SARS-Coronavirus-2 Nsp13 Possesses NTPase and RNA Helicase Activities That Can Be Inhibited by Bismuth Salts显示文摘The ongoing outbreak of Coronavirus Disease 2019(COVID-19)has become a global public health emergency.SARScoronavirus-2(SARS-CoV-2),the causative pathogen of COVID-19,is a positive-sense single-stranded RNA virus belonging to the family Coronaviridae. For RNA viruses,virus-encoded RNA helicases have long been recognized to play pivotal roles during viral life cycles by facilitating the correct folding and replication of viral RNAs.Here, our studies show that SARS-CoV-2-encoded nonstructural protein 13(nsp13)possesses the nucleoside triphosphate hydrolase(NTPase)and RNA helicase activities that can hydrolyze all types of NTPs and unwind RNA helices dependently of the presence of NTP,and further characterize the biochemical characteristics of these two enzymatic activities associated with SARS-CoV-2 nsp13.Moreover,we found that some bismuth salts could effectively inhibit both the NTPase and RNA helicase activities of SARS-CoV-2 nsp13 in a dose-dependent manner. Thus, our findings demonstrate the NTPase and helicase activities of SARS-CoV-2 nsp13,which may play an important role in SARS-CoV-2 replication and serve as a target for antivirals. | Ting Shu Muhan Huang Di Wu Yujie Ren Xueyi Zhang Yang Han Jingfang Mu Ruibing Wang Yang Qiu Ding-Yu Zhang Xi Zhou | 2020 | Virologica Sinica2020,35,3: | 11
The ORF3a protein of SARS-CoV-2 induces apoptosis in cells显示文摘Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)has caused the ongoing pandemic of Coronavirus Disease 2019.SARS-CoV-2 belongs to the genus Betacoronavirus of the Coronaviridae family, which includes SARS-CoV and Middle East respiratory syndrome coronavirus.1,2 Coronavirus-encoded accessory proteins play critical roles in virus–host interactions and the modulation of host immune responses, thereby contributing to coronaviral pathogenicity via different strategies.3 However, the functions of SARS-CoV-2-encoded accessory proteins are not well understood.Apoptosis is a predominant type of programmed cell death, and has been recognized as an important host antiviral defense mechanism that controls viral infection and regulates the inflammatory response.4,5 Previous studies have reported that the SARS-CoV-encoded accessory protein ORF3a can induce apoptosis in cells,6,7 leading to the question of whether SARS-CoV-2 ORF3a also has pro-apoptotic activity. Here,we investigated the potential apoptosis-inducing activity of SARS-CoV-2 ORF3a in different cell lines and compared the pro-apoptotic activities of SARS-CoV-2 ORF3a with those of SARS-CoV ORF3a using the same system. | Yujie Ren Ting Shu Di Wu Jingfang Mu Chong Wang Muhan Huang Yang Han Xue-Yi Zhang Wei Zhou Yang Qiu Xi Zhou | 2020 | Cellular & Molecular Immunology2020,17,8: | 11

Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis. The symptoms of COVID-19 range from mild to severe, but the physiological changes associated with COVID-19 are barely understood. In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms. We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients, indicating that the development of COVID-19 affected their whole-body metabolism. In particular, malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction, respectively. It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms. And,more importantly, guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases. In addition, dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19. | Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou | 2020 | National Science Review2020,7,7: | 9
Impact factors of fractal analysis of porous structure显示文摘Characterization of pore structure is one of the key problems for fabrication and application research on porous materials. But, complexity of pore structure makes it difficult to characterize pore structure by Euclidean geometry and traditional experimental methods. Fractal theory has been proved effective to characterize the complex pore structure. The box dimension method based on fractal theory was applied to characterizing the pore structure of fiber porous materials by analyzing the electronic scanning microscope (SEM) images of the porous materials in this paper. The influences of image resolution, threshold value, and image magnification on fractal analysis were investigated. The results indicate that such factors greatly affect fractal analysis process and results. The appropriate magnification threshold and fractal analysis are necessary for fractal analysis. | TANG HuiPing, ZHU JiLei, XI ZhengPing, DI XiaoBo, WANG JianYong & AO QingBo State Key Laboratory of Porous Metal Materials, Northwest Institute for Nonferrous Metal Research, Xi’an 710016, China | 2010 | Science China(Technological Sciences)2010,53,2: | 6
Lipoprotein(a) as a Marker for Predicting the Presence and Severity of Coronary Artery Disease in Untreated Chinese Patients Undergoing Coronary Angiography显示文摘Objective Low-density lipoprotein cholesterol(LDL-C) has been well known as the risk factor of coronary artery disease(CAD). However, the role of lipoprotein(a) [Lp(a)] in the development of CAD is of great interest but still controversial. Thus, we aim to explore the effect of Lp(a) on predicting the presence and severity of CAD in Chinese untreated patients, especially in combination with LDL-C. Methods We consecutively recruited 1,980 non-treated patients undergoing coronary angiography, among which 1,162 patients were diagnosed with CAD. Gensini score(GS) was used to assess the severity of CAD. Lp(a) was measured by immunoturbidimetric method. Results Patients with CAD had higher level of LDL-C and Lp(a) compared with non-CAD(P < 0.05). Multivariable logistic regression revealed that Lp(a) > 205 mg/L(highest tertile) predicted 1.437-fold risk for CAD(95% CI: 1.108-1.865, P = 0.006) and 1.480-fold risk for high GS(95% CI: 1.090-2.009, P = 0.012) respectively. Interestingly, concomitant elevated level of Lp(a) and LDL-C conferred the highest risk for both presence [OR = 1.845, 95% CI: 1.339-2.541, P < 0.001] and severity [OR = 1.736, 95% CI: 1.188-2.538, P = 0.004] of CAD. Conclusion Lipoprotein(a) is a useful marker for predicting the presence and severity of CAD, especially combined with LDL-C. | SUN Di ZHAO Xi LI Sha ZHANG Yan WU Na Qiong ZHU Cheng Gang GUO Yuan Lin GAO Ying QING Ping LIU Geng DONG Qian LI Jian Jun | 2018 | Biomedical and Environmental Sciences2018,31,4: | 6
Natural gas leakage of Mizhi gas reservoir in Ordos Basin, recorded by natural gas fluid inclusion显示文摘Abundant natural gas inclusions were found in calcite veins filled in fractures of Central Fault Belt across the centre of Ordos Basin. Time of the calcite veins and characteristics of natural gas fluid inclusion were investigated by means of dating of thermolum luminescence (TL) and analyzing stable isotope of fluid inclusion. Results show that natural gas inclusion formed at 130―140℃ with salinity of 5.5 wt%―6.0 wt% NaCl. It indicates that natural gas inclusion is a kind of thermal hydrocarbon fluid formed within the basin. Method of opening inclusion by heating was used to analyze composition of fluid inclusion online, of which the maximal hydrocarbon gas content of fluid inclusion contained in veins is 2.4219 m3/t rock and the maximal C1/Σci ratio is 91%. Laser Raman spectroscopy (LRS) was used to analyze chemistry of individual fluid inclusion in which the maximal hydrocarbon gas content is 91.6% compared with little inorganic composition. Isotope analysis results of calcite veins show that they were deposited in fresh water, in which the δ13CPDB of calcite veins is from -5.75‰ to 15.23‰ andδ18OSMOW of calcite veins is from 21.33‰ to 21.67‰. Isotope results show thatδ13C1 PDB of natural gas fluid inclusion is from -21.36‰ to -29.06‰ and δDSMOW of that is from -70.89‰ to -111.03‰. It indicates that the gas of fluid inclusion formed from coal source rocks and it is the same as that of natural gas of Mizhi gas reservoir. Results of TL dating show that time of calcite vein is (32.4±3.42)×104 a, which is thought to be formation time of gas inclusion. It indicated that natural gas inclusion contained in calcite veins recorded natural gas leakage from Mizhi gas reservoir through the Central Fault Belt due to Himalayan tectonic movement. | LI RongXi DI LingJun XI ShengLi | 2007 | Science China Earth Sciences2007,50,z2: | 5

800 Gbit/s transmission over 1 km single-mode fiber using a four-channel silicon photonic transmitter显示文摘We demonstrate the optical transmission of an 800 Gbit/s(4×200 Gbit/s)pulse amplitude modulation-4(PAM-4)signal and a 480 Gbit/s(4×120 Gbit/s)on–off-keying(OOK)signal by using a high-bandwidth(BW)silicon photonic(SiP)transmitter with the aid of digital signal processing(DSP).In this transmitter,a four-channel SiP modulator chip is co-packaged with a four-channel driver chip, with a measured 3 dB BW of 40 GHz.DSP is applied in both the transmitter and receiver sides for pre-/post-compensation and bit error rate(BER)calculation.Back-to-back(B2B)BERs of the PAM-4 signal and OOK signal are first measured for each channel of the transmitter with respect to a variety of data rates.Similar BER performance of four channels shows good uniformity of the transmitter between different channels. The BER penalty of the PAM-4 and OOK signals for 500 m and 1 km standard single-mode fiber(SSMF)transmission is then experimentally tested by using one channel of the transmitter. For a 200 Gbit/s PAM-4 signal, the BER is below the hard-decision forward error correction(HD-FEC)threshold for B2B and below the soft-decision FEC(SD-FEC)threshold after 1 km transmission. For a 120 Gbit/s OOK signal, the BER is below SD-FEC threshold for B2B.After 500 m and 1 km transmission, the data rate of the OOK signal shrinks to 119 Gbit/s and 118 Gbit/s with the SD-FEC threshold, respectively. Finally, the 800 Gbit/s PAM-4 signal with 1 km transmission is achieved with the BER of all four channels below the SD-FEC threshold. | Hongguang Zhang Miaofeng Li Yuguang Zhang Di Zhang Qiwen Liao Jian He Shenglei Hu Bo Zhang Lei Wang Xi Xiao Nan Qi Shaohua Yu | 2020 | Photonics Research2020,8,11: | 5
Effects of Hydrogen on Behaviour of Low Cycle Fatigue of 2.25Cr-1Mo Steel显示文摘The effects of hydrogen atoms on behaviour of low cycle fatigue of 2.25Cr-1Mo steel have been investigated in present work. The results indicate that the cyclic softening rate and low cycle fatigue life are respectively increased and reduced remarkably by hydrogen atoms. In addition, hydrogen atoms make the original stress amplitude of low cycle fatigue increase, which is because of the drag effect of hydrogen atoms on the moving dislocations. Analyses using electron microscopy show that hydrogen atoms accelerate crack initiation of low cycle fatigue from inclusion and transfer the source of low cycle fatigue crack from the surface of specimen to the inclusion, which results in the marked decrease of low cycle fatigue life. The increase of cyclic softening rate for hydrogen charged specimen is due to hydrogen atoms accelerating the initiating and growing of microvoids from the secondary phase particles in the steel. The reducing of the drag effect of hydrogen atoms on moving dislocations is also helpful to the increase of the cyclic softening rate. | Guangwei HAN and Di FENG (Central Iron & Steel Research Institute, Beijing, 100081, China)(To whom correspondence should be addressed)Yujiu SONG(Dept. of Mater. | 15 | Credit-based scheme for security-aware and fairness-aware resource allocation in cloud computing显示文摘Cloud computing systems include different types of participants with varied requirements for resources and multiple tasks; these varying requirements must be considered in the design of fairness-aware resource allocation schemes for better resources sharing. However, some participants may be malicious with a goal to damage the resource allocation fairness and increase their own utility. Hence, the resource scheduling policy must guarantee allocation fairness among the participants; further, it must ensure that fairness is not affected by the malicious usage of resources, that could cause resource exhaustion and lead to denial of service. In order to address this challenge, we propose a credit-based mechanism for resource allocation that will avoid the malicious usage of resources and, simultaneously, guarantee allocation fairness. In our scheme, a credit factor is introduced for each participant in order to evaluate the history of resource utilization and determine future resource allocation. Our model encourages a participant to release the occupied resources in timely manner after the completion of a task and imposes a punishment for malicious occupation of resources. We prove the fairness of our model and provide linear and variable gradient approaches to determine the credit factor for different scenarios. We simulate our model and perform experiments on a real cloud computing platform. The results prove the rationality, effectiveness and correctness of our approaches. | Di LU Jianfeng MA Cong SUN Xindi MA Ning XI | 2017 | Science China(Information Sciences)2017,60,5: | 4

A review of common-path off-axis digital holography:towards high stable optical instrument manufacturing显示文摘Digital holography possesses the advantages of wide-field,non-contact,precise,and dynamic measurements for the complex amplitude of object waves.Today, digital holography and its derivatives have been widely applied in interferometric measurements,three-dimensional imaging, and quantitative phase imaging, demonstrating significant potential in the material science, industry, and biomedical fields, among others. However,in conventional off-axis holographic experimental setups, the object and reference beams propagate in separated paths, resulting in low temporal stability and measurement sensitivity.By designing common-path configurations where the two interference beams share the same or similar paths, environmental disturbance to the two beams can be effectively compensated. Therefore, the temporal stability of the experimental setups for hologram recording can be significantly improved for time-lapsing measurements. In this review,we categorise the common-path models as lateral shearing, point diffraction, and other types based on the different approaches to generate the reference beam.Benefiting from compact features,common-path digital holography is extremely promising for the manufacture of highly stable optical measurement and imaging instruments in the future. | Jiwei Zhang Siqing Dai Chaojie Ma Teli Xi Jianglei Di Jianlin Zhao | 2021 | Light(Advanced Manufacturing)2021,2,3: | 4
HS/PCs 的现出症状之前的潜的移植和安全从人的脐的绳索血膨胀了显示文摘 AIM: To expand hematopoietic/progenitor stem cells (HS/PCs) from umbilical cord blood (UCB) and prepare the HS/PC product, and analyze preclinical transplantation and safety of HS/PC product. METHODS: Human bone marrow-derived mesenchymal stem cells (MSCs) were used as feeder cells to expand HS/PCs from UCB in a serum-free culture system. The proliferation potential of HS/PCs was analyzed. The expanded HS/PCs were suspended in the L-15 medium to prepare the HS/PC product. The contamination of bacteria, fungi and mycoplasmas, the infection of exogenous virus, the concentration of bacterial endotoxin, and the SCF residual in HS/PC product were determined. Finally, cells from the HS/PC product with or without bone marrow-derived mesenchymal stem cells (BM-MSCs) were transplanted into the irradiated NOD/SCID mice to determine the in vivo engraftment potential. RESULTS: After co-culture for 10 d, the total nuclear cells (TNCs) increased 125-fold, and CD34 + cells increased 43-fold. The granulocyte-macrophage colonyforming cells (GM-CFCs) and erythroid colony-forming cells (E-CFCs) increased 3.3and 4.7-fold respectively. The expanded cells were collected and prepared as the expanded product of HS/PCs by re-suspending cells in L-15 medium. For preclinical safety, the HS/PC product was analysed for contamination by bacteria, fungi and mycoplasmas, the bacterial endotoxin concentration and the SCF content. The results showed that the HS/PC product contained no bacteria, fungi or mycoplasmas. The bacterial endotoxin concentration was less than the detection limit of 6 EU/mL, and residual SCF was 75 pg/mL. Based on clinical safety, the HS/PC product was qualified for clinical transplantation. Finally, the HS/PC product was transplanted the irradiated mice where it resulted in rapid engraftment of hematopoietic cells. CONCLUSION: HSPC product prepared from UCB in the serum-free culture system with hMSCs as feeder cells should be clinically safe and effective for clinical transplantation. | Chun-Juan Guo Ying Gao Di Hou Dong-Yan Shi Xiang-Min Tong Dan Shen Yong-Mei Xi Jin-Fu Wang | 2011 | World Journal of Stem Cells2011,3,5: | 4
Combating maternal mortality in China:experience, challenges and opportunities显示文摘Since the People’s Republic of China founded, especially over the last 15 years, maternal mortality rate(MMR)has fallen dramatically. In 2014,China has achieved the target of a reduction of 75%MMR from 1990 to 2015(MDG 5)ahead of schedule. This achievement can be attributed to the improvement of legislation and policy, government commitment and investment on maternal health, extensive international cooperation, implementation of the maternal health program, development of maternal and child health(MCH)service system and development of MCH surveillance systems.Although China has achieved MDG 5,there are many kinds of difficulties and challenges in achieving the post-2015 Sustainable Development Goals(SDGs).Therefore,multi-strategy actions need to be considered to seize the opportunities to further reduce the MMR in the future. | Jiangli Di Qi Yang Xi Jin | 2017 | Global Health Journal2017,1,3: | 4
