1Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10

6Carbon fluxes and soil carbon dynamics along a gradient of biogeomorphic succession in alpine wetlands of Tibetan Plateau显示文摘Hydrological changes under climate warming drive the biogeomorphic succession of wetlands and may trigger substantial carbon loss from the carbon-rich ecosystems.Although many studies have explored the responses of wetland carbon emissions to short-term hydrological change,it remains poorly understood how the carbon cycle evolves with hydrology-driven wetland succession.Here,we used a space-for-time approach across hydrological gradients on the Tibetan Plateau to examine the dynamics of ecosystem carbon fluxes(carbon dioxide(CO_(2))and methane(CH4))and soil organic carbon pools during alpine wetland succession.We found that the succession from mesic meadow to fen changed the seasonality of both CO_(2) and CH4 fluxes,which was related to the shift in plant community composition,enhanced regulation of soil hydrology and increasing contribution of spring-thaw emission.The paludification caused a switch from net uptake of gaseous carbon to net release on an annual timescale but produced a large accumulation of soil organic carbon.We attempted to attribute the paradox between evidence from the carbon fluxes and pools to the lateral carbon input and the systematic changes of historical climate,given that the wetlands are spatially low-lying with strong temporal climate-carbon cycle interactions.These findings demonstrate a systematic change in the carbon cycle with succession and suggest that biogeomorphic succession and lateral carbon flows are both important for understanding the long-term dynamics of wetland carbon footprints.Hao Wang Lingfei Yu Litong Chen Zhenhua Zhang Xuefei Li Naishen Liang Changhui Peng Jin-Sheng He 2023Fundamental Research2023,3,2:1
7Effects of laser powder bed fusion process parameters on microstructure and hydrogen embrittlement of high-entropy alloy显示文摘Hydrogen embrittlement behavior, micro-deformation, and crack propagation mechanism of CoCrFeNiMn high-entropy alloy (HEA) fabricated by laser powder bed fusion (LPBF) under different parameters were investigated by slow strain rate tensile tests (at room temperature) with/without electrochemical hydrogen pre-charging. The LPBF CoCrFeNiMn HEA shows excellent resistance to hydrogen embrittlement. Unsuitable LPBF parameters are accompanied by many microcracks and holes, resulting in a slight decrease in the hydrogen embrittlement resistance of the material. The electron backscatter diffraction (EBSD), electron channeling contrast image (ECCI) techniques, and transmission electron microscope (TEM) were carried out to research the main influencing factors of hydrogen on the deformation mechanism and crack propagation. Compared with un-charged samples, a larger number of deformation twins (DTs) appear in the deformation process of hydrogen-charged LPBF CoCrFeNiMn, attributing to the reduction of stacking fault energy (SFE) due to the ingress of hydrogen. The nano DTs and crossing twin system contribute to the extra work hardening, and a strain hardening platform is observed for all hydrogen-charged samples, resulting in the increase of strain hardening rate or the mitigation of the loss of strain hardening. 8Aptamer-Based Fluorescent Assay for Sensitive Detection of Cardiac Troponin Ⅰ显示文摘Acute myocardial infarction(AMI)is a major health problem leading to high rates of mortality and morbidity.Biomarker cardiac troponin I(cTnI)has shown high sensitivity and specificity towards AMI detection,and has been regarded as'gold standard'.An ultrasensitive method to detectcTnI with low concentration in human fluid is essential.In this paper,we developed an aptamer-based assay coupled with rolling circle amplification(RCA)and molecular beacon probe for sensitive detection ofcTnI.In this strategy,aptamer acts as a bridge to communicate between oligonucleotides andcTnI.RCA reaction produces a single-stranded tandem repeated copy of the circular template,which are recognized by fluorescence molecular beacon probe.With this strategy,highly sensitive and specific detection of cTnI was realized with the lowest detectable concentration of 7.24 pg/m L.The developed aptamer-RCA assay can be a promising tool in clinical samples analysis.The assay can also analyze other disease-related biomarkers by replacing the aptamer.Yu Li Yuanzhan Yang Xuefei Lu Yulin Deng 2020Journal of Beijing Institute of Technology2020,29,1:0

10Dynamic computation offloading in time-varying environment for ultra-dense networks:a stochastic game approach显示文摘To meet the demands of large-scale user access with computation-intensive and delay-sensitive applications,combining ultra-dense networks(UDNs)and mobile edge computing(MEC)are considered as important solutions.In the MEC enabled UDNs,one of the most important issues is computation offloading.Although a number of work have been done toward this issue,the problem of dynamic computation offloading in time-varying environment,especially the dynamic computation offloading problem for multi-user,has not been fully considered.Therefore,in order to fill this gap,the dynamic computation offloading problem in time-varying environment for multi-user is considered in this paper.By considering the dynamic changes of channel state and users’queue state,the dynamic computation offloading problem for multi-user is formulated as a stochastic game,which aims to optimize the delay and packet loss rate of users.To find the optimal solution of the formulated optimization problem,Nash Q-learning(NQLN)algorithm is proposed which can be quickly converged to a Nash equilibrium solution.Finally,extensive simulation results are presented to demonstrate the superiority of NQLN algorithm.It is shown that NQLN algorithm has better optimization performance than the benchmark schemes.Xie Renchao Liu Xu Duan Xuefei Tang Qinqin Yu Fei Richard Huang Tao 2021The Journal of China Universities of Posts and Telecommunications2021,28,2:0
11Optimal planning of high voltage distribution substations显示文摘Aimed at solving the problem of optimal planning for high voltage distribution substations,an efficient method is put forward.The method divides the problem into two sub-problems:source locating and combina tional optimization.The algorithm of allocating and locating alternatively(ALA)is widely used to deal with the source lo cating problem,but it is dependent on the initial location to a large degree.Thus,some modifications were made to the ALA algorithm,which could greatly improve the quality of solutions.In addition,considering the non-convex and nonconcave nature of the sub-problem of combinational optimization,the branch-and-bound technique was adopted to obtain or approximate a global optimal solution.To improve the efficiency of the branch-and-bound technique,some heuristic principles were proposed to cut those branches that may generate a global optimization solution with low probability.Examples show that the proposed algorithm meets the requirement of engineering and it is an effective approach to rapidly solve the problem of optimal planning for high voltage distribution substations.YU Yixin YAN Xuefei ZHANG Yongwu 2007Frontiers of Electrical and Electronic Engineering in China2007,2,3:0

13Comparative Proteomics Based on 18O Labeling to Evaluate the Hepatotoxicity of Tris(2,3-dibromopropyl) Isocyanuratein Rats显示文摘Detecting protein expression levels in organisms exposed to environmental pollutants can help us understand the action mechanism of toxicity. In the present study, adult female Sprague-Dawley (SD) rats received a consecutive intraperitoneal injection of tris(2,3-dibromopropyl) isocyanurate (TBC) for seven days, and the ^16O/^18O labeling comparative proteomic approach was used to study the change of liver proteome in these rats. Thirty six differential proteins were identified between the control group and the high-dose-exposed group (8 mg/kg), of which, twenty eight proteins were down-regulated and eight proteins were up-regulated. Bioinformatics analysis revealed that most of the differential proteins were related with the metabolic and cellular processes. In addition, the contents of malondialdehyde (MDA), catalase (CAT), and glutathione (GSH) in liver were measured to evaluate the oxidative stress status induced by TBC exposure. Together, these findings showed that TBC might be toxic to liver by disrupting metabolic process and apoptosis. 14Function of arsATorf7orf8 of Bacillus sp. CDB 3 in arsenic resistance显示文摘Bacillus sp. CDB3 isolated from an arsenic contaminated cattledip site possesses an uncommon arsenic resistance (ars) operon bearing eight genes in the order of arsRYCDATorf7orf8. We investigated the functions of arsA, arsT, orf7 and orf8 in arsenic resistance using a plasmid-based gene knockout approach in the ars genedeficient Escherichia coli strain AW3110. The CDB3 arsA genewas shown to play a significant role in resistance, suggesting that the encoded ArsA may couplewith the arsenite transporter, forming an ArsAY complex that can enhance arsenite extrusion efficiency. Thedisruption of either arsT or orf7was not observed to affect arsenic resistance in the heterologous E. coli host, but their involvement in arsenic resistance can not be excluded. The orf8 gene is predicted to encode a putativedual-specificity protein phosphatasewhich also shares certain homology to arsenate reductases. The function loss of orf8 resulted in a remarkabledecrease in resistance to arsenate, though not arsenite. To examine if this effectwasdue to the reduction of arsenate by orf8, the arsC genewithin the 8-gene operonwasdisrupted. 15Technologies for pollutant removal and resource recovery from blackwater:a review显示文摘Blackwater(BW),consisting of feces,urine,flushing water and toilet paper,makes up an important portion of domestic wastewater.The improper disposal of BW may lead to environmental pollution and disease transmission,threatening the sustainabie development of the world.Rich in nutrients and organic matter,BW could be treated for resource recovery and reuse through various approaches.Aimed at providing guidance for the future development of BW treatment and resource recovery,this paper presented a literature review of BWs produced in different countries and types of toilets,including their physiochemical characteristics,and current treatment and resource recovery strategies.The degradation and utilization of carbon(C),nitrogen(N)and phosphorus(P)within BW are underlined.The performance of different systems was classified and summarized.Among all the treating systems,biological and ecological systems have been long and widely applied for BW treatment,showing their universality and operability in nutrients and energy recovery,but they are either slow or ineffective in removal of some refractory pollutants.Novel processes,especially advanced oxidation processes(AOPs),are becoming increasingly extensively studied in BW treatment because of their high efficiency,especially for the removal of micropollutants and pathogens.This review could serve as an instructive guidance for the design and optimization of BW treatment technologies,aiming to help in the fulfilment of sustainable human excreta management.Wei Zhang Huaqiang Chu Libin Yang Xiaogang You Zhenjiang Yu Yalei Zhang Xuefei Zhou 2023Frontiers of Environmental Science & Engineering2023,17,7:0

18Oxygen-contained amorphous MoS_(x) cocatalyst by one-step photodeposition to enhance H-adsorption affinity for efficient photocatalytic H_(2)generation显示文摘Traditional bulk MoS_(2) as an effective H_(2)-evolution cocatalyst is mainly subjected to the weak hydrogen-adsorption ability of highporpotion saturated S,resulting in a slow interfacial H_(2)-evolution reaction.In this paper,an efficient strategy for enhancing hydrogen adsorption of saturated S by manipulating electron density through O atoms is proposed to boost photocatalytic performance of CdS.Simultaneously,amorphization of MoS_(2) can further increase the unsaturated active S sites.Herein,oxygencontained amorphous MoS_(x)(a-MoOS_(x))nanoparticles(10-30 nm)were tightly loaded on the CdS surface through a mild photoinduced deposition method by using(NH_(4))_(2)[MoO(S_(4))_(2)]solution as the precursor at room temperature.The photocatalytic H_(2)-evolution result showed that the a-MoOS_(x)/CdS performed the superior H_(2)-production activity(382μmol·h^(-1),apparent quantum efficiencies(AQE)=11.83%)with a lot of visual H_(2)bubbles,which was 54.6,2.5,and 5.1 times as high as that of CdS,MoS_(x)/CdS,and annealed a-MoOS_(x)/CdS,respectively.Characterizations and density functional theory(DFT)calculations revealed the mechanism of improved H_(2)-evolution activity is that the O heteroatom in amorphous MoOS_(x) can enhance the atomic H-adsorption ability by manipulating the electron density to form electron-deficient S^((2-δ)-)sites.This study provides a new idea to improve the efficiency and number of H_(2)-evolution active sites for developing efficient cocatalysts in the field of photocatalytic hydrogen evolution.Pinsi Deng Ping Wang Xuefei Wang Feng Chen Huogen Yu 2023Nano Research2023,16,7:0

20Basic properties of saline soil in Da'an, western Jilin, China显示文摘This paper studied the basic properties of saline soil at different depths of a sampling site in Da'an, China, through field reconnaissance and laboratory analysis. A series of experiments which comprised the analyses of grain size distribution, mineral composition, soil physical properties, soluble salt concent, p H, organic content and cation exchange capacity were conducted. Through these experiments, the distribution rule of each property and their causes are discussed.
