| 1 | Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 2 | Characterization and Fine Mapping of a Novel Rice Narrow Leaf Mutant nal9显示文摘A narrow leaf mutant was isolated from transgenic rice(Oryza sativa L.)lines carrying a T-DNA insertion.The mutant is characterized by narrow leaves during its whole growth period,and was named nal9(narrow leaf 9).The mutant also has other phenotypes,such as light green leaves at the seedling stage,reduced plant height,a small panicle and increased tillering.Genetic analysis revealed that the mutation is controlled by a single recessive gene.A hygromycin resistance assay showed that the mutation was not caused by T-DNA insertion,so a map-based cloning strategy was employed to isolate the nal9 gene.The mutant individuals from the F2generations of a cross between the nal9 mutant and Longtepu were used for mapping.With 24 F2mutants,the nal9 gene was preliminarily mapped near the marker RM156 on the chromosome 3.New INDEL markers were then designed based on the sequence differences between japonica and indica at the region near RM156.The nal9 gene was fnally located in a 69.3 kb region between the markers V239B and V239G within BAC OJ1212_C05 by chromosome walking.Sequence and expression analysis showed that an ATP-dependent Clp protease proteolytic subunit gene(ClpP)was most likely to be the nal9 gene.Furthermore,the nal9 mutation was rescued by transformation of the ClpP cDNA driven by the 35S promoter.Accordingly,the ClpP gene was identifed as the NAL9 gene.Our results provide a basis for functional studies of NAL9 in future work. | Wei Li Chao Wu Guocheng Hu Li Xing Wenjing Qian Huamin Si Zongxiu Sun Xingchun Wang Yaping Fu Wenzhen Liu | 2013 | Journal of Integrative Plant Biology2013,55,11: | 8 |
| 3 | Metallogenic model and prognosis of the Shuiyindong super-large strata-bound Carlin-type gold deposit, southwestern Guizhou Province, China显示文摘The Shuiyindong deposit is one of the largest (more than 100 tonnes of Au) and highest grade (more than 7×10-6-10×10-6), strata-bound Carlintype gold deposits in southwestern Guizhou Province, China. The deposit is controlled by both structure and favorable lithology. It is situated near the axis of the striking Huijiabao anticline and is hosted in bioclastic limestone of the Permian Longtan Formation. Gold mineralization occurred under low temperature with Th of 220℃± and is closely associated with decarbonation, silicification, sulfidation and dolomitization. The deposit has a characteristic elemental assemblage of Au-As-Hg-Tl. Studies of geochemistry and isotope compositions indicated that the ore-bearing materials and fluids of the gold deposit mainly originated from a plutonic source, and possess a mixing feature with the strata matter during transportation from mantle to crust. Fluid inclusions in vein quartz from the gold deposit are rich in volatile flux, indicating that metallogenic fluid is an overpres-sured one. The activity and geothermal state of the Earth's crust in the long period of time are favorable for the formation of overpressured fluids in a large area, and extensive structures would drive the fluids into ore-forming sys-tem and make gold deposits formed. The complexity of structural movement in the upper crust of southwestern Guizhou Province resulted in complicated gold mineralization. | 5 | Algorithm for Calculating Torque Base in Vehicle Traction Control System显示文摘Existing research on the traction control system(TCS) mainly focuses on control methods, such as the PID control, fuzzy logic control, etc, aiming at achieving an ideal slip rate of the drive wheel over long control periods. The initial output of the TCS (referred to as the torque base in this paper), which has a great impact on the driving performance of the vehicle in early cycles, remains to be investigated. In order to improve the control performance of the TCS in the first several cycles, an algorithm is proposed to determine the torque base. First, torque bases are calculated by two different methods, one based on states judgment and the other based on the vehicle dynamics. The confidence level of the torque base calculated based on the vehicle dynamics is also obtained. The final torque base is then determined based on the two torque bases and the confidence level. Hardware-in-the-loop(HIL) simulation and vehicle tests emulating sudden start on low friction roads have been conducted to verify the proposed algorithm. The control performance of a PID-controlled TCS with and without the proposed torque base algorithm is compared, showing that the proposed algorithm improves the performance of the TCS over the first several cycles and enhances about 5% vehicle speed by contrast. | 10 | Spatial and temporal dynamics of microbial community composition and factors influencing the surface water and sediments of urban rivers显示文摘The temporal and spatial characteristics of urban river bacterial communities help us understand the feedback mechanism of bacteria to changes in the aquatic environment.The Fuhe River plays an important role in determining the water ecological environment of Baiyangdian Lake.16S rRNA gene sequencing was used to study the microbial distribution characteristics in the Fuhe River in different seasons.The results showed that some environmental factors of the surface water(ammonia nitrogen(NH_(3)^(-)N),total nitrogen(TN),and total phosphorus(TP))were different on the spatial and temporal scales.Moreover,there were no seasonal differences in the contents of TN,TP,total organic carbon(TOC),or heavy metals in the sediments.The distributions of Cyanobacteria,Actinomycetes and Firmicutes in the water and Actinomycetes and Planctomycetes in the sediments differed significantly among seasons(P<0.05).There were significant spatial differences in bacteria in the surface water,with the highest abundance of Proteobacteria recorded in the river along with the highest nutrient concentration,while the abundance of Bacteroidetes was higher in the upstream than the downstream.Microbial communities in the water weremost sensitive to temperature(T)and the TP concentration(P<0.01).Moreover,differences in the bacterial community were better explained by the content of heavy metals in the sediments than by the chemical characteristics.A PICRUStmetabolic inference analysis showed that the effect of high summer temperatures on the enzyme action led to an increase in the abundances of the metabolic-related genes of the river microorganisms. | Hongjie Wang Xingchun Liu Yali Wang Shengqi Zhang Guangming Zhang Yangyang Han Mengxiang Li Ling Liu | 2023 | Journal of Environmental Sciences2023,,2: | 2 |

| 18 | Germanium/silica ratio and trace element composition of Early Cambrian siliceous rocks in Keping:implications for the siliceous rocks'formation and paleoenvironment interpretations显示文摘This study used the germanium/silica(Ge/Si)ratios,together with rare earth elements and other trace elements to infer the siliceous source and sedimentary environment of the siliceous rocks located at the bottom of Yuertusi Formation in Northwestern Tarim Basin,Keping,China.Previous studies have shown that this siliceous rock stratum formed at the edge of the carbonate platform on the continental shelf.Researchers suggest that these siliceous rocks were formed by hydrothermal activity,but some still draw different conclusions.Understanding the silicon source and depositional environment of these siliceous rocks would help us learn the processes of environmental changes and the causes of biological explosions during this period.The value of germanium/silica ratios of these siliceous rocks is from 0.15 to 0.37 pmol/mol and much lower than above 10|imol/mol values in siliceous rocks that are known formed by hydrothermal activity.All samples are rich in HREE,which differ from hydrothermal siliceous rocks that are rich in LREE.Most samples lack hydrothermal related elements.All these features show that the source of these siliceous rocks'siliceous is not hydrothermal flu ids.The samples'Ce/Ce*range from 0.88 to 1,and Th/U ratios range from 0.01 to 0.36.These features suggest these siliceous rocks were formed in an anoxic environment.Considering all the evidence,we conclude that the siliceous rock stratum at the bottom of the Lower Cambrian Yuertusi Formation in northwest Tarim Basin.Keping,was formed in anoxic seawater at the edge of the carbonate platform on the continental shelf.Its silicon source is seawater instead of hydrothermal fluid. | Zixuan Guan Shibiao Deng Peixian Liu Yiqiu Jin Xingchun Cao | 2020 | Acta Geochimica2020,39,6: | 0 |
