1Responses of the Toll-like receptor and melanoma differentiation-associated protein 5 signaling pathways to avian infectious bronchitis virus infection in chicks显示文摘Avian infectious bronchitis virus(IBV) is a Gammacoronavirus in the family Coronaviridae and causes highly contagious respiratory disease in chickens. Innate immunity plays significant roles in host defense against IBV. Here, we explored the interaction between IBV and the host innate immune system. Severe histopathological lesions were observed in the tracheal mucosa at 3–5days post inoculation(dpi) and in the kidney at 8 dpi, with heavy viral loads at 1–11 and 1–28 dpi,respectively. The expression of m RNAs encoding Toll-like receptor(TLR) 3 and TLR7 were upregulated at 3–8 dpi, and that of TIR-domain-containing adapter-inducing interferon(IFN) β(TRIF) was upregulated at 21 dpi in the trachea and kidney. Myeloid differentiation primary response protein 88(My D88) was upregulated in the trachea during early infection. Tumor necrosis factor receptor-associated factor(TRAF) 3 and TRAF6 were upregulated expression in both tissues.Moreover, melanoma differentiation-associated protein 5(MDA5), laboratory of genetics and physiology 2(LGP2), stimulator of IFN genes(STING), and mitochondrial antiviral signaling protein(MAVS), as well as TANK binding kinase 1(TBK1), inhibitor of kappa B kinase(IKK) ?, IKKα, IKKβ,IFN regulatory factor(IRF) 7, nuclear factor of kappa B(NF-κB), IFN-α, IFN-β, various interleukins(ILs), and macrophage inflammatory protein-1β(MIP-1β) were significantly upregulated in the trachea and downregulated in the kidney. These results suggested that the TLR and MDA5 signaling pathways and innate immune cytokine were induced after IBV infection. 2Topic enhanced deep structured semantic models for knowledge base question answering显示文摘Knowledge Base Question Answering(KBQA) is a hot research topic in natural language processing(NLP). The most challenging problem in KBQA is how to understand the semantic information of natural language questions and how to bridge the semantic gap between the natural language questions and the structured fact triples in knowledge base. This paper focuses on simple questions which can be answered by a single fact triple in knowledge base. We propose a topic enhanced deep structured semantic model for KBQA. The proposed method considers the task of KBQA as a matching problem between questions and the subjects and predicates in knowledge base. And the proposed model consists of two stages to match the subjects and predicates, respectively. In the first stage, we propose a Convolutional based Topic Entity Extraction Model(CTEEM) to extract topic entities mentioned in questions. With the extracted entities, we can retrieve the relevant candidate fact triples from knowledge base and obviously decrease the amount of noising candidates. In the second stage, we employ Deep Structured Semantic Models(DSSMs) to compute the semantic relevant score between questions and predicates in the candidates. And we combine the semantic level and the lexical level scores to rank the candidates. We evaluate the proposed method on KBQA dataset released by NLPCC-ICCPOL 2016. The experimental results show that our proposed method achieves the third place among the 21 submitted systems.Furthermore, we also extend the DSSM by using Bi LSTM and integrate a convolutional structure on the top of Bi LSTM layers. 5mTOR pathway regulates the differentiation of peripheral blood Th2/Treg cell subsets in patients with pemphigus vulgaris显示文摘Pemphigus vulgaris(PV)is a chronic and potentially life-threatening autoimmune blistering disease.Aberrant mTOR pathway activity is involved in many autoimmune diseases.This study investigated the correlation of mTOR pathway(PI3K/AKT/mTOR/p70S6K)activity with the loss of balance in T helper 2/regulatory T(Th2/Treg)cells in the peripheral blood of PV patients.CD4^(+)T cells were isolated from 15 PV patients and 15 healthy controls(HCs),the ratios of Th2/CD4^(+)T cells and Treg/CD4^(+)T cells,the activity of the mTOR pathway(PI3K/AKT/mTOR/p70S6K),the transcription factors and cytokines of Th2 and Treg cells were detected.Primary CD4^(+)T cells from PV patients were cultured under Th2-or Treg-polarizing conditions with or without rapamycin in vitro.We found that PV patients showed significantly elevated serum IL-4 when compared with HCs,and serum IL-4 level was positively correlated with the titer of anti-Dsg1/3 antibody and disease severity,while the serum TGF-βlevel was negatively correlated with the titer of anti-Dsg3 antibody and disease severity.Meanwhile,PV patients showed increased Th2/CD4^(+)T cell ratio;decreased Treg/CD4^(+)T cell ratio;elevated mRNA of PI3K,AKT,mTOR and protein of PI3K(P85),AKT,p-AKT(Ser473),mTOR,p-mTOR(Ser2448),p-p70S6K(Thr389),GATA3;reduced protein of forkhead box protein 3.Rapamycin inhibited Th2 cell differentiation and promoted Treg cell differentiation in vitro.These data suggest a close association between mTOR pathway activation and the loss of balance in Th2/Treg cells in peripheral blood of PV patients.Inhibiting mTORC1 can help restore the Th2/Treg balance.Kuan Lai Wenjing Zhang Songshan Li Zhiwen Zhang Shuangde Xie Meinian Xu Changxing Li Kang Zeng 2021Acta Biochimica et Biophysica Sinica2021,53,4:4
6Development and Evaluation of a Loop-mediated Isothermal Amplification Assay for the Rapid Detection and Identification of Pectobacterium carotovorum on Celery in the Field显示文摘Pectobacterium carotovorum is the causal agent of bacterial soft rot in a wide range of vegetable host species.Once P.carotovorum infects the plant,the spread of the disease is difficult to control.In this study,a rapid and sensitive method based on loop-mediated isothermal amplification(LAMP)was developed for detecting P.carotovorum in celery with soft rot using a primer set designed from the pmrA conserved sequence of P.carotovorum.The specificity of the LAMP primer set for P.carotovorum was extensively validated on both P.carotovorum strains and nontarget strains.The sensitivity was 1 pg of P.carotovorum genomic DNA,which demonstrated 10 times more sensitive than the conventional PCR assay.LAMP was also used to detect P.carotovorum in bacterial suspension.The lowest detection concentration was 104 CFU·mL^−1.In addition,a LAMP assay,in conjunction with a crude DNA extraction method,was successfully performed on P.carotovorum-infected samples derived from both artificially and naturally infected plants.In summary,the LAMP assay established in this study constitutes a simple,sensitive,and rapid method for the detection of P.carotovorum,and has potential application in the control of celery soft rot disease through early detection.Yanxia Shi Zhiwen Jin Xianglong Meng Lixue Wang Xuewen Xie Ali Chai Baoju Li 2020Horticultural Plant Journal2020,6,5:3
7Different Ablation Models for the Wall-plasma Interaction Process in Pulsed Plasma Thruster显示文摘Ablation excited by current pulses is a very critical physical process in pulse plasma thrusters(PPT).Its effects on wall-plasma interaction directly determine the PPT performances.In order to reveal the process of the ablated wall interaction with the discharge plasma in PPT,ablation models formulated by three different boundary conditions at the wall-plasma interface are studied.These are the two widely used high-speed evaporation models(Model-L and Model-M),and the recently developed Keida-Zaghloul model(Model-K)of the Knudsen layer that takes into account the internal degrees of freedom on the energy flux conservation.First,fundamental mechanisms of the three ablation models are clarified by comparative analysis in order to gain a comprehensive understanding of the wall-plasma interaction.Then,the applicability of different ablation models with the numerical solutions of LES-6 PPT is investigated in detail using magnetohydrodynamic(MHD)modeling.Results show that Model-L and Model-M are actually special cases of Model-K when a simplified jump conditions limited by high velocity at the vapor/plasma interface is used;A ratio of ablation rate in Model-L to that in Model-M is about 0.8at the same wall surface temperature,while it rises to 1 at different surface temperature determined by Model-L and Model-M in PPT.Even though Model-K solution requires significant computational time,it shows more accurate ablation feature for the wall-plasma interaction and possesses better computing precision of impulse bit during post-pulse which is useful for future studies of the late time ablation.YANG Lei LIU Xiangyang WANG Siyu XIE Kan WU Zhiwen WANG Ningfei 2013高电压技术2013,39,9:3

12Organising a juvenile ratio monitoring programme for 10 key waterbird species in the Yangtze River floodplain:analysis and proposals显示文摘Background:In the face of continued degradation and loss of wetlands in the Yangtze River floodplain(YRF),there is an urgent need to monitor the abundance and distribution of wintering waterbirds.To understand fully observed annual changes,we need to monitor demographic rates to understand factors affecting global population size.Annual reproduction success contributes to dynamic changes in population size and age structure,so an assessment of the juvenile ratio(i.e.first winter birds as a proportion of total number aged)of overwintering waterbirds can be an important indicator of the reproductive success in the preceding breeding season.Methods:During 2016-2019,we sampled juvenile ratios among 10 key waterbird species from the wetlands in the YRF.Based on these data,we here attempt to establish a simple,efficient,focused and reliable juvenile ratio monitoring scheme,to assess consistently and accurately relative annual breeding success and its contribution to the age structure among these waterbird species.Results:We compared juvenile ratio data collected throughout the winter and found that the optimal time for undertaking these samples was in the early stages of arrival for migratory waterbirds reaching their wintering area(early to mid-December).We recommend counting consistently at key points(i.e.those where>1%biogeographical flyway population were counted)at sites of major flyway importance(Poyang Lake,East Dongting Lake,Shengjin Lake,Caizi Lake,Longgan Lake and Chen Lake).Based on this,the error rate of the programme(155 planned points,the count of 10 waterbird species is 826-8955)is less than 5%.Conclusions:We established a juvenile ratio monitoring programme for 10 key waterbird species in the wetlands of the YRF,and discuss the feasibility and necessity of implementing such a future programme,and how to use these data in our monitoring and understanding of the population dynamics of these waterbird populations.Yuxi Wang Iderbat Damba Qingshan Zhao Yanbo Xie Xueqing Deng Rdi Ga Guanhua Liu Zhiwen Xu Yue Li Dali Gao Wenbin Xu Guoxun Chen Lei Cao 2021Avian Research2021,12,4:1

15Survival after radical cystectomy for bladder cancer:Multicenter comparison between minimally invasive and open approaches显示文摘Objective:To investigate oncological outcomes in patients with bladder cancer who underwent minimally invasive radical cystectomy(MIRC)or open radical cystectomy(ORC).Methods:We identified patients with bladder cancer who underwent radical cystectomy(RC)in 13 centers of the Chinese Bladder Cancer Consortium(CBCC).Perioperative outcomes were compared between MIRC and ORC.The influence of surgical approaches on overall survival(OS)and cancer-specific survival(CSS)in the entire study group and subgroups classified according to pathologic stage or lymph node(LN)status was assessed with the log-rank test.Multivariable Cox proportional hazard models were used to evaluate the association among OS,CSS and risk factors of interest.Results:Of 2098 patients who underwent RC,1243 patients underwent MIRC(1087 laparoscopic RC and 156 robotic-assisted RC,respectively),while 855 patients underwent ORC.No significant differences were noted in positive surgical margin rate and 90-day postoperative mortality rate.MIRC was associated with less estimated blood loss,more LN yield,higher rate of neobladder diversion,longer operative time,and longer length of hospital stay.There was no significant difference in OS and CSS according to surgical approaches(pZ0.653,and 0.816,respectively).Subgroup analysis revealed that OS and CSS were not significantly different regardless of the status of extravesical involvement or LN involvement.Multivariable Cox regression analyses showed that the surgical approach was not a significant predictor of OS and CSS.Conclusions:Our study showed that MIRC was comparable to conventional ORC in terms of OS and CSS.Weibin Xie Junming Bi Qiang Wei Ping Han Dongkui Song Lei Shi Dingwei Ye Yijun Shen Xin Gou Weiyang He Shaogang Wang Zheng Liu Jinhai Fan Kaijie Wu Zhiwen Chen Xiaozhou Zhou Chuize Kong Yang Liu Chunxiao Liu Abai Xu Baiye Jin Guanghou Fu Wei Xue Haige Chen Tiejun Pan Zhong Tu Tianxin Lin Jian Huang 2020Asian Journal of Urology2020,7,3:1

18A preliminary study on spatial unmasking of virtual separated sources显示文摘An experimental method with headphone virtual reproduction is proposed and a series of experiments to study forward masking effect when the masker and the masked signal are spatially separated in azimuth are conducted. Then, the masking thresholds are compared with those when the masker and the masked signal source are at the same place. The results show that, although both the thresholds of 0° and ±30° sound images increase with the sound pressure level (SPL) of the masker, spatial unmasking may be really observed. The maximum unmasking is as large as 15 dB. 19First experimental comparisons of laser-plasma interactions between spherical and cylindrical hohlraums at SGⅢ laser facility显示文摘We present our recent laser-plasmas instability(LPI)comparison experiment at the SGIII laser facility between the spherical and cylindrical hohlraums.Three kinds of filling are considered:vacuum,gas-filling with or without a capsule inside.A spherical hohlraum of 3.6 mm in diameter,and a cylindrical hohlraum of 2.4 mm?4.3 mm are used.The capsule diameter is 0.96 mm.A flat-top laser pulse with 3 ns duration and up to 92.73 kJ energy is used.The experiment has shown that the LPI level in the spherical hohlraum is close to that of the outer beam in the cylindrical hohlraum,while much lower than that of the inner beam.The experiment is further simulated by using our 2-dimensional radiation hydrodynamic code LARED-Integration,and the laser back-scattering fraction and the stimulated Raman scatter(SRS)spectrum are post-processed by the high efficiency code of laser interaction with plasmas HLIP.According to the simulation,the plasma waves are strongly damped and the SRS is mainly developed at the plasma conditions of electron density from 0.08 n_(c) to 0.1 n_(c) and electron temperature from 1.5 keV to 2.0 keV inside the hohlraums.However,obvious differences between the simulation and experiment are found,such as that the SRS back-scattering is underestimated,and the numerical SRS spectrum peaks at a larger wavelength and at a later time than the data.These dif-ferences indicate that the development of a 3D radiation hydrodynamic code,with more accurate physics models,is mandatory for spherical hohlraum study.Yaohua Chen Zhichao Li Xufei Xie Chunyang Zheng Chuanlei Zhai Liang Hao Dong Yang Wenyi Huo Guoli Ren Jie Liu Xiaoshi Peng Tao Xu Yulong Li Sanwei Li Zhiwen Yang Liang Guo Lifei Hou Yonggang Liu Huiyue Wei Xiangming Liu Weiyi Cha Yukun Li Keli Deng Zheng Yuan Xiayu Zhan Haijun Zhang Baibin Jiang Wei Zhang Kai Du Xuewei Deng Yongkun Ding Xiaofeng Wei Wanguo Zheng Xiaodong Chen Xiantu He Ke Lan 2017Matter and Radiation at Extremes2017,2,2:0
