1Discrete Variable Structural Optimization based on Multidirectional Fuzzy Genetic Algorithm显示文摘Round method is the common method for discrete variable optimization in optimal design of complex mechanical structures;however,it has some disadvantages such as poor precision,simple model and lacking of working conditions' description,etc.To solve these problems,a new model is constructed by defining parameterized fuzzy entropy,and the rationality of parameterized fuzzy entropy is verified.And a new multidirectional searching algorithm is further put forward,which takes information of actual working conditions into consideration and has a powerful local searching capability.Then this new algorithm is combined with the GA by the fuzzy clustering algorithm(FCA).With the application of FCA,the optimal solution can be effectively filtered so as to retain the diversity and the elite of the optimal solution,and avoid the structural re-analysis phenomenon between the two algorithms.The structure design of a high pressure bypass-valve body is used as an example to make a structural optimization by the proposed HGA and finite element method(FEM),respectively.The comparison result shows that the improved HGA fully considers the characteristic of discrete variable and information of working conditions,and is more suitable to the optimal problems with complex working conditions.Meanwhile,the research provides a new approach for discrete variable structure optimization problems.LAI Yinan DAI Ye BAI Xue CHEN Dongyan 2012Chinese Journal of Mechanical Engineering2012,25,2:12
2A mass balanced model of trophic structure and energy flows of a semi-closed marine ecosystem显示文摘The marine ecosystem of the Jiaozhou Bay has degraded significantly in fisheries productivity and its ecological roles as spawning and nursery ground for many species of commercial importance has been declining in recent years. A mass-balanced trophic model was developed using Ecopath with Ecosim to evaluate the trophic structure of the Jiaozhou Bay for improving ecosystem management. The model were parameterized based on the fisheries survey data in the Jiaozhou Bay in 2011, including 23 species groups and one detritus group according to their ecological roles. The trophic levels of these ecological groups ranged from 1(primary producers and detritus) to4.3(large demersal fishes). The estimated total system throughput was 12 917.10 t/(km^2·a), with 74.59% and25.41% contribution of the total energy flows from phytoplankton and detritus, respectively. Network analyses showed that the overall transfer efficiency of the ecosystem was 14.4%, and the mean transfer efficiency was 14.5%for grazing food chain and 13.9% for detritus food chain. The system omnivory index(SOI), Finn's cycled index(FCI) and connectance index(CI) were relatively low in this area while the total primary production/total respiration(TPP/TR) was high, indicating an immature and unstable status of the Jiaozhou Bay ecosystem. Mixed trophic impact analysis revealed that the cultured shellfish had substantial negative impacts on most functional groups. 3Beak Measurements of Octopus (Octopus variabilis) in Jiaozhou Bay and Their Use in Size and Biomass Estimation显示文摘Cephalopods play key roles in global marine ecosystems as both predators and preys.Regressive estimation of original size and weight of cephalopod from beak measurements is a powerful tool of interrogating the feeding ecology of predators at higher trophic levels.In this study,regressive relationships among beak measurements and body length and weight were determined for an octopus species(Octopus variabilis),an important endemic cephalopod species in the northwest Pacific Ocean.A total of 193 individuals(63 males and 130 females) were collected at a monthly interval from Jiaozhou Bay,China.Regressive relationships among 6 beak measurements(upper hood length,UHL;upper crest length,UCL;lower hood length,LHL;lower crest length,LCL;and upper and lower beak weights) and mantle length(ML),total length(TL) and body weight(W) were determined.Results showed that the relationships between beak size and TL and beak size and ML were linearly regressive,while those between beak size and W fitted a power function model.LHL and UCL were the most useful measurements for estimating the size and biomass of O.variabilis.The relationships among beak measurements and body length(either ML or TL) were not significantly different between two sexes;while those among several beak measurements(UHL,LHL and LBW) and body weight(W) were sexually different.Since male individuals of this species have a slightly greater body weight distribution than female individuals,the body weight was not an appropriate measurement for estimating size and biomass,especially when the sex of individuals in the stomachs of predators was unknown.These relationships provided essential information for future use in size and biomass estimation of O.variabilis,as well as the estimation of predator/prey size ratios in the diet of top predators.XUE Ying REN Yiping MENG Wenrong LI Long MAO Xia HAN Dongyan MA Qiuyun 2013Journal of Ocean University of China2013,12,3:2

9Effect of mechanical stretching and substrate stiffness on the morphology,cytoskeleton and nuclear shape of corneal endothelial cells显示文摘Due to the limited capacity of corneal endothelial cells(CECs)division,corneal endothelial diseases have become a great challenge.The cornea is subjected to various mechanical stimuli in vivo,which may have a positive or negative influence.Thus,it is significant to gain an insight into the mechanism of mechanobiology of CECs for seeking more possible treatment.The purpose of this study was to determine the impacts of mechanical stretch and substrate stiffness on the morphology and fundamental cell behavior of CECs.Rabbit corneal endothelial cells(RCECs)were subjected to a 5%mechanical stretch or cultured on substrates of different stiffness.The impacts of mechanical stimulus on cell area,aspect ratio,circularity,cell density,nuclear shape,cytoskeleton,and cell viability were investigated.The expressions of the corneal endothelium-related markers ZO-1 and Na^(+)/K^(+) ATPase were also evaluated by confocal immunofluorescence microscopy in the stiffness group.Our results suggested that mechanical stretch promoted the rearrangement of the cytoskeleton while decreasing the cell circularity,nuclear area,and cell density as well as cell viability.RCECs cultured on 10 kPa substrates,which was close to the physiological stiffness of rabbit Descemet's membrane(DM),showed better cell morphology,more stable actin cytoskeleton assembly,and more robust expression of the functional marker compared with other softer or stiffer substrates.In summary,mechanical stretch and substrate stiffness have profound influences on the morphology and function of CECs,which may have implications for the understanding and possible treatment of corneal endothelial diseases.Ruotian Du Dongyan Li Yan Huang Hui Xiao Jindong Xue Jing Ji Yun Feng Yubo Fan 2022Medicine in Novel Technology and Devices2022,,4:0
10Impacts of sample size for stomach content analysis on the estimation of ecosystem indices显示文摘This study used Ecopath model of the Jiaozhou Bay as an example to evaluate the effect of stomach sample size of three fish species on the projection of this model. The derived ecosystem indices were classified into three categories:(1) direct indices, like the trophic level of species, influenced by stomach sample size directly;(2)indirect indices, like ecology efficiency(EE) of invertebrates, influenced by the multiple prey-predator relationships;and(3) systemic indices, like total system throughout(TST), describing the status of the whole ecosystem. The influences of different stomach sample sizes on these indices were evaluated. The results suggest that systemic indices of the ecosystem model were robust to stomach sample sizes, whereas specific indices related to species were indicated to be with low accuracy and precision when stomach samples were insufficient.The indices became more uncertain when the stomach sample sizes varied for more species. This study enhances the understanding of how the quality of diet composition data influences ecosystem modeling outputs. 11Ancient dormant virus remnant ERVW-1 drives ferroptosis via degradation of GPX4 and SLC3A2 in schizophrenia显示文摘Human endogenous retroviruses(HERVs)are remnants of retroviral infections in human germline cells from millions of years ago.Among these,ERVW-1(also known as HERV-W-ENV,ERVWE1,or ENVW)encodes the envelope protein of the HERV-W family,which contributes to the pathophysiology of schizophrenia.Additionally,neuropathological studies have revealed cell death and disruption of iron homeostasis in the brains of individuals with schizophrenia.Here,our bioinformatics analysis showed that differentially expressed genes in the human prefrontal cortex RNA microarray dataset(GSE53987)were mainly related to ferroptosis and its associated pathways.Clinical data demonstrated significantly lower expression levels of ferroptosis-related genes,particularly Glutathione peroxidase 4(GPX4)and solute carrier family 3 member 2(SLC3A2),in schizophrenia patients compared to normal controls.Further in-depth analyses revealed a significant negative correlation between ERVW-1 expression and the levels of GPX4/SLC3A2 in schizophrenia.Studies indicated that ERVW-1 increased iron levels,malondialdehyde(MDA),and transferrin receptor protein 1(TFR1)expression while decreasing glutathione(GSH)levels and triggering the loss of mitochondrial membrane potential,suggesting that ERVW-1 can induce ferroptosis.Ongoing research has shown that ERVW-1 reduced the expression of GPX4 and SLC3A2 by inhibiting their promoter activities.Moreover,Ferrostatin-1(Fer-1),the ferroptosis inhibitor,reversed the iron accumulation and mitochondrial membrane potential loss,as well as restored the expressions of ferroptosis markers GSH,MDA,and TFR1 induced by ERVW-1.In conclusion,ERVW-1 could promote ferroptosis by downregulating the expression of GPX4 and SLC3A2,revealing a novel mechanism by which ERVW-1 contributes to neuronal cell death in schizophrenia.Dongyan Zhang Xiulin Wu Xing Xue Wenshi Li Ping Zhou Zhao Lv Kexin Zhao Fan Zhu 2024Virologica Sinica2024,39,1:0
12Domesticated HERV-W env contributes to the activation of the small conductance Ca^(2+)-activated K^(+)type 2 channels via decreased 5-HT4 receptor in recent-onset schizophrenia显示文摘The human endogenous retroviruses type W family envelope(HERV-W env)gene is located on chromosome 7q21-22.Our previous studies show that HERV-W env is elevated in schizophrenia and HERV-W env can increase cal-cium influx.Additionally,the 5-HTergie system and particularly 5-hydroxytryptamine(5-HT)receptors play a prominent role in the pathogenesis and treatment of schizophrenia.5-hydroxytryptamine receptor 4(5-HT4R)agonist can block calcium channels.However,the underlying relationship between HERV-W env and 5-HT4R in the etiology of schizophrenia has not been revealed.Here,we used enzyme-linked immunosorbent assay to detect the concentration of HERV-W env and 5-HT4R in the plasma of patients with schizophrenia and we found that there were decreased levels of 5-HT4R and a negative correlation between 5-HT4R and HERV-W env in schizophrenia.Overexpression of HERV-W env decreased the transcription and protein levels of 5-HT4R but increased small conductance Ca^(2+)-activated K^(+)type 2 channels(SK2)expression levels.Further studies revealed that HERV-w env could interact with 5-HT4R.Additionally,luciferase assay showed that an essential region(-364 to-176 from the transcription start site)in the SK2 promoter was required for HERV-W env-induced SK2 expression.Importantly,5-HT4R participated in the regulation of SK2 expression and promoter activity.Electrophysiological recordings suggested that HERV-Wenv could increase SK2 channel currents and the increase of SK2 currents was inhibited by 5-HT4R.In condusion,HERV-W env could activate SK2 channels via decreased 5-HT4R,which might exhibit a novel mechanism for HERV-Wenv to influence neuronal activity in schizophrenia.Xiulin Wua Qiujin Yan Lianzhong Liu Xing Xue Wei Yao Xuhang Li Wenshi Li Shuang Ding Yaru Xia Dongyan Zhang Fan Zhu 2023Virologica Sinica2023,38,1:0
