1The chromosome-level reference genome assembly for Panax notoginseng and insights into ginsenoside biosynthesis显示文摘Panax notoginseng,a perennial herb of the genus Panax in the family Araliaceae,has played an important role in clinical treatment in China for thousands of years because of its extensive pharmacological effects.Here,we report a high-quality reference genome of P.notoginseng,with a genome size up to 2.66 Gb and a contig N50 of 1.12 Mb,produced with third-generation PacBio sequencing technology.This is the first chromosome-level genome assembly for the genus Panax.Through genome evolution analysis,we explored phylogenetic and whole-genome duplication events and examined their impact on saponin biosynthesis.We performed a detailed transcriptional analysis of P.notoginseng and explored genelevel mechanisms that regulate the formation of characteristic tubercles.Next,we studied the biosynthesis and regulation of saponins at temporal and spatial levels.We combined multi-omics data to identify genes that encode key enzymes in the P.notoginseng terpenoid biosynthetic pathway.Finally,we identified five glycosyltransferase genes whose products catalyzed the formation of different ginsenosides in P.notoginseng.The genetic information obtained in this study provides a resource for further exploration of the growth characteristics,cultivation,breeding,and saponin biosynthesis of P.notoginseng.Zhouqian Jiang Lichan Tu Weifei Yang Yifeng Zhang Tianyuan Hu Baowei Ma Yun Lu Xiuming Cui Jie Gao Xiaoyi Wu Yuru Tong Jiawei Zhou Yadi Song Yuan Liu Nan Liu Luqi Huang Wei Gao 2021Plant Communications2021,2,1:11

4Identification of aeolian loess deposits on the Indo-Gangetic Plain(India)and their significance显示文摘Aeolian loess deposits contain abundant information about the evolution of the paleoenvironment.For example,paleoclimate changes recorded in Chinese loess area obtained significant achievement in the past few decades.Compared to Chinese loess,research on Indian loess is lacking.Currently,most studies focus on the Kashmir area located in the southern Himalayas,and studies on other areas are rare.However,field observations demonstrate that the sediments around the New Delhi-Agra-Jaipur Plain are similar to Chinese loess-paleosol sequences.For example,the boundary between two strata is transitional and without horizontal bedding.Moreover,obvious pedogenic horizons developed among sediment sequences,probably indicating unrecognised aeolian deposits in the Indo-Gangetic Plain(IGP).To confirm this,pilot samples were obtained from the IGP and detailed indoor measurements conducted.The results indicate that the distribution patterns of particle size and rare earth elements(REE) of the pilot samples are similar to Chinese loess.Furthermore,the scanning electronic microscopy(SEM) images of pilot samples show obvious conchoidal fractures,dash-shaped concavities,and abundant small pits that usually form through mechanical impact.These are typical characteristics of aeolian particles.In addition,environmental and rock magnetic measurements indicate that the dominant magnetic minerals in the pilot samples are magnetite and maghemite,and that they likely contain small amounts of hematite.Furthermore,conventional magnetic parameters are comparable with Chinese loess.Based on this,aeolian loess deposits are widely distributed in the IGP,which may have promoted the development of Indian farming and contributed towards the prosperity of ancient Indian civilisation.This study also provides a new and valuable record for the research on paleoclimate changes in the study area in the future.LIU XiuMing MA MingMing WU HaiBin ZHOU ZiBo 2017Science China Earth Sciences2017,60,3:2

8Electrodeposited synthesis of self-supported Ni-P cathode for efficient electrocatalytic hydrogen generation显示文摘One of the key challenges for electrochemical water splitting is the development of low-cost and efficient hydrogen evolution cathode. In this work, a self-supported Ni-P cathode was synthesized by a facile electrodeposition method. The composition and morphology were characterized by X-ray diffraction,X-ray photoelectron spectroscopy, scanning electron microscopy and transmission electron microscopy.The Ni-P cathode performed low onset over-potential, good catalytic activity and long-term stability under neutral and alkaline conditions. The mechanism of Ni-P electrode for hydrogen production was discussed by electrochemical impedance spectroscopy. 12Hyperosmolarity promotes macrophage pyroptosis by driving the glycolytic reprogramming of corneal epithelial cells in dry eye disease显示文摘Tear film hyperosmolarity plays a core role in the development of dry eye disease(DED)by mediating the disruption of ocular surface homeostasis and triggering inflammation in ocular surface epithelium.In this study,the mechanisms involving the hyperosmolar microenvironment,glycolysis mediating metabolic reprogramming,and pyroptosis were explored clinically,in vitro,and in vivo.Data from DED clinical samples indicated that the expression of glycolysis and pyroptosis-related genes,including PKM2 and GSDMD,was significantly upregulated and that the secretion of IL-1βsignificantly increased.In vitro,the indirect coculture of macrophages derived from THP-1 and human corneal epithelial cells(HCECs)was used to discuss the interaction among cells.The hyperosmolar environment was found to greatly induce HCECs’metabolic reprogramming,which may be the primary cause of the subsequent inflammation in macrophages upon the activation of the related gene and protein expression.2-Deoxy-d-glucose(2-DG)could inhibit the glycolysis of HCECs and subsequently suppress the pyroptosis of macrophages.In vivo,2-DG showed potential efficacy in relieving DED activity and could significantly reduce the overexpression of genes and proteins related to glycolysis and pyroptosis.In summary,our findings suggested that hyperosmolar-induced glycolytic reprogramming played an active role in promoting DED inflammation by mediating pyroptosis.Yu Han Yu Zhang Kelan Yuan Yaying Wu Xiuming Jin Xiaodan Huang 2023Frontiers of Medicine2023,17,4:0
13Identification of potential key genes in resveratrol biosynthesis via transcriptional analyses of berry development in grapevine(Vitis spp.)genotypes varying in trans-resveratrol content显示文摘Resveratrol is an important secondary metabolite not only owing to its function as a phytoalexin,but also its potential benefits to human health.In this study,the content of trans-resveratrol was documented in seven accessions of grapevine,in the seed,pulp and skin of berries,and at three developmental stages.The highest amount(2.99μg g^(−1) FW)was found in the skin of berries at the ripe stage from V.amurensis‘Tonghua-3'.Resveratrol was not detected in several samples,including skin of berries at the green hard or véraison stage from V.davidii‘Tangwei'.We carried out transcriptional profiling of developing‘Tonghua-3'and‘Tangwei'berries to identify gene expression patterns that may be linked with the difference in resveratrol content between these accessions.The expression levels of several differentially expressed genes(DEGs)with presumed function in resveratrol biosynthesis,including STILBENE SYNTHASEs(STSs),CINNAMATE 4-HYDROXYLASEs(C4Hs)and 4-COUMARATE-COA LIGASEs(4CLs),were significantly higher in‘Tonghua-3',than in'Tangwei'during the véraison and ripe stages.Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses suggested that these DEGs were enriched for multiple biological processes at the three stages of fruit development.Additionally,we identified a total of 36 transcription factors,including MYBs,WRKYs,ERFs,bHLHs and bZIPs,that were coexpressed with 17 STSs via a weighted gene co-expression network analysis,suggesting roles as regulators of resveratrol biosynthesis.Overall,these findings provide insight into genotypic differences in resveratrol biosynthesis in grapevine,as well as the molecular genetics of its regulation.Xiuming Zhang Yifei Wu Yichu Zhang Xiangjing Yin Steve van Nocker Junqiang Guo Zhi Li Min Gao Junyang Song Xiping Wang 2022Fruit Research2022,2,1:0
14Morphology and strain control of hierarchical cobalt oxide nanowire electrocatalysts via solvent effect显示文摘Designing highly efficient and low-cost electrocatalysts for oxygen evolution reaction is important for many renewable energy applications.In particular,strain engineering has been demonstrated as a powerful strategy to enhance the electrochemical performance of catalysts;however,the required complex catalyst preparation process restricts the implementation of strain engineering.Herein,we report a simple self-template method to prepare hierarchical porous Co_(3)O_(4)nanowires(PNWs)with tunable compressive strain via thermal-oxidation-transformation of easily prepared oxalic acid-cobalt nitrate(Co(NO_(3))_(2))composite nanowires.Based on the complementary theoretical and experimental studies,the selection of proper solvents in the catalyst preparation is not only vital for the hierarchical structural evolution of Co_(3)O_(4) but also for regulating their compressive surface strain.Because of the rich surface active sites and optimized electronic Co d band centers,the PNWs exhibit the excellent activity and stability for oxygen evolution reaction,delivering a low overpotential of 319 mV at 10 mA·cm^(−2)in 1 M KOH with a mass loading 0.553 mg·cm^(−2),which is even better than the noble metal catalyst of RuO_(2).This work provides a cost-effective example of porous Co_(3)O_(4)nanowire preparation as well as a promising method for modification of surface strain for the enhanced electrochemical performance.Xiuming Bu Xiongyi Liang Kingsley O.Egbo Zebiao Li You Meng Quan Quan Yang Yang Li Kin Man Yu Chi-Man Lawrence Wu Johnny C.Ho 2020Nano Research2020,13,11:0
