6A general strategy to quantify analytes through fluorescence chromaticity and luminosity显示文摘To realize a fast,easy-operation and precise way using fluorescence probes to quantify analytes is a goal to facilitate detection,especially in situ.Herein,we are reporting an approach which can be generally employed for the differentiation and quantitation of analytes through fluorescence chromaticity and luminosity.Seven representative fluorescent probes,targeting p H,cysteine,hydrogen sulfide,hydrogen peroxide,palladium and hydrazine,were synthesized and tested.Without utilizing costly instrumentations,portable devices were employed to collect data of photographs from the fluorescence samples in responses to different analytes.Subsequently,the photographic images were digitally processed to generate calibration curves between chromaticity/luminosity verse concentrations after mapping to the CIE 1931 xy Y standard color space.Good linear calibration curves and quantitative analysis of unknown samples with low errors through the spectral technology demonstrated the reliability of this method.Thus,we showed the analytical method with a simple and on-site constructible/portable device which is promising for applications in more fluorescence probes.Tianyu Liang Peiwei Yang Tianhong Wu Menghan Shi Xiayu Xu Taotao Qiang Xiaolong Sun 2020Chinese Chemical Letters2020,31,11:0

8Using iTRAQ-Based Quantitative Proteomics Analysis to Identify Differentially Expressed Proteins Related to Larval Development of Portunus trituberculatus显示文摘The swimming crab Portunus trituberculatus is an economically important marine crustacean species.Here isobaric tags for relative and absolute quantitation(iTRAQ)analysis were used to identify proteins that are differentially expressed during larval development of P.trituberculatus to elucidate the underlying mechanisms.In comparison with the first zoea stage(Z1),3980 proteins were identified from 32789 peptides,which were matched with 115497 spectrums.A total of 241 proteins were screened with significantly differential expressions in all development stages.These 241 proteins are involved in various biological processes,such as cytoskeleton organization,protein synthesis,energy production and substance metabolism,physiological activities,and transport.Cluster analyses of the 241 differentially expressed proteins led to the generation of four protein clusters based on the overall similarity in protein expression patterns.Exactly 54,70,36,and 45 proteins clustered in profiles 10(0,0,0,−1,0,0),15(0,1,0,1,0,1),18(0,1,2,2,1,0),and 19(0,1,2,3,4,5),respectively.Muscle development and exoskeleton renewal were important processes throughout the development stages.In addition,protein synthesis,degradation,and digestion actively occurred,especially at the Z4 stage.These results provide novel insights into the mechanisms underlying larval development of swimming crab and can assist in larval rearing.CHEN Xiayue MU Changkao LI Ronghua YE Yangfang SONG Weiwei SHI Ce LIU Lei WANG Huan WANG Chunlin 2021Journal of Ocean University of China2021,20,4:0
9Lipopolysaccharide-induced podocyte injury is regulated by calcineurin/NFAT and TLR4/MyD88/NF-κB signaling pathways through angiopoietin-like protein 4显示文摘Podocyte injury is an important cause of proteinuria.Angiopoietin-like protein 4(Angptl4)is a secreted glycoprotein and has a role in proteinuria.However,the exact role of Angptl4 in podocyte injury and its upstream regulators has not been clarified.In this study,we used lipopolysaccharide(LPS)-induced mice and cultured podocytes as podocyte injury models.Our results indicated that LPS increased the expression of podocyte Angptl4 in vivo and in vitro.Furthermore,we showed that Angptl4 overexpression deteriorated LPS-induced podocyte injury by inducing podocyte cytoskeleton rearrangement,reducing the expression of synaptopodin while Angptl4 knockdown alleviated LPS-induced podocyte injury.In addition,we found that inhibitors and siRNA targeting TLR4/MyD88/NF-κB signaling inhibited the upregulation of Angptl4 in LPS-induced podocytes.Moreover,inhibitors and siRNA targeting calcineurin/NFAT signaling also relieved LPS-induced Angptl4 expression and podocyte injury in vivo and in vitro.Taken together,our study has elucidated that both of the TLR4/MyD88/NF-κB and calcineurin/NFAT signaling mediate the upregulation of Angptl4 in LPS-induced podocytes,which has important implications for further understanding the molecular mechanism of LPS-induced podocyte injury.Xiujin Shen Chunhua Weng Yucheng Wang Cuili Wang Shi Feng Xiayu Li Heng Li Hong Jiang Haibing Wang Jianghua Chen 2022Genes & Diseases2022,9,2:0
10Novel seven-membered ring-fused naphthalimide derivatives with potentials for cancer theranostics显示文摘Naphthalimide derivatives have good planarity and large conjugated structure and therefore possess photophysical properties and biological activities. Previously, our group discovered seven-membered heterocyclic derivatives via modifying 4-and 5-positions of naphthalimide skeleton and found the derivatives had good water solubility and showed large stokes shift and strong fluorescence in water. In this article, we designed and synthesized more seven-membered ring-fused naphthalimide derivatives(Y1-Y16)by introducing different substitutions on the imide group. Among them, Y1, Y5, Y9 were found to show similar cytotoxic activities with Amonafide against A549 and HL60 cells, with IC50values at 10^(-6)mol/L.What is more, the asymmetry derivatives(Y1 and Y5) showed high fluorescent quantum yields in the aqueous phase(Ф = 0.47).
