| 2 | Early inoculation with caecal fermentation broth alters small intestine morphology,gene expression of tight junction proteins in the ileum,and the caecal metabolomic profiling of broilers显示文摘Background:The establishment of stable microbiota in early life is beneficial to the individual.Changes in the intestinal environment during early life play a crucial role in modulating the gut microbiota.Therefore,early intervention to change the intestinal environment can be regarded as a new regulation strategy for the growth and health of poultry.However,the effects of intestinal environmental changes on host physiology and metabolism are rarely reported.This study was conducted to investigate the effects of early inoculation with caecal fermentation broth on small intestine morphology,gene expression of tight junction proteins in the ileum,and cecum microbial metabolism of broilers.Results:Our data showed that early inoculation with caecal fermentation broth could improve intestine morphology.The small intestine villus height was significantly increased(P<0.05)in the intervened broilers compared to the control group,especially on day 28.A similar result was observed in the ratio of villus height to crypt depth(P<0.05).Meanwhile,we found early inoculation significantly increased(P<0.05)the expression levels of zonula occludens-1(ZO1)on days 14 and 28,claudin-1(CLDN1)on day 28,whereas the gene expression of claudin-2(CLDN2)was significantly decreased(P<0.05)on days 14 and 28.Gas chromatography time-of-flight/mass spectrometry(GC-TOF/MS)technology was further implemented to systematically evaluate the microbial metabolite profiles.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)displayed a distinct trend towards separation between the fermentation broth group(F group)and the control group(C group).The differentially expressed metabolites were identified,and they were mainly functionally enriched in beta-alanine metabolism and biosynthesis of unsaturated fatty acids.In addition,1,3-diaminopropane was selected as a key biomarker that responded to early inoculation with caecal fermentation broth.Conclusions:These results provide insight into intestinal metabolomics and confirm that early inoculation with caecal fermentation broth can be used as a potential strategy to improve intestinal health of broilers. | Yujie Gong Wenrui Xia Xueting Wen Wentao Lyu Yingping Xiao Hua Yang Xiaoting Zou | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 3 |
| 3 | Intrauterine growth restriction alters nutrient metabolism in the intestine of porcine offspring显示文摘Background:Intrauterine growth restriction(IUGR)has negative impacts on the postnatal survival,growth and development of humans and animals,with not only on newborns but also adulthood.However,the characteristics for nutrient digestion and absorption in IUGR offspring are still largely unknown.Therefore,the normal birth weight(NBW)and IUGR growing pigs were used in this study to investigate their differences in nutrient utilization,with an expectition for further nutritional optimization of the IUGR offspring during their later life.Methods:Twelve IUGR and 12 NBW growing pigs were fitted with catheters in their portal vein to measure blood flow rate as well as nutrients and metabolites in plasma.The digestibilities of nutrients in different intestinal segments,and bacterial fermentation in the large intestine were examined to reveal the characteristics of nutrients utilization in IUGR versus NBW pigs.Results:The rate of portal venous blood flow did not differ beween IUGR and NBW pigs.Plasma concentrations of total cholesterol,triglycerides and glucose were much lower but those of urea were higher in the portal vein of IUGR pigs,compared with the NBW pigs.The ileal digestibility of dry matter,gross energy and starch were lower in IUGR pigs than in NBW pigs.IUGR increased hindgut microbial diversity and bacterial fermentation activity in the caecum.In vitro cross-fermentation of ileal digesta by caecal microbes of NBW and IUGR pigs showed that gas production was much higher for IUGR ileal digesta regardless of the source of caecal inocula.Conclusion:IUGR impairs the nutrient digestion and absorption in small intestine,reduces caecal microbial diversity and promotes bacterial fermentation in the large intestine during the growing phase.These findings aid in our understanding of nutrient metabolism in IUGR pigs and provide the basis for future nutritional interventions. | Tiantian Li Shimeng Huang Long Lei Shiyu Tao Yi Xiong Guoyao Wu Jie Hu Xiongkun Yuan Shengjun Zhao Bin Zuo Hongjian Yang Yingping Xiao Gang Lin Junjun Wang | 2021 | Journal of Animal Science and Biotechnology2021,12,2: | 2 |
| 4 | Donor age and body weight determine the effects of fecal microbiota transplantation on growth performance,and fecal microbiota development in recipient pigs显示文摘Background:The application of fecal microbiota transplantation(FMT)to improve swine growth performance has been sporadically studied.Most of these studies used a single microbiota source and thus the effect of donor characteristics on recipient pigs’fecal microbiota development and growth performance is largely unknown.Results:In this study,we collected feces from six donors with heavy(H)or light(L)body weight and different ages(d 42,nursery;d 96,growing;and d 170,finisher)to evaluate their effects on the growth performance and fecal microbiota development of recipient pigs.Generally,recipients that received two doses of FMT from nursery and finisher stages donor at weaning(21±2 days of age)inherited the donor’s growth pattern,while the pigs gavaged with grower stage material exerted a numerically greater weight gain than the control pigs regardless of donor BW.FMT from heavier donors(NH,GH,and FH)led to the recipients to have numerically increased growth compared to their lighter counterparts(NL,GL,and FL,respectively)throughout the growing and most finishing stages.This benefit could be attributed to the enrichment of ASV25 Faecalibacterium,ASV61 Faecalibacterium,ASV438 Coriobacteriaceae_unclassified,ASV144 Bulleidia,and ASV129 Oribacterium and decrease of ASV13 Escherichia during nursery stage.Fecal microbiota transplantation from growing and finishing donors influenced the microbial community significantly in recipient pigs during the nursery stage.FMT of older donors’gut microbiota expedited recipients’microbiota maturity on d 35 and 49,indicated by increased estimated microbiota ages.The ageassociated bacterial taxa included ASV206 Ruminococcaceae,ASV211 Butyrivibrio,ASV416 Bacteroides,ASV2 Streptococcus,and ASV291 Veillonellaceae.The body weight differences between GL and GH pigs on d 104 were associated with the increased synthesis of the essential amino acid,lysine and methionine,mixed acid fermentation,expedited glycolysis,and sucrose/galactose degradation.Conclusions:Overall,our study provided insights into how donor age and body weight affect FMT outcomes regarding growth performance,microbiota community shifts,and lower GI tract metabolic potentials.This study also provided guidance to select qualified donors for future fecal microbiota transplantation. | Xiaofan Wang Tsungcheng Tsai Bin Zuo Xiaoyuan Wei Feilong Deng Ying Li Charles V.Maxwell Hua Yang Yingping Xiao Jiangchao Zhao | 2022 | Journal of Animal Science and Biotechnology2022,13,5: | 1 |
| 5 | Association of growth rate with hormone levels and myogenic gene expression profile in broilers显示文摘Background: The growth rate often varies among individual broilers of the same breed under a common management condition. To investigate whether a variation in the growth rate is associated with a difference in hormone levels and myogenic gene expression profile in broilers, a feeding trial was conducted with 10,000 newly hatched Ross 308 chicks in a commercial production facility under standard management. At 38 d of age,30 fast-, 30 medium-, and 30 slow-growing broilers were selected among 600 healthy male individuals. The levels of insulin-like growth factor-1(IGF-1), triiodothyronine(T3), thyroxine(T4), and growth hormone in the serum or breast muscle were assayed by ELISA or RIA kits, and the expression levels of several representative pro-and anti-myogenic genes in the breast muscle were also measured by real-time PCR.Results: Results showed that both absolute and relative weights of the breast muscle were in linear positive correlations with the body weight of broilers(P < 0.001). Fast-growing broilers had higher concentrations of IGF-1 than slow-growing broilers(P < 0.05) in both the serum and breast muscle. The serum concentration of T3 was significantly higher in fast-growing birds than in slow-growing birds(P < 0.05). However, no difference was observed in growth hormone or T4 concentration among three groups of birds. Additionally, a decreased expression of an anti-myogenic gene(myostatin) and increased expressions of pro-myogenic genes such as myogenic differentiation factor 1, myogenin, muscle regulatory factor 4, myogenic factor 5, IGF-1, and myocyte enhancer factor 2B, C, and D were observed in fast-growing broilers(P < 0.05), relative to slow-growing broilers.Conclusions: Collectively, these findings suggested that the growth rate is linked to the hormone and myogenic gene expression levels in broiler chickens. | 9 | Large-scale roll-to-roll printed,flexible and stable organic bulk heterojunction photodetector显示文摘A flexible and stable photodetector shows great potential applications in intelligent wearable devices,health monitoring,and biological sensing.The high-output fabrication of flexible and stable photodetector via the large-scale printing process would accelerate its commercialization.Herein,a high performance,flexible organic bulk heterojunction(BHJ)photodetector with good stability is designed and fabricated via a large-scale roll-to-roll(R2R)micro-gravure printing technique on polyethylene terephthalate(PET)or paper substrate,in which the organic BHJ active layer is structured with[6,6]-phenyl C_(61)butyric acid methyl ester(PCBM)and a donor-acceptor copolymer,i.e.,employing 4,8-bis(2-ethylhexylthiophene)benzo[1,2-b;3,4-b′]dithiophene(BDTT)as the donor unit and 5,8-bis(5-thiophen-2-yl)-6,7-difluoro-2,3-bis(4-ethylhexyloxy-1-mata-luorophenyl)quinoxaline(ffQx)as the acceptor unit(PBDTT-ffQx).The PBDTT-ffQx/PCBM BHJ photodetector shows a broad photoresponse in ultraviolet and visible light,a high detectivity(D^(*))value up to 6.19×10^(11)Jones,and an excellent Iphoto/Idark as high as 5.6×10^(2).It exhibits excellent flexibility and stability.Its performance parameters could maintain over 80%of original values after bending 10,000 cycles or exposing in ambient condition(humidity~50%,temperature~30℃)for 50 days without any encapsulation.More importantly,the R2R micro-gravure printed PBDTT-ffQx/PCBM BHJ active layer is great homogeneous,and the responsivity(R)values of photodetector arrays show a very narrow distribution.The research results show that a high-performance PBDTT-ffQx/PCBM BHJ photodetector with well reliability and reproducibility can be fabricated via the R2R micro-gravure printing technique,which provides an available strategy for fabricating large-area and flexible electronic and optoelectronic devices. | Sichao Tong Jun Yuan Chujun Zhang Chunhua Wang Baoxing Liu Jianqiang Shen Huayan Xia Yingping Zou Haipeng Xie Jia Sun Si Xiao Jun He Yongli Gao Junliang Yang | 2018 | npj Flexible Electronics2018,2,1: | 0 |
| 10 | Diflurobenzothiadiazole core-based noncovalently fused small molecule acceptor exhibiting over 12% efficiency and high fill factor显示文摘The versatility and flexibility of organic photoelectric materials endow organic photovoltaic cells fine function modulation and huge commercial potential. In this work, a new noncovalent fused-ring small molecule acceptor(SMA) BID-4 F has been synthesized for high-efficient organic solar cells(OSCs). BID-4 F consists of a diflurobenzothiadiazole(DFBT) core, ladder-like indacenodithiophene(IDT) spacers, and dicyanoindanone electron-withdrawing end groups, which are supposed to be conformationally interlocked by noncovalent interactions, leading to good molecular planarity. In addition, compact solid state stacking was revealed by UV–vis–NIR absorption spectrum. The optimized PM6:BID-4 F based device delivers an eminent power conversion efficiency(PCE) of 12.30% with a high open-circuit voltage(Voc) of 0.92 V and a high fill factor(FF) of 74.3%. Most importantly, the PCE and FF are among one of the highest values reported for the OSCs based on the unfused-ring SMAs.
