| 1 | Laser-assisted grinding of RB-SiC composites:Laser ablation behavior and mechanism显示文摘Laser ablation is an important process during Laser-Assisted Grinding(LAG)of hard and brittle materials.To realize controllable material removal during laser ablation of RB-SiC composites,ablation experiments under different Laser Energy Density(LAED)and LAG experiments are conducted.Evolution rules and mechanism of physical phase,ablation morphology and crack characteristics caused by laser irradiation are investigated.The forces of LAG and Conventional Grinding(CG)are compared.The results show that ablation surface changes from slight oxidation to obvious material removal with LAED increasing,and ablation depth increases gradually.The ablation products change from submicron SiO_(2)particles to nanoscale particles and floccule.High LAED promotes SiC decomposition and sublimation,which leads to the increase of C element.The SiC phase forms corrugated shape in recast layer and columnar shape in Heat Affected Zone(HAZ)at 56 J/mm^(2).The cold and heat cycle leads to formation of fishbone crack.For ablation specimen under 30 J/mm^(2),the grinding force can be reduced by a maximum of 39%and brittle damage region is reduced.The material removal and microcrack generated will significantly reduce the hardness and improve machinability,which can promote grinding efficiency. | Zhigang DONG Wenchao SUN Xintong CAI Feng YANG Yan BAO Renke KANG Dongjiang WU Guangyi MA Fangyong NIU | 2024 | Chinese Journal of Aeronautics2024,37,1: | 0 |
| 2 | Discovery and druggability evaluation of pyrrolamide-type GyrB/ParE inhibitor against drug-resistant bacterial infection显示文摘The bacterial ATP-competitive GyrB/ParE subunits of type II topoisomerase are important anti-bacterial targets to treat super drug-resistant bacterial infections.Herein we discovered novel pyrrolamide-type GyrB/ParE inhibitors based on the structural modifications of the candidate AzD5099 that was withdrawn from the clinical trials due to safety liabilities such as mitochondrial toxicity.The hydroxyisopropyl pyridazine compound 28 had a significant inhibitory effect on Gyrase(GyrB,IC_(50)=49 nmol/L)and a modest inhibitory effect on TopoⅣ(ParE,IC_(50)=1.513μmol/L)of Staphylococcus aureus.It also had significant antibacterial activities on susceptible and resistant Gram-positive bacteria with a minimum inhibitory concentration(MIC)of less than 0.03μg/mL,which showed a time-dependent bactericidal effect and low frequencies of spontaneous resistance against S.aureus.Compound 28 had better protective effects than the positive control drugs such as DS-2969(5)and AZD5099(6)in mouse models of sepsis induced by methicillin-resistant Staphylococcus aureus(MRSA)infection.It also showed better bactericidal activities than clinically used vancomycin in the mouse thigh MRSA infection models.Moreover,compound 28 has much lower mitochondrial toxicity than AZD5099(6)as well as excellent therapeutic indexes and pharmacokinetic properties.At present,compound 28 has been evaluated as a pre-clinical drug candidate for the treatment of drug-resistant Gram-positive bacterial infection.On the other hand,compound 28 also has good inhibitory activities against stubborn Gram-negative bacteria such as Escherichia coli(MIC=1μg/mL),which is comparable with the most potent pyrrolamide-type GyrB/ParE inhibitors reported recently.In addition,the structure-activity relationships of the compounds were also studied. | Xintong Zhao Jing Feng Jie Zhang Zunsheng Han Yuhua Hu Hui-Hui Shao Tianlei Li Jie Xia Kangfan Lei Weiping Wang Fangfang Lai Yuan Lin Bo Liu Kun Zhang Chi Zhang Qingyun Yang Xinyu Luo Hanyilan Zhang Chuang Li Wenxuan Zhang Song Wu | 2023 | Acta Pharmaceutica Sinica B2023,13,12: | 0 |
| 3 | Two-step model of paleohexaploidy, ancestral genome reshuffling and plasticity of heat shock response in Asteraceae显示文摘An ancient hexaploidization event in the most but not all Asteraceae plants,may have been responsible for shaping the genomes of many horticultural,ornamental,and medicinal plants that promoting the prosperity of the largest angiosperm family on the earth.However,the duplication process of this hexaploidy,as well as the genomic and phenotypic diversity of extant Asteraceae plants caused by paleogenome reorganization,are still poorly understood.We analyzed 11 genomes from 10 genera in Asteraceae,and redated the Asteraceae common hexaploidization(ACH)event∼70.7–78.6 million years ago(Mya)and the Asteroideae specific tetraploidization(AST)event∼41.6–46.2 Mya.Moreover,we identified the genomic homologies generated from the ACH,AST and speciation events,and constructed a multiple genome alignment framework for Asteraceae.Subsequently,we revealed biased fractionations between the paleopolyploidization produced subgenomes,suggesting the ACH and AST both are allopolyplodization events.Interestingly,the paleochromosome reshuffling traces provided clear evidence for the two-step duplications of ACH event in Asteraceae.Furthermore,we reconstructed ancestral Asteraceae karyotype(AAK)that has 9 paleochromosomes,and revealed a highly flexible reshuffling of Asteraceae paleogenome.Of specific significance,we explored the genetic diversity of Heat Shock Transcription Factors(Hsfs)associated with recursive whole-genome polyploidizations,gene duplications,and paleogenome reshuffling,and revealed that the expansion of Hsfs gene families enable heat shock plasticity during the genome evolution of Asteraceae.Our study provides insights on polyploidy and paleogenome remodeling for the successful establishment of Asteraceae,and is helpful for further communication and exploration of the diversification of plant families and phenotypes. | Xiangming Kong Yan Zhang Ziying Wang Shoutong Bao Yishan Feng Jiaqi Wang Zijian Yu Feng Long Zejia Xiao Yanan Hao Xintong Gao Yinfeng Li Yue Ding Jianyu Wang Tianyu Lei Chuanyuan Xu Jinpeng Wang | 2023 | Horticulture Research2023,10,6: | 0 |
| 4 | Long noncoding RNA GAS5 attenuates cigarette smoke-induced airway remodeling by regulating miR-217-5p/PTEN axis显示文摘Airway remodeling is a remarkable pathological characteristic of chronic obstructive pulmonary disease(COPD),and long noncoding RNAs have been demonstrated to participate in COPD development and pathogenesis.Here,we investigate the role of long noncoding RNA GAS5 in cigarette smoke(CS)-induced airway remodeling.GAS5 expression is significantly lower in lung tissues of CS-exposed mice than in tissues of control mice without exposure to CS.Forced GAS5 overexpression suppresses CS-induced airway inflammation and remodeling.GAS5 overexpression also inhibits CS extract-induced inflammatory-cytokine expression and fibroblast activation in vitro.Regarding the mechanism,GAS5 acts as a sponge of miR-217-5p,thereby increasing PTEN expression.MiR-217-5p overexpression and PTEN knockdown separately reverse the inhibitory effects of GAS5 overexpression on the inflammatory-cytokine expression and fibroblast activation.Collectively,these results suggest that GAS5 can suppress airway inflammation and fibroblast activation by regulating miR-217-5p/PTEN axis,which may help develop novel therapeutic strategies against COPD. | Yong Du Yi Ding Tianyun Shi Wei He Jingjing Feng Zhoufang Mei Xuru Chen Xintong Feng Xiaohua Zhang Zhijun Jie | 2022 | Acta Biochimica et Biophysica Sinica2022,54,7: | 0 |
| 5 | A Study of Rust Layer of Ultra-Thin Cast Strip Steel Containing 0.10%Sb in Simulated Industrial Atmosphere显示文摘A new type of steel containing Sb based on the CASTRIP process had been designed to improve the corrosion resistance of high-strength steels in industrial atmospheres.The properties of the electrochemistry and rust layer(RL)were analyzed to evaluate the corrosion resistance of the ultra-thin cast strip(UCS)steel containing 0.10%Sb.Experimental results showed that the corrosion resistance of UCS steel containing Sb was doubled compared to that of traditional hot rolling strip(HRS)steel.It could be interpreted as an integral effect of CASTRIP process and alloying elements.The optimization for microstructure and inclusion of UCS steel can reduce the localized corrosion effects in initial corrosion processes.Cu,P and Sb reduced the anodic reaction and CuFeO2/Cu2O,CuO,FePO4 and Sb2O3 were formed,promoting the transition fromγ-FeOOH toα-FeOOH.A(Cu,Sb,P)-enriched inner RL was generated and significantly improved protective ability of the RL. | Long Chen Xintong Lian Zhong Xi Tengshi Liu Qingxiao Feng Hualong Li Yixin Shi Han Dong | 2023 | Acta Metallurgica Sinica(English Letters)2023,36,8: | 0 |

| 7 | Ultrasound-responsive spherical nucleic acid against c-Myc/PD-L1 to enhance anti-tumoral macrophages in triple-negative breast cancer progression显示文摘Triple-negative breast cancer(TNBC)is the most challenging breast cancer subtype because of its aggressive behavior and limited therapeutic targets.c-Myc is hyperactivated in the majority of TNBC tissues,however,it has been considered an“undruggable”target due to its disordered structure.Herein,we developed an ultrasound-responsive spherical nucleic acid(SNA)against c-Myc and PD-L1 in TNBC.It is a self-assembled and carrier-free system composed of a hydrophilic small-interfering RNA(si RNA)shell and a hydrophobic core made of a peptide nucleic acid(PNA)-based antisense oligonucleotide(ASO)and a sonosensitizer.We accomplished significant enrichment in the tumor by enhanced permeability and retention(EPR)effect,the controllable release of effective elements by ultrasound activation,and the combination of targeted therapy,immunotherapy and physiotherapy.Our study demonstrated significant anti-tumoral effects in vitro and in vivo.Mass cytometry showed an invigorated tumor microenvironment(TME)characterized by a significant alteration in the composition of tumor-associated macrophages(TAM)and decreased proportion of PD-1-positive(PD-1+)T effector cells after appropriate treatment of the ultrasound-responsive SNA(USNA).Further experiments verified that tumor-conditioned macrophages residing in the TME were transformed into the anti-tumoral population.Our finding offers a novel therapeutic strategy against the“undruggable”c-Myc,develops a new targeted therapy for c-Myc/PD-L1 and provides a treatment option for the TNBC. | Runtian Wang Gaigai Li Fangyan Gao Feng Xu Xintong Li Jian Zhang Jinbo Li Xiaoxiang Guan | 2024 | Science China(Life Sciences)2024,67,4: | 0 |
