1Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5

3High-density SNP genetic linkage map construction and quantitative trait locus mapping for resistance to cucumber mosaic virus in tobacco(Nicotiana tabacum L.)显示文摘Genetic linkage maps are essential for studies of genetics, genomic structure, and genomic evolution, and for mapping quantitative trait loci (QTL). Identification of molecular markers and construction of genetic linkage maps in tobacco (Nicotiana tabacum L.), a classical model plant and important economic crop, have remained limited. In the present study we identified a large number of single nucleotide polymorphism (SNP) markers and constructed a high-density SNP genetic map for tobacco using restriction site-associated DNA sequencing. In 1216.30 Gb of clean sequence obtained using the Illumina HiSeq 2000 sequencing platform, 99,647,735 SNPs were identified that differed between 203 sequenced plant genomes and the tobacco reference genome. Finally, 13,273 SNP markers were mapped on 24 high-density tobacco genetic linkage groups. The entire linkage map spanned 3421.80 cM, with a mean distance of 0.26 cM between adjacent markers. Compared with genetic linkage maps published previously, this version represents a considerable improvement in the number and density of markers. Seven QTL for resistance to cucumber mosaic virus (CMV) in tobacco were mapped to groups 5 and 8. 7Formulated nano-liposomes for reversal of cisplatin resistance in NSCLC with nucleus-targeting peptide显示文摘Cell membrane-engineered nano-delivery systems have evolved as a promising strategy to enhance drug bioavailability,offering an alternative for reversing drug resistance in cancer therapy.Herein,a formulated nano-liposome that fabricated by hybridizing cisplatin-resistant A549 cell line(A549/cis)cancer cell membrane and phospholipids for co-delivery of cisplatin and nuclear protein zeste homolog 2(EZH2)-targeting peptide EIP103,referred to as cLCE,was developed.In vitro results indicated that the formulated nano-liposome can efficiently inhibit A549/cis cancer cell invasion and metastasis through the down-regulation of Ncadherin and vimentin proteins.Mechanistic studies demonstrated that the reduction of nerve growth factor receptor(NGFR)levels and the increase of peroxisome proliferator-activated receptorγ(PPARγ)levels achieved by EIP103 may contribute to the reversal of cisplatin resistance.In vivo results demonstrated that the encapsulation of both cisplatin and EIP103 within cLCE leads to increased intratumoral accumulation and prolonged survival in A549/cis cancer-bearing mice as compared to the individual drugs alone.This can be attributed to the enhanced tumor homing capability of cLCE achieved through the presence of inherited membrane proteins derived from A549/cis cells.Taken together,this study may provide a highly promising therapeutic strategy to improve clinical treatments for cisplatin-resistance non-small-cell lung cancer(NSCLC)as well as other malignant cancers.Minxian Li Mei Jiang Mengting Chen Lilusi Ma Xiaocui Fang Yanlian Yang Chen Wang 2023Nano Research2023,16,11:0

9Short-term Effects of Nitrogen Deposition on Soil Respiration of Cinnamomum camphora Plantation显示文摘The nitrogen deposition experiments on Cinnamomum camphora plantation of Hunan provincial botanical garden were simulated from June the 7th to October the 7th,2010,the nitrogen deposition levels was respectively as control(CK, 0 g·m-2 a-1),low nitrogen(LN,5 g·m-2 a-1),medium nitrogen(MN,15 g·m-2 a-1) and high nitrogen(HN, 30 g·m -2 a-1).The soil respiration speed and soil surface temperature and the surface water content of the soil were determined by LI-8100 measuring instrument.The results show that the average values of soil respiration speed were CK (4.09±0.66μmol·m-2 s-1),LN(2.39±029μmol·m-2 s-1), MN(2.18±0.19μmol·m-2s-1),HN (2.28±0.25μmol·m-2 s-1),and the treatment of the CK was obviously higher than the other three treatments(P<0.01).With different nitrogen concentrations deposition treatments,the Q10 (temperature sensitivity coefficients) for soil respiration of CK,LN,MN and HN treatments were 1.84,1.71,1.83 and 1.56.It was positive correlation between the respiration speed and the surface(≤10 cm) temperature of tested soil.It was negative correlation between the respiration speed and the surface(≤5 cm) water content of tested soil(P>0.05).Volumetric water content of 5-cm soil was between 0.266 6-0.294 4(m3·m-3), and the monthly content did not vary too much during the research period(1.8%-9.4%).The findings suggest that the initial stage of nitrogen deposition obviously influenced the soil respiration of C.camphora plantation.Zhang Xuyuan Yan Wende Ma Xiuhong Zheng Wei Wang Guangjun Liang Xiaocui 2012Chinese Forestry Science and Technology2012,11,3:0
10β-Catenin Deletion in Regional Neural Progenitors Leads to Congenital Hydrocephalus in Mice显示文摘Congenital hydrocephalus is a major neurological disorder with high rates of morbidity and mortality;however,the underlying cellular and molecular mechanisms remain largely unknown.Reproducible animal models mirroring both embryonic and postnatal hydrocephalus are also limited.Here,we describe a new mouse model of congenital hydrocephalus through knockout ofβ-catenin in Nkx2.1-expressing regional neural progenitors.Progressive ventriculomegaly and an enlarged brain were consistently observed in knockout mice from embryonic day 12.5 through to adulthood.Transcriptome profiling revealed severe dysfunctions in progenitor maintenance in the ventricular zone and therefore in cilium biogenesis afterβ-catenin knockout.Histological analyses also revealed an aberrant neuronal layout in both the ventral and dorsal telencephalon in hydrocephalic mice at both embryonic and postnatal stages.Thus,knockout ofβ-catenin in regional neural progenitors leads to congenital hydrocephalus and provides a reproducible animal model for studying pathological changes and developing therapeutic interventions for this devastating disease.Lin Ma Yanhua Du Xiangjie Xu Hexi Feng Yi Hui Nan Li Guanyu Jiang Xiaoqing Zhang Xiaocui Li Ling Liu 2022Neuroscience Bulletin2022,38,1:0
11Effect of Ab_(1-40) on membrane permeability and intracellular free Ca^(2+) of nerve cells显示文摘The effects of soluble and fibrillar Ab1-40 on membrane permeability and intracellular free Ca2+ of nerve cells were investigated by the laser confocal microscopy. Results indicate that: i) Effects of soluble and fibrillar Ab1-40 on cell membrane permeability are both concentration-de- pendent. Soluble Ab1-40 increases membrane permeability only at concentration of 3 mmol/L, while the toxic effect of fibrillar Ab1-40 is much stronger, its evident effect begins from 1 mmol/L. When its concentration rose to 3 mmol/L, not only the membrane permeability increased, but also the nuclear membrane broke seriously. ii) Both soluble and fibrillar Ab1-40 at high concentrations increased the intracellular free Ca2+, and the increased amplitudes are concentration-dependent. However, the fibrillar one induces the increase of intracellular Ca2+ much quicker and synchronously.
