| 1 | In vivo transfection of enhanced green fluorescent protein in rat retinal ganglion cells mediated by ultrasound-induced microbubbles显示文摘BACKGROUND:Studies have demonstrated that ultrasound-mediated microbubble destruction significantly improves transfection efficiency of enhanced green fluorescent protein(EGFP) in in vitro cultured retinal ganglial cells(RGCs). OBJECTIVE:To investigate the feasibility of ultrasound-mediated microbubble destruction for EGFP transfection in rat RGCs,and to compare efficiency and cell damage with traditional transfection methods. DESIGN,TIME AND SETTING:In vivo,gene engineering experiment.The study was performed at the Central Laboratory,Institute of Ultrasonic Imaging,Chongqing Medical University from March to July 2008. MATERIALS:Eukaryotic expression vector plasmid EGFP and microbubbles were prepared by the Institute of Ultrasonic Imaging,Chongqing Medical University.The microbubbles were produced at a concentration of 8.7×10^(11)/L,with a 2-4μm diameter,and 10-hour half-life in vitro. METHODS:A total of 50 Sprague Dawley rats were randomly assigned to four groups.Normal controls(n=5) were infused with 5μL normal saline to the vitreous cavity;the naked plasmid group (n=15) was infused with 5μL EGFP plasmid to the vitreous cavity;in the plasmid with ultrasound group(n=15),the eyes were irradiated with low-energy ultrasound wave(0.5 W/cm^2) for a total of 60 seconds(irradiated for 5 seconds,at 10-second intervals) immediately following infusion of EGFP plasmids to the vitreous cavities.In the microbubble-ultrasound group(n=15),the eyes were irradiated with the same power of ultrasonic wave immediately following infusion of microbubbles containing EGFP plasmids to the vitreous cavities. MAIN OUTCOME MEASURES:After 7 days,retinal preparations and EGFP expression in RGCs were observed by fluorescence microscopy.RGC quantification in the retinal ganglion cell layer was performed.In addition,EGFP mRNA expression was semi-quantitatively determined by RT-PCR. RESULTS:The transfection efficiency of EGFP to RGCs by microbubbles with ultrasound was significantly greater than the other groups,and no obvious damage was detected in the RGCs. | 5 | Lycium barbarum polysaccharides ameliorate canine acute liver injury by reducing oxidative stress,protecting mitochondrial function,and regulating metabolic pathways显示文摘The development of acute liver injury can result in liver cirrhosis,liver failure,and even liver cancer,yet there is currently no effective therapy for it.The purpose of this study was to investigate the protective effect and therapeutic mechanism of Lycium barbarum polysaccharides(LBPs)on acute liver injury induced by carbon tetrachloride(CCl_(4)).To create a model of acute liver injury,experimental canines received an intraperitoneal injection of 1 mL/kg of CCl_(4)solution.The experimental canines in the therapy group were then fed LBPs(20 mg/kg).CCl_(4)-induced liver structural damage,excessive fibrosis,and reduced mitochondrial density were all improved by LBPs,according to microstructure data.By suppressing Kelch-like epichlorohydrin(ECH)-associated protein 1(Keap1),promoting the production of sequestosome 1(SQSTM1)/p62,nuclear factor erythroid 2-related factor 2(Nrf2),and phaseⅡdetoxification genes and proteins downstream of Nrf2,and restoring the activity of anti-oxidant enzymes like catalase(CAT),LBPs can restore and increase the antioxidant capacity of liver.To lessen mitochondrial damage,LBPs can also enhance mitochondrial respiration,raise tissue adenosine triphosphate(ATP)levels,and reactivate the respiratory chain complexes I–V.According to serum metabolomics,the therapeutic impact of LBPs on acute liver damage is accomplished mostly by controlling the pathways to lipid metabolism.9-Hydroxyoctadecadienoic acid(9-HODE),lysophosphatidylcholine(Lyso PC/LPC),and phosphatidylethanolamine(PE)may be potential indicators of acute liver injury.This study confirmed that LBPs,an effective hepatoprotective drug,may cure acute liver injury by lowering oxidative stress,repairing mitochondrial damage,and regulating metabolic pathways. | JIANJIA HUANG YUMAN BAI WENTING XIE RONGMEI WANG WENYUE QIU SHUILIAN ZHOU ZHAOXIN TANG JIANZHAO LIAO RONGSHENG SU | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,2: | 2 |
| 6 | Arginine methylation of ribose-5-phosphate isomerase A senses glucose to promote human colorectal cancer cell survival显示文摘Cancer cells remodel their metabolic network to adapt to variable nutrient availability. Pentose phosphate pathway(PPP) plays protective and biosynthetic roles by oxidizing glucose to generate reducing power and ribose. How cancer cells modulate PPP activity in response to glucose supply remains unclear. Here we show that ribose-5-phosphate isomerase A(RPIA), an enzyme in PPP, directly interacts with co-activator associated arginine methyltransferase 1(CARM1) and is methylated at arginine 42(R42). R42 methylation up-regulates the catalytic activity of RPIA. Furthermore, glucose deprivation strengthens the binding of CARM1 with RPIA to induce R42 hypermethylation. Insufficient glucose supply links to RPIA hypermethylation at R42, which increases oxidative PPP flux. RPIA methylation supports ROS clearance by enhancing NADPH production and fuels nucleic acid synthesis by increasing ribose supply. Importantly, RPIA methylation at R42 significantly potentiates colorectal cancer cell survival under glucose starvation.
