1Temporal changes in SSR allelic diversity of major rice cultivars in China显示文摘Forty simple sequence repeats (SSRs) were used to assess the changes of diversity in 310 major Chinese rice cultivars grown during the 1950s-1990s. Of the 40 SSR loci, 39 were polymorphic. A total of 221 alleles were detected with an average of 5.7 alleles per locus (Na). The Nei’s genetic diversity index (He) varied drastically among the loci (0.207 to 0.874, mean 0.625). Comparing the temporal changes in Na and He, the cultivars from the 1950s had more alleles and higher He scores than the cultivars from the other four decades Analysis of molecular variance (AMOVA) indicated that the genetic differentiation among the five decades was not significant in the whole set, but significant within indica and japonica. More changes among the decades were revealed in indica cultivars than in japonica cultivars. Some alleles had been lost in current rice cultivars in the 1990s, occurring more frequently in indica. 2Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8

In this study, we constructed a rice-codon optimized adenine base editor(ABE)-nC as9 tool that induced targeted A·T to G·C point mutation of a key single nucleotide polymorphism site in an important agricultural gene. 4Zonal Differences in Correlation Patterns Between Soil Organic Carbon and Climate Factors at Multi-extent显示文摘Studying the relationship between climate factors and soil organic carbon(SOC) is vitally important. However, how SOC responses to climate(temperature and precipitation) at cohesive extents is poorly studied. Two transects of approximately the same length(transect P and transect T) were selected to examine the variation of SOC content in relation to mean annual temperature(MAT) and mean annual precipitation(MAP). The coefficients of partial correlation between SOC density and MAT(Rt) and MAP(Rp) were determined to quantify the relationships between SOC density and the two climate factors. The results indicated that for transect T, Rt was statistically significant once the extent level was greater than or equal to two fundamental extent units, while for transect P, Rp showed statistical significance only at extent levels which were greater than two fundamental extent units. At the same extent levels but in different transects, Rts exhibited no zonal difference, but Rps did once the extent level was greater than two fundamental extent units. Therefore, to study the relationship between SOC density and different climate factors, different minimum extent levels should be examined. 5Effect of ALA-PDT on the expressions of MMP-9, MMP-13 and TIMP-1 of hypertrophic scar model in rabbit ears显示文摘Objective: To investigate the effect of 5-aminolevulinic(ALA)-photodynamic therapy(PDT) on the expressions of MMP-9, MMP-13 and TIMP-1 of hypertrophic scar model in rabbit ears, and analyze the possible therapeutic mechanisms of ALA-PDT treatment to hypertrophic scars of rabbit ears. Methods: The experimental animals were randomly divided into normal control, negative control, high concentration of ALA-PDT, low concentration of ALA-PDT and PDT groups. The latter three groups received ALA-PDT treatment or PDT treatment once a week for 3 weeks. The specimens of the rabbits were collected respectively 1, 2 and 3 months after treatment to be used for RT-PCR and Western-blot test. Results: 1, 2 and 3 months after PDT treatment, the expressions of MMP-9 and MMP-13(including mRNA and protein) in hypertrophic scar tissues of three treatment groups were significantly higher than those of the negative control group(P<0.01), and the expression of TIMP-1 mRNA and protein of three treatment groups were significantly lower than that of the negative control group(P<0.01). There were also significant differences between high-concentration ALA-PDT treatment group and the low one(P<0.05). Conclusion: ALA-PDT is effective in treating hypertrophic scars of rabbit ears, and its possible therapeutic mechanisms are that ALA-PDT treatment generates oxidation activation effect to activate the activity of MMPs and induces the photoaging of fibroblasts of hypertrophic scar tissues of rabbit ears to inhibit the activity of TIMPs, which causes the up-regulation of MMPs and the down-regulation of TIMPs. 8Influence of Climate on Soil Organic Carbon in Chinese Paddy Soils显示文摘Soil organic carbon(SOC) is a major component of the global carbon cycle and has a potentially large impact on the greenhouse effect. Paddy soils are important agricultural soils worldwide, especially in Asia. Thus, a better understanding of the relationship between SOC of paddy soils and climate variables is crucial to a robust understanding of the potential effect of climate change on the global carbon cycle. A soil profile data set(n = 1490) from the Second National Soil Survey of China conducted from 1979 to 1994 was used to explore the relationships of SOC density with mean annual temperature(MAT) and mean annual precipitation(MAP) in six soil regions and eight paddy soil subgroups. Results showed that SOC density of paddy soils was negatively correlated with MAT and positively correlated with MAP(P < 0.01). The relationships of SOC density with MAT and MAP were weak and varied among the six soil regions and eight paddy soil subgroups. A preliminary assessment of the response of SOC in Chinese paddy soils to climate indicated that climate could lead to a 13% SOC loss from paddy soils. Compared to other soil regions, paddy soils in Northern China will potentially more sensitive to climate change over the next several decades. Paddy soils in Middle and Lower Yangtze River Basin could be a potential carbon sink. 10Local Anomalies of the Load-Unload Response of the Geomagnetic Field to Solar Wind and Earthquakes in Southwest China显示文摘Many earthquakes occurred during the period 1994 -1996 in Sichuan and Yunnan Provinces, Southwest China. Taking the process of the initial main phase recovery phase of the magnetic storm as the process of load-unload response of the geomagnetic field to the solar wind, we have estimated and analyzed the distribution in time and space of the load-unload response ratio P(z) of the storm time disturbance daily variation of the vertical component Z of the geomagnetic field at ten stations in Southwest China. We found that the area with high ratio P(z) was just the area where moderately strong earthquakes would occur from 44 days to 15 months later. 11U1 small nuclear ribonucleoprotein is essential for early larval development in silkworm, Bombyx mori显示文摘U1 small nuclear ribonucleoproteins(U1 snRNP)associates with 5ʹsplice sites in the form of ribonucleoprotein particles and is highly conserved among species.The physiological functions of U1 snRNP in a lepidopteran model insect Bombyx mori is mostly unknown.Here,we showed that U1 snRNP plays an important role in the development of silkworm.Knockout of U1 snRNP in silkworm showed either delayed or stationary 1st instar larva development compared with the wild-type group.U1 snRNP deletion mutants exhibited abnormal cellular phenotypes with enlarged cell nucleus,scant cytoplasm and enlarged nuclei.RNA-seq analysis revealed that genes involved in metabolic pathway,biosynthesis of secondary metabolites and steroid hormone biosynthesis were significantly affected by U1 snRNP depletion.Taken together,our study suggests that U1 snRNP homeostasis plays an important role in silkworm development.Xiaoqian Zhang Dehong Yang Xu Yang Xinran Zhang Yong Zhang Yongping Huang Shengxiang Zhang Zulian Liu 2022Insect Science2022,29,2:0
12Uncertainty and Sensitivity Analyses for Modeling Long-Term Soil Organic Carbon Dynamics of Paddy Soils Under Different Climate-Soil-Management Combinations显示文摘Reporting modeling results with uncertainty information can benefit decision making by decreasing the extent that variability exerts a disproportionate influence on the options selected. For making decisions with more confidence, the uncertainty interval should be as narrow as possible. Here, the soil organic carbon(SOC) dynamics of the major paddy soil subgroup from 4 different paddy field regions of China(located in 4 counties under different climate-soil-management combinations) were modeled using the De NitrificationDe Composition(DNDC) model for the period from 1980 to 2008. Uncertainty intervals associated with the SOC dynamics for these 4 subgroups were estimated by a long-term global sensitivity and uncertainty analysis(i.e., the Sobol′method), and their sensitivities to 7 influential factors were quantified using the total effect sensitivity index. The results, modeled with high confidence, indicated that in the past 29 years, the studied paddy soils in Xinxing, Yixing, and Zhongjiang counties were carbon(C) sinks, while the paddy soil in Helong County was a C source. The 3 C sinks sequestered 12.2(5.4, 19.6), 17.1(8.9, 25.0), and 16.9(-1.2, 33.6) t C ha^(-1)(values in the parentheses are the 5th and 95th percentiles, respectively). Conversely, the C source had a loss of -5.4(-14.2, 0.06) t C ha^(-1) in the past 29 years. The 7 factors, which changed with the climate-soil-management context, exhibited variable influences on modeled SOC.
