1Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 10Cytochrome P450 Monooxygenase Immobilization as a Model of Herbicide Metabolism in vitro显示文摘To investigate herbicide metabolism in vitro by cytochrome P450 with stable enzymatic activity, cytochrome P450 is immobilized in silk fibroin. The enzymatic activity of immobilized cytochrome P450 is maintained above 80% after repeated batch experiments for 10 times. Moreover, the enzymatic activity of immobilized cytochrome P450 is kept as relatively high as 73.8% after storage for two months at 4℃. In addition, immobilization can improve the temperature and pH stability of cytochrome P450. Immobilized cytochrome P450 has the similar affinity Km values for herbicide chlorsulfuron and triasulfuron as the free cytochrome P450. In the case of chlorosulfuron, affinity Km value is 53μmol/L for free cytochrome P450, and 63μmol/L for immobilized cytochrome P450, respectively. In the case of triasulfuron affinity, Km value is 36μmol/L for free cytochrome P450, and 44μmol/L for immobilized cytochrome P450, respectivily. 11Characteristics of intracellular polyphosphate granules and phosphorus-absorption of a marine polyphosphateaccumulating bacterium, Halomonas sp. YSR-3显示文摘Halomonas sp.YSR-3 was isolated from the Yellow Sea and identified as a polyphosphate-accumulating bacterium and the characteristics of its intracellular polyphosphate(polyP)granules and phosphorus absorption were studied.Most YSR-3 cells stored one or two polyP granules in regular appearance and high-density.The diameter of the granules was about 400 nm measuring by a transmission electron microscope(TEM).After stained with 4,6-diamidino-2-phenylindole(DAPI)and visualized by a fluorescence microscope,the cells turned blue and the granules were bright yellow.The composition of granules includes P(major ingredient),Mg,S,K,and Ca as detected by an energy dispersive X-ray spectrometer(EDS).When inorganic phosphorus(po34-)and ferric ion(Fe3+)were added into media,the biomass increased and the cells formed intracellular polyP granules owing to the phosphorus assimilation from media.The YSR-3 obtained higher biomass by adding 0.02 g/L FePO4 than 0.005 g/L and 0.01 g/L FePO4;however,the phosphorus absorption was higher with 0.01 g/L FePO4 than 0.005 g/L and 0.02 g/L FePO4.The optical density at wavelength 480 nm(OD480nm)was 0.79 and 100%cells could form intracellular polyP granules.These results show that strain YSR-3 is able to acquire higher biomass and absorb more inorganic phosphorus when 0.01 g/L FePO4 is added.The characteristics of absorbing and storing phosphorus as intracellular inorganic polyP granules have a potential for application in high-efficiency phosphorus removal in wastewater treatment.REN Shiying LI Xiangqian YIN Xiulian LUO Chuping LIU Fei 2020Journal of Oceanology and Limnology2020,38,1:0
