| 1 | Selection and development of representative simple sequence repeat primers and multiplex SSR sets for high throughput automated genotyping in maize显示文摘In the current study, 1900 maize simple sequence repeat (SSR) primers published in MaizeGDB were screened utilizing reference literature, 15 representative Chinese maize inbred lines and 15 Chinese maize hybrids from national regional testing. In total, 500 highly polymorphic primers were identified and used to construct a genetic map. 100 evenly distributed primers, 10 primers per chromosome, were further selected as a set of universal SSR core primers, recommended as preferred primers for general studies. These core primers were then redesigned and used to construct a high throughput multiplex PCR system based on a five-color fluorescence capillary detection system. | 4 | An improved Landsat Image Mosaic of Antarctica显示文摘A revised Landsat Image Mosaic of Antarctica (LIMA) is presented, using the 1073 multi-band scenes of the original Landsat-7 ETM+ LIMA image collection available at the United States Geological Survey (USGS: http://gffzza64998f73d704ee0hpf0xu0cwuk5566cv.ffgz.tsg.suse.edu.cn/). Three improvements have been applied during the data processing: (1) DN saturation is adjusted by adopting a linear regression, which has a lower root mean square error than the ratio regression used by LIMA; (2) solar elevation angle is calculated using pixel-level latitude/longitude and the acquisition time and date of the central pixel of the scene, improving slightly upon the bilinear interpolation of the solar elevation angles of scene corners applied in LIMA; and (3) two additional image bands, Band 5 and Band 7, are sharpened using the panchromatic band (Band 8) and a Gram-Schmidt Spectral Sharpening algorithm to more easily distinguish snow, cloud and exposed rocks. The final planetary reflectance product is stored in 16-bit bands to preserve the full radiometric content of the scenes. | 19 | Alkaline activation of endogenous latent TGFβ1 by an injectable hydrogel directs cell homing for in situ complex tissue regeneration显示文摘Utilization of the body’s regenerative potential for tissue repair is known as in situ tissue regeneration.However,the use of exogenous growth factors requires delicate control of the dose and delivery strategies and may be accompanied by safety,efficacy and cost concerns.In this study,we developed,for the first time,a biomaterial-based strategy to activate endogenous transforming growth factor beta 1(TGFβ1)under alkaline conditions for effective in situ tissue regeneration.We demonstrated that alkaline-activated TGFβ1 from blood serum,bone marrow fluids and soaking solutions of meniscus and tooth dentin was capable of increasing cell recruitment and early differentiation,implying its broad practicability.Furthermore,we engineered an injectable hydrogel(MS-Gel)consisting of gelatin microspheres for loading strong alkaline substances and a modified gelatin matrix for hydrogel click crosslinking.In vitro models showed that alkaline MS-Gel controllably and sustainably activated endogenous TGFβ1 from tooth dentin for robust bone marrow stem cell migration.More importantly,infusion of in vivo porcine prepared root canals with alkaline MS-Gel promoted significant pulp-dentin regeneration with neurovascular stroma and mineralized tissue by endogenous proliferative cells.Therefore,this work offers a new bench-to-beside translation strategy using biomaterial-activated endogenous biomolecules to achieve in situ tissue regeneration without the need for cell or protein delivery. | Sainan Wang Yuting Niu Peipei Jia Zheting Liao Weimin Guo Rodrigo Cotrim Chaves Khanh-Hoa Tran-Ba Ling He Hanying Bai Sam Sia Laura J.Kaufman Xiaoyan Wang Yongsheng Zhou Yanmei Dong Jeremy J.Mao | 2022 | Bioactive Materials2022,7,9: | 1 |
