1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19
2Chinese expert consensus statement on the diagnosis and treatment of fulminant myocarditis显示文摘Fulminant myocarditis is primarily caused by infection with any number of a variety of viruses. It arises quickly, progresses rapidly, and may lead to severe heart failure or circulatory failure presenting as rapid-onset hypotension and cardiogenic shock,with mortality rates as high as 50%–70%. Most importantly, there are no treatment options, guidelines or an expert consensus statement. Here, we provide the first expert consensus, the Chinese Society of Cardiology Expert Consensus Statement on the Diagnosis and Treatment of Fulminant Myocarditis, based on data from our recent clinical trial(NCT03268642). In this statement, we describe the clinical features and diagnostic criteria of fulminant myocarditis, and importantly, for the first time,we describe a new treatment regimen termed life support-based comprehensive treatment regimen. The core content of this treatment regimen includes(i) mechanical life support(applications of mechanical respirators and circulatory support systems,including intraaortic balloon pump and extracorporeal membrane oxygenation),(ii) immunological modulation by using sufficient doses of glucocorticoid, immunoglobulin and(iii) antiviral reagents using neuraminidase inhibitor. 3External Characteristics and Internal Flow Features of a Centrifugal Pump during Rapid Startup显示文摘Centrifugal pumps always work under steady conditions,and many researches focus on the steady operation.But transient conditions,such as sudden startup and shutdown,are inevitable.The researches on the inner flow of centrifugal pumps under transient conditions have been done,and they show that the transient operation is different from the steady operation.In order to research the evolution of unsteady flow in a centrifugal pump under transient conditions,and to investigate the mechanism of transient effects by analyzing the unsteady flow in a centrifugal pump,the external characteristic experiment and the internal flow numerical calculation of the centrifugal pump with an open impeller during startup is presented.The relationships of the rotation speed,capacity and head between start-time are obtained by the external characteristics experiment.The numerical calculations under startup process are carried out by using the k-e model and N-S equation.The distribution of velocity and pressure in the inner channel of the tested pump was obtained by choosing fourteen start-time points and twelve geometrical points in the impeller channel during startup.The calculation results show that the velocity and the pressure increase linearly with the start-time before rotation's speed gets steady,then changes almost horizontally after rotation speed becomes steady,then fluctuates until being steady.The internal flow characteristics are in good agreement with the external characteristic experimental results and numerical calculation.The simulation methods and results make the basis for the diagnosis and optimization of under flow in the centrifugal pump during transient operation.ZHU Zuchao GUO Xiaomei CUI Baoling LI Yi 2011Chinese Journal of Mechanical Engineering2011,24,5:14
4Sorption of tylosin and sulfamethazine on solid humic acid显示文摘Tylosin(TYL) and sulfamethazine(SMT) are ionizable and polar antimicrobial compounds,which have seeped into the environment in substantial amounts via fertilizing land with manure or sewage. Sorption of TYL and SMT onto humic acid(HA) may affect their environmental fate. In this study, the sorption of TYL and SMT on HA at different conditions(pH, ionic strength) was investigated. All sorption isotherms fitted well to the Henry and Freundlich models and they were highly nonlinear with values of n between 0.5 and 0.8, which suggested that the HA had high heterogeneity. The sorption of TYL and SMT on HA decreased with increasing p H(2.0–7.5), implying that the primary sorption mechanism could be due to cation exchange interactions between TYL^+/SMT^+ species and the functional groups of HA.Increasing ionic strength resulted in a considerable reduction in the K_d values of TYL and SMT,hinting that interactions between H bonds and π–π EDA might be an important factor in the sorption of TYL and SMT on HA. Results of Fourier transform infrared(FT-IR) and ~13C-nuclear magnetic resonance(NMR) analysis further demonstrated that carboxyl groups and O-alkyl structures in the HA could interact with TYL and SMT via ionic interactions and H bonds,respectively. 5Ocean color products retrieval and validation around China coast with MODIS显示文摘Waters along China coast are very turbid with high concentrations of suspended sediment nearly all the time,especially at the Hangzhou Bay,the Changjiang (Yangtze) River Estuary and the shoal along Jiangsu Province.In these turbid and optically complex waters,the standard MODIS ocean color products tend to have invalid values.Because the water-leaving radiances in the near-infrared (NIR) are significant resulting from the strong scattering of suspended particles,the standard MODIS atmospheric correction algorithm often gets no results or produces significant errors.And because of the complex water optical properties,the OC3 model used in the standard MODIS data processing tends to get extremely high chlorophyll-a (Chl-a) concentrations.In this paper,we present an atmospheric correction approach using MODIS short wave infrared (SWIR) bands based on the fact that water-leaving radiances are negligible in the SWIR region because of the extreme strong absorption of water even in turbid waters.A regional Chl-a concentration estimation model is also constructed for MODIS from in situ data.These algorithms are applied to MODIS Aqua data processing in the China coastal regions.In situ data collected in the Yellow Sea and the East China Sea in spring and autumn,2003 are used to validate the performance.Reasonably good results have been obtained.It is noted that water-leaving reflectance in the NIR bands are significant in waters along the China coast with high sediment loadings.The satellite derived and in-situ reflectance spectra can match in the turbid waters along China coast,and there is relatively good linear relationship between satellite derived and in-situ reflectance.The RMSE value of Rrs(λ) is 0.0031 sr ?1 for all the nine ocean color bands (412 to 869 nm).The satellite-derived Chl-a value is in the reasonable range and the root mean square percentage difference is 46.1%.SUN Lingt GUO Maohua WANG Xiaomei 2010Acta Oceanologica Sinica2010,29,4:5
6Fouling corrosion in aluminum heat exchangers显示文摘Fouling deposits on aluminum heat exchanger reduce the heat transfer efficiency and cause corrosion to the apparatus.This study focuses on the corrosive behavior of aluminum coupons covered with a layer of artificial fouling in a humid atmosphere by their weight loss,Tafel plots,electrochemical impedance spectroscopy(EIS),and scanning electron microscope(SEM)observations.The results reveal that chloride is one of the major elements found in the fouling which damages the passive film and initiates corrosion.The galvanic corrosion between the metal and the adjacent carbon particles accelerates the corrosive process.Furthermore,the black carbon favors the moisture uptake,and gives the dissolved oxygen greater chance to migrate through the fouling layer and form a continuous diffusive path.The corrosion rate decreasing over time is conformed to electrochemistry measurements and can be verified by Faraday’s law.The EIS results indicate that the mechanism of corrosion can be interpreted by the pitting corrosion evolution mechanism,and that pitting was observed on the coupons by SEM after corrosive exposure.Su Jingxin Ma Minyu Wang Tianjing Guo Xiaomei Hou Liguo Wang Zhiping 2015Chinese Journal of Aeronautics2015,28,3:5
7Anti-cavitation performance of a splitter-bladed inducer under different flow rates and different inlet pressures显示文摘The anti-cavitation performance of a high-speed centrifugal pump with a splitter-bladed inducer is investigated under different flow rates and different inlet pressures. Simulations and external characteristics experiments are carried out. Static pressure and the vapor volume fraction distributions on the inducer and the impeller of the pump under various operation conditions are obtained. The results show that the cavitation developments on the impeller and on the inducer with the flow rates are reverse, while the development of the inlet pressure on the inducer and the impeller is the same. Cavitation on the impeller increases with the increase of flow rates, and it extends to the near passages with rotating, while cavitation on the inducer is more complex than that on the impeller. Cavitation at the inlet of the inducer decreases with the increase of flow rates, while cavitation at the outlet of the inducer is opposite. The results also show that cavitation development on the impeller and on the inducer with the inlet pressure is the same. Cavitation both decreases with the increase of the inlet pressure at the same flow rate. Furthermore, asymmetric cavitation on the impeller and on the inducer is both observed. 8Suspended moxibustion at Tianshu (ST25) inhibits prokineticin 1 and prokineticin receptor 1 expression in the spinal cord of rats with chronic visceral hypersensitivity显示文摘Suspended moxibustion can decrease the expression of prokineticin 1 and its receptor in colonic tissue from rats modeling chronic visceral hyperalgesia. This study aimed to verify if rat spinal cord prokineticin 1 and its receptor contribute to the analgesic effect of suspended moxibustion in a rat model of irritable bowel syndrome where rats display chronic visceral hypersensitivity. Results showed that suspended moxibustion at Tianshu (ST25) point significantly decreased visceral sensitivity to colorectal distention in a chronic visceral hyperalgesia rat model; also protein and mRNA expression of prokineticin 1 and prokineticin receptor 1 in the spinal cord of rats was significantly decreased. 12Duplication and transcriptional divergence of three Kunitz protease inhibitor genes that modulate insect and pathogen defenses in tea plant(Camellia sinensis)显示文摘Kunitz protease inhibitors(KPIs)are ubiquitous in plants and act as crucial compounds in defense responses against insect attack and pathogen infection.However,the influence of gene duplication on the postdivergence of the CsKPI genes involved in biotic stresses in tea plant is not well known.Here,we identified three CsKPI genes from tea plant(Camellia sinensis)and characterized their expression and evolutionary patterns among plant species.We found that CsKPI1,CsKPI2,and CsKPI3 diverged from their common ancestor 72.94 million years ago(MYA),and the tandem duplication of CsKPI2 and CsKPI3 occurred 26.78 MYA.An in vitro protein assay showed that the three CsKPI proteins were functional and inhibited the production of p-nitroanilide(PNA)from an artificial substrate.The three CsKPI-GFP fusion proteins localized to the cytoplasm.We showed that salicylic acid(SA)and transcripts of CsKPI2 and CsKPI3 significantly accumulated after infection with Glomerella cingulata.The application of exogenous SA stimulated the high expression of both CsKPI2 and CsKPI3 by activating cis-elements within their promoters.Under Ectropis oblique attack,CsKPI1 expression and jasmonic acid(JA)levels were more abundant in both insect-damaged leaf tissues and undamaged neighboring leaves.The application of jasmonic acid methyl ester elicited high expression levels of CsKPI1,suggesting that CsKPI1 accumulation requires JA production in tea plant.The overall findings suggest that the transcriptional divergence of KPI genes after duplication led to the specialized role of CsKPI1 in the physiological response to insect stress;the functional conservation between CsKPI2 and CsKPI3 confers resistance to pathogen infection in tea plant.Junyan Zhu Yaxian He Xiaomei Yan Lu Liu Rui Guo Xiaobo Xia Daojie Cheng Xiaozeng Mi Lidiia Samarina Shenrui Liu Enhua Xia Chaoling Wei 2019Horticulture Research2019,6,1:2
