| 1 | Olfactory ecto-mesenchymal stem cell-derived exosomes ameliorate murine Sjögren’s syndrome by modulating the function of myeloid-derived suppressor cells显示文摘Sjögren’s syndrome(SS)is a systemic autoimmune disease characterized by progressive inflammation and tissue damage in salivary glands and lacrimal glands.Our previous studies showed that myeloid-derived suppressor cells(MDSCs)exhibited impaired immunosuppressive function during disease progression in patients with SS and mice with experimental Sjögren’s syndrome(ESS),but it remains unclear whether restoring the function of MDSCs can effectively ameliorate the development of ESS.In this study,we found that murine olfactory ecto-mesenchymal stem cell-derived exosomes(OE-MSC-Exos)significantly enhanced the suppressive function of MDSCs by upregulating arginase expression and increasing ROS and NO levels.Moreover,treatment with OE-MSC-Exos via intravenous injection markedly attenuated disease progression and restored MDSC function in ESS mice.Mechanistically,OE-MSC-Exo-secreted IL-6 activated the Jak2/Stat3 pathway in MDSCs.In addition,the abundant S100A4 in OE-MSC-Exos acted as a key factor in mediating the endogenous production of IL-6 by MDSCs via TLR4 signaling,indicating an autocrine pathway of MDSC functional modulation by IL-6.Taken together,our results demonstrated that OE-MSC-Exos possess therapeutic potential to attenuate ESS progression by enhancing the immunosuppressive function of MDSCs,possibly constituting a new strategy for the treatment of Sjögren’s syndrome and other autoimmune diseases. | Ke Rui Yue Hong Qiugang Zhu Xiaofei Shi Fan Xiao Hailong Fu Qing Yin Yida Xing Xinfeng Wu Xiaodan Kong Huaxi Xu Jie Tian Shengjun Wang Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,2: | 12 |
| 2 | A NAC transcription factor, NOR-like1, is a new positive regulator of tomato fruit ripening显示文摘Ripening of the model fruit tomato(Solanum lycopersicum)is controlled by a transcription factor network including NAC(NAM,ATAF1/2,and CUC2)domain proteins such as No-ripening(NOR),SlNAC1,and SlNAC4,but very little is known about the NAC targets or how they regulate ripening.Here,we conducted a systematic search of fruit-expressed NAC genes and showed that silencing NOR-like1(Solyc07g063420)using virus-induced gene silencing(VIGS)inhibited specific aspects of ripening.Ripening initiation was delayed by 14 days when NOR-like1 function was inactivated by CRISPR/Cas9 and fruits showed obviously reduced ethylene production,retarded softening and chlorophyll loss,and reduced lycopene accumulation.RNA-sequencing profiling and gene promoter analysis suggested that genes involved in ethylene biosynthesis(SlACS2,SlACS4),color formation(SlGgpps2,SlSGR1),and cell wall metabolism(SlPG2a,SlPL,SlCEL2,and SlEXP1)are direct targets of NOR-like1.Electrophoretic mobility shift assays(EMSA),chromatin immunoprecipitation-quantitative PCR(ChIP-qPCR),and dual-luciferase reporter assay(DLR)confirmed that NOR-like1 bound to the promoters of these genes both in vitro and in vivo,and activated their expression.Our findings demonstrate that NOR-like1 is a new positive regulator of tomato fruit ripening,with an important role in the transcriptional regulatory network. | Ying Gao Wei Wei Xiaodan Zhao Xiaoli Tan Zhongqi Fan Yiping Zhang Yuan Jing Lanhuan Meng Benzhong Zhu Hongliang Zhu Jianye Chen Cai-Zhong Jiang Donald Grierson Yunbo Luo Da-Qi Fu | 2018 | Horticulture Research2018,5,1: | 9 |
| 3 | Effects of Plant Growth-Promoting Rhizobacteria and N Source on Plant Growth and N and P Uptake by Tomato Grown on Calcareous Soils显示文摘Introducing specific microorganisms into the soil ecological system is an important strategy for improving nutrient use efficiency.Two pot experiments were conducted in the greenhouse from December 3, 2012 to January 25, 2013(Experiment 1) and March 11 to April 23, 2013(Experiment 2) to evaluate the effect of nitrogen(N) source and inoculation with plant growth-promoting rhizobacteria(PGPR) on plant growth and N and phosphorus(P) uptake in tomato(Lycopersicon esculentum Mill.) grown on calcareous soils from South Florida, USA.Treatments included urea, controlled release urea(a controlled release fertilizer, CRF) each at low and high N rates and with or without inoculation of PGPR.A mixture of PGPR strains Bacillus amyloliquefaciens IN937 a and Bacillus pumilus T4 was applied to the soil during growing periods of tomato.Treatments with PGPR inoculation increased plant height compared to treatments without PGPR in both experiments.Inoculation with PGPR increased shoot dry weight and shoot N uptake for the same N rate and N source.In both experiments, only at high N rate, CRF and urea treatments with PGPR had significantly(P < 0.05)greater shoot biomass than those without PGPR.Only at high N rate, CRF treatment with PGPR significantly increased shoot N uptake by 39.0% and 10.3% compared to that without PGPR in Experiments 1 and 2, respectively.Meanwhile, presence of PGPR in the soil increased shoot P uptake for all treatments in Experiment 1 and for most treatments in Experiment 2.In Experiment 1, only at low N rate, CRF treatment with PGPR significantly increased shoot P uptake compared with that without PGPR.In Experiment2, a significant increase in shoot P uptake by inoculation of PGPR was only observed in CRF treatment at high N rate.Results from this study indicate that inoculation with PGPR may increase plant growth and N and P uptake by tomato grown on calcareous soils.However, the effect of PGPR varied and was influenced by many factors such as N source, N rate, and soil fertility.Further investigations are warranted to confirm the effect of PGPR under different soil conditions. | Xiaohui FAN Shouan ZHANG Xiaodan MO Yuncong LI Yuqing FU Zhiguang LIU | 2017 | Pedosphere2017,27,6: | 8 |
| 4 | Improvements of corner frequency and scaling factor for stochastic finite-fault modeling显示文摘In this paper, three existing source spectral models for stochastic finite-fault modeling of ground motion were reviewed. These three models were used to calculate the far-field received energy at a site from a vertical fault and the mean spectral ratio over 15 stations of the Northridge earthquake, and then compared. From the comparison, a necessary measure was observed to maintain the far-field received energy independent of subfault size and avoid overestimation of the long-period spectral level. Two improvements were made to one of the three models (i.e., the model based on dynamic corner frequency) as follows: (i) a new method to compute the subfault corner frequency was proposed, where the subfault corner frequency is determined based on a basic value calculated from the total seismic moment of the entire fault and an increment depending on the seismic moment assigned to the subfault; and (ii) the difference of the radiation energy from each subfault was considered into the scaling factor. The improved model was also compared with the unimproved model through the far-field received energy and the mean spectral ratio. | 6 | Eco-friendly preparation of chitooligosaccharides with different degrees of deacetylation from shrimp shell waste and their effects on the germination of wheat seeds显示文摘Production of chitosan and its derivatives by traditional methods involves the excessive use of a reaction solution comprisedof sodium hydroxide and hydrochloric acid. Waste water resulting from this process has lim让ed the application of chitosanas a fertilizer as the process causes serious environmental pollution. Specifically, the resulting waste water contains highlevels of dissolved nitrogen and minerals from shrimp shells. In this study, an eco-friendly method was established to produce chitooligosaccharides (COS) with different degrees of deacetylation (DDAs) from shrimp shell waste. At a solid-tosolventratio of 1:6, the degree of demineralization was above 90% with the treatment of 30 g-L_1 H3PO4, and the degree ofdeproteinization was above 80% when treated with 30 g-L_1 KOH at 70 °C. Chitosans with different DDAs were obtainedby microwave-assisted KOH metathesis and the COS with Mw approximately 1500 Da were then prepared by oxidativedegradation. In summary, 33.73 kg H3PO4,12.77 kg, and 241.31 kg KOH were supplied during the processes of demineralization,deproteinization, and deacetylation of 100 kg shrimp shell waste, respectively. The process water was totally recycled,demonstrating that the shrimp shell could be wholly transformed into fertilize The entire process created a product withthe fractions of N:P2O5:K2O:COS = 7.94:24.44:10.72:1 &27. The test on the germination promotion of wheat seeds revealedthat the COS with 72」2% DDA significantly promoted germination. | 9 | Effect of seasonal snow on the start of growing season of typical vegetation in Northern Hemisphere显示文摘Under global warming, seasonal snow takes faster melting rate than before, which greatly changes the hydro-logical cycle. In this study, by targeting three typical seasonal snow-covered land types (i.e., open shrubland,evergreen needleleaf forest and mixed forest) in the Northern Hemisphere, the start of growing season (SGS) hasbeen found obviously advanced in the past years, greatly contributed by the faster melting rate of seasonal snow.It is manifested that significantly positive correlation has been found between SGS and May snow depth for openshrubs, March and April snow depth for evergreen needleleaf forests and March snow depth for mixed forests.However, such close association is not appeared in all the climate conditions of same vegetation. | 14 | Response of Salmonella enterica serovar Typhimurium to alginate oligosaccharides fermented with fecal inoculum:integrated transcriptomic and metabolomic analyses显示文摘Alginate oligosaccharides(AOS),extracted from marine brown algae,are a common functional feed additive;however,it remains unclear whether they modulate the gut microbiota and microbial metabolites.The response of Salmonella enterica serovar Typhimurium,a common poultry pathogen,to AOS fermented with chicken fecal inocula was investigated using metabolomic and transcriptomic analyses.Single-strain cultivation tests showed that AOS did not directly inhibit the growth of S.Typhimurium.However,when AOS were fermented by chicken fecal microbiota,the supernatant of fermented AOS(F-AOS)exhibited remarkable antibacterial activity against S.Typhimurium,decreasing the abundance ratio of S.Typhimurium in the fecal microbiota from 18.94 to 2.94%.Transcriptomic analyses showed that the 855 diferentially expressed genes induced by F-AOS were mainly enriched in porphyrin and chlorophyll metabolism,oxidative phosphorylation,and Salmonella infection-related pathways.RT-qPCR confrmed that F-AOS downregulated key genes involved in fagellar assembly and the type III secretory system of S.Typhimurium,indicating metabolites in F-AOS can infuence the growth and metabolism of S.Typhimurium.Metabolomic analyses showed that 205 microbial metabolites were signifcantly altered in F-AOS.Among them,the increase in indolelactic acid and 3-indolepropionic acid levels were further confrmed using HPLC.This study provides a new perspective for the application of AOS as a feed additive against pathogenic intestinal bacteria. | Jiaying Cheng Mengshi Xiao Xinmiao Ren Francesco Secundo Ying Yu Shihao Nan Weimiao Chen Changliang Zhu Qing Kong Youtao Huang Xiaodan Fu Haijin Mou | 2023 | Marine Life Science & Technology2023,5,2: | 0 |
