2Chinese expert consensus statement on the diagnosis and treatment of fulminant myocarditis显示文摘Fulminant myocarditis is primarily caused by infection with any number of a variety of viruses. It arises quickly, progresses rapidly, and may lead to severe heart failure or circulatory failure presenting as rapid-onset hypotension and cardiogenic shock,with mortality rates as high as 50%–70%. Most importantly, there are no treatment options, guidelines or an expert consensus statement. Here, we provide the first expert consensus, the Chinese Society of Cardiology Expert Consensus Statement on the Diagnosis and Treatment of Fulminant Myocarditis, based on data from our recent clinical trial(NCT03268642). In this statement, we describe the clinical features and diagnostic criteria of fulminant myocarditis, and importantly, for the first time,we describe a new treatment regimen termed life support-based comprehensive treatment regimen. The core content of this treatment regimen includes(i) mechanical life support(applications of mechanical respirators and circulatory support systems,including intraaortic balloon pump and extracorporeal membrane oxygenation),(ii) immunological modulation by using sufficient doses of glucocorticoid, immunoglobulin and(iii) antiviral reagents using neuraminidase inhibitor. 3External Characteristics and Internal Flow Features of a Centrifugal Pump during Rapid Startup显示文摘Centrifugal pumps always work under steady conditions,and many researches focus on the steady operation.But transient conditions,such as sudden startup and shutdown,are inevitable.The researches on the inner flow of centrifugal pumps under transient conditions have been done,and they show that the transient operation is different from the steady operation.In order to research the evolution of unsteady flow in a centrifugal pump under transient conditions,and to investigate the mechanism of transient effects by analyzing the unsteady flow in a centrifugal pump,the external characteristic experiment and the internal flow numerical calculation of the centrifugal pump with an open impeller during startup is presented.The relationships of the rotation speed,capacity and head between start-time are obtained by the external characteristics experiment.The numerical calculations under startup process are carried out by using the k-e model and N-S equation.The distribution of velocity and pressure in the inner channel of the tested pump was obtained by choosing fourteen start-time points and twelve geometrical points in the impeller channel during startup.The calculation results show that the velocity and the pressure increase linearly with the start-time before rotation's speed gets steady,then changes almost horizontally after rotation speed becomes steady,then fluctuates until being steady.The internal flow characteristics are in good agreement with the external characteristic experimental results and numerical calculation.The simulation methods and results make the basis for the diagnosis and optimization of under flow in the centrifugal pump during transient operation.ZHU Zuchao GUO Xiaomei CUI Baoling LI Yi 2011Chinese Journal of Mechanical Engineering2011,24,5:14
4miR-188-5p regulates proliferation and invasion via PI3K/Akt/MMP-2/9 signaling in keloids显示文摘Keloids(KDs)and hypertrophic scars(HSs),two forms of pathological scars,seriously affect the physical and psychological health of patients.Despite many similarities with HSs,KDs are characterized by invasion and a high rate of recurrence after surgery,features they share in common with tumors.The underlying molecular mechanisms of this phenomenon have not been fully elucidated.In this study,we used microRNA(miRNA)array analysis to search for invasion-associated miRNAs in KDs.The expression of miR-188-5p in KDs,HSs,normal skin(NS)tissues,and cell lines was measured by quantitative real-time polymerase chain reaction.Furthermore,cell proliferation,migration,and invasion were detected in KD fibroblasts(KFs)and HS fibroblasts(HSFs),and interrelated proteins were ascertained by western blot analysis.It was found that miR-188-5p was significantly decreased in KD tissue compared with HS and NS tissues.Upregulated expression of miR-188-5p suppressed KF proliferation,migration,and invasion;and decreased expression of miR-188-5p also promoted HSF proliferation,migration,and invasion.The protein levels of MMP-2,MMP-9,PI3K,and p-Akt in miR-188-5p mimic-transfected KFs were repressed.In contrast,after transfection with miR-188-5p inhibitor,the protein levels of MMP-2,MMP-9,PI3K,and p-Akt were higher than the control in HSFs.Treatment with PI3K/Akt inhibitor LY294002 in KFs with miR-188-5p inhibitor did not further reduce their proliferation,migration,and invasion.The upregulation of MMP-2 and MMP-9 by miR-188-5p inhibitor could be abolished by LY294002.These findings together demonstrate a tumor-suppressive role of miR-188-5p in KD proliferation and invasion via PI3K/Akt/MMP-2/9 signaling,indicating that miR-188-5p may be a potential prognostic marker and therapeutic target for KDs.Wenyan Zhu Xiaoyan Wu Bo Yang Xiaodong Yao Xiaomei Cui Pan Xu Xiaodong Chen 2019Acta Biochimica et Biophysica Sinica2019,51,2:9
5Identification and Characterization of microRNA during Bemisia tabaci Infestations in Solanum lycopersicum and Solanum habrochaites显示文摘MicroRNAs(miRNAs) are a class of small non-coding RNAs that can regulate target gene expression during many plant growth and development processes. In recent years, several studies identified the miRNAs involved in fruit development, leaf development, and abiotic responses in tomato. However, little is known about the miRNAs that respond to insect attack. Here, miRNAs were identified by high-throughput sequencing at different stages after infections by the whitefly Bemisia tabaci in Solanum lycopersicum and Solanum habrochaites, which are susceptible and resistant to whitefly, respectively. A total of 44 known miRNA families were identified, and 33 were shared between the two species. Among these, 13 miRNA families were newly reported in tomato. After strict filtering, some novel miRNAs were also discovered. The global expression patterns of the miRNAs exhibited were different between the two species, reflecting their different responses and resistance levels to whitefly attack. Some of the predicted target genes of differentially expressed miRNAs may be involved in responding to, and defending against, diseases and insects. 6Molecular characteristics and evolutionary analysis of a very virulent infectious bursal disease virus显示文摘Infectious bursal disease virus(IBDV) poses a significant threat to the poultry industry. Viral protein 2(VP2), the major structural protein of IBDV, has been subjected to frequent mutations that have imparted tremendous genetic diversity to the virus. To determine how amino acid mutations may affect the virulence of IBDV, we built a structural model of VP2 of a very virulent strain of IBDV identified in China, vv IBDV Gx, and performed a molecular dynamics simulation of the interaction between virulence sites. The study showed that the amino acid substitutions that distinguish vv IBDV from attenuated IBDV(H253Q and T284A) favor a hydrophobic and flexible conformation of ?-barrel loops in VP2, which could promote interactions between the virus and potential IBDV-specific receptors. Population sequence analysis revealed that the IBDV strains prevalent in East Asia show a significant signal of positive selection at virulence sites 253 and 284. In addition, a signal of co-evolution between sites 253 and 284 was identified. 7Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats.Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang 2022Science China(Life Sciences)2022,65,10:5
8Anti-cavitation performance of a splitter-bladed inducer under different flow rates and different inlet pressures显示文摘The anti-cavitation performance of a high-speed centrifugal pump with a splitter-bladed inducer is investigated under different flow rates and different inlet pressures. Simulations and external characteristics experiments are carried out. Static pressure and the vapor volume fraction distributions on the inducer and the impeller of the pump under various operation conditions are obtained. The results show that the cavitation developments on the impeller and on the inducer with the flow rates are reverse, while the development of the inlet pressure on the inducer and the impeller is the same. Cavitation on the impeller increases with the increase of flow rates, and it extends to the near passages with rotating, while cavitation on the inducer is more complex than that on the impeller. Cavitation at the inlet of the inducer decreases with the increase of flow rates, while cavitation at the outlet of the inducer is opposite. The results also show that cavitation development on the impeller and on the inducer with the inlet pressure is the same. Cavitation both decreases with the increase of the inlet pressure at the same flow rate. Furthermore, asymmetric cavitation on the impeller and on the inducer is both observed. 9A single mutation in the PBC loop of VP2 is involved in the in vitro replication of infectious bursal disease virus显示文摘To test whether amino acid mutations in the PBC and PHI loops of VP2 are involved in the replication and virulence of infectious bursal disease virus(IBDV), a pair of viruses, namely the moderately virulent IBDV(rG x-F9VP2) and the attenuated strain(rGt), were used. Residue mutations A222P(P_(BC)) and S330R(PHI), selected by sequence comparison, were introduced individually into r Gx-F9VP2 by using a reverse genetics system. In addition, the reverse mutation of either P222 A or R330 S was introduced into r Gt. The four modified viruses were then rescued and evaluated in vitro(CEF cells) and in vivo(SPF chickens). Results showed that A222 P elevated the replication efficiency of rG x-F9VP2 while P222 A reduced that of rG t in CEF cells. A mutation at residue 330 did not alter IBDV replication. In addition, animal experiments showed that a single mutation at either residue 222 or 330 did not significantly influence the virulence of IBDV. 10Experimental and numerical investigations on fatigue behavior of aluminum alloy 7050-T7451 single lap four-bolted joints显示文摘The fatigue behavior of aluminum alloy 7050-T7451 single lap four-bolted joints was studied by highfrequency fatigue test and finite element(FE) methods. The fatigue test results showed that a better enhancement of fatigue life was achieved for the joints with high-locked bolts by employing the combinations of cold expansion, interference fit, and clamping force. The fractography revealed that fatigue cracks propagated tortuously; more fatigue micro-cliffs, tearing ridges, lamellar structure were observed,and fatigue striation spacing was simultaneously reduced. The evaluation of residual stress conducted by FE methods confirmed the experimental results and locations of fatigue crack initiation. 15Spontaneous immunomodulation and regulation of angiogenesis and osteogenesis by Sr/Cu-borosilicate glass (BSG) bone cement to repair critical bone defects显示文摘Injectable bone biomaterials like bone cement should be designed and fabricated with certain biological criteria,which include:1)recruitment and polarization of the macrophages from M1(pro-inflammatory)to M2(anti-inflammatory)phenotype,2)enhance vascularization,and 3)activate osteogenic differentiation of bone marrow-derived stem cells to promote bone healing.So far,no injectable biomaterials could spontaneously regulate the entire bone healing process that involves inflammation,angiogenesis,and osteogenesis.Therefore,in this study,we designed bone cement comprised of strontium and copper-incorporated borosilicate glass(Sr/Cu-BSG)in the liquid phase of chitosan to modulate bone healing.In vitro studies showed that the controlled release of Sr and Cu ions up-regulated anti-inflammatory genes(IL-1Ra and TGF-β1)while down-regulating pro-inflammatory genes(IL-1βand IL-6)in macrophages at 3 days.Sr and Cu ions also increased the expressions of angiogenic genes(VEGF and bFGF)in HUVECs at 5 days and osteogenic genes(Runx-2,OCN,and OPN)in hBMSCs at 7,14,and 21 days.5Sr3Cu-BSG bone cement exhibited the best anti-inflammatory,angiogenic,and osteogenic properties among the bone cement groups with different Sr and Cu ratios.Short-term and long-term implantation of Sr/Cu-BSGs in femoral condylar bone defects of rats and rabbits confirmed the in vitro results,where the degradation rate of Sr/Cu-BSG matched the bone healing rate.Similar to in vitro,the 5Sr3Cu-BSG group also showed the highest bone formation in vivo.Excellent physical and chemical properties,along with its bone repairing ability,make the Sr/Cu-BSG bone cement a good candidate biomaterial for treating bone defects.Shuaijie Li Liyan Zhang Chunyu Liu Jua Kim Kun Su Tingli Chen Limin Zhao Xiaomei Lu Hao Zhang Yinglin Cui Xu Cui Feng Yuan Haobo Pan 2023Bioactive Materials2023,,5:1
