Molecular mapping of QTLs for root response to phosphorus deficiency at seedling stage in wheat (Triticum aestivum L.)显示文摘Phosphorus (P) deficiency in the soil is one of the major abiotic stresses that limit plant growth and crop productivity throughout the world. Development of cultivars with improved P-deficiency tolerance is an efficient strategy for sustainable agriculture. Plant roots play an important role in crop growth and development, especially in nutrient uptake and improvement of P-efficiency. Mapping quantitative trait loci (QTLs) for root traits and their response to low P stress at seedling stage will facilitate the development of P-efficient wheat cultivars. In this study, 30 QTLs (LOD>2.0) were mapped for the three root traits, such as root length, root number and root dry matter under different P supply conditions and their response to P-stress. These QTLs were distributed on 14 chromosomes, with each of the 5 QTLs explaining more than 10% phenotype variance. Analyses showed that root traits and their response to P-deficiency were controlled by different QTLs. In addition, alleles with positive effects were separated on both parents, and wheat cultivars with improved P-efficiency could be developed by accumulating these positive effect alleles together.Li Zhenxing Ni Zhongfu Peng Huiru Liu Zhiyong Nie Xiuling Xu Shengbao Liu Gang Sun Qixin 2007Progress in Natural Science:Materials International2007,17,10:8
Molecular dissection of plant height QTLs using recombinant inbred lines from hybrids between common wheat (Triticum aestivum L.) and spelt wheat (Triticum spelta L.)显示文摘In wheat, plant height is an important agronomic trait, and a number of quantitative trait loci (QTLs) controlling plant height have been located. In this study, using the conditional and unconditional QTL mapping methods, combined with data from five different growth stages over two years of field trials, the developmental behavior for plant height in wheat was dissected. Nine unconditional QTLs and 8 conditional QTLs were identified, of which 6 were detected by both methods. None of the 11 QTLs was detected at all of the 5 investigated developmental stages, but 7 QTLs were detected at certain stages in both years. Further analysis identified 9 unconditional QTLs at different stages, which could explain the phenotypic variation from 4.81% to 17.35%. It was noteworthy that one major QTL designated QHt-4B-2, which was located on chromosome 4B, was detected on May 18 and 25 in both years, and its genetic contributions to plant height ranged from 13.42% to 16.13%. Moreover, of the 8 conditional QTLs identified, six were detected in both years, in the order of QHt-3B→QHt-4B-1→QHt-4B-2→QHt-4D→QHt-5A and QHt-2B expressed at the same developmental stage. The results indicate that QTL expression during plant height development is selective and in a temporal order.LIU Gang XU ShengBao NI ZhongFu XIE ChaoJie QIN DanDan LI Jing LU LaHu ZHANG JinPing PENG HuiRu SUN QiXin 2011Chinese Science Bulletin2011,56,18:6

Clinical features and outcome of 15 patients with recurrent endometrial cancer or atypical endometrial hyperplasia received primary fertility-sparing therapy followed by hysterectomy显示文摘Objective To investigate the clinical features and outcome of recurrent endometrial cancer(EC)or atypical endometrial hyperplasia(AEH)patients who underwent hysterectomy after fertility-sparing therapy. Methods Clinical data was retrospectively collected for 15 recurrent endometrial cancer or atypical endometrial hyperplasia patients who underwent hysterectomy in six hospitals from 2003 to 2019.According to the indicators of hysterectomy, patients were divided into four groups:7 patients who underwent direct hyesterectomy after the first relapse,4 due to re-treatment failure,1 after full term live birth, and 3 because of multiple recurrence.Clinical coexisting conditions, regimen and outcome of fertility-sparing therapy,pre-and post-operative pathological results, and prognosis were analyzed.Results(1)Fertility-spraring treatment was given to a total of 15 eligible patients, including 6 with EC and 9 with AEH.Median time interval from remission to recurrence was 12 months(range 3–92).Oral progestin-based medicine was the main fertility-sparing therapy in both primary treatment and re-treatment after recurrence.Six(6/8)patients received progestin combined with gonadotrophin releasing hormone agonist or metformin in three re-treatment groups.(2)Nine patients underwent hysterectomy and 6 staging surgery.Three patients had pathological upgrade after surgery.Five(5/10)EC patients had superficial myometrial invasion in postoperative pathology. All patients showed no evidence of disease in the median follow-up of 17 months(range 3–118).(3)Among seven patients who failed to re-treatment and had multiple recurrence, six were overweight/obesity and six had insulin resistance. Two of these patients had synchronous ovarian cancer in the postoperative pathology. Conclusion For the patients with recurrent EC or AEH after primary fertility-sparing therapy,overweight/obesity and insulin resistance might be the risk factors for re-treatment failure.Hysterectomy is recommended when patients had re-treatment failure or multiple recurrence;and generally the prognosis is favorable.Yuanting Li Yiqin Wang Yijiao He Rong Zhou Huiru Tang Lijiang Xu Mian He Weili Li Chunlin Chen Weifeng Zhang Jianliu Wang 2021Gynecology and Obstetrics Clinical Medicine2021,1,1:0
Age of the Marwar Supergroup,NW India:A note on the U–Pb geochronology of Jodhpur Group felsic volcanics显示文摘The Marwar Supergroup(NW Peninsular India)is thought to be of Ediacaran-Cambrian age, based on previous paleontological and geochronological studies. However, direct constraints on the onset of sedimentation within the Marwar basin are still scarce. In this study,we report U–Pb zircon,LA-ICP-MS,and SIMS ages from the Chhoti Khatu felsic volcanic rocks, interlayered with the Jodhpur Group sandstones(Lower Marwar Supergroup). The cathodoluminescence images of the zircons indicate complex morphologies, and core-rim textures coupled with the wide range of ages indicate that they are likely inherited or in the case of thin poorly indurated ash-beds,detrital in origin. The age spectra of 68 zircon analyses from our sampling display a dominant 800–900 Ma age peak corresponding to the age of basement'Erinpura granite'rocks in the region. The youngest inherited zircon from a felsic ash layer yielded a U–Pb age of651 Ma±18 Ma that, together with previous studies and paleontological evidence, indicates a postCryogenian age for the initiation of Marwar sedimentation following a~125 Ma hiatus between the end of Malani magmatism and Marwar deposition.Huiru Xu Joseph G.Meert Manoj K.Pandit 2022Geoscience Frontiers2022,13,1:0
Circulating Lipoproteins Mediate the Association Between Cardiovascular Risk Factors and Cognitive Decline:A Community‑Based Cohort Study显示文摘Cardiovascular health metrics are now widely recognized as modifable risk factors for cognitive decline and dementia.Metabolic perturbations might play roles in the linkage of cardiovascular diseases and dementia.Circulating metabolites profling by metabolomics may improve understanding of the potential mechanism by which cardiovascular risk factors contribute to cognitive decline. In a prospective community-based cohort in China(n=725),312 serum metabolic phenotypes were quantifed, and cardiovascular health score was calculated including smoking, exercise, sleep, diet, body mass index, blood pressure, and blood glucose.Cognitive function assessments were conducted in baseline and follow-up visits to identify longitudinal cognitive decline.A better cardiovascular health was signifcantly associated with lower risk of concentration decline and orientation decline(hazard ratio(HR):0.84–0.90;p<0.05).Apolipoprotein-A1,high-density lipoprotein(HDL)cholesterol, cholesterol ester, and phospholipid concentrations were signifcantly associated with a lower risk of longitudinal memory and orientation decline(p<0.05 and adjusted-p<0.20).Mediation analysis suggested that the negative association between health status and the risk of orientation decline was partly mediated by cholesterol ester and total lipids in HDL-2 and-3(proportion of mediation:7.68–8.21%,both p<0.05).Cardiovascular risk factors were associated with greater risks of cognitive decline, which were found to be mediated by circulating lipoproteins, particularly the medium-size HDL components. These fndings underscore the potential of utilizing lipoproteins as targets for early stage dementia screening and intervention.Jialin Li Qingxia Huang Yingzhe Wang Mei Cui Kelin Xu Chen Suo Zhenqiu Liu Yanpeng An Li Jin Huiru Tang Xingdong Chen Yanfeng Jiang 2024Phenomics2024,4,1:0
Transcriptomic Analysis and Comparison of the Gene Expression Profiles in Fast- and Slow-Growing Pearl Oysters Pinctada fucata martensii显示文摘The pearl oyster Pinctada fucata martensii is an economically valuable shellfish that is cultured for seawater pearl pro-duction,which mainly depends on oyster growth. However, the growth mechanisms of the pearl oyster are still poorly understood. In this study, oysters were grouped with relative growth rate, including fast-growing(FG)group and slow-growing(SG)group.Oxford Nanopore Technologies(ONT)long-read sequencing was applied to investigate the molecular mechanisms involved in the growth of this species.Five alternative splicing(AS)types were analyzed in both FG and SG groups, which include alternative 3’splice site, alternative 5’splice site, exon skipping, intron retention, and mutually exclusive exon.Transcriptome analysis showed that four of five different AS events(excluding mutually exclusive exons)occurred more frequently in FG than in SG oysters, and the five main AS types exhibited different characteristics. The AS events that were detected may be involved in growth, and the difference in ex-pression of AS events between FG and SG oysters may be involved in the mechanism underlying the difference in growth.Fifty dif-ferentially expressed genes(DEGs)were identified between the FG and SG oysters. The results showed that 40 genes were signifi-cantly up-regulated in FG oysters, while 10 genes were significantly down-regulated in SG oyster.Several genes related to nutrient metabolism, shell formation, and immunity were more highly expressed in FG oysters than in SG oysters. In summary,FG oysters exhibited higher metabolic and biomineralization activities and had a more powerful immune system than SG oysters. These results provide insight into the growth of P.f.martensii that can be used to improve breeding programs.XU Hanzhi LIU Huiru ZHANG Hua HE Maoxian 2022Journal of Ocean University of China2022,21,1:0
Reduced temporal activation during a verbal fluency test in clinical high risk of psychosis: a functional near-infrared spectroscopy-based study显示文摘Background Clinical high risk(CHR)of psychosis is a state in which positive symptoms cause the subjects distress but do not approach a severity level that fulfils the criteria for a psychotic episode.CHR exhibits cognitive deficits;however, the underlying neurobiological mechanisms remain unclear. This study aimed to investigate whether brain activation measured by the levels of oxygenated hemoglobin(oxy-Hb)in CHR subjects could be correlated with cognitive deficits. Methods Fifty-eight CHR individuals who fulfilled the criteria for attenuated positive syndrome as specified in the Structured Interview for Prodromal Syndrome(SIPS)and the Scale of Prodromal Syndrome(SOPS)and 58 age-and sex-matched healthy participants were included in the study. All subjects completed the Measurement and Treatment Research to Improve Cognition in Schizophrenia(MATRICS)Consensus Cognitive Battery(MCCB)that includes tests measuring attention, verbal memory, verbal fluency, executive function, and general intelligence.Functional near-infrared spectroscopy(fNIRS)was used to measure the level of oxy-Hb in the dorsolateral prefrontal and frontotemporal cortices. Results We observed significantly decreased oxy-Hb levels in channel 32(located in the right superior temporal gyrus,rSTG)within the CHR individuals compared with that in the healthy controls(HCs)(t=−3.44,Bonferroni-corrected p=0.002),indicating lower brain activity.A significant positive correlation was observed between task-relatedβvalues and working memory in the CHR group(r=0.35,p=0.008).Conclusions The brain activation of rSTG is abnormal among subjects at clinicial high risk for psychosis. This abnormality is probably associated with the neural mechanisms of deficits in the working memory during the early stage of psychosis.Yanyan Wei Xiaochen Tang Tingyu Zhang Wenjun Su Lihua Xu Huiru Cui Zhenying Qian Tianhong Zhang Jijun Wang 2022General Psychiatry2022,35,2:0
TaANR1-TaMADS25 module regulates lignin biosynthesis and root development in wheat(Triticum aestivum L.)显示文摘Common wheat is a staple food for 35%of the global population, therefore increasing wheat yield in an ever-changing environment is essential for food security in the present day(Peng et al.,2011).Root system is responsible for water and nutrient acquisition, thus crucial for competitive fitness and crop yield in challenging environments(Karlova et al.,2021;Liu et al.,2022).Over the past decades, extensive research has focused on identifying genes accountable for root growth and development in plants(Rogers and Benfey,2015).However,only a limited number of genes have been cloned in wheat(Li et al.,2021).Weiya Xu Yongming Chen Bin Liu Qiuyuan Li Yilan Zhou Xuanshuang Li Weilong Guo Zhaorong Hu Zhenshan Liu Mingming Xin Yingyin Yao Mingshan You Huiru Peng Zhongfu Ni Jiewen Xing 2023Journal of Genetics and Genomics2023,50,11:0
The role of Smad6 in immunity of the pearl oyster Pinctada fucata martensii显示文摘Inhibitory Smads(I-Smads),which belong to the Smad family and inhibit bone morphogenic protein 2(BMP2)signaling by a variety of mechanisms, can suppress innate immunity responses in vertebrates. However, there are no reports for the role of Smad6 in immunity in mollusks. In this study,we showed that Smad6 of the pearl oyster Pinctada fucata martensii was located in the Smad6 cluster of the phylogenetic tree;mRNA expression of Smad6 and Smad3 was up-regulated after lipopolysaccharide and polyinosinic:polycytidylic challenge;and transcript levels of Smad6 and Smad3 showed opposite patterns during wound healing.Under salinity stress, water inflow and outflow in the gills appear to be regulated by BMP2-Smads signals, and BMP2-Smads signaling may be closely related to the immune response. Our results indicate that Smad6 is involved in immunity, that it plays a positive role in the response to immune challenge and an inhibitory role during wound healing, and that Smad6 and Smad3 may work against each other.Yu SHI Xiaolan PAN Meng XU Huiru LIU Hanzhi XU Maoxian HE 2022Journal of Oceanology and Limnology2022,40,3:0
Relationships between selfreflectiveness and clinical symptoms in individuals during pre-morbid and early clinical stages of psychosis显示文摘Background Self-reflectiveness,one dimension of cognitive insight, plays a protective role in an individual’s mental state. Both high and low levels of self-reflectiveness have been reported in patients with schizophrenia and individuals at clinical high risk for the illness.Aims This study aimed to explore the relationship patterns between self-reflectiveness and clinical symptoms in individuals during the pre-morbid and early clinical stages of psychosis. Methods A total of 181 subjects, including individuals with attenuated positive symptoms(APS,n=122)and patients with first-episode psychosis(FEP,n=59),completed the Beck Cognitive Insight Scale and were evaluated using the Schedule of Assessment of Insight and Positive and Negative Syndrome Scale. All subjects were classified into three groups according to their level of selfreflectiveness:low level(LSR,n=59),medium level(MSR,n=67)and high level(HSR,n=55).Both linear and nonlinear relationships between self-reflectiveness and clinical symptoms were explored. Results More individuals with APS were classified into the MSR group, while more patients with FEP were classified into the LSR group. The LSR group demonstrated less awareness of illness than the MSR and HSR groups, more stereotyped thinking and poorer impulse control but less anxiety than the MSR group, and lower levels of blunted affect and guilt feelings than the HSR group. The MSR group demonstrated lower stereotyped thinking than the HSR group.Compared to the LSR group, the MSR group had increased selfreflectiveness, improved awareness of illness, decreased stereotyped thinking, and better impulse control, but increased feelings of guilt. The HSR group showed increased stereotyped thinking when compared to the MSR group, but the other variables did not change significantly between these two groups.Overall,self-reflectiveness demonstrated an approximately inverse S-shaped relationship with the awareness of illness,a U-shaped relationship with stereotyped thinking and poor impulse control, and an almost linear relationship with anxiety and guilt feelings. Conclusions Self-reflectiveness demonstrates complex relationships with clinical symptoms and fails to exert significant positive effects when reaching a certain high level.Lihua Xu Huiru Cui Yanyan Wei Zhenying Qian Xiaochen Tang Yegang Hu Yingchan Wang Hao Hu Qian Guo Yingying Tang Tianhong Zhang Jijun Wang 2022General Psychiatry2022,35,3:0

The TaTCP4/10-B1 cascade regulates awn elongation in wheat(Triticum aestivum L.)显示文摘Awns are important morphological markers for wheat and exert a strong physiological effect on wheat yield. The awn elongation suppressor B1 has recently been cloned through association and linkage analysis in wheat. However, the mechanism of awn inhibition centered around B1 remains to be clarified. Here,we identified an allelic variant in the coding region of B1 through analysis of re-sequencing data;this variant causes an amino acid substitution and premature termination, resulting in a long-awn phenotype.Transcriptome analysis indicated that B1 inhibited awn elongation by impeding cytokinin-and auxinpromoted cell division.Moreover,B1 directly repressed the expression of TaRAE2 and TaLks2,whose orthologs have been reported to promote awn development in rice or barley.More importantly,we found that TaTCP4 and TaTCP10 synergistically inhibited the expression of B1,and a G-to-A mutation in the B1 promoter attenuated its inhibition by TaTCP4/10.Taken together, our results reveal novel mechanisms of awn development and provide genetic resources for trait improvement in wheat.Wensheng Ke Jiewen Xing Zhaoyan Chen Yidi Zhao Weiya Xu Lulu Tian Jinquan Guo Xiaoming Xie Dejie Du Zihao Wang Yufeng Li Jin Xu Mingming Xin Weilong Guo Zhaorong Hu Zhenqi Su Jie Liu Huiru Peng Yingyin Yao Qixin Sun Zhongfu Ni 2023Plant Communications2023,4,4:0
Changes in target ability of nanoparticles due to protein corona composition and disease state显示文摘Many studies have shown the influence of protein corona(PC)on the active targeting capability of ligand-modified nanoparticles;however, the influence of clinical status on PC composition and targeting capacity is rarely discussed. In this study, when transferrinmodified PEGylated polystyrene nanoparticles(Tf-PNs)is intravenously injected into mice with non-small cell lung cancer(NSCLC)comorbid with type 2 diabetes mellitus(T2DM),more Tf-PNs accumulated in the tumor tissue than in those of NSCLC model mice. This indicated that PC derived fromdifferent states of disease changed the active targeting ability of Tf-PNs.To explain the occurrence of this phenomenon, our analysis of PC from different disease states revealed that Tf(transferrin)modification had no significant effect on the formation of PC,and that the PC from the NSCLC comorbid with T2DM model contained more proteins like fibrin and clusterin. This work demonstrates the impacts of comorbidity, such as with T2DM,on the active targeting capability of ligand-modified nanoparticles, and the results promote the application of nanoparticles for precision medicine.Wenwen Xu Mingyu Xu Yumeng Xiao Lu Yu Huiru Xie Xuehua Jiang Meiwan Chen Huile Gao Ling Wang 2022Asian Journal of Pharmaceutical Sciences2022,17,3:0
PIFs interact with SWI2/SNF2-related 1 complex subunit 6 to regulate H2A.Z deposition and photomorphogenesis in Arabidopsis显示文摘Light is an essential environmental signal perceived by a broad range of photoreceptors in plants. Among them, the red/far-red light receptor phytochromes function to promote photomorphogenesis, which is critical to the survival of seedlings after seeds germination. The basic-helix-loop-helix transcription factors phytochrome-interacting factors (PIFs) are the pivotal direct downstream components of phytochromes. H2A.Z is a highly conserved histone variant regulating gene transcription, and its incorporation into nucleosomes is catalyzed by SWI2/SNF2-related 1 complex, in which SWI2/SNF2-related 1 complex subunit 6 (SWC6) and actin-related protein 6 (ARP6) serve as core subunits. Here, we show that PIFs physically interact with SWC6 in vitro and in vivo, leading to the disassociation of HY5 from SWC6. SWC6 and ARP6 regulate hypocotyl elongation partly through PIFs in red light. PIFs and SWC6 coregulate the expression of auxin-responsive genes such as IAA6, IAA19, IAA20, and IAA29 and repress H2A.Z deposition at IAA6 and IAA19 in red light. Based on previous studies and our findings, we propose that PIFs inhibit photomorphogenesis, at least in part, through repression of H2A.Z deposition at auxin-responsive genes mediated by the interactions of PIFs with SWC6 and promotion of their expression in red light.Huiru Chen Wanting Wang Xiao Chen Yake Niu Yuanyuan Qi Ze Yu Minyu Xiong Pengbo Xu Wenxiu Wang Tongtong Guo Hong-Quan Yang Zhilei Mao 2023Journal of Genetics and Genomics2023,50,12:0
Changes in concentrations and transcripts of plant hormones in wheat seedling roots in response to Fusarium crown rot显示文摘Fusarium crown rot(FCR) is a soilborne disease causing severe yield losses in many wheat-growing areas of the world. Diseased plants show browning and necrosis of roots and stems causing white heads at maturity. Little is known about the molecular processes employed by wheat roots to respond to the disease. We characterized morphological, transcriptional and hormonal changes in wheat seedling roots following challenge with Fusarium pseudograminearum(Fp), the main pathogen of FCR. The pathogen inhibited root development to various extents depending on plants' resistance level. Many genes responsive to FCR infection in wheat roots were enriched in plant hormone pathways. The contents of compounds involved in biosynthesis and metabolism of jasmonic acid, salicylic acid, cytokinin and auxin were drastically changed in roots at five days post-inoculation. Presoaking seeds in methyl jasmonate for 24 h promoted FCR resistance, whereas presoaking with cytokinin 6-benzylaminopurine made plants more susceptible. Overexpression of TaOPR3, a gene involved in jasmonic acid biosynthesis, enhanced plant resistance as well as root and shoot growth during infection.Yutian Gao Xuejun Tian Weidong Wang Xiangru Xu Yuqing Su Jiatian Yang Shuonan Duan Jinlong Li Mingming Xin Huiru Peng Qixin Sun Chaojie Xie Jun Ma 2023The Crop Journal2023,11,5:0
