1Genome editing for plant research and crop improvement显示文摘The advent of clustered regularly interspaced short palindromic repeat(CRISPR) has had a profound impact on plant biology, and crop improvement. In this review, we summarize the state-of-the-art development of CRISPR technologies and their applications in plants, from the initial introduction of random small indel(insertion or deletion) mutations at target genomic loci to precision editing such as base editing, prime editing and gene targeting. 2The GAMYB-like gene SlMYB33 mediates flowering and pollen development in tomato显示文摘GAMYBs are positive GA signaling factors that exhibit essential functions in reproductive development,particularly in anther and pollen development.However,there is no direct evidence of the regulation of any GAMYB in these biological processes in tomato(Solanum lycopersicum).Here,we identified a tomato GAMYB-like gene,SlMYB33,and characterized its specific roles.SlMYB33 is predominately expressed in the stamens and pistils.During flower development,high mRNA abundance of SlMYB33 is detected in both male and female organs,such as microspore mother cells,anthers,pollen grains,and ovules.Silencing of SlMYB33 leads to delayed flowering,aberrant pollen viability,and poor fertility in tomato.Histological analyses indicate that SlMYB33 exerts its function in pollen development in the mature stage.Further transcriptomic analyses imply that the knockdown of SlMYB33 significantly inhibits the expression of genes related to flowering in shoot apices,and alters the transcription of genes controlling sugar metabolism in anthers.Taken together,our study suggests that SlMYB33 regulates tomato flowering and pollen maturity,probably by modulating the expression of genes responsible for flowering and sugar metabolism,respectively.Yan Zhang Bo Zhang Tongwen Yang Jie Zhang Bin Liu Xiangqiang Zhan Yan Liang 2020Horticulture Research2020,7,1:5
3A novel PIK3CD C896T mutation detected in bilateral sudden sensorineural hearing loss using next generation sequencing:An indication of primary immunodeficiency显示文摘Objective: To investigate immune-related genetic background in bilateral sudden sensorineural hearing loss(SSNHL).Case report and methods: The case is a 45-year-old man presenting with a 7-year history of bilateral profound SSNHL. Blood biochemical testing demonstrated increased levels of total cholesterol(5.88 mmol/L). Tests for hepatitis B showed a positive antibody against the hepatitis B core antigen. Complement C3 was below the normal value, and complement C4 and Ig G were in the lower range of normal values. CT images showed a normal inner ear and vestibular aqueduct but round window membranous ossification on both sides. A total number of 232 immuneassociated genes were sequenced using the next generation sequencing technique.Results: Mutations were detected in 5 genes, including the phosphoinositide 3-kinase catalytic subunit delta(PIK3CD), caspase recruitment domain-containing protein 9(CARD9), complement factor H-related(CFHR2), immunoglobulin lambda-like polypeptide 1 Protein(IGLL1),and transmembrane channel-like gene family 8(TMC8). In the PIK3 CD gene, a C896 T substitute in exon 7 was detected. 4The tomato 2-oxoglutarate-dependent dioxygenase gene SlF3HL is critical for chilling stress tolerance显示文摘Low temperature is a major stress that severely affects plant development,growth,distribution,and productivity.Here,we examined the function of a 2-oxoglutarate-dependent dioxygenase-encoding gene,SlF3HL,in chilling stress responses in tomato(Solanum lycopersicum cv.Alisa Craig[AC]).Knockdown(KD)of SlF3HL(through RNA interference)in tomato led to increased sensitivity to chilling stress as indicated by elevated levels of electrolyte leakage,malondialdehyde(MDA)and reactive oxygen species(ROS).In addition,the KD plants had decreased levels of proline and decreased activities of peroxisome and superoxide dismutase.The expression of four cold-responsive genes was substantially reduced in the KD plants.Furthermore,seedling growth was significantly greater in AC or SlF3HLoverexpression plants than in the KD plants under either normal growth conditions with methyl jasmonate(MeJA)or chilling stress conditions.SlF3HL appears to positively regulate JA accumulation and the expression of JA biosynthetic and signaling genes under chilling stress.Together,these results suggest that SlF3HL is a positive regulator of chilling stress tolerance and functions in the chilling stress tolerance pathways,possibly by regulating JA biosynthesis,JA signaling,and ROS levels.Tixu Hu Yuqin Wang Qiqi Wang Ningning Dang Ling Wang Chaochao Liu Jianhua Zhu Xiangqiang Zhan 2019Horticulture Research2019,6,1:1

16Molecular mapping of two semidwarf genes in an indica rice variety Aitaiyin3(Oryza sativa L.)显示文摘Genetic analysis established that Aitaiyin3,a dwarf rice variety derived from a semidwarf cultivar Taiyin1,carries two recessive semidwarf genes.By using simple sequence repeat(SSR)markers,we mapped the two semidwarf genes,sd-1 and sd-t2 on chromosomes 1 and 4,respectively.Sd-t2 was thus named because the semidrawf gene sd-t has already been identified from Aitaiyin 2 whose origin could be traced back to Taiyin1.The result of the molecular mapping of sd-1 gene revealed it is linked to four SSR markers found on chromosome 1.These markers are:RM297,RM302,RM212,and OSR3 spaced at 4.7 cM,0 cM,0.8cM and 0 cM,respectively.Sd-t2 was found to be located on chromosome 4 using five SSR markers:two markers,SSR332 and RM1305 located proximal to sd-t2 are spaced 11.6 cM,3.8 cM,respectively,while the three distally located primers,RM5633,RM307,and RM401 are separated by distances of 0.4 cM,0.0 cM,and 0.4 cM,respectively.Zhao Xiangqiang Liang Guohua Zhou Jingsong Yan Changjie Cao Xiaoying Gu Minghong 2006Frontiers in Biology2006,1,2:0

194-methylumbelliferone (4-MU) enhances drought tolerance of apple by regulating rhizosphere microbial diversity and root architecture显示文摘The dwarfing rootstocks-mediated high-density apple orchard is becoming the main practice management.Currently,dwarfing rootstocks are widely used worldwide,but their shallow root system and drought sensitivity necessitate high irrigation requirements.Here,the root transcriptome and metabolome of dwarfing(M9-T337,a drought-sensitive rootstock)and vigorous rootstocks(Malus sieversii,a drought-tolerant species,is commonly used as a rootstock)showed that a coumarin derivative,4-Methylumbelliferon(4-MU),was found to accumulate significantly in the roots of vigorous rootstock under drought condition.When exogenous 4-MU was applied to the roots of dwarfing rootstock under drought treatment,the plants displayed increased root biomass,higher root-to-shoot ratio,greater photosynthesis,and elevated water use efficiency.In addition,diversity and structure analysis of the rhizosphere soil microbial community demonstrated that 4-MU treatment increased the relative abundance of putatively beneficial bacteria and fungi.Of these,Pseudomonas,Bacillus,Streptomyces,and Chryseolinea bacterial strains and Acremonium,Trichoderma,and Phoma fungal strains known for root growth,or systemic resistance against drought stress,were significantly accumulated in the roots of dwarfing rootstock after 4-MU treatment under drought stress condition.Taken together,we identified a promising compound—4-MU,as a useful tool,to strengthen the drought tolerance of apple dwarfing rootstock.Dehui Zhang Jieqiang He Pengda Cheng Yutian Zhang Abid Khan Shicong Wang Zhongxing Li Shuang Zhao Xiangqiang Zhan Fengwang Ma Xuewei Li Qingmei Guan 2023Horticulture Research2023,10,7:0
20Identification and fine mapping of a fertility restorer gene for wild abortive cytoplasmic male sterility in the elite indica rice non-restorer line 9311显示文摘The wild abortive(WA)-type cytoplasmic male sterility(CMS)derived from the wild rice species Oryza rufipogon Griff.is used widely in three-line indica hybrids.The identification and mapping of restorer of fertility(Rf)genes aided in the development of WA-type hybrids.Here we report that testcross F1 plants from the WA-type CMS line and 9311 exhibited stainable pollen grains with no seed set,indicating that 9311 carries minor-effect Rfs for WA-type CMS.We developed an advanced backcross population consisting of plants harboring small regions of donor chromosomal segments from 9311 in the WATianfeng A genetic background with moderate seed setting rates.Genetic analysis showed that the pollen fertility levels of the backcross individuals are governed by a single gene from 9311 that we named Rf19(t).By use of the RICE 40 K gene chip,three introduced segments were identified in the fertile lines,and a candidate region spanning 4.37–8.29 Mb on chromosome 1 was identified for Rf19(t).Finally,Rf19(t)was fine-mapped to a region of 90 kb between the DNA marker loci STS1-163 and STS1-183,in which eight ORFs were predicted.Also,using relative expression analyses,comparative sequence analyses and functional domain analyses,we identified LOC_Os01g10530 as the most likely candidate gene for Rf19(t).Furthermore,Rf19(t)was found to function in fertility restoration,most probably by regulating the degradation of m RNA transcribed from the mitochondrial gene WA352.These results increase our knowledge of fertility restoration in WA-type CMS lines and will facilitate the development of high-quality pairs of WAtype CMS and maintainer lines.Zuopeng Xu Yuanyue Du Xixu Li Ruixuan Wang Zhi'ai Chen Xiangqiang Zhao Qiaoquan Liu Shuzhu Tang Honggen Zhang 2023The Crop Journal2023,11,3:0
