| 1 | The evidence for the existence of methane seepages in the northern South China Sea:abnormal high methane concentrations in bottom waters显示文摘The methane concentration of water samples at five stations collected by the CTD rosette water sampler in the areas of southwest Dongsha Islands and the Xisha Trough was analyzed by the gas-stripping method on aboard ship. It shows abnormal high methane concentrations in near bottom water samples at three stations. In the southwest Dongsha Islands area, the methane concentration of 4.25 and 10.64 nmol/dm3 occurs in near bottom water samples at Stas E105A and E106, respectively. In the Xisha Trough area, the high methane concentrations of 5.17, 8.48 and 8.70 nmol/dm3 in water depths of 1 750, 1 900 and 2 050 m, respectively, have been observed at Sta. E413. It is believed that the abnormal high methane concentrations are generated from the leakage of methane from sediments. Combining with previous geophysical and geochemical data from these two areas, this was probably related to the submarine gas hydrates decomposition and cold seep system. In May 2007, gas hydrate samples were successfully obtained by the drilling in the Shenhu Sea area located in the southwest Dongsha Islands area. | 2 | Analysis and external validation of a nomogram to predict peritoneal dissemination in gastric cancer显示文摘Objective:Peritoneal dissemination is difficult to diagnose by conventional imaging technologies.We aimed to construct a nomogram to predict peritoneal dissemination in gastric cancer(GC)patients.Methods:We retrospectively analyzed 1,112 GC patients in Sun Yat-sen University Cancer Center between2001 and 2010 as the development set and 474 patients from The Sixth Affiliated Hospital,Sun Yat-sen University between 2010 and 2016 as the validation set.The clinicopathological variables associated with gastric cancer with peritoneal dissemination(GCPD)were analyzed.We used logistic regression analysis to identify independent risk factors for peritoneal dissemination.Then,we constructed a nomogram for the prediction of GCPD and defined its predictive value with a receiver operating characteristic(ROC)curve.External validation was performed to validate the applicability of the nomogram.Results:In total,250 patients were histologically identified as having peritoneal dissemination.Logistic regression analysis demonstrated that age,sex,tumor location,tumor size,signet-ring cell carcinoma(SRCC),T stage,N stage and Borrmann classification IV(Borrmann IV)were independent risk factors for peritoneal dissemination.We constructed a nomogram consisting of these eight factors to predict GCPD and found an optimistic predictive capability,with a C-index of 0.791,an area under the curve(AUC)of 0.791,and a 95%confidence interval(95%CI)of 0.762-0.820.The results found in the external validation set were also promising.Conclusions:We constructed a highly sensitive nomogram that can assist clinicians in the early diagnosis of GCPD and serve as a reference for optimizing clinical management strategies. | Xijie Chen Shi Chen Xinyou Wang Runcong Nie Dongwen Chen Jun Xiang Yijia Lin Yingbo Chen Junsheng Peng | 2020 | Chinese Journal of Cancer Research2020,32,2: | 6 |
| 3 | Source and spatial distributions of particulate organic carbon and its isotope in surface waters of Prydz Bay, Antarctica, during summer显示文摘In this study, we investigated the distributions of sea-surface suspended particulate organic carbon(POC) and its stable isotope(δ13CPOC) in Prydz Bay, Antarctica, and examined the factors infl uencing their distribution, sources, and transport. We used measurements collected from 61 stations in Prydz Bay during the 29 th Chinese National Antarctic Research Expedition, in combination with remote sensing data on sea surface temperature(SST), chlorophyll a concentration, and sea ice coverage. The POC concentration in the surface waters of Prydz Bay was 0.28–0.84 mg·L-1, with an average concentration of 0.48 mg·L-1. The δ13CPOC value ranged from-29.68‰ to-26.30‰, with an average of-28.01‰. The concentration of suspended POC was highest in near-shore areas and in western Prydz Bay. The POC concentration was correlated with chlorophyll a concentration and sea ice coverage, suggesting that POC was associated with phytoplankton production in local water columns, while the growth of phytoplankton was obviously affected by sea ice coverage. The δ13CPOC value in suspended particles decreased gradually towards the outer waters of Prydz Bay, while in eastern Prydz Bay the δ13CPOC value become gradually more negative from nearshore to deep-water areas, suggesting that δ13CPOC was mainly infl uenced by CO2 fixation by phytoplankton. | 4 | Denitrification in Qi'ao Island coastal zone,the Zhujiang Estuary in China显示文摘Samples of sediments and the overlying water were collected in the Qi'ao Island coastal zone,the Zhujiang(Pearl River) Estuary(ZE).Denitrification rates,sediment oxygen demand(SOD),and fluxes of inorganic nitrogen compounds were investigated with N2 flux method,using a self-designed continuous flow through and auto-sampling system.The results indicate that the denitrification rates varied between 222 and 908 μmol/(m2·h) with an average of 499 μmol /(m2·h).During incubation,the sediments absorbed dissolved oxygen in the overlying water with SOD ranging from 300 to 2 363 μmol/(m2·h).The denitrification rates were highly correlated with the SOD(r2=0.77) regardless of the NO-3+NO-2 concentrations in the overlying water,organic carbon contents in sediments and water temperature,suggesting that the SOD was probably the main environmental factor controlling the denitrification in the Qi'ao Island coastal zone.There was a net flux of NO-3+NO-2 into the sediments from the overlying water.The NH+4 flux from sediments into water as the result of mineralization was between 12.3 and 210.3 μmol/(m2·h),which seems limited by both organic carbon content in sediment and dissolved oxygen concentration in the overlying water. | WANG Hu ZHOU Huaiyang PENG Xiaotong YANG Qunhui QIN Chaomei YIN Xijie CHEN Guangqian | 2009 | Acta Oceanologica Sinica2009,28,1: | 4 |
| 5 | Growth model of a hydrothermal low-temperature Si-rich chimney:Example from the CDE hydrothermal field,Lau Basin显示文摘The CDE hydrothermal field was first discovered during a Chinese cruise to the East Lau Basin Spreading Centre in 2007.Apart from significant amounts of loose Fe-Si-Mn(oxyhydr) oxide(referred to as oxide below) precipitates,a small Si-rich oxide chimney was also recovered on this cruise.In this study,we report on the mineralogical and geochemical analyses of this chimney and a model for its growth that has been developed.Based on the mineralogy and O isotope results,the chimney walls can be divided into four growth generations(layers) from the inner to the outer layers:amorphous opal and barite layer(precipitation temperature 68.5°C based on oxygen isotope determinations),a rod-like amorphous layer(precipitation temperature 39.6°C),a filamentous Fe-Si oxide layer,and an outer Fe-Mn oxide layer.Investigations based on SEM and EDS showed that neutrophilic Fe-oxidizing bacteria play an important role in the formation of this chimney,particularly in the outer two generations.In the first stage,the metabolic activity of the microbes results in the pervasive precipitation of the filamentous Fe-rich oxides inside a ring formed by some amorphous opal and barite;therefore,a loose porous layer forms.In the second stage,amorphous opal then precipitates inside this wall as a result of conductive cooling and gradually controls the mixing between the hydrothermal fluids and ambient seawaters.In the third stage,barite and some amorphous opal form from the higher temperature fluids at the summit of the chimney growth history.In the last stage,the chimney wall becomes thicker and denser and the exchange of hydrothermal fluids and seawater ceases.As a result,a Fe-Mn oxide layer precipitates onto the outer surface of the chimney wall as neutrophilic Fe-oxidizing bacteria reoccupy the surface of the chimney.This mineral sequence and the resultant growth generations are confirmed by the chemical characteristics of the chimney wall.Sr isotopes extracted from the Fe oxides of the four-generation wall generally show a decreasing trend of the 87 Sr/86 Sr ratios from the second layer to the inner layer(from 0.707008 to 0.705877) except for the outer layer(0.706502).The Sr isotope and chondrite normalized REE patterns of the corresponding bulk samples from the chimney wall also display a similar trend.Our study shows that the biogenic filament network plays a key role in the formation of the chimney in contrast to previous growth models of higher temperature chimneys,which often ignore the influence of biogenic factors. | SUN ZhiLei ZHOU HuaiYang YANG QunHui YIN XiJie WANG Hu YAO HuiQiang DONG CongFang | 2012 | Science China Earth Sciences2012,55,10: | 4 |

| 9 | Enhanced but hypofunctional osteoclastogenesis in an autosomal dominant osteopetrosis type II case carrying a c.1856C>T mutation in CLCN7显示文摘Type II autosomal dominant osteopetrosis(ADO2), which is the most common form of osteopetrosis, is caused by heterozygous mutations in the chloride channel 7(CLCN7) gene. The osteopetrosis of ADO2 has been attributed to hypofunctional osteoclasts. The mechanism underlying the abnormality in osteoclast function remains largely unknown. This study was designed to investigate gene mutations and osteoclast function in a case that was clinically diagnosed as ADO2. Genomic DNA was extracted from blood samples of this patient, and the 25 exons of CLCN7 were amplified. Peripheral blood from the ADO2 subject and a healthy age- and sex-matched control was used to evaluate osteoclastogenesis, osteoclast morphology, and bone resorption. Analysis of DNA from the patient showed a germline heterozygous missense mutation,c.1856C>T(p.P619L), in exon 20 of CLCN7. A similar homozygous mutation at this site was previously reported in a patient with autosomal recessive osteopetrosis. When cultured, the peripheral blood mononuclear cells(PBMCs) from the ADO2 patient spontaneously differentiated into mature osteoclasts in vitro. The ADO2 patient's PBMCs formed enhanced, but heterogeneous, osteoclasts in both the presence and absence of macrophage-colony stimulating factor, and nuclear factor-?B ligand. Bone resorption was reduced in the ADO2 patient's osteoclasts, which exhibited aberrant morphology and abnormal distribution of integrin avβ3. Gene analysis found increased c-fos expression and reduced Rho A and integrin beta 3expression in ADO2 cells. | 11 | SY-1530, a highly selective BTK inhibitor, effectively treats B-cell malignancies by blocking B-cell activation显示文摘Objective:B-cell antigen receptor(BCR)signaling is required to maintain the physiological functions of normal B cells and plays an important pathogenic role in B-cell malignancies.Bruton tyrosine kinase(BTK),a critical mediator of BCR signaling,is an attractive target for the treatment of B-cell malignancies.This study aimed to identify a highly potent and selective BTK inhibitor.Methods:Homogeneous time-resolved fluorescence assays were used to screen BTK inhibitors.Typhoon fluorescence imaging and Western blot analysis were used to confirm the effects of SY-1530 on the BCR signaling pathway.Additionally,the anti-tumor activities of SY-1530 were evaluated in TMD8 xenografts and spontaneous canine B-cell lymphoma.Results:We found a novel irreversible and non-competitive inhibitor of BTK,SY-1530,which provided dose-dependent and timedependent inhibition.SY-1530 selectively bound to BTK rather than inducible T-cell kinase;consequently,it did not significantly affect T-cell receptor signaling and caused limited off-target effects.SY-1530 blocked the BCR signaling pathway through downregulation of BTK activity,thus leading to impaired phosphorylation of BTK and its downstream kinases.Moreover,SY-1530 induced apoptosis in a caspase-dependent manner and efficaciously inhibited tumor growth in mouse xenograft models of B-cell malignancy(P<0.001).SY-1530 also induced positive clinical responses in spontaneous canine B-cell lymphoma.Conclusions:SY-1530 is an irreversible and selective BTK inhibitor that shows inhibitory effects on B-cell malignancies by blocking the BCR signaling pathway.Therefore,it may be a promising therapeutic approach for the treatment of B-cell malignancies. | Liao Wang Yinghui Sun Xijie Liu Hongjuan Li Chang Lu Ronghui Yang Chuanzhen Yang Binghui Li | 2022 | Cancer Biology & Medicine2022,19,7: | 0 |

| 13 | Non-Ischemic, Non-Hypoxic Myocardial Injury, and Long-Term Mortality in Patients with Coronavirus Disease 2019: A Retrospective Cohort Study显示文摘Objective:Cardiac damage is commonly reported in patients with coronavirus disease 2019(COVID-19)but its prevalence and impact on the long-term survival of patients remain uncertain.This study aimed to explore the prevalence of myocardial injury and assess its prognostic value in patients with COVID-19.Methods:A single-center,retrospective cohort study was performed at the Affiliated Hospital of Jianghan University.Data from 766 patients with confirmed COVID-19 who were hospitalized from December 27,2019 to April 25,2020 were collected.Demographic,clinical,laboratory,electrocardiogram,treatment data and all-cause mortality during follow-up were collected and analyzed.Results:Of the 766 patients with moderate to critically ill COVID-19,86(11.2%)died after a mean follow-up of 72.8 days.Myocardial injury occurred in 94(12.3%)patients.The mortality rate was 64.9%(61/94)and 3.7%(25/672)in patients with and without myocardial injury,respectively.Cox regression showed that myocardial injury was an independent risk factor for mortality(hazard ratio:8.76,95%confidence interval:4.76–16.11,P<0.001).Of the 90 patients with myocardial injury with electrocardiogram results,sinus tachycardia was present in 29,bundle branch block in 26,low voltage in 10,and abnormal T-wave in 53.Conclusions:COVID-19 not only involves pneumonia but also cardiac damage.Myocardial injury is a common complication and an independent risk factor for mortality in COVID-19 patients. | Fajiu Li Xijie Zhu Ziyang Zhu Yinjian Yang Zhuang Tian Duolao Wang Shi Chen Xiaoyan Gao Yalin Xu Bo Zhang Wei Yu Min Liu Xiqi Xu Chenghong Li Shuyang Zhang | 2022 | Cardiology Discovery2022,2,2: | 0 |
| 14 | Trophic gauntlet effects on fisheries recovery:a case study in Sansha Bay,China显示文摘Low biological productivity causes ecosystem energy deficiency.Sansha Bay is an important spawning and nursery ground for migratory fish in the East China Sea(ECS).There is significant mariculture in this area,but stock enhancement programs and seasonal fishery closures have failed to recover populations of the commercially important species,Larimichthys crocea.We want to figure out the potential reason of unsuccessful L.crocea resource in recent 30 years.In this study,the trophic status of L.crocea,the food source proportions of L.crocea and zooplankton,and the food web structure and functioning of Sansha Bay was analyzed.A high nutrient low chlorophyll phenomenon was observed:this prevented harmful algal blooms,and phytoplankton growth was restrained by intensive macroalgal culture,resulting in a low abundance of zooplankton in Sansha Bay.Phytoplankton was the most important food source of zooplankton,and zooplankton was the greatest food source of juvenile L.crocea.Analyzed L.crocea suffered from starvation.Crucially,most of the phytoplankton was not used efficiently in the Sansha Bay ecosystem.This study suggests that trophic bottleneck,caused by food limitation,is a potential reason for unsuccessful fishery recovery in enclosed bay with macroalgal culture. | Bin Xie Cheng Huang Ya Wang Xijie Zhou Guogan Peng Yancheng Tao Jiaju Huang Xiaoqing Lin Lingfeng Huang | 2021 | Ecosystem Health and Sustainability2021,7,1: | 0 |
| 15 | Animal biodistribution, safety and validation study of dopamine transporter PET imaging agent ^(18)F-FECNT显示文摘This work was to investigate the pharmacologic characteristics of 18F-FECNT (2β-carbomethoxy-3β- (4-chlorophenyl)-8-(2-[18F]fluoroethyl)nortropane) as a dopamine transporter (DAT) PET imaging agent. Its partition coefficients were determined in n-octanol and phosphate buffer (PB) (pH 7.0 and pH 7.4). 6-Hydroxydopamine (6-OHDA) left-sided lesioned Parkinsonian rats were established and validated by rotational behavior tests. Biodistribution in vivo in mice, autoradiography in normal and hemi-Parkinsonian rat brains, and toxicity test were performed. The results showed that partition coefficients were 34.14 (pH 7.0) and 56.41 (pH 7.4), respectively. Biodistribution exhibited rapid uptake and favorable retention in the mice brains. The major radioactivity was metabolized by the hepatic system. The autoradiography showed that 18F-FECNT was highly concentrated in striatum, and that the left and the right striatal uptake were symmetrical in normal SD rat brains. In left-sided lesioned PD rat brains, the striatal uptake of 18F-FECNT bilaterally decreased in comparison with normal rats. No significant uptake was visible in the 6-OHDA lesioned-sided striatal areas. The results demonstrated that 18F-FECNT binds to DAT was specific. Toxicity trial displayed that the acceptable dose per kilogram to mice was 625 times greater than that to human.
