1Identification of genetic alterations associated with primary resistance to EGFR-TKIs in advanced non-small-cell lung cancer patients with EGFR sensitive mutations显示文摘Background:Identification of activated epidermal growth factor receptor(EGFR)mutations and application of EGFR-tyrosine kinase inhibitors(EGFR-TKIs)have greatly changed the therapeutic strategies of non-small-cell lung cancer(NSCLC).However,the long-term efficacy of EGFR-TKI therapy is limited due to the development of drug resistance.The aim of this study was to investigate the correlation between the aberrant alterations of 8 driver genes and the primary resistance to EGFR-TKIs in advanced NSCLC patients with activated EGFR mutations.Methods:We retrospectively reviewed the clinical data from 416 patients with stage III/IV or recurrent NSCLC who received an initial EGFR-TKI treatment,from April 2004 and March 2011,at the Sun Yat-sen University Cancer Center.Several genetic alterations associated with the efficacy of EGFR-TKIs,including the alterations in BIM,ALK,KRAS,PIK3CA,PTEN,MET,IGF1R,and ROS1,were detected by the routine clinical technologies.The progression-free survival(PFS)and overall survival(OS)were compared between different groups using Kaplan-Meier survival analysis with the log-rank test.A Cox regression model was used to estimate multivariable-adjusted hazard ratios(HRs)and their 95%confi-dence intervals(95%CIs)associated with the PFS and OS.Results:Among the investigated patients,169 NSCLC patients harbored EGFR-sensitive mutations.EGFR-mutant patients having PTEN deletion had a shorter PFS and OS than those with intact PTEN(P=0.003 for PFS,and P=0.034 for OS).In the combined molecular analysis of EGFR signaling pathway and resistance genes,we found that EGFR-mutant patients coexisted with aberrant alterations in EGFR signaling pathway and those having resistant genes had a statistically poorer PFS than those without such alterations(P<0.001).A Cox proportional regression model determined that PTEN deletion(HR=4.29,95%CI=1.72-10.70)and low PTEN expression(HR=1.96,95%CI=1.22-3.13),MET FISH+(HR=2.83,95%CI=1.37-5.86)were independent predictors for PFS in patients with EGFR-TKI treatment after adjustment for multiple factor.Conclusions:We determined that the coexistence of genetic alterations in cancer genes may explain primary resistance to EGFR-TKIs.Fang Wang Xia-Yao Diao Xiao Zhang Qiong Shao Yan-Fen Feng Xin An Hai-Yun Wang 2019Cancer Communications2019,39,1:10
2Paleomagnetic results of Late Paleozoic rocks from northern Qiangtang Block in Qinghai-Tibet Plateau, China显示文摘Tectonic evolution of the Tethys and the boundary between the Gondwanaland and the Eurasia during the Carboniferous and Permian remain hotly debated. Qiangtang region in the Qinghai-Tibet Plateau may be a key place to study these problems. A paleomagnetic study was conducted on the Late Paleozoic rocks in the northern Qiangtang region (33.7°N, 86.7°W), Tibet. Two sites (21 samples) in the Upper Carboniferous, eleven sites (101 samples) in the Permian, and two sites (16 samples) in the Lower Triassic were investigated. The rock magnetic data revealed hematite and magnetite as the main magnetic carriers. In stepwise thermal demagnetization and/or combined alternating field (AC) demagnetization, two characteristic components in the majority of the samples were identified as (1) the Low-temperature Component (LTC), characterized by northerly decli- nation and moderate to steep inclination, corresponding to a pole position overlay with the present North Pole. A minority of the samples present single component, and their directions are the same as (2) the High-temperature Component (HTC) of double components. The combined single-component and HTC data of the Permian can pass the R-test at 95% level and the F-test at 99% level, as well as the BC-test. The pole position from the Late Carboniferous is at 31.8°S, 45.7°E with dp=2.1, dm=3.9, that from the Early and Middle (Late) Permian is at 31.7°S, 46.8°E with dp=9.2, dm=16.9 (34.4°N, 54.1°E with dp=6.9, dm=12.5) respectively, and that from the Early Triassic is at 16.9°S, 22.5°E with dp=4.9, dm=9.2. 3Paleomagnetic data from the Late Carboniferous-Late Permian rocks in eastern Tibet and their implications for tectonic evolution of the northern Qiangtang-Qamdo block显示文摘We report paleomagnetic results from the Late Carboniferous-Late Permian strata in eastern Tibet (China), and aim to clarify the tectonic and paleogeographic evolution of the northern Qiangtang-Qamdo block, which is the key to the study of plate boundary between the Gondwanaland and the Eurasia during the late Paleozoic. Two hundred and nineteen samples-including limestone, muddy siltstone, basalt, lava, and tuff-were collected at 24 sites in the Upper Carboniferous and Middle-Upper Permian successions. A systematic study of rock magnetism and paleomagnetism yields three reliable paleomagnetic pole positions. Both hematite and magnetite occurred in the Late Carboniferous limestone samples. The demagnetization curve shows a characteristic double-component, with the remanent magnetization (ChRM) exhibiting a positive polarity (negative inclination). In the Late Permian limestone, tuff, and basalt, magnetic information were recorded primarily in magnetite, although a small fraction of them was found in hematite in basalt. The demagnetization curve illustrates a double or single component, with the ChRM showing a negative polarity (positive inclination), which has passed the classic fold test successfully. The single polarity features of the ChRM directions of the Late Carboniferous and Middle-Late Permian rocks are respectively related to the Kiaman positive and reversed polarities under the stratigraphic coordinates. This, in turn, indicates that both ChRMs directions represent the original remanence directions. By comparison with the previously published paleomagnetic results from the late Paleozoic rocks in the northern Qiangtang Range, we suggest that: (1) Qamdo and northern Qiangtang block were independent of each other during the Late Carboniferous to the Early Permian periods. The north Lancangjiang ocean basin between the two blocks may have closed before the Middle Permian and been involved in the continent-continent collision stage in the Late Permian-Early Triassic periods. 4Activation and coreceptor expression of T lymphocytes induced by highly active antiretroviral therapy in Chinese HIV/AIDS patients显示文摘Background At the end of 2005, 650 000 people lived with human immunodeficiency virus type-1 (HIV-1) in China, of whom 75 000 were AIDS patients. Many AIDS patients received highly active antiretroviral therapy (HAART) supported by the “China CARES” program but the immune responses of HAART were seldom reported. This study investigated the effect of HAART on the activation and coreceptor expression of T lymphocytes in Chinese HIV/AIDS patients and evaluated its effect on immune reconstitution. Methods Seventeen HIV/AIDS patients were enrolled and three-color-flow cytometry was used to detect the activation of HLA-DR CD38 and the coreceptor CCR5, CXCR4 expression on T lymphocytes in whole blood samples taken from the patients before and after 3- or 6-month HAART. Results The activation percents of CD4+, CD8+ T lymphocytes were significantly higher before therapy than the normal controls (HLA-DR/CD4: 40.47±18.85 vs 11.54±4.10; CD38/CD4: 81.34±10.86 vs 53.34±11.44; HLA-DR/CD8: 63.94±12.71 vs 25.67±9.18; CD38/CD8: 86.56±11.41 vs 58.84±6.16, all P<0.01). After 6-month combined antiretroviral treatment, the activation of T lymphocytes in HIV/AIDS patients was significantly decreased (HLA-DR/CD4: 28.31±13.48; CD38/CD4: 69.88±12.64; HLA-DR/CD8: 46.56±18.64; CD38/CD8: 70.17±14.54, all P<0.01 compared with the pre-treatment values). Before the treatment, CCR5 expression on CD8+ T lymphocytes was up-regulated while CXCR4 expression on CD8+ T lymphocytes downregulated in HIV/AIDS patients compared with the normal controls (CD8/CCR5: 70.91±10.03 vs 52.70± 7.68; CD8/CXCR4: 24.14±11.08 vs 50.05±11.68, all P<0.01). After 6-month HAART, CCR5 expression on CD8+ T lymphocytes significantly decreased (56.35±12.96, P<0.01), while CXCR4 expression on CD8+ T lymphocytes increased (36.95±9.96, P<0.05) compared with the pre-treatment and the normal controls. A significant statistical relationship was observed between the expression of activation markers, CCR5 and the CD4+ T lymphocyte counts after HAART (P<0.05). Conclusions Reduced activation of T lymphocytes and a normalization of coreceptor expression were observed in Chinese HIV/AIDS patients after HAART. 5Protective effect of nitric oxide on hepatopulmonary syndrome from ischemia-reperfusion injury显示文摘AIM:To evaluate immunological protection of nitric oxide(NO) in hepatopulmonary syndrome and probable mechanisms of ischemia-reperfusion(IR) injury in rat liver transplantation.METHODS:Sixty-six healthy male Wistar rats were randomly divided into three groups(11 donor/recipi-ent pairs).In group Ⅱ,organ preservation solution was lactated Ringer's solution with heparin 10 000/L at 4 ℃.In groups Ⅰ and Ⅲ,the preservation solution added,respectively,L-arginine or N G-L-arginine methyl ester(L-NAME)(1 mmol/L) based on group Ⅱ,and recipients were injected with L-arginine or L-NAME(50 mg/kg) in the anhepatic phase.Grafted livers in each group were stored for 6 h and implanted into recipients.Five rats were used for observation of postoperative survival in each group.The other six rats in each group were used to obtain tissue samples,and executed at 3 h and 24 h after transplantation.The levels of alanine aminotransferase(ALT),tumor necrosis factor(TNF)-and NO metabolites(NOx) were detected,and expression of NO synthase,TNF-and intercellular adhesion molecule 1(ICAM-1) was examined by triphosphopyridine nucleotide diaphorase histochemical and immunohistochemical staining.RESULTS:By supplementing L-arginine to strengthen the NO pathway,a high survival rate was achieved and hepatic function was improved.One-week survival rate of grafted liver recipients in group Ⅰ was significantly increased(28.8 ± 36.6 d vs 4 ± 1.7 d,P < 0.01) as compared with groups Ⅱ and Ⅲ.Serum levels of ALT in group Ⅰ were 2-7 times less than those in groups Ⅱ and Ⅲ(P < 0.01).The cyclic guanosine monophosphate(cGMP) levels in liver tissue and NOx in group Ⅰ were 3-4 times higher than those of group Ⅱ after 3 h and 24 h reperfusion,while in group Ⅲ,they were significantly reduced as compared with those in group Ⅱ(P < 0.01).The levels of TNF-in group Ⅰ were significantly lower than in group Ⅱ after 3 h and 24 h reperfusion(P < 0.01),while being significantly higher in group Ⅲ than group Ⅱ(P < 0.01).Histopathology revealed more severe tissue damage in graft liver and lung tissues,and a more severe inflammatory response of the recipient after using NO synthase inhibitor,while the pathological damage to grafted liver and the recipient's lung tissues was signifi-cantly reduced in group Ⅰ after 3 h and 24 h reperfusion.A small amount of constitutive NO synthase(cNOS) was expressed in liver endothelial cells after 6 h cold storage,but there was no expression of inducible NO synthase(iNOS).Expression of cNOS was particularly significant in vascular endothelial cells and liver cells at 3 h and 24 h after reperfusion in group Ⅱ,but expression of iNOS and ICAM-1 was low in group Ⅰ.There was diffuse strong expression of ICAM-1 and TNF-in group Ⅲ at 3 h after reperfusion.CONCLUSION:The NO/cGMP pathway may be critical in successful organ transplantation,especially in treating hepatopulmonary syndrome during cold IR injury in rat orthotopic liver transplantation.Tong-Jin Diao, Xin Chen Li-Hua Deng, Han-Xiang ChenI Yan Liang Xiao-Dong Zhao Qing-Hua Wang, Wei-Sheng Yuan Bai-Chun Gao Yong Ye 2012World Journal of Gastroenterology2012,18,25:7

8Foxtail millet supplementation improves glucose metabolism and gut microbiota in rats with high-fat diet/streptozotocin-induced diabetes显示文摘Foxtail millet(FM)whole grain has received special attention in recent years.To confirm the hypoglycemic effects of FM,we investigated the effects of FM supplementation on glucose metabolism and gut microbiota in rats with high-fat diet/streptozotocin(HFD/STZ)-induced diabetes.Specifically,we fully assessed the blood biochemical profiles,pancreatic histopathology,insulin-glucagon immunofluorescence,short-chain fatty acids,and gut microbiota composition of rats with HFD/STZ-induced diabetes before and after FM supplementation.Results showed that both 30% and 48% FM supplementation significantly decreased concentrations of fasting blood glucose,60-min blood glucose,and blood triglycerides(P<0.05);additionally,48% FM supplementation significantly improved blood glucose tolerance and insulin resistance(P<0.05).However,FM supplementation could not effectively repair damage to β-cells over a short period of time.In addition,4 weeks of 48% FM supplementation siginificantly increased the relative abundance of Bifidobacterium and concentration of butyrate,suggesting that the hypoglycemic effects of FM supplementation might be partially mediated by gut microbiota.Collectively,we found a dose-dependent relationship between FM supplementation and improvement of blood glucose metabolism,but did not find a synergistic effect between FM supplementation and metformin(Met)treatment.Our findings provide further support that consuming more whole-grain FM might be beneficial to individuals suffering from type 2 diabetes.Xin Ren Linxuan Wang Zenglong Chen Min Zhang Dianzhi Hou Yong Xue Xianmin Diao Ruihai Liu Qun Shen 2022Food Science and Human Wellness2022,11,1:3
9Different self-assembly behaviors of mono-modified β-cyclodextrin substituted by benzoic acid derivatives显示文摘The mono-modified β-cyclodextrin derivative,6-O-(4-aminobenzoyl)-β-cyclodextrin(1) ,was synthesized and characterized. We compared its self-assembling behavior with structurally related β-cyclodextrin derivative,6-O-(4-hydroxybenzoyl)-βcyclodextrin(2) ,in aqueous solution and in the solid state using NMR spectroscopy and single crystal X-ray structure analysis. The two complexes displayed different self-assembling behaviors.Complex 1 formed a head-to-tail helical columnar superstructure in which the substituent group penetrated deeply into the hydrophobic cavity of the adjacent β-cyclodextrin from the second side and aligned along the screw axis,whereas 2 exhibited a channel superstructure with a tail-to-tail dimer as the repeating motif.Further NMR studies in aqueous solution supported the result of the solid state.The influence of substituent groups on the self-assembly behavior was examined in mono-modified β-cyclodextrins by means of substituting an amino group for a hydroxyl group,which indicates that the weak intermolecular interaction is a critical factor in determining the self-assembling behavior.Zhi Fan MinJie Guo Bin Dong ChunHua Diao ZuoLiang Jing Xin Chen 2010Science China Chemistry2010,53,5:2
10Bioinformatics analysis of structure and function in the MRP gene family and its expression in response to various drugs in Bursaphelenchus xylophilus显示文摘Genes homologous to members of the MRP gene family in Caenorhabditis elegans are important in drug resistance.To further explore the molecular mechanism of drug resistance in pine wood nematode(Bursaphelenchus xylophilus),we used bioinformatics approaches to analyze genomic data for B.xylophilus and identified Bx-MRP genes.We predicted the structure and function of the genes and encoded proteins.Using bioinformatics programs to predict and analyze various properties of the predicted proteins,including hydrophobicity,transmembrane regions,phosphorylation sites,and topologically isomeric structures,of these Bx-MRP genes,we determined that they function in transmembrane transport.From the results of RT-qPCR,the Bx-MRP family members confer significant differential resistance to different drug treatments.After treatment with different concentrations of emamectin benzoate,avermectin and matrine,the expression of each gene increased with increasing drug concentrations,indicating that the family members play a positive role in the regulation of multidrug resistance.Jian Diao Xin Hao Wei Ma Ling Ma 2021Journal of Forestry Research2021,32,2:2

12Effects of Transition Metal Oxides on Thermal Conductivity of Mould Fluxes显示文摘The thermal conductivity of the mould fluxes containing transition metal oxides was measured by hotline method at different temperatures.The relationship between the thermal conductivity of mold fluxes and the contents of transition metal oxides was discussed.The synthetic slags were composed of 30.0%-35.4%CaO,34.7%-38.6%SiO2,6%Al2O3,9%Na2O,14.4%CaF2,0-4%Cr2O3and 0-8%MnO in mass percent.The results indicated that Cr2O3and MnO had a negative effect on thermal conductivity of mold fluxes.The thermal conductivity of mold fluxes was about 0.25-0.55W/(m·K)when the temperature reached 1 300℃,and it increased sharply to about 1.32-1.99W/(m·K)when the temperature reduced from 1 300to 1 000℃.The thermal conductivity of mold fluxes containing Cr2O3and MnO was 10%-25%lower than those of original fluxes.The decrease in thermal conductivity was attributed to the change of molecular structure of mold fluxes.In addition,the poor integrity and regulation of polycrystal structure,complexity of crystal structure,and effects of impurities in the boundary and lattice distortion leaded to the reduction in the thermal conductivity.Na2CrO4,Mn2SiO4and other minor phases were also found in the samples containing Cr2O3and MnO,respectively.QIU Xin XIE Bing QING Xue-mei DIAO Jiang HUANG Qing-yun WANG Shui-bo 2013Journal of Iron and Steel Research(International)2013,20,11:2
13Arabidopsis AIP1-1 regulates the organization of apical actin filaments by promoting their turnover in pollen tubes显示文摘Apical actin filaments are highly dynamic structures that are crucial for rapid pollen tube growth, but the mechanisms regulating their dynamics and spatial organization remain incompletely understood. We here identify that AtAIP1-1 is important for regulating the turnover and organization of apical actin filaments in pollen tubes. AtAIP1-1 is distributed uniformly in the pollen tube and loss of function of AtAIP1-1 affects the organization of the actin cytoskeleton in the pollen tube. Specifically, actin filaments became disorganized within the apical region of aip1-1 pollen tubes. Consistent with the role of apical actin filaments in spatially restricting vesicles in pollen tubes, the apical region occupied by vesicles becomes enlarged in aip1-1 pollen tubes compared to WT. Using ADF1 as a representative actin-depolymerizing factor, we demonstrate that AtAIP1-1 enhances ADF1-mediated actin depolymerization and filament severing in vitro, although AtAIP1-1 alone does not have an obvious effect on actin assembly and disassembly. The dynamics of apical actin filaments are reduced in aip1-1 pollen tubes compared to WT.
