1Laminin-coated multifilament entubulation, combined with Schwann cells and glial cell line-derived neurotrophic factor, promotes unidirectional axonal regeneration in a rat model of thoracic spinal cord hemisection显示文摘Biomaterial bridging provides physical substrates to guide axonal growth across the lesion.To achieve efficient directional guidance,combinatory strategies using permissive matrix,cells and trophic factors are necessary.In the present study,we evaluated permissive effect of poly(acrylonitrile-co-vinyl chloride)guidance channels filled by different densities of laminin-precoated unidirectional polypropylene filaments combined with Schwann cells,and glial cell line-derived neurotrophic factor for axonal regeneration through a T10 hemisected spinal cord gap in adult rats.We found that channels with filaments significantly reduced the lesion cavity,astrocytic gliosis,and inflammatory responses at the graft-host boundaries.The laminin coated low density filament provided the most favorable directional guidance for axonal regeneration which was enhanced by co-grafting of Schwann cells and glial cell line-derived neurotrophic factor.These results demonstrate that the combinatorial strategy of filament-filled guiding scaffold,adhesive molecular laminin,Schwann cells,and glial cell line-derived neurotrophic factor,provides optimal topographical cues in stimulating directional axonal regeneration following spinal cord injury.This study was approved by Indiana University Institutional Animal Care and Use Committees(IACUC#:11011)on October 29,2015.Ling-Xiao Deng Nai-Kui Liu Ryan Ning Wen Shuang-Ni Yang Xuejun Wen Xiao-Ming Xu 2021Neural Regeneration Research2021,16,1:4

3Challenges and strategies in high-accuracy manufacturing of the world’s largest SiC aspheric mirror显示文摘In the process of manufacturing the world’s largest silicon carbide(SiC)aspheric mirror,the primary difficulties are mirror blank preparation,asphere fabrication,and testing,as well as cladding and coating.Specifically,the challenges include the homogeneity of the complicated structure casting,accuracy and efficiency of the fabrication process,print-through effect,fidelity and precision of test procedure,stress and denseness of cladding process,the dynamic range of interferometric measurement,and air turbulence error due to the long optical path.To break through such a barrier of difficulties,we proposed the water-soluble room temperature vanishing mold and gel casting technology,homogeneous microstructure reaction-formed joint technology,nano-accuracy efficient compound fabrication,gravity unloading technology,high-denseness low-defect physical vapor deposition(PVD)Si-cladding technology,test data fusion method,and time-domain averaging method,etc.Based on the proposed technologies and methods,we have accomplished the world’s largest SiC aspheric mirror with a size of φ4.03 m.The impressive performance of the SiC aspheric mirror is validated by the characteristics of the fabricated SiC aspheric mirror.The aerial density of the SiC blank is less than 120 kg/m^(2),surface shape test accuracy is better than 6 nm RMS,thickness inhomogeneity of the cladding layer is less than 5%,and the final surface figure error and roughness are 15.2 nm RMS and 0.8 nm RMS,respectively.Xuejun Zhang Haixiang Hu Xiaokun Wang Xiao Luo Ge Zhang Wenxing Zhao Xiaoyi Wang Zhenyu Liu Ling Xiong Erhui Qi Congcong Cui Yanchao Wang Yingjie Li Xu Wang Longxiang Li Yang Bai Qiang Cheng Zhiyu Zhang Ruigang Li Wa Tang Xuefeng Zeng Weijie Deng Feng Zhang 2022Light(Science & Applications)2022,11,12:3
4HER3 intracellular domains play a crucial role in HER3/HER2 dimerization and activation of downstream signaling pathways显示文摘Dimerization among the EGFR family of tyrosine kinase receptors leads to allosteric activation of the kinase domains of the partners.Unlike other members in the family,the kinase domain of HER3 lacks key amino acid residues for catalytic activity.As a result,HER3 is suggested to serve as an allosteric activator of other EGFR family members which include EGFR,HER2 and HER4.To study the role of intracellular domains in HER3 dimerization and activation of downstream signaling pathways,we constructed HER3/HER2 chimeric receptors by replacing the HER3 kinase domain(HER3-2-3)or both the kinase domain and the C-terminal tail(HER3-2-2)with the HER2 counterparts and expressed the chimeric receptors in Chinese hamster ovary(CHO)cells.While over expression of the intact human HER3 transformed CHO cells with oncogenic properties such as AKT/ERK activation and increased proliferation and migration,CHO cells expressing the HER3-2-3 chimeric receptor showed significantly reduced HER3/HER2 dimerization and decreased phosphorylation of both AKT and ERK1/2 in the presence of neuregulin-1(NRG-1).In contrast,CHO cells expressing the HER3-2-2 chimeric receptor resulted in a total loss of downstream AKT activation in response to NRG-1,but maintained partial activation of ERK1/2.The results demonstrate that the intracellular domains play a crucial role in HER3’s function as an allosteric activator and its role in downstream signaling.Byung-Kwon Choi Xiumei Cai Bin Yuan Zhao Huang Xuejun Fan Hui Deng Ningyan Zhang Zhiqiang An 2012Protein & Cell2012,3,10:3

9Increased expression of Notch1 in temporal lobe epilepsy: animal models and clinical evidence显示文摘Temporal lobe epilepsy is associated with astrogliosis.Notch1 signaling can induce astrogliosis in glioma.However,it remains unknown whether Notch1 signaling is involved in the pathogenesis of epilepsy.This study investigated the presence of Notch1,hairy and enhancer of split-1,and glial fibrillary acidic protein in the temporal neocortex and hippocampus of lithium-pilocarpine-treated rats.The presence of Notch1 and hairy and enhancer of split-1 was also explored in brain tissues of patients with intractable temporal lobe epilepsy.Quantitative electroencephalogram analysis and behavioral observations were used as auxiliary measures.Results revealed that the presence of Notch1,hairy and enhancer of split-1,and glial fibrillary acidic protein were enhanced in status epilepticus and vehicle-treated spontaneous recurrent seizures rats,but remain unchanged in the following groups:control,absence of either status epilepticus or spontaneous recurrent seizures,and zileuton-treated spontaneous recurrent seizures.Compared with patient control cases,the presences of Notch1 and hairy and enhancer of split-1 were upregulated in the temporal neocortex of patients with intractable temporal lobe epilepsy.Therefore,these results suggest that Notch1 signaling may play an important role in the onset of temporal lobe epilepsy via astrogliosis.Furthermore,zileuton may be a potential therapeutic strategy for temporal lobe epilepsy by blocking Notch1 signaling.Xijin Liu Zhiyong Yang Yaping Yin Xuejun Deng 2014Neural Regeneration Research2014,9,5:1

17Proliferation changes in hippocampal neurons following pilocarpine-induced intractable epilepsy in adult rats显示文摘Epilepsy can lead to the changes in neurons residing in the dentate gyrus. The present study aimed to observe the cell dividing features following epilepsy in adult rat hippocampi,and to study difference in cell proliferation between adult rats with common epilepsy and intractable epilepsy. Adult,male,Sprague Dawley rats were randomly divided into control(n = 8,treatment with normal saline) and three experimental groups: common epilepsy(n = 33) ,intractable epilepsy(n = 11) ,and drug-responsive(n = 25) . Pilocarpine(15 mg/kg) was intraperitoneally administered to establish epilepsy in the three experimental groups. Rats that developed epilepsy were treated with chloral hydrate. Rats that did not exhibit spontaneous seizures were enrolled in the common epilepsy group,and rats with spontaneous seizure were included in the spontaneous seizure group. At 6 hours after epileptic attack termination,rats were intraperitoneally injected with bromodeoxyuridine(BrdU;50 mg/kg) ,an optimal marker for labeling cell proliferation in vivo,four times. Immunohistochemistry results at 48 hours after BrdU injection indicated that the number of BrdU-positive cells was the highest in the common epilepsy group,followed by the control group,and lastly the intractable group(P < 0.01) . In addition,the number of BrdU-positive cells in the common epilepsy group was similar to the drug-responsive group. 19Analysis of mesoscopic mechanical dynamic characteristics of ballast bed with under sleeper pads显示文摘The meso-dynamical behaviour of a high-speed rail ballast bed with under sleeper pads(USPs)was studied.The geometrically irregular refined discrete element model of the ballast particles was constructed using 3D scanning techniques,and the 3D dynamic model of the rail-sleeper-ballast bed was constructed using the coupled discrete element method-multiflexible-body dynamics(DEM-MFBD)approach.We analyse the meso-mechanical dynamics of the ballast bed with USPs under dynamic load on a train and verify the correctness of the model in laboratory tests.It is shown that the deformation of the USPs increases the contact area between the sleeper and the ballast particles,and subsequently the number of contacts between them.As the depth of the granular ballast bed increases,the contact area becomes larger,and the contact force between the ballast particles gradually decreases.Under the action of the elastic USPs,the contact forces between ballast particles are reduced and the overall vibration level of the ballast bed can be reduced.The settlement of the granular ballast bed occurs mainly at the shallow position of the sleeper bottom,and the installation of the elastic USPs can be effective in reducing the stress on the ballast particles and the settlement of the ballast bed.Xiong Yang Liuyang Yu Xuejun Wang Zhigang Xu Yu Deng Houxu Li 2024Railway Engineering Science2024,32,1:0
20Tomato leaf curl New Delhi virus:an emerging plant begomovirus threatening cucurbit production显示文摘Tomato leaf curl New Delhi virus(ToLCNDV),a bipartite begomovirus,was first reported to infect tomato and has recently spread rapidly as an emerging disease to Cucurbitaceae crops.To date,the virus has been reported to infect more than 11 cucurbit crops,in 16 countries and regions,causing severe yield losses.In autumn 2022,ToLCNDV was first isolated from cucurbit plants in Southeastern coastal areas of China.Phylogenetic analysis established that these isolates belong to the Asian ToLCNDV clade,and shared high nucleotide identity and closest genetic relationship with the DNA-A sequence from the Chinese tomato-infecting ToLCNDV isolate(Accession no.OP356207)and the tomato New Delhi ToLCNDV-Severe isolate(Accession no.HM159454).In this review,we summarize the occurrence and distribution,host range,detection and diagnosis,control strategies,and genetic resistance of ToLCNDV in the Cucurbitaceae.We then summarize pathways that could be undertaken to improve our understanding of this emerging disease,with the objective to develop ToLCNDV-resistant cucurbit cultivars.Lingmin Cai Yuzhen Mei Ruyi Ye Yun Deng Xuejun Zhang Zhongyuan Hu Xueping Zhou Mingfang Zhang Jinghua Yang 2023aBIOTECH2023,4,3:0
