| 1 | Soil surface roughness change and its effect on runoff and erosion on the Loess Plateau of China显示文摘As an important parameter in the soil erosion model, soil surface roughness(SSR) is used to quantitatively describe the micro-relief on agricultural land. SSR has been extensively studied both experimentally and theoretically; however, no studies have focused on understanding SSR on the Loess Plateau of China. This study investigated changes in SSR for three different tillage practices on the Loess Plateau of China and the effects of SSR on runoff and erosion yield during simulated rainfall. The tillage practices used were zero tillage(ZT), shallow hoeing(SH) and contour ploughing(CP). Two rainfall intensities were applied, and three stages of water erosion processes(splash erosion(I), sheet erosion(II) and rill erosion(III)) were analyzed for each rainfall intensity. The chain method was used to measure changes in SSR both initially and after each stage of rainfall. A splash board was used to measure the splash erosion at stage I. Runoff and sediment data were collected continuously at 2-min intervals during rainfall erosion stages II and III. We found that SSR of the tilled surfaces ranged from 1.0% to 21.9% under the three tillage practices, and the order of the initial SSR for the three treatments was ZT0.49) for the ZT, SH and CP treatments. The runoff initiation time was longer in the SH and CP treatments than in the ZT treatment. There were no significant differences in the total runoff yields among the ZT, SH and CP treatments. | 4 | A stringent dual control system overseeing transcription and activity of the Cre recombinase for the liver-specific conditional gene knock-out mouse model显示文摘Liver cancer is one of the most threatening diseases in Chinese population. Just like in other tissues, tumor initiation and development in liver involve multiple steps of genetic and epigenetic alterations with several unknown details. However, unlike in other tissues, a tis-sue specific inducible Cre recombinase system that allows temporal and spatial deletion of a target DNA fragment is still not available for in vivo functional gene annotation in hepatocytes. In our pursuit to establish such a mouse model, we designed a dual inducible Cre transgene system and tested it in cultured cells. | 8 | Effect of bacterial intra-species community interactions on the production and activity of volatile organic compounds显示文摘Microorganisms experience intra-and inter-species interactions in the soil,and how these interactions affect the production of microbial volatile organic compounds(VOCs)is still not well-known.Here we evaluated the production and activity of microbial VOCs as driven by bacterial intra-species community interactions.We set up bacterial communities of increasing biodiversity out of 1–4 strains each of the Gram-positive Bacillus and Gram-negative Pseudomonas genera.We evaluated the ability of each community to provide two VOCmediated services,pathogen suppression and plant-growth promotion and then correlated these services to the production of VOCs by each community.The results showed that an increase in community richness from 1 to 4 strains of both genera increased VOC-mediated pathogen suppression and plant-growth promotion on agar medium and in the soil,which was positively correlated with the production of pathogen suppressing and plant growth-promoting VOCs.Pseudomonas strains maintained while Bacillus strains reduced community productivity with an increase in community richness and produced eight novel VOCs compared with the monocultures.These results revealed that intra-species interactions may vary between Gram-negative and Gram-positive species but improved VOC-mediated functioning with respect to pathogen suppression and plant-growth promotion by affecting the amount and diversity of produced VOCs potentially affecting plant disease outcomes. | Jianing Wang Xinlan Mei Zhong Wei Waseem Raza Qirong Shen | 2021 | Soil Ecology Letters2021,3,1: | 1 |

| 18 | Knockout of ISCA 1 causes early embryonic death in rats显示文摘Background : Iron-sulfur cluster assembly 1( ISCA 1) is an iron-sulfur(Fe/S) carrier protein that accepts Fe/S from a scaffold protein and transfers it to target proteins including the mitochondrial Fe/S containing proteins. ISCA 1 is also the newly identified causal gene for multiple mitochondrial dysfunctions syndrome( MMDS). However, our knowledge about the physiological function of ISCA 1 in vivo is currently limited. In this study, we generated an ISCA 1 knockout rat line and analyzed the embryo development. Methods : ISCA 1 knockout rats were generated by replacing the exon1 of ISCA 1 gene with the mC herry-Cre fusion gene using CRISPR-Cas9 technology. The ISCA 1 expression pattern was analyzed by fluorescence imaging using ISCA 1 promotor driven Cre and mC herry expression. The embryonic morphology was examinated by microscope and mitochondrial proteins were tested by Western blot. Results : An ISCA 1 knockout rat line was obtained, which expressed mC herry-Cre fusion protein. Both of the fluorescence images from mC herry and Cre induced mC herry in a reporter rat strain, showing that ISCA 1 expressed in most of the tissues in rats. The ISCA 1 knockout resulted in abnormal development at 8.5 days, with a significant decrease of NDUFA 9 protein and an increase of aconitase 2( ACO 2) in rat embryos. Conclusion : Deletion of ISCA 1 induced early death in rats.
