1Cocrystallization of N-donor type compounds with 5-sulfosalicylic acid:The effect of hydrogen-bonding supramolecular architectures显示文摘Benzotriazole,N,N'-dimethylpiperazine and N-methylpiperazine were applied to crystallize with 5-sulfosalicylic acid(5-H2SSA),affording three new binary molecular cocrystals [(C6H6N3+).(C7H5O6S-)].H2O(1),[(C6H16N22+)1/2.(C7H5O6S-)].H2O(2) and [(C5H14N22+).(C7H5O6S-)2].3H2O(3) under general conditions.Proton-transferring occurs from acid to nitrogen of N-donor compounds in all compounds 1,2 and 3.Analysis of the hydrogen-bonding synthons and their effects on crystal packing were also presented in the context of crystal engineering and host-guest chemistry.In compound 1,1-D infinite chains are extended to a 2-D layered architecture via strong O-H...O hydrogen bonds and then to a 3-D network by N-H...O interactions.Compound 2 and 3 both have the 1-D chain which is formed by O-H...O bonds and weak C-H...O hydrogen bonds.A common intramolecular S(6) [synthon I] ring is formed by the hydroxyl with the carboxyl group in all three compounds.WANG Lei XU LingYan XUE RuiFeng LU XiFeng CHEN RuiXin TAO XuTang 2012Science China Chemistry2012,55,1:8
2KIF18B promotes tumor progression in osteosarcoma by activating β-catenin显示文摘Objective:Osteosarcoma is a common primary highly malignant bone tumor.Kinesin family member 18B(K1F18B)has been identified as a potential oncogene involved in the development and metastasis of several cancer types.While KIF18B overexpression in osteosarcoma tissue is clearly detected,its specific function in the disease process remains to be established.Methods:K IF18B expression was assessed in osteosarcoma tissues and cells.We additionally evaluated the effects of KIF18B on proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.Results:Our results showed overexpression of KIF18B in osteosarcoma tissues and cells.Knockdown of K IF18B induced G1/S phase arrest and significantly inhibited proliferation,migration,and invasion of osteosarcoma cells,both in vitro and in vivo.K IF18B regulated P-catenin expression at the transcriptional level by controlling nuclear aggregation of ATF2 and at the post-transcriptional level by interacting with the adenomatous polyposis coli(APC)tumor suppressor gene in osteosarcoma cells.Conclusions:KIF18B plays a carcinogenic role in osteosarcoma by regulating expression ofβ-catenin transcriptionally via decreasing nuclear aggregation of ATF2 or post-transcriptionally through interactions with APC.Our collective findings support the potential utility of KIF18B as a novel prognostic biomarker for osteosarcoma.Tian Gao Ling Yu Zhiwei Fang Jiayong Liu Chujie Bai Shu Li Ruifeng Xue Lu Zhang Zhichao Tan Zhengfu Fan 2020Cancer Biology & Medicine2020,17,2:7
3Efficacy of vinorelbine combined with low-dose methotrexate for treatment of inoperable desmoid tumor and prognostic factor analysis显示文摘Objective: To assess the efficacy of conservative chemotherapy for inoperable desmoid tumor(DT) and analyze the prognostic factors.Methods:From November 2008 to April 2016,71 patients of inoperable DT were treated with vinorelbine and low-dose methotrexate in the Department of Bone and Soft Tissue Tumors,Peking University Cancer Hospital&Institute,and enrolled in this retrospective study.The chemotherapy duration is one year.The efficacy of chemotherapy and the prognosis were observed.Results:Of the 71 patients,55% were female.Age of onset varied from 1 to 47 years,and the median age was 14years.Only 11(15.5%)cases suffered primary tumor.The distribution of the site of tumors was:31(43.7%)in the trunk,36(50.7%)in the limbs,and 4(5.6%)in the peritoneal and pelvic cavity.The size of tumor(the maximum diameter)differed from 2 to 37 cm with a mean of 9.3 cm.The median follow-up duration was 28(range,6–87)months.Common side effects included:nausea and vomiting,liver injury,bone marrow suppression and oral ulcers.When the chemotherapy finished,1(1.4%)case achieved complete response,24(33.8%)achieved partial response,37(52.1%)achieved stable disease and 9(12.7%)had progressive disease.The overall response rate was 87.3%.The progression-free survival(PFS)of the participants were from 6 to 87 months,and the 2-,3-and 5-year PFS was 79.9%,68.4% and 36.3%,respectively.No significant difference was identified in PFS in subgroups of gender,age of onset,age of chemotherapy,tumor site and tumor size.Conclusions:For recurrent,inoperable and progressive DT,enough course of chemotherapy with vinorelbine combined with low-dose methotrexate was an optional choice for local control.Shu Li Zhengfu Fan Zhiwei Fang Jiayong Liu Chujie Bai Ruifeng Xue Lu Zhang Tian Gao 2017Chinese Journal of Cancer Research2017,29,5:6
4A viral protein orchestrates rice ethylene signaling to coordinate viral infection and insect vector-mediated transmission显示文摘Arthropod-borne viruses cause serious threats to human health and global agriculture by rapidly spreading via insect vectors. Southern rice black-streaked dwarf virus (SRBSDV) is the most damaging rice-infecting virus that is frequently transmitted by planthoppers. However, the molecular mechanisms underlying its propagation in the host plants and epidemics in the field are largely unknown. Here, we showed that the SRBSDV-encoded P6 protein is a key effector that regulates rice ethylene signaling to coordinate viral infection and transmission. In early SRBSDV infection, P6 interacts with OsRTH2 in the cytoplasm to activate ethylene signaling and enhance SRBSDV proliferation;this also repels the insect vector to reduce infestation. In late infection, P6 enters the nucleus, where it interacts with OsEIL2, a key transcription factor of ethylene signaling. The P6-OsEIL2 interaction suppresses ethylene signaling by preventing the dimerization of OsEIL2, thereby facilitating viral transmission by attracting the insect vector. 5Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. 8Direct observation of nodeless superconductivity and phonon modes in electron-doped copper oxide Sr_(1-X)Nd_(X)CuO_(2)显示文摘The microscopic understanding of high-temperature superconductivity in cuprates has been hindered by the apparent complexity of crystal structures in these materials.We used scanning tunneling microscopy and spectroscopy to study the electron-doped copper oxide compound Sr_(1-x)Nd_(x)CuO_(2),which has only bare cations separating the CuO_(2) planes and thus the simplest infinite-layer structure of all cuprate superconductors.Tunneling conductance spectra of the major CuO_(2) planes in the superconducting state reve aled direct evidence for a nodeless pairing gap,regardless of variation of its magnitude with the local doping of trivalent neodymium.Furthermore,three distinct bosonic modes are observed as multiple peak-dip-hump features outside the superconducting gaps and their respective energies depend little on the spatially varying gaps.As well as the bosonic modes,with energies identical to those of the external,bending and stretching phonons of copper oxides,our findings reveal the origin of the bosonic modes in lattice vibrations rather than spin excitations.Jia-Qi Fan Xue-Qing Yu Fang-Jun Cheng Heng Wang Ruifeng Wang Xiaobing Ma Xiao-Peng Hu Ding Zhang Xu-Cun Ma Qi-Kun Xue Can-Li Song 2022National Science Review2022,9,4:0
9Nuclear peripheral chromatin-lamin B1 interaction is required for global integrity of chromatin architecture and dynamics in human cells显示文摘The eukaryotic genome is folded into higher-order conformation accompanied with constrained dynamics for coordinated genome functions.However,the molecular machinery underlying these hierarchically organized three-dimensional(3D)chromatin architecture and dynamics remains poorly understood.Here by combining imaging and sequencing,we studied the role of lamin B1 in chromatin architecture and dynamics.We found that lamin B1 depletion leads to detachment of lamina-associated domains(LADs)from the nuclear periphery accompanied with global chromatin redistribution and decompaction.Consequently,the interchromosomal as well as inter-compartment interactions are increased,but the structure of topologically associating domains(TADs)is not affected.Using live-cell genomic loci tracking,we further proved that depletion of lamin B1 leads to increased chromatin dynamics,owing to chromatin decompaction and redistribution toward nucleoplasm.Taken together,our data suggest that lamin B1 and chromatin interactions at the nuclear periphery promote LAD maintenance,chromatin compaction,genomic compartmentalization into chromosome territories and A/B compartments and confine chromatin dynamics,supporting their crucial roles in chromatin higher-order structure and chromatin dynamics.Lei Chang Mengfan Li Shipeng Shao Chen Li Shanshan Ai Boxin Xue Yingping Hou Yiwen Zhang Ruifeng Li Xiaoying Fan Aibin He Cheng Li Yujie Sun 2022Protein & Cell2022,13,4:0
10Synthesis and characterization of a new open-framework zinc phosphite (4,4-(C_5H_4N)_2(CH_2)_3)·[Zn_3(HPO_3)_4]显示文摘Employing 4,4-trimethylenedipyridine as a template,a new two-dimensional zinc phosphite (4,4-(C5H4N)2(CH2)3)· [Zn3(HPO3)4 ] has been prepared at room temperature and characterized by single-crystal X-ray diffraction,FTIR,elemental analysis,powder X-ray diffraction,and thermogravimetric analysis.The compound crystallizes in the monoclinic space group P2 1 /c,with cell parameters,a=9.3309(19),b=14.798(3),c=33.637(7),β=91.11(3),V=4643.8(16) nm 3 and Z=4.The connectivity of the ZnO4 tetrahedra and HPO 3 pseudo pyramids results in infinite corner-sharing 4-membered ring chains as second building units which are further linked by Zn O P bonds to form a 2-D layer that with interesting 8-membered ring channels along the [100] direction.The diprotonated 4,4-trimethylenedipyridine molecules sit in the middle of the layers along the [100] direction and interact with the framework via hydrogen bonds.WANG Lei XUE RuiFeng CHEN RuiXin LU XiFeng LI GuangHua TAO XuTang 2012Science China Chemistry2012,55,3:0
11MAX-DOAS observation in the midlatitude marine boundary layer: Influences of typhoon forced air mass显示文摘As a passive remote sensing technique,MAX-DOAS method was widely used to investigate the vertical profiles of aerosol and trace gases in the lower troposphere.However,the measurements for midlatitude marine boundary layer are rarely reported,especially during the storm weather system.In this study,the MAX-DOAS was used to retrieve the aerosol,HCHO and NO_(2) vertical distribution at Huaniao Island of East China Sea in summer 2018,during which a strong tropical cyclone developed and passed through the measurement site.The observed aerosol optical depth(AOD),HCHO-and NO_(2)-VCDs(Vertical Column Density)were in the range of 0.19-0.97,(2.57-12.27)×10^(15) molec/cm^(2),(1.24-4.71)×10^(15) molec/cm^(2),which is much higher than remote ocean area due to the short distance to continent.The vertically resolved aerosol extinction coefficient(AEC),HCHO and NO_(2) presented the decline trend with the increase of height.After the typhoon passing through,the distribution of high levels of aerosol and HCHO stretched to about 1 kmand the abundances of the bottom layer were found as double higher than before,reaching 0.51 km^(−1) and 2.44 ppbv,while NO_(2) was still constrained within about 300 m with 2.59 ppbv in the bottom layer.The impacts of typhoon process forced air mass were also observed at the suburban site in Shanghai in view of both the aerosol extinction and chemical components.The different changes on air quality associated with typhoon and its mechanism in two different environments:coastal island and coastal city are worthy of further investigation as it frequent occurred in East Asia during summer and fall.Ruifeng Zhang Shanshan Wang Sanbao Zhang Ruibin Xue Jian Zhu Bin Zhou 2022Journal of Environmental Sciences2022,34,10:0
