1Comparative pharmacokinetics of six major compounds in normal and insomnia rats after oral administration of Ziziphi Spinosae Semen aqueous extract显示文摘Ziziphi Spinosae Semen(ZSS),a traditional Chinese medicine,is used in clinics for the treatment of insomnia in China and other Asian countries.Herein,we described for the first time a comparative pharmacokinetics study of the six major compounds of ZSS in normal control(NC)and para-chlorophenylalanine(PCPA)-induced insomnia model(IM)rats that were orally administered the aqueous extract of ZSS.An ultra-high-performance liquid chromatography coupled with quadrupole orbitrap mass(UHPLC-Q-Orbitrap-MS)method was developed and validated for the simultaneous determination of coclaurine,magnoflorine,spinosin,6000-feruloylspinosin,jujuboside A(JuA),and jujuboside B(JuB)in ZSS in rat plasma.The established approach was successfully applied to a comparative pharmacokinetic study.The systemic exposures of spinosin and 6000-feruloylspinosin were decreased in the IM group compared to the NC group,while plasma clearance(CL)was significantly increased.The Tmax values of JuA and JuB in IM rats were significantly lower than those in NC rats.The T1/2 of JuA in the IM group was significantly accelerated.The pharmacokinetic parameters of coclaurine and magnoflorine were not evidently affected between the two groups.These results indicate that the pathological state of insomnia altered the plasma pharmacokinetics of spinosin,6000-feruloylspinosin,JuA,and JuB in the ZSS aqueous extract,providing an experimental basis for the role of ZSS in insomnia treatment.The comparative pharmacokinetics-based UHPLC-Q-Orbitrap-MS using full-scan mode can therefore provide a reliable and suitable means for the screening of potentially effective substances applied as quality markers of ZSS.Chenhui Du Yan Yan Chenxi Shen Xiaofang Cui Xiangping Pei Xuemei Qin 2020Journal of Pharmaceutical Analysis2020,10,4:14

4Fault Tolerant Design of Large-Scale Digital Beam Forming in SRAM-FPGAs for Software Defined Satellite Platforms显示文摘Large scale digital beamforming(LS-DBFs)are widely used in satellite communications for spectrum reuse and transmission enhancement.SRAM-FPGAs are a popular option for software defined satellite platforms due to their rich computation resources and high flexibility.However,they are sensitive to soft errors,which limit their application in space.This paper discusses the application of coding based fault tolerance schemes for the protection of LS-DBFs on software defined payloads.Since multiple FPGAs are usually needed to support the whole LS-DBFs system,different decomposition schemes are compared in terms of resource efficiency and reliability when the coding based scheme is applied to protect the DBFs on a FPGA.Theoretical analysis and hardware experiments shows that resource efficiency and reliability are a pair of contradictory requirements for decomposition schemes.The protection with vertical decomposition could improve the reliability by 96%with 1.5x redundancy.And the protection with horizontal decomposition could improve the reliability by 85%with 1.2 x redundancy.Zhen Gao Jinhua Zhu Tong Yan Linghua Guo Xiangping Chen Yinqiao Li Xiaolei Wan 2020China Communications2020,17,7:2

6Numerical simulation of CO_2-ionic liquid flow in a stirred tank显示文摘Ionic liquids(ILs) are new solvents that represent a breakthrough for absorption and conversion of CO2. However, there has been little research on the hydrodynamics of gas-IL systems in stirred tanks, and this has become a bottleneck for development of IL-based reactors. In the present study, a CO2-IL([bmim][BF4]) flow in a stirred tank was studied using computational fluid dynamics(CFD). A new drag coefficient model that is specific to a gas-ionic liquid system and a population balance model(PBM) were adopted to describe the bubble behavior, such as gas holdup and bubble size evolution, precisely. The predicted results for the total gas holdup and local Sauter diameter agree well with the experimental data. The influences of the gassing rate, agitation speed, and temperature on local gas holdup, bubble size distribution, and interfacial area for the CO2-[bmim][BF4] system were also investigated. 13Aqueous-soluble components of sporoderm-removed Ganoderma lucidum spore powder promote ferroptosis in oral squamous cell carcinoma显示文摘Objective: Ferroptosis is a novel cell death process which displays a promising role in cancer treatment.However, clinically available drugs targeting ferroptosis are rarely used, and yet there are no studies reporting on inducing ferroptosis via Chinese herbal extracts. Here we explored the tumor inhibition effects of Ganoderma lucidum(G. lucidum) on oral squamous cell carcinoma(OSCC). Specifically, we aimed to clarify the biological mechanism of components in the dietary, aqueous-soluble sporoderm-removed G. lucidum spore powder(A-GSP).Methods: Preliminary transcriptome analysis revealed the significant enrichment of the ferroptosis pathway.Cellular Fe2+, glutathione(GSH), malondialdehyde(MDA), reactive oxygen species(ROS) and lipid peroxide levels were measured to identify ferroptosis occurrence. Western blotting was used to measure ferroptosis-related proteins. Changes in mitochondria morphology and function were observed with transmission electron microscopy(TEM) and ATP detection assays. Ferroptosis inhibitor ferrostatin-1 was then used to verify the anti-tumor effects of A-GSP. Finally, nude mice xenograft models of oral cancer confirmed that A-GSP inhibited tumor growth.Results: A-GSP promoted ferroptosis in oral cancer cells by inducing Fe2+influx, GSH depletion, as well as lipid peroxide and ROS accumulation. Ferroptosis-related proteins exhibited corresponding changes, particularly Acylco A synthetase long chain family member 4(ACSL4) increase and glutathione peroxidase 4(GPX4) decrease. A-GSP considerably lowered mitochondrial volume and ridge number, while significantly decreasing ATP production. Ferrostatin-1 reversed all of these A-GSP-induced changes. 14Multi-objective optimization of biomass to biomethane system显示文摘The superstructure optimization of biomass to biomethane system through digestion is conducted in this work. The system encompasses biofeedstock collection and transportation, anaerobic digestion, biogas upgrading, and digestate recycling. We propose a multicriteria mixed integer nonlinear programming(MINLP) model that seeks to minimize the energy consumption and maximize the green degree and the biomethane production constrained by technology selection, mass balance, energy balance, and environmental impact. A multi-objective MINLP model is proposed and solved with a fast nondominated sorting genetic algorithm Ⅱ(NSGA-Ⅱ). The resulting Pareto-optimal surface reveals the trade-off among the conflicting objectives. 15Discovery of novel covalent selective estrogen receptor degraders against endocrine-resistant breast cancer显示文摘Endocrine-resistance remains a major challenge in estrogen receptorαpositive(ERα^(+))breast cancer(BC)treatment and constitutively active somatic mutations in ERαare a common mechanism.There is an urgent need to develop novel drugs with new mode of mechanism to fight endocrineresistance.Given aberrant ERαactivity,we herein report the identification of novel covalent selective estrogen receptor degraders(cSERDs)possessing the advantages of both covalent and degradation strategies.A highly potent cSERD 29c was identified with superior anti-proliferative activity than fulvestrant against a panel of ERa+breast cancer cell lines including mutant ERα.Crystal structure of ERα-29c complex alongside intact mass spectrometry revealed that 29c disrupted ERa protein homeostasis through covalent targeting C530 and strong hydrophobic interaction collied on H11,thus enforcing a unique antagonist conformation and driving the ERαdegradation.These significant effects of the cSERD on ERαhomeostasis,unlike typical ERαdegraders that occur directly via long side chains perturbing the morphology of H12,demonstrating a distinct mechanism of action(MoA).In vivo,29c showed potent antitumor activity in MCF-7 tumor xenograft models and low toxicity.This proof-of-principle study verifies that novel cSERDs offering new opportunities for the development of innovative therapies for endocrine-resistant BC.Yubo Wang Jian Min Xiangping Deng Tian Feng Hebing Hu Xinyi Guo Yan Cheng Baohua Xie Yu Yang Chun-Chi Chen Rey-Ting Guo Chune Dong Hai-Bing Zhou 2023Acta Pharmaceutica Sinica B2023,13,12:0
16Polymorphism of apolipoprotein E gene and post-stroke vascular dementia显示文摘BACKGROUND: Studies have indicated that the more certain etiological factors of vascular dementia are the sites, size and number of cerebrovascular lesions, but there are arguments all the time in the relationship of genetic factors, especially apolipoprotein E gene, with the occurrence and development of vascular dementia. OBJECTIVE: To analyze the relationship between the polymorphism of apolipoprotein E gene and vascular dementia in stroke patients. DESIGN: A non-randomized grouped comparative observation at the same time. SETTING: The Laboratory of Neurology, the Second Affiliated Hospital of Chongqing University of Medical Sciences. PARTICIPANTS: Totally 121 inpatients with stroke were selected from the Department of Neurology, the Second Affiliated Hospital of Chongqing University of Medical Sciences from January 2000 to December 2001. All the patients were accorded with the diagnostic standards for cerebrovascular diseases set by the Second National Academic Meeting for Cerebrovascular Disease, and confirmed by cranial CT or MRI. According to with vascular dementia or not, they were divided into vascular dementia group (n =58) and non-vascular dementia group (n =63). In the vascular dementia group, there were 37 males and 21 females, the average age was (59±8) years. They were all in accordance with the standard of the third edition of Diagnostic and Statistical Manual of Mental Disorders (DMS-Ⅲ), without conscious disturbance and language disorder; hospitalized within 3 days after the attack of cerebrovascular disease. In the non-vascular dementia group, there were 37 males and 26 females, the average age was (59±9.5) years. They all had no intellectual disturbance within 3 months after the attack of cerebrovascular disease. All the subjects agreed to supply blood samples for the experiment. METHODS: ①Fasting venous blood (2 mL) was drawn from each patient in the morning to extract DNA. The frequencies of apolipoprotein E genotypes and alleles were detected with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). ② Gene frequency was calculated using the gene calculation, genotype or allele frequency were compared using the Chi-square test. MAIN OUTCOME MEASURES: The frequencies of apolipoprotein E genotypes and alleles were compared between the two groups. RESULTS: All the 121 patients were involved in the analysis of results. Five genotypes of apolipoprotein E gene were detected, and ε2/2 genotype was not detected. The frequency of ε3/3 was the highest in both groups, the frequencies of ε4/4 genotype and ε4 allele were obviously higher in the vascular dementia group [9%(5/58), 24.1%] than in the non-vascular dementia group [3% (2/58), 19.0%, P < 0.05]. The genotype frequencies of ε2/2,ε2/4, ε3/3, ε2/3 and ε3/4 and the allele frequencies of ε2 and ε3 in the vascular dementia group [0, 2%(1/58), 57%(33/58), 3%(2/58), 29%(17/58); 2.6%, 73.3%] were not obviously different from those in the non-vascular dementia group [0, 2%(1/58), 59%(37/63), 6%(4/63), 30%(19/63); 4.0%, 77.0%, P > 0.05]. 17Cylindrical vector beam revealing multipolar nonlinear scattering for superlocalization of silicon nanostructures显示文摘The resonant optical excitation of dielectric nanostructures offers unique opportunities for developing remarkable nanophotonic devices.Light that is structured by tailoring the vectorial characteristics of the light beam provides an additional degree of freedom in achieving flexible control of multipolar resonances at the nanoscale.Here,we investigate the nonlinear scattering of subwavelength silicon(Si)nanostructures with radially and azimuthally polarized cylindrical vector beams to show a strong dependence of the photothermal nonlinearity on the polarization state of the applied light.The resonant magnetic dipole,selectively excited by an azimuthally polarized beam,enables enhanced photothermal nonlinearity,thereby inducing large scattering saturation.In contrast,radially polarized beam illumination shows no observable nonlinearity owing to off-resonance excitation.Numerical analysis reveals a difference of more than 2 orders of magnitude in photothermal nonlinearity under two types of polarization excitations.Nonlinear scattering and the unique doughnut-shaped focal spot generated by the azimuthally polarized beam are demonstrated as enabling far-field high-resolution localization of nanostructured Si with an accuracy approaching 50 nm.Our study extends the horizons of active Si photonics and holds great potential for label-free superresolution imaging of Si nanostructures.BIN WANG YING CHE XIANGCHAO ZHONG WEN YAN TIANYUE ZHANG KAI CHEN YI XU XIAOXUAN XU XIANGPING LI 2021Photonics Research2021,9,6:0
18Modification to solution-diffusion model for performance prediction of nanofiltration of long-alkyl-chain ionic liquids aqueous solutions based on ion cluster显示文摘Mathematical modeling for nanofiltration of ionic liquids(ILs) solutions could assist to understand transfer mechanism and predict experimental values. In this work, modeling by solution-diffusion model for nanofiltration of long-alkyl-chain ILs aqueous solutions was proposed. Molecular simulations were performed to validate the existence of ion cluster in long-alkyl-chain ILs aqueous solution. Based on the results of simulations, parameters used in the solution-diffusion model were modified, such as concentration of ILs and diameter of ion cluster.The modeling process was developed for three long-alkyl-chain ILs aqueous solutions with different concentrations(1-alkyl-3-methylimidazolium chloride: [C6 mim]Cl, [C8 mim]Cl, [C10 mim]Cl). 19Plant above-ground biomass and litter quality drive soil microbial metabolic limitations during vegetation restoration of subtropical forests显示文摘Changes in litter quality(carbon:nitrogen,C:N)and above-ground biomass(AGB)following vegetation restoration significantly impact soil physicochemical properties,yet their effects on soil microbial metabolic limitations remain unclear.We measured litter quality,AGB,soil physicochemical properties,and extracellular enzyme activity(EEA)along a vegetation restoration gradient(7,14,49,70 years,and nearly climax evergreen broadleaved forests)in southern China.We also evaluated soil microbial metabolic limitations by a vector analysis of the EEA.Results revealed the soil microbial metabolisms were co-limited by C and phosphorus(P).The microbial C limitation initially decreased(before 14 years)and then increased,while the microbial P limitation initially increased(before 49 years)and then decreased.Partial least squares path modeling(PLS-PM)showed that the microbial C limitation was mainly attributed to microbial C use efficiency induced by litter quality,suggesting that microorganisms may transfer cellular energy between microbial growth and Cacquiring enzyme production.The microbial P limitation was primarily correlated with AGB-driven change in soil elements and their stoichiometry,highlighting the importance of nutrient stoichiometry and balance in microbial metabolism.The shifts between microbial C and P limitations and the strong connections of plant–soil-microbe processes during vegetation restoration revealed here will provide us with helpful information for optimal management to achieve forest restoration success.Shuo Zhang Ying-Ping Wang Xi Fang Jinlei Chen Nannan Cao Pingping Xu Mengxiao Yu Xin Xiong Xiangping Tan Qi Deng Junhua Yan 2023Soil Ecology Letters2023,5,2:0
