| 1 | Coronavirus MHV-A59 infects the lung and causes severe pneumonia in C57BL/6 mice显示文摘It remains challenging to develop animal models of lung infection and severe pneumonia by severe acute respiratory syndrome coronavirus(SARS-CoV) and Middle East respiratory syndrome cornavirus(MERS-Co V) without high level of containment. This inevitably hinders understanding of virushost interaction and development of appropriate countermeasures. Here we report that intranasal inoculation of sublethal doses of murine coronavirus mouse hepatitis virus A-59(MHV-A59), a hepatic and neuronal tropic coronavirus, can induce acute pneumonia and severe lung injuries in C57BL/6 mice. Inflammatory leukocyte infiltrations, hemorrhages and fibrosis of alveolar walls can be observed 2-11 days after MHV-A59 infection. This pathological manifestation is associated with dramatical elevation of tissue IP-10 and IFN-γ and moderate increase of TNF-α and IL-1β, but inability of anti-viral type I interferon response. | 2 | Dialdehyde starch nanoparticles: Preparation and application in drug carrier显示文摘Dialdehyde starch nanoparticles (DASNP) were prepared by the redox reaction of NaIO4 and starch in water-in-oil microemulsion. IR spectrum showed that DASNP had aldehyde groups, and quantitative alkali consumption showed that its dialdehyde content was about (50±5)%. The average diameter of DASNP determined by SEM was about 100 nm. TGA-DTA showed that its thermal stability was better than starch nanoparticle (StNP) and dialdehyde starch (DAS). Its low biological toxicity was detected by cell experiment. Also the best mass ratio of doxorubicin (DOX) to combined DASNP detected by UV-VIS was 15 : 1, and the product was effective for controlled release of DOX. The cell experiment showed that the drug-carrier particle (DOX-DASNP) can release DOX for a long time and strengthened the effect of the anticancer drug. | 3 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |

| 5 | From Symbols to Embeddings:A Tale of Two Representations in Computational Social Science显示文摘Computational Social Science(CSS),aiming at utilizing computational methods to address social science problems,is a recent emerging and fast-developing field.The study of CSS is data-driven and significantly benefits from the availability of online user-generated contents and social networks,which contain rich text and network data for investigation.However,these large-scale and multi-modal data also present researchers with a great challenge:how to represent data effectively to mine the meanings we want in CSS?To explore the answer,we give a thorough review of data representations in CSS for both text and network.Specifically,we summarize existing representations into two schemes,namely symbol-based and embeddingbased representations,and introduce a series of typical methods for each scheme.Afterwards,we present the applications of the above representations based on the investigation of more than 400 research articles from 6 top venues involved with CSS.From the statistics of these applications,we unearth the strength of each kind of representations and discover the tendency that embedding-based representations are emerging and obtaining increasing attention over the last decade.Finally,we discuss several key challenges and open issues for future directions.This survey aims to provide a deeper understanding and more advisable applications of data representations for CSS researchers. | Huimin Chen Cheng Yang Xuanming Zhang Zhiyuan Liu Maosong Sun Jianbin Jin | 2021 | Journal of Social Computing2021,2,2: | 4 |
| 6 | Ectopic expression of fungal EcGDH improves nitrogen assimilation and grain yield in rice显示文摘NADP(H)-dependent glutamate dehydrogenases(GDH) in lower organisms have stronger ammonium affinity than those in higher plants. Here we report that transgenic rice overexpressing the EcGDH from Eurotium cheralieri exhibited significantly enhanced aminating activities. | 7 | Distinct contribution of PD-L1 suppression by spatial expression of PD-L1 on tumor and non-tumor cells显示文摘Programmed cell death receptor 1(PD-1)and its ligand,PD-L1,are important immune checkpoint proteins.Although antibodies that block PD-1/PD-L1 have shown promising clinical efficacy in a subset of cancer patients,the detailed cellular and molecular mechanisms behind anti-PD-1 and anti-PD-L1 immunotherapy are not well defined.Specifically,the way in which PD-L1 contributes to immune suppression on tumor and non-tumor cells remains controversial.By selectively blocking PD-L1 on either tumor or non-tumor cells,we demonstrated that PD-L1 from both sources suppressed the anti-tumor T-cell response.Blocking PD-L1 on either tumor cells or non-tumor cells inhibited tumor growth and enhanced immune cell infiltration,as well as the tumor-specific T-cell response.Further,simultaneously blocking tumor-and non-tumor-derived PD-L1 maximized anti-tumor T-cell responses and demonstrated synergy.In addition,the relative contribution of PD-L1 on tumor and non-tumor cells to immune suppression depended on the PD-L1 expression level.Lastly,we found that the F4/80 receptor was involved in the anti-tumor effect of PD-L1 blockade.Taken together,our data indicate that PD-L1 on both tumor and non-tumor cells is critical for T-cell inhibition,which provides new directions for the optimization of PD-L1-blocking antibodies and the development of clinical biomarker strategies. | Xiaoqing Zhang Chen Cheng Jiyan Hou Xinyue Qi Xin Wang Ping Han Xuanming Yang | 2019 | Cellular & Molecular Immunology2019,16,4: | 3 |
| 8 | The osmolyte-producing endophyte Streptomyces albidoflavus OsiLf-2 induces drought and salt tolerance in rice via a multi-level mechanism显示文摘Drought and salinity are major environmental stresses that impair crop growth and productivity worldwide. Improving drought and salt tolerance of crops with microbial mutualists is an effective and environmentally sound strategy to meet the demands of the ever-growing world population. In the present study, we found that the Streptomyces albidoflavus Osi Lf-2, a moderately salt-tolerant endophytic actinomycete, produced abundant osmolytes, including proline, polysaccharides, and ectoine. Inoculation with Osi Lf-2 increased the osmotic-adjustment ability of the rice host by increasing the proline content(by250.3% and 49.4%) and soluble sugar(by 20.9% and 49.4%) in rice under drought and salt conditions, relative to the uninoculated control. Osi Lf-2 increased stress responses in the rice host at the physiological and biochemical levels(photosynthesis efficiency, osmolytes and antioxidant content), and the gene level(osmolytes synthesis, stress-responsive and ion-transport related genes), raising rice yields under both greenhouse and saline–alkaline soil conditions. | 10 | Tartary buckwheat(Fagopyrum tataricum(L.)Gaertn)protein-derived antioxidant peptides:mechanisms of action and structure-activity relationship in Caco-2 cell models显示文摘Excessive reactive oxygen species(ROS)can cause oxidative damage and lead to various metabolic disease.Tartary buckwheat(Fagopyrum tataricum(L.)Gaertn)is a new kind of protein-rich functional food,the protein in which has been proved to have good antioxidant capacity.In this study,in order to further explore the antioxidant mechanism of Tartary buckwheat protein,4 peptides(CR-8,LR-8,GK-10 and SR-12)were isolated and identified from it.H2 O2 was used to induce oxidative damage to Caco-2 cells to evaluate antioxidant capacity of these peptides.The results of superoxide dismutase(SOD),total antioxidant capacity(T-AOC)and mitochondrial membrane potential etc.showed that these peptides have superior antioxidant capacity.CR-8 has the best antioxidant capacity.In order to further clarify the antioxidant mechanism of CR-8,metabolomics was used to analyze related metabolites and metabolic pathways.The results showed that after CR-8 intervention,the content of metabolites such as L-acetyl carnitine has increased.This indicated that CR-8 can improve the antioxidant capacity of damaged cells by intervening in multiple metabolic pathways.This also revealed the anti-oxidant mechanism of tartary buckwheat protein.In conclusion,it provided a theoretical basis for further studying the activity of tartary buckwheat portein and utilizing buckwheat resources. | Yiming Zhou Xuanming She Zhidong Chen Yun Wei Ying Xiao Xiaoli Zhou | 2022 | Food Science and Human Wellness2022,11,6: | 2 |

However, how different internal and external environmental signals modulate the activity of PP2 CAs are not well known. The transmembrane kinase(TMK) protein4(TMK4), one member of a previously identified receptor kinase subfamily on the plasma membrane that plays vital roles in plant cell growth,directly interacts with PP2 CAs member(ABAInsensitive 2, ABI2). tmk4 mutant is hypersensitive to ABA in both ABA-inhibited seed germination and primary root growth, indicating that TMK4 is a negative regulator in ABA signaling pathway.Further analyses indicate that TMK4 phosphorylates ABI2 at three conserved Ser residues,thus enhancing the activity of ABI2. The phosphorylation-mimic ABI2~(S139DS140DS266D)can complement butnon-phosphorylated form ABI2~(S139AS140AS266) Acannot complement ABA hypersensitive phenotype of the loss-of-function mutant abi1-2 abi2-2.
