1Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
2Responses of Patients with Disorders of Consciousness to Habit Stimulation:A Quantitative EEG Study显示文摘Whether habit stimulation is effective in DOC patient arousal has not been reported. In this paper, we analyzed the responses of DOC patients to habit stimulation. Nineteen DOC patients with alcohol consumption or smoking habits were recruited and 64-channel EEG signals were acquired both at the resting state and at three stimulation states. Wavelet transformation and nonlinear dynamics were used to extract the features of EEG signals and four brain lobes were selected to investigate the degree of EEG response to habit stimulation. Results showed that the highest degree of EEG response was from the callname stimulation, followed by habit and music stimulations. Significant differences in EEG wavelet energy and response coefficient were found both between habit and music stimulation, and between habit and call-name stimulation. 3Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
4Response surface modeling-based source contribution analysis and VOC emission control policy assessment in a typical ozone-polluted urban Shunde,China显示文摘To develop a sound ozone(O_3) pollution control strategy,it is important to well understand and characterize the source contribution due to the complex chemical and physical formation processes of O_3.Using the 'Shunde' city as a pilot summer case study,we apply an innovative response surface modeling(RSM) methodology based on the Community Multi-Scale Air Quality(CMAQ) modeling simulations to identify the O_3 regime and provide dynamic analysis of the precursor contributions to effectively assess the O_3 impacts of volatile organic compound(VOC) control strategy.Our results show that Shunde is a typical VOC-limited urban O_3 polluted city.The 'Jiangmen' city,as the main upper wind area during July 2014,its VOCs and nitrogen oxides(NO_x) emissions make up the largest contribution(9.06%).On the contrary,the contribution from local(Shunde) emission is lowest(6.35%) among the seven neighbor regions.The local VOCs industrial source emission has the largest contribution comparing to other precursor emission sectors in Shunde.The results of dynamic source contribution analysis further show that the local NO_x control could slightly increase the ground O_3 under low(10.00%) and medium(40.00%)reduction ratios,while it could start to turn positive to decrease ground O_3 under the high NO_x abatement ratio(75.00%).The real-time assessment of O_3 impacts from VOCs control strategies in Pearl River Delta(PRD) shows that the joint regional VOCs emission control policy will effectively reduce the ground O_3 concentration in Shunde.Zhiqiang You Yun Zhu Carey Jang Shuxiao Wang Jian Gao Che-Jen Lin Minhui Li Zhenghua Zhu Hao Wei Wenwei Yang 2017Journal of Environmental Sciences2017,29,1:7
5OsPKpa_1 encodes a plastidic pyruvate kinase that affects starch biosynthesis in the rice endosperm显示文摘Pyruvate kinase(PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice(Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpa_1, which encodes a plastidic isoform of PK(PKp). OsPKpa_1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpa_1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, atthe early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein–protein interaction analysis. 6Mass‑Spring‑Damping Theory Based Equivalent Mechanical Model for Cylindrical Lithium‑ion Batteries under Mechanical Abuse显示文摘An equivalent mechanical model with the equivalent physical meaning of mass-spring-damping is proposed for cylindrical lithium-ion batteries through experiments and theory.The equivalent mechanical model of a cylindrical lithium-ion battery consists of a spring-damping parallel unit.Therefore,a spring-damping parallel unit connecting a damping unit in series is selected to construct the constitutive characteristics of the battery under mechanical abuse.Comparison results show that the equivalent mechanical model can more effectively describe the mechanical properties of the batteries than most cubic fitting models,of which the average relative error of the equivalent mechanical model under different states-of-charge is less than 6.75%.Combined with the proposed equivalent mechanical model,the failure process of the batteries was simulated and analyzed using LS-Dyna and HyperWorks.Under rigid rod tests,failure occurred at the core and bottom of the batteries;under hemispherical punch tests,failure occurred at the core and top,consistent with the experimental results.The average prediction error for the failure displacement under different abuse conditions is less than 4%in the simulations.The equivalent mechanical model requires only a few parameters and can be recognized easily.In the future,the model can be used in safety warning devices based on mechanical penetration.Wenwei Wang Yiding Li Cheng Lin Sheng Yang 2020Chinese Journal of Mechanical Engineering2020,33,2:3
7Ischemic postconditioning enhances glycogen synthase kinase-3β expression and alleviates cerebral ischemia/reperfusion injury显示文摘The present study established global brain ischemia using the four-vessel occlusion method.Following three rounds of reperfusion for 30 seconds,and occlusion for 10 seconds,followed by reperfusion for 48 hours,infarct area,the number of TUNEL-positive cells and Bcl-2 expression were significantly reduced.However,glycogen synthase kinase-3β activity,cortical Bax and caspase-3 expression significantly increased,similar to results following ischemic postconditioning.Our results indicated that ischemic postconditioning may enhance glycogen synthase kinase-3β activity,a downstream molecule of the phosphatase and tensin homolog deleted on chromosome 10/phosphatidylinositol 3-kinase/protein kinase B signaling pathway,which reduces caspase-3 expression to protect the brain against ischemic injury.Bo Zhao Wenwei Gao Jiabao Hou Yang Wu Zhongyuan Xia 2012Neural Regeneration Research2012,7,19:2
8Effects of Space Radiation Mutation on Germination and Growth of Sugarcane Hybrid Seeds显示文摘[Objectives] This study was conducted to investigate the mutagenic effects of space radiation mutagenesis on sugarcane hybrid seeds. [Methods] Sugarcane hybrid spikes were subjected to space radiation mutagenesis treatment to identify the effects on the germination, seedling growth and plant growth and development of sugarcane hybrid seeds. [Results] The space radiation treatment had a great impact on the germination and growth of sugarcane hybrid seeds, especially on the germination of sugarcane seeds. The germination number per gram of seeds increased significantly, and the survival rate of seedlings increased significantly. The effect on the growth status was relatively small. The average number of tillers decreased slightly, the percentage of available stems increased, and the plants became higher, which resulted in a greater effect on sugarcane yield. Sugarcane sucrose was increased. The seed germination and late growth status indicated that space radiation promoted the germination and growth of sugarcane hybrid seeds. 13Source identification and prediction of nitrogen and phosphorus pollution of Lake Taihu by an ensemble machine learning technique显示文摘Effective control of lake eutrophication necessitates a full understanding of the complicated nitrogen and phosphorus pollution sources,for which mathematical modeling is commonly adopted.In contrast to the conventional knowledge-based models that usually perform poorly due to insufficient knowledge of pollutant geochemical cycling,we employed an ensemble machine learning(ML)model to identify the key nitrogen and phosphorus sources of lakes.Six ML models were developed based on 13 years of historical data of Lake Taihu’s water quality,environmental input,and meteorological conditions,among which the XGBoost model stood out as the best model for total nitrogen(TN)and total phosphorus(TP)prediction.The results suggest that the lake TN is mainly affected by the endogenous load and inflow river water quality,while the lake TP is predominantly from endogenous sources.The prediction of the lake TN and TP concentration changes in response to these key feature variations suggests that endogenous source control is a highly desirable option for lake eutrophication control.Finally,one-month-ahead prediction of lake TN and TP concentrations(R2 of 0.85 and 0.95,respectively)was achieved based on this model with sliding time window lengths of 9 and 6 months,respectively.Our work demonstrates the great potential of using ensemble ML models for lake pollution source tracking and prediction,which may provide valuable references for early warning and rational control of lake eutrophication.Yirong Hu Wenjie Du Cheng Yang Yang Wang Tianyin Huang Xiaoyi Xu Wenwei Li 2023Frontiers of Environmental Science & Engineering2023,17,5:1

19Transient-Spatial Pattern Mining of Eddy Current Pulsed Thermography Using Wavelet Transform显示文摘Eddy current pulsed thermography(ECPT) is an emerging Non-destructive testing and evaluation(NDT & E) technique, which uses hybrid eddy current and thermography NDT & E techniques that enhances the detectability from their compensation. Currently, this technique is limited by the manual selection of proper contrast frames and the issue of improving the efficiency of defect detection of complex structure samples remains a challenge. In order to select a specific frame from transient thermal image sequences to maximize the contrast of thermal variation and defect pattern from complex structure samples, an energy driven approach to compute the coefficient energy of wavelet transform is proposed which has the potential of automatically selecting both optimal transient frame and spatial scale for defect detection using ECPT. According to analysis of the variation of different frequency component and the comparison study of the detection performance of different scale and wavelets, the frame at the end of heating phase is automatically selected as an optimal transient frame for defect detection. In addition, the detection capabilities of the complex structure samples can be enhanced through proper spatial scale and wavelet selection.
