| 1 | Survival of transplanted neurotrophin-3 expressing human neural stem cells and motor function in a rat model of spinal cord injury显示文摘<正>BACKGROUND:Many methods have been attempted to repair nerves following spinal cord injury, including peripheral nerve transplantation,Schwann cell transplantation,olfactory ensheathing cell transplantation,and embryonic neural tissue transplantation.However,there is a need for improved outcomes. OBJECTIVE:To investigate the repair feasibility for rat spinal cord injury using human neural stem cells(hNSCs) genetically modified by lentivirus to express neurotrophin-3. DESIGN,TIME AND SETTING:In vitro cell biological experiment and in vivo randomized,controlled, genetic engineering experiment were performed at the Third Military Medical University of Chinese PLA and First People's Hospital of Yibin,China from March 2006 to December 2007. MATERIALS:A total of 64 adult,female,Wistar rats were used for the in vivo study.Of them,48 rats were used to establish models of spinal cord hemisection,and were subsequently equally and randomly assigned to model,genetically modified hNSC,and normal hNSC groups.The remaining 16 rats served as normal controls. METHODS:hNSCs were in vitro genetically modified by lentivirus to secrete both green fluorescence protein and neurotrophin-3.Neurotrophin-3 expression was measured by Western blot. Genetically modified hNSC or normal hNSC suspension(5×10~5) was injected into the rat spinal cord following T_(10) spinal cord hemisection.A total of 5μL Dulbecco's-modified Eagle's medium was infused into the rat spinal cord in the model group.Transgene expression and survival of transplanted hNSCs were determined by immunohistochemistry.Motor function was evaluated using the Basso,Beattie,and Bresnahan(BBB) scale. MAIN OUTCOME MEASURES:The following parameters were measured:expression of neurotrophin-3 produced by genetically modified hNSCs,transgene expression and survival of hNSCs in rats,motor function in rats. RESULTS:hNSCs were successfully genetically modified by lentivirus to stably express neurotrophin-3.The transplanted hNSCs primarily gathered at,or around,the injection site two weeks following transplantation,and gradually migrated towards the surrounding tissue. Transplanted hNSCs were observed 7.0-8.0 mm away from the injection site.In addition,hNSCs were observed 10 weeks after transplantation.At week 4,BBB locomotor scores were significantly greater in the genetically modified hNSC and normal hNSC groups,compared with the model group (P<0.05),and scores were significantly greater in the genetically modified hNSC group compared with the normal hNSC group(P<0.05). | 2 | Genomic Epidemiology of Carbapenemaseproducing Klebsiella pneumoniae in China显示文摘The rapid spread of carbapenemase-producing Klebsiella pneumoniae(cpKP)poses serious threats to public health;however,the underlying genetic basis for its dissemination is still unknown.We conducted a comprehensive genomic epidemiology analysis on 420 cpKP isolates collected from 70 hospitals in 24 provinces/autonomous regions/municipalities of China during 2009–2017 by short-/long-read sequencing.The results showed that most cpKP isolates were categorized into clonal group 258(CG258),in which ST11 was the dominant clone.Phylogenetic analysis revealed three major clades including the top one of Clade 3 for CG258 cpKP isolates.Additionally,carbapenemase gene analysis indicated that blaKPC was dominant in the cpKP isolates,and most blaKPC genes were located in five major incompatibility(Inc)groups of blaKPC-harboring plasmids.Importantly,three advantageous combinations of host–blaKPC-carrying plasmid(Clade 3.1+3.2–IncFIIpHN7A8,Clade 3.1+3.2–IncFIIpHN7A8:IncR,and Clade 3.3–IncFIIpHN7A8:IncpA1763-KPC)were identified to confer cpKP isolates the advantages in both genotypes(strong correlation/coevolution)and phenotypes(resistance/growth/competition)to facilitate the nationwide spread of ST11/CG258 cpKP.Intriguingly,Bayesian skyline analysis illustrated that the three advantageous combinations might be directly associated with the strong population expansion during 2007–2008 and subsequent maintenance of the population of ST11/CG258 cpKP after 2008.We then examined drug resistance profiles of these cpKP isolates and proposed combination treatment regimens for CG258/non-CG258 cpKP infections.Thus,the findings of our systematical analysis shed light on the molecular epidemiology and genetic basis for the dissemination of ST11/CG258 cpKP in China,and much emphasis should be given to the close monitoring of advantageous cpKP–plasmid combinations. | Cuidan Li Xiaoyuan Jiang Tingting Yang Yingjiao Ju Zhe Yin Liya Yue Guannan Ma Xuebing Wang Ying Jing Xinhua Luo Shuangshuang Li Xue Yang Fei Chen Dongsheng Zhou | 2022 | Genomics, Proteomics & Bioinformatics2022,20,6: | 1 |

| 5 | Electronic structure of LaIrIn5and f-electron character in its related Ce-115 compounds显示文摘LaIrIn5 is a reference compound of the heavy-fermion superconductor LaIrIn5.The lack of f electrons in LaIrIn5 indicates that there should not be any f electron participating in the construction of its Fermi surface.Thus the electronic structure comparison between LaIrIn5 and LaIrIn5 provides a good platform to study the properties of f electrons.Here angle-resolved photoemission spectroscopy(ARPES)measurements and density functional theory(DFT)calculations are performed to study the electronic structures of LaIrIn5 and LaIrIn5.We find the valence band structures of the two materials are similar to each other,except for the absence of f bands in LaIrIn5.By analyzing the Fermi crossings of the three conduction bands of the two materials quantitatively,we find the volumes of the electron pocketsαandβaround the M′point become larger from LaIrIn5 to LaIrIn5,while the hole pocketγaround theΓ′point becomes smaller.Together with the calculation results,we confirm that this is mainly originated from the f-electron contribution,while the lattice-constant difference between LaIrIn5 and LaIrIn5 only has a finite influence.We also give a summary of the f-electron character in its related Ce-115 heavy fermion compounds.Our results may be essential for the complete microscopic understanding of the 115 compounds and the related heavy-fermion systems. | Rui Zhou XueBing Luo ZhaoFeng Ding Lei Shu XingYu Ji ZiHao Zhu YaoBo Huang DaWei Shen ZhengTai Liu ZhongHao Liu Yun Zhang QiuYun Chen | 2020 | Science China(Physics,Mechanics & Astronomy)2020,63,11: | 0 |

| 8 | Electronic structure evolution accompanying heavy fermion formation in CeCu2Si2显示文摘The cooper pairs in the heavy-fermion superconductor CeCu2Si2 are formed of heavy fermions.Therefore,the heavy fermions are fundamental to the emergence of unconventional superconductivity and associated non-Fermi-liquid behavior in the normal state.The interplay between localization and itinerancy manifested on the electronic structure is key for understanding the heavyfermion behavior.Here,via the first-principle density functional theory(DFT)combined with single-site dynamical mean-field theory(DMFT),we investigate the temperature(T)evolution of the electronic structure of CeCu2Si2 in the normal state,focusing on the role of the 4f states in the low energy regime.Two characteristic temperature scales of this evolution,which accompanied the heavy-fermion formation,are established.The coherence onset temperature is around 130K,whereas the heavy-fermion band formation temperature is between 40 and 80K;both characteristic temperature scales are higher than the transport coherence temperature.Furthermore,the heavy-fermion formation is confirmed by calculating its effective mass variation with the temperature.Based on the calculated T-dependent evolution of the 4 f orbital occupancy and electronic structure,an explanation on the behavior of the temperature evolution of the correlation strength of CeCu2Si2 is provided.Our results offer a comprehensive microscopic picture of the heavy-fermion formation in CeCu2Si2,which is essential for further understanding the emergent superconducting pairing mechanism. | XueBing Luo Yun Zhang QiuYun Chen Qin Liu LiZhu Luo ShiYong Tan XieGang Zhu XinChun Lai | 2020 | Science China(Physics,Mechanics & Astronomy)2020,63,8: | 0 |

| 10 | Knowledge Mining of Low Specific Speed Centrifugal Pump Impeller Based on Proper Orthogonal Decomposition Method显示文摘To clarify the complex relation between the pump blade shape and its corresponding hydraulic performance,the knowledge mining method of centrifugal pump impeller based on proper orthogonal decomposition(POD)was proposed.The pump blade shape was parameterized by cubic Bezier curve.The Latin hypercube design method was employed to supply the necessary samples for producing the perturbations of blade wrap angle,and blade angle at inlet and outlet.The hydraulic efficiency and head were optimized by NSGA-II and RBF hybrid algorithm.The Pareto-optimal solutions were obtained.In order to further illustrate the relationship between the centrifugal pump blade shape and its hydraulic performance,the POD method was used to discover the effects of optimized blade shape to the flow solutions.For the optimization of centrifugal pump MH48-12.5,blade shape and relative velocity field in impeller from Pareto-optimal solutions were analyzed.The results demonstrate that larger blade angle and smaller wrap angle increase the average kinetic energy in impeller,resulting in higher pump head design.Smaller blade angle and larger wrap angle decrease the velocity gradient from the pressure side to suction side,resulting in smaller hydraulic loss and higher efficiency design. | ZHANG Renhui CHEN Xuebing LUO Jiaqi | 2021 | Journal of Thermal Science2021,30,3: | 0 |
