1Anti-diabetic vanadyl complexes reduced Alzheimer's disease pathology independent of amyloid plaque deposition显示文摘Association of Alzheimer's disease(AD) with cerebral glucose hypometabolism, likely due to impairments of insulin signaling,has been reported recently, with encouraging results when additional insulin is provided to AD patients. Here, we tested the potential effects of the anti-diabetic vanadium, vanadyl(IV) acetylacetonate(VAC), on AD in vitro and in vivo models. The experimental results showed that VAC at sub-micromolar concentrations improved the viability of neural cells with or without increased β-amyloid(Aβ) burden; and in APP/PS1 transgenic mice, VAC treatment(0.1 mmol kg-1 d-1) preserved cognitive function and attenuated neuron loss, but did not reduce brain Aβ plaques. Further studies revealed that VAC attenuated Aβpathogenesis by(i) activation of the PPARγ-AMPK signal transduction pathway, leading to improved glucose and energy metabolism;(ii) up-regulation of the expression of glucose-regulated protein 75(Grp75), thus suppressing p53-mediated neuronal apoptosis under Aβ-related stresses; and(iii) decreasing toxic soluble Aβ peptides. 2Optimization and control of a reactive distillation process for the synthesis of dimethyl carbonate显示文摘Dimethyl carbonate is an environmentally benign and biodegradable chemical.Based on integration of reactive distillation and pressure-swing distillation technologies,a novel process for synthesis of dimethyl carbonate through transesterification with propylene carbonate and methanol has been developed by Huang et al.In this work,the optimization of this process was performed by minimizing the total TAC.The results show that the optimal design flowsheet can save energy consumption by 18.6% with the propylene carbonate conversion of 99.9%.Then,an effective plant-wide control structure for the process was developed.Dynamic simulation results demonstrate that the temperature/flow rate cascade control plus with simple temperature control can keep not only product purity but also the conversion of the reactant at their desired values in the face of the disturbance in reactant feed flow rate and feed composition.Zhixian Huang Yixiong Lin Xiaoda Wang Changshen Ye Ling Li 2017Chinese Journal of Chemical Engineering2017,25,8:8
3Transesterification of palm oil to biodiesel using Brφnsted acidic ionic liquid as high-efficient and eco-friendly catalyst显示文摘The transesterification of palm oil and methanol catalyzed by Br?nsted acidic ionic liquids was investigated. Four eco-friendly Br?nsted acidic ionic liquids were prepared and their structures were characterized by NMR, FT-IR and TG–DTG. The results demonstrated that [CyN_(1,1)PrSO_3H][p-TSA] was more efficient than the other ionic liquids and chosen as catalyst for further research. The influences of various reaction parameters on the conversion of palm oil to biodiesel were performed, and the orthogonal test was investigated to seek the optimum reaction conditions, which were illustrated as follows: methanol to oil mole ratio of 24:1, catalyst dosage of 3.0 wt% of oil, reaction temperature of 120 °C, reaction time of 150 min, and the biodiesel yield achieved 98.4%. In addition, kinetic study was established for the conversion process, with activation energy and preexponential factor of 122.93 k J·mol^(-1) and 1.83 × 10^(15), respectively. Meanwhile, seven-time recycling runs of ionic liquid were completed with ignorable loss of its catalyst activity. 4Prussian blue nanoparticle-loaded microbubbles for photothermally enhanced gene delivery through ultrasound-targeted microbubble destruction显示文摘By adsorbing chitosan(CS)-functionalized Prussian blue(PB) nanoparticles(CS/PB NPs) complexing DNA onto the surface of gas encapsulated microbubbles(MBs), a multifunctional gene delivery system of MBs@CS/PB/DNA was fabricated for photothermally enhanced gene transfection through ultrasound-targeted microbubble destruction. CS/PB NPs of(2.69 ± 0.49) nm could complex DNA effectively when the mass ratio was2:1. It was found that MBs@CS/PB/DNA could enhance ultrasound imaging greatly both in vitro and in vivo. In addition, MBs@CS/PB/DNA could be disrupted by applying a higher-intensity ultrasound irradiation to release CS/PB/DNA, which could effectively transform the nearinfrared(NIR) light into heat to assist the uptake of CS/PB/DNA by cells. With the aid of ultrasound irradiation and NIR light irradiation, the gene transfection efficiency was significantly enhanced to(43.08 ± 1.13) %, much higher than polyethylenimine. 5Identification of diseases for soybean seeds by computer vision applying BP neural network显示文摘The use of computer vision for estimating quality in agriculture products has become wide spread in recent years and the composition,variety,or ripeness can be estimated.On the other hand,the appearance is one of the most worrying issues for producers due to its influence on quality.In this research,computer vision technology combined with BP artificial neural network(ANN)was developed to identify soybean frogeye,mildewed soybean,worm-eaten soybean and damaged soybean.Thirty-nine characteristic parameters from color,texture and shape characteristics were computed after preprocessing the acquired soybean images.The dimensionality of the characteristic parameters was reduced from 39 dimensionalities to 12 dimensionalities using the method of principal component analysis(PCA).MALAB software was used to build a prediction model according to 12 characteristic parameters.The identification accuracies of soybean frogeye,mildewed soybean,damaged soybean and worm-eaten soybean are 96%,95%,92%,and 92%,respectively.And the accuracy for heterogeneous soybean seeds with several diseases is 90%.The results show that the prediction model constructed by BP neural network can identify the diseases of soybean seeds.And it is useful to estimate appearance quality of soybean by computer vision applying BP neural network.Tan Kezhu Chai Yuhua Song Weixian Cao Xiaoda 2014International Journal of Agricultural and Biological Engineering2014,7,3:4
6Brain-derived neurotrophic factor protects neurons from GdCl_3-induced impairment in neuron-astrocyte co-cultures显示文摘Gadolinium (Gd3+) complexes are important contrast agents in medical magnetic resonance imaging (MRI) and of great potential value in brain research. In order to better understand the mechanisms of the action of Gd3+ on neurons in the complex central nervous system (CNS), the neurotoxic actions of GdCl3 have been investigated in both neuron monoculture and astrocyte-neuron co-culture systems. Measurements of lactate dehydrogenase release showed that GdCl3 causes significant cell death of monocultured neurons as a result of reactive oxygen species (ROS) generation and down-regulation of brain-derived neurotrophic factor (BDNF). However, GdCl3 does not affect the viability and BDNF expression of astrocytes. Both co-culturing of neurons with astrocytes and addition of BDNF ameliorated GdCl3-induced neurotoxicity by decreasing ROS generation and facilitating recovery of BDNF levels. The results obtained suggest that astrocytes in the CNS may protect neurons from GdCl3-induced impairment through secreting BDNF and thus up-regulating BDNF expression and interfering with Gd3+-induced cell signaling in neurons. 7Vanadyl ions binding to GroEL (HSP60) and inducing its depolymerization显示文摘Several vanadium compounds have been known for the hypoglycemic and anticancer effects. However, the mechanisms of the pharmacological and toxicological effects were not clear. In this work, we in- vestigated the potential targets of vanadium in mitochondria. Vanadyl ions were found to bind to mi- tochondria from rat liver with a stoichiometry of 244±58 nmol/mg protein and an apparent dissocia- tion constant (Kd) of (2.0±0.8)×10·16 mol/L. Using size exclusion chromatography, a vanadium-binding protein was isolated and identified to be the 60-kDa heat shock protein (HSP60) by mass spectrometry analysis and immunoassays. Additionally, binding of vanadyl ions was found to result in depolymeri- zation of homo-oligomeric HSP60 (GroEL). HSP60 is an indispensable molecular chaperone and in- volved in many kinds of pathogenesis of inflammatory and autoimmune diseases, e.g. type 1 diabetes. 18Copper ions inactivate S-ade-nosylhomocysteine hydrolase显示文摘S-adenosylhomocysteine (AdoHcy) hydrolase is an enzyme that regulates biomethylation and some other physiological processes. Recombinant AdoHcy hydrolase was overexpressed in E. coli JM109 and purified with ion exchange and gel filtration chromatographies. The effects of copper ions (Cu2+) on the activity of AdoHcy hydrolase were investigated and the results showed that Cu2+ inhibited the enzyme’s activity by a concentration and time-dependent process. The inhibition constant (Ki) and the apparent rate constant (Kapp) were calculated to be (14±4) nmol·L-1 and (1.08±0.15) min-1, respectively. The existence of the natural substrate Ado could to some extent prevent Cu2+ from inactivating the enzyme, suggesting that copper ions possibly could compete with the natural substrate on enzyme’s substrate binding site.
