4, CD8, CD4/CD8 began to decrease, whereas CD8 increased in the experimental group, which were very significantly different from those in the control group (t =2.77–3.26, P < 0.01), and began to recover at 7 days, which were significantly different from those in the control group (t = 2.06–2.24, P < 0.05), and generally recovered to the normal levels at 14 days (P > 0.05). ② Conditions of pulmonary infections: At 4 days after injury, the rate of pulmonary infection was significantly different between the experimental group and control group [73% (22/30), 0, χ2=37.29, P < 0.01]. | 6 | Allosteric inhibition reveals SHP2-mediated tumor immunosuppression in colon cancer by single-cell transcriptomics显示文摘Colorectal cancer(CRC), a malignant tumor worldwide consists of microsatellite instability(MSI) and stable(MSS) phenotypes. Although SHP2 is a hopeful target for cancer therapy, its relationship with innate immunosuppression remains elusive. To address that, single-cell RNA sequencing wasperformed to explore the role of SHP2 in all cell types of tumor microenvironment(TME) from murine MC38 xenografts. Intratumoral cells were found to be functionally heterogeneous and responded significantly to SHP099, a SHP2 allosteric inhibitor. The malignant evolution of tumor cells was remarkably arrested by SHP099. Mechanistically, STING-TBK1-IRF3-mediated type I interferon signaling was highly activated by SHP099 in infiltrated myeloid cells. Notably, CRC patients with MSS phenotype exhibited greater macrophage infiltration and more potent SHP2 phosphorylation in CD68;macrophages than MSI-high phenotypes, suggesting the potential role of macrophagic SHP2 in TME. | 10 | Establishment of NOD/SCID mouse models of human hepatocellular carcinoma via subcutaneous transplantation of histologically intact tumor tissue显示文摘Hepatocellular carcinoma (HCC) is one of the most deadly human cancers, but it is very difficult to establish an animal model by using surgical specimens. In the present experiment, histologically intact fresh surgical specimens of HCC were subcutaneously transplanted in non-obese diabetic/severe combined immunodeficienccy (NOD/SCID) mice. The biological characteristics of the original and the corresponding transplanted tumors and cell lines were investigated. The results showed that 5 new animal models and 2 primary cell lines were successfully established from surgical specimens. Hematoxylin-eosin staining showed that xenografts retained major histological features of the original surgical specimens. The two new cell lines had been cultivated for 3 years and successively passaged for more than 100 passages in vitro. The morphological characteristics and biologic features of the two cell lines were genetically similar to the original tumor. | 11 | Loss of hnRNP A1 in murine skeletal muscle exacerbates high-fat diet-induced onset of insulin resistance and hepatic steatosis显示文摘Impairment of glucose(Glu)uptake and storage by skeletal muscle is a prime risk factor for the development of metabolic diseases.Heterogeneous nuclear ribonucleoprotein A1(hnRNP Al)is a highly abundant RNA-binding protein that has been implicated in diverse cellular functions.The aim of this study was to investigate the function of hnRNP A1 on muscle tissue insulin sensitivity and systemic Glu homeostasis.Our results showed that conditional deletion of hnRNP Al in the muscle gave rise to a severe insulin resistance phenotype in mice fed a high-fat diet(HFD).Conditional knockout mice fed a HFD showed exacerbated obesity,insulin resistance,and hepatic steatosis.In vitro interference of hnRNP Al in C2C12 myotubes impaired insulin signal transduction and inhibited Glu uptake,whereas hnRNP Al overexpression in C2C12 myotubes protected against insulin resistance induced by supraphysiological concentrations of insulin.The expression and stability of glycogen synthase(gysl)mRNA were also decreased in the absence of hnRNP A l.Mechanistically,hnRNP Al interacted with gys l and stabilized its mRNA,thereby promoting glycogen synthesis and maintaining the insulin sensitivity in muscle tissue.Taken together,our findings are the first to show that reduced expression of hnRNP Al in skeletal muscle affects the metabolic properties and systemic insulin sensitivity by inhibiting glycogen synthesis. | Mingxia Zhao Lihong Shen Zijun Ouyang Manru Li Guoliang Deng Chenxi Yang Wei Zheng Lingdong Kong Xuefeng Wu Xudong Wu Wenjie Guo Ye Yin Qiang Xu Yang Sun | 2020 | Journal of Molecular Cell Biology2020,12,4: | 2 |

| 14 | Identification of serum metabolites enhancing inflammatory responses in COVID-19显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is characterized by a strong production of inflammatory cytokines such as TNF and IL-6,which underlie the severity of the disease.However,the molecular mechanisms responsible for such a strong immune response remains unclear.Here,utilizing targeted tandem mass spectrometry to analyze serum metabolome and lipidome in COVID-19 patients at different temporal stages,we identified that 611 metabolites(of 1,039)were significantly altered in COVID-19 patients.Among them,two metabolites,agmatine and putrescine,were prominently elevated in the serum of patients;and 2-quinolinecarboxylate was changed in a biphasic manner,elevated during early COVID-19 infection but levelled off.When tested in mouse embryonic fibroblasts(MEFs)and macrophages,these 3 metabolites were found to activate the NF-κB pathway that plays a pivotal role in governing cytokine production.Importantly,these metabolites were each able to cause strong increase of TNF and IL-6 levels when administered to wildtype mice,but not in the mice lacking NF-κB.Intriguingly,these metabolites have little effects on the activation of interferon regulatory factors(IRFs)for the production of type I interferons(IFNs)for antiviral defenses.These data suggest that circulating metabolites resulting from COVID-19 infection may act as effectors to elicit the peculiar systemic inflammatory responses,exhibiting severely strong proinflammatory cytokine production with limited induction of the interferons.Our study may provide a rationale for development of drugs to alleviate inflammation in COVID-19 patients. | Chen-Song Zhang Bingchang Zhang Mengqi Li Xiaoyan Wei Kai Gong Zhiyong Li Xiangyang Yao Jianfeng Wu Cixiong Zhang Mingxia Zhu Lei Zhang Xiufeng Sun Yi-Hong Zhan Zhengye Jiang Wenpeng Zhao Wei Zhong Xinguo Zhuang Dawang Zhou Hai-Long Piao Sheng-Cai Lin Zhanxiang Wang | 2022 | Science China(Life Sciences)2022,65,10: | 1 |
| 15 | Honeycomb-like polyaniline for flexible and folding all-solid-state supercapacitors显示文摘Porous polyaniline (PANI) was prepared through an efficient and costeffective method by polymerization of aniline in the NaCl solution at room temperature. The resulting PANI provided large surface area due to its highly porous structure and the intercrossed nanorod, resulting in good electrochemical performance. The porous PANI electrodes showed a high specific capacitance of 480 F·g^-1, 3 times greater than that of PANI without using the NaCl solution. We also make chemically crosslinked hydrogel film for hydrogel polymer electrolyte as well as the flexible supercapacitors (SCs) with PANI. The specific capacitance of the device was 234 F·g^-1 at the current density of 1 A·g^-1. | 18 | Decoding an Old 1907 Prescription of Dr.Ing Hay(“Doc Hay”):One of the Earliest Chinese Medicine Doctors in the United States显示文摘Objectives:Chinese medicine doctors have practiced in the United States since April of 1854,the world's first Chinese medicine doctor's memorial is Kam Wah Chung Museum in John Day,Oregon,U.S.,to commemorate Dr.Ing Hay(“Doc Hay”伍于念)and his business partner Lung On(梁光荣).There were few studies to explore Doc Hay's real practice and prescription,while most of papers focused on Doc Hay's legend life.This paper aims to analyze and interpret one of the intriguing herbal prescriptions that Doc Hay had hand-written.Materials and Methods:Dr.Arthur Yin Fan made an academic travel to Kam Wah Chung museum on August 5,2018 and examined the books and hand-written prescriptions of Doc Hay,which were not exhibited to the public at that time.This paper analyzes and interprets one of herbal prescriptions that Doc Hay had hand-written in 1907.Interpretation process included four steps:(1):Transform Doc Hay's hand-written prescription(with original herb names)to standard herb names in Chinese.(2):Induct and rearrange the herb name into groups based on herb property characteristics and then translate them into both PinYin names and English names.Analyzation of the prescription may include in what classic formula(s)routinely introduced in Chinese medicine textbooks.(3):Analyze the herbal action for each group and potential symptoms or conditions the patient may have had.An analysis of classic formula(s)used in the prescription may represent what clinical condition was being treated at that time.(4):Combine the analysis to give a comprehensive picture of the patient.Results:The prescription consists of 67 herbs,total 934.6 grams,and including Yin Qiao Powder(银翘散),Sang Ju Decoction(桑菊饮),Zhi Sou Powder(止嗽散),Qing Ying Decoction(清营汤),Xi Jiao Di Huang Decoction(犀角地黄汤),Long Dan Xie Gan Decoction(龙胆泻肝汤)and Chai Hu Shu Gan Powder(柴胡疏肝散).Speculation indicated that the prescription was for a patient who had obvious pulmonary infection accompanied by severe cough,and probably had prolonged pulmonary tuberculosis(TB)also with acute respiratory infection caused by other bacteria or viruses.Based on the analysis of composition of this prescription,it can be used for the treatment of epidemic diseases.Conclusion:The majority of herbs used in this prescription are commonly included in the herbal medicines in China used to effectively counter severe acute respiratory syndrome(SARS),H1N1 and currently COVID-19,it might be Doc Hay's basic formula for patients with“Spanish Flu”during the 1918-1920 pandemic. | Arthur Yin Fan Louis Lei Jin Boling Huang Mingxia Yu Xiaoyin Zhao Sarah Faggert Alem | 2020 | Chinese Medicine and Culture2020,3,4: | 1 |

| 20 | Laboratory Diagnosis and Monitoring the Viral Shedding of SARS-CoV-2 Infection显示文摘The worldwide epidemic of coronavirus disease 2019(COVID-19)is ongoing.Rapid and accurate detection of the causative virus SARSCoV-2 is vital for the treatment and control of COVID-19.In this study,the comparative sensitivity of different respiratory specimen types were retrospectively analyzed using 3,552 clinical samples from 410 COVID-19 patients confirmed by Guangdong CDC(Center for Disease Control and Prevention).Except for bronchoalveolar lavage fluid(BALF),the sputum possessed the highest positive rate(73.4%–87.5%),followed by nasal swabs(53.1%–85.3%)for both severe and mild cases during the first 14 days after illness onset(d.a.o.).Viral RNA could be detected in all BALF samples collected from the severe group within 14 d.a.o.and lasted up to 46 d.a.o.Moreover,although viral RNA was negative in the upper respiratory samples,it was also positive in BALF samples in most cases from the severe group during treatment.Notably,no viral RNA was detected in BALF samples from the mild group.Despite typical ground-glass opacity observed via computed tomographic scans,no viral RNA was detected in the first three or all upper respiratory tract specimens from some COVID-19 patients.In conclusion,sputum is most sensitive for routine laboratory diagnosis of COVID-19,followed by nasal swabs.Detection of viral RNA in BALF improves diagnostic accuracy in severe COVID-19 patients. | Yang Yang Minghui Yang Jing Yuan Fuxiang Wang Zhaoqin Wang Jinxiu Li Mingxia Zhang Jinli Wei Ling Peng Gary Wong Haixia Zheng Weibo Wu Chenguang Shen Mingfeng Liao Kai Feng Jianming Li Qianting Yang Juanjuan Zhao Lei Liu Yingxia Liu | 2020 | The Innovation2020,1,3: | 1 |
