| 2 | Effect of Dy^(3+) on osteogenic and adipogenic differentiation of mouse primary bone marrow stromal cells and adipocytic trans-differentiation of mouse primary osteoblasts显示文摘A series of experimental methods including 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bro-mide (MTT) test,alkaline phosphatase (ALP) activity measurement,mineralized function,Oil Red O stain and measurement were employed to assess the effect of Dy3+ on the osteogenic and adipogenic differentiation of mouse primary bone marrow stromal cells (BMSCs) and the adipogenic trans-differ-entiation of mouse primary osteoblasts (OBs). The results showed that Dy3+ had no effect on BMSC proliferation at concentrations of 1×10-8 and 1×10-5 mol/L,but inhibited BMSC proliferation at other concentrations. Dy3+ had no effect on OB proliferation at concentrations of 1×10-10 and 1×10-9 mol/L,but inhibited OB proliferation at other concentrations. Dy3+ had no effect on the osteogenic differentia-tion of BMSCs at concentrations of 1×10-9 and 1×10-7 mol/L,and promoted osteogenic differentiation of BMSCs at other concentrations at the 7th day. The osteogenic differentiation of BMSCs was inhibited by Dy3+ at concentration of 1×10-5 mol/L at the 14th day,but promoted osteogenic differentiation of BMSCs at concentrations of 1×10-9,1×10-8,1×10-7 and 1×10-6 mol/L with the maximal effect at concen-tration of 10-6 mol/L. Dy3+ promoted mineralized function of BMSCs at any concentration. Dy3+ had no effect on adipogenic differentiation of BMSCs at concentration of 1×10-7 mol/L,but inhibited adipogenic differentiation of BMSCs at other concentrations. Dy3+ inhibited adipocytic trans-differentiation of OBs at any concentration,suggesting that Dy3+ had protective effect on bone and the protective effect on bone may be mediated by modulating differentiation of BMSCs away from the adipocyte and inhibiting adipocytic trans-differentiation of OBs which may promote differentiation and mineralization of OBs. | 3 | Effects of genistein, daidzein and glycitein on the osteogenic and adipogenic differentiation of bone marrow stromal cells and on the adipogenic trans-differentiation of osteoblasts显示文摘The 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT), alkaline phosphatase (ALP) activity and oil red O assays were used to examine the effects of genistein, daidzein and glycitein on the osteogenic and adipogenic differentiation of primary mouse bone marrow stromal cells (MSCs) and the adipogenic trans-differentiation of primary mouse osteoblasts. The results indicated that daidzein, genistein and glycitein at concentrations from 1×10 -8 mol/L to 1×10 -5 mol/L promoted the proliferation of MSCs and osteoblasts; genistein, daidzein and glycitein promoted osteogenic differentiation and inhibited adipogenic differentiation of MSCs, and inhibited adipocytic transdifferentiation of osteoblasts at appropriate concentrations as 17β-estradiol. It suggests that genistein, daidzein and glycitein regulate a dual differentiational process of MSCs into the osteogenic and adipogenic lineages, and trans-differentiational process of primary osteoblasts into the adipocyte lineages, causing a lineage shift toward osteoblast. Protective effects of them on bone may be mediated by a reversal of adipogenesis which may promote the proliferation, differentiation and mineralization of osteoblasts, and make adipocytes secrete less cytokines which may promote osteoclast formation and activation. | 4 | The influence of ^(18)F-fluorodeoxyglucose positron emission tomography/computed tomography on the N-and M-staging and subsequent clinical management of intrahepatic cholangiocarcinoma显示文摘Background:Intrahepatic cholangiocarcinoma(ICC)is a highly metastatic cancer.^(18)F-fluorodeoxyglucose positron emission tomography/computed tomography(^(18)F-FDG PET/CT)enables sensitive tumor and metastasis detection.Our aim is to evaluate the influence of pre-treatment PET/CT on the N-and M-staging and subsequent clinical management in ICC patients.Methods:Between August 2010 and August 2018,660 consecutive ICC patients,without prior anti-tumor treatments nor other malignancies,were enrolled.The diagnostic performance of PET/CT on the N-and M-staging was compared with conventional imaging,and the preoperative staging accuracy and treatment re-allocation by PET/CT were retrospectively calculated.Survival difference was compared between patients receiving PET/CT or not after propensity score matching.Results:Patients were divided into group A(n=291)and group B(n=369)according to whether PET/CT was performed.Among 291 patients with both PET/CT and conventional imaging for staging in group A,PET/CT showed significantly higher sensitivity(83.0%vs.70.5%,P=0.001),specificity(88.3%vs.74.9%,P<0.001)and accuracy(86.3%vs.73.2%,P<0.001)than conventional imaging in diagnosing regional lymph node metastasis,as well as higher sensitivity(87.8%vs.67.6%,P<0.001)and accuracy(93.5%vs.89.3%,P=0.023)in diagnosing distant metastasis.Overall,PET/CT improved the accuracy of preoperative staging from 60.1%to 71.8%(P<0.001),and modified clinical treatment strategy in 5.8%(17/291)of ICC patients,with unique roles in different tumor-node-metastasis(TNM)stages.High tumor-to-non-tumor ratio(TNR)predicted poor overall survival[hazard ratio(HR)=2.17;95%confidence interval(CI):1.49-3.15;P<0.001].Furthermore,patients performing PET/CT had longer overall survival compared with those without PET/CT(HR=0.74;95%CI:0.58-0.93;P=0.011)after propensity score matching.Conclusions:PET/CT was valuable for diagnosing regional lymph node metastasis and distant metastasis in ICC patients,and facilitated accurate tumor staging and optimal treatment allocation. | Youpei Lin Huanhuan Chong Guohe Song Chenhao Zhang Liangqing Dong Ling Aye Fei Liang Shuaixi Yang Mengsu Zeng Guangyu Ding Shu Zhang Jieyi Shi Aiwu Ke Xiaoying Wang Jian Zhou Jia Fan Qiang Gao | 2022 | Hepatobiliary Surgery and Nutrition2022,11,5: | 3 |
| 5 | The effects of gold nanoparticles on the proliferation, differentiation, and mineralization function of MC3T3-E1 cells in vitro显示文摘This study has investigated the effects of gold nanoparticles (Au NPs) on the proliferation, differentiation, and mineralization of a murine preosteoblast cell line MC3T3-E1 in vitro. The results show that Au NPs with diameters of both 20 and 40 nm promoted the proliferation, differentiation, and mineralization of MC3T3-E1 cells in a time- and dose-dependent manner at the concentra-tions of 1.5×10-5, 3.0×10-5, and 1.5×10-4 μmol/L. The reverse transcriptase polymerase chain reaction (RT-PCR) indicates that the expressions of runt-related transcription factor 2 (Runx2), bone morphogenetic protein 2 (BMP-2), alkaline phosphatase (ALP), and osteocalcin (OCN) genes increased after the 20 and 40 nm Au NP treatments, and the expression levels were higher than those of the NaF group. The above results suggest that Au NPs have the potential to promote the osteogenic differentiation and miner- alization of MC3T3-E1 cells and the particle size plays a significant role in the process. | 12 | Synthesis and antitumor activity of iodo-bridged binuclear platinum complex显示文摘Iodo-bridged binuclear platinum(II) com- plex[Pt( -NH2)I2]2(BPA) has been synthesized and characterized by elemental analysis, conductivity, differential thermal analysis, IR, UV and 1HNMR spectra techniques. The cytotoxicity of the complex was tested by MTT and SRB assays. The results show that complex BPA demonstrates better cyto-toxicity than that of the clinically established cisplatin against EJ, HCT-8, BGC-823, HL-60 and MCF-7 cell lines. The complex BPA at concentrations of 1.00 and 2.00 μmol/L induces G1 cell cycle arrest in HL-60 cells. The level of total platinum bound to DNA in HL-60 cells is significantly higher than that of cisplatin under the same experimental conditions. Acute tox-icity experimental results indiacte that LD50 of BPA is 815.3 mg/kg by intraperitoneal administration. BPA at dose of 12 mg/kg significantly inhibits the growth of nude mice implanted by human A2780 and HCT-116 carcinomas, and inhibition rate is similar to that of cisplatin at dose of 4 mg/kg by intraperitoneal admini-stration. | 16 | Synergistic effect of trace elements and flavonoids from Epimedium koreanum Nakai on primary osteoblasts显示文摘Several trace elements,particularly,manganese(Mn)and zinc(Zn),are essential in bone metabolism as cofactors for specific enzymes.It has been reported that there exists the relationship between osteo-porosis and trace element-deficiency and the efficacy of Ca,Mn and Zn supplementation on spinal bone mineral density in postmenopausal women.Traditional Chinese medicines(TCM),such as Herba epimedii,were proved to be effective for prevention of osteoprosis in vivo;however,the efficacy of the main constituents and/or crude extract was not ideal in vitro,which suggested that they may work in another way.The purpose of the present study was to examine whether the combination of icariin and total flavonoids(TF)from Herba epimedii with mineral elements,which were abundant in Herba epimedii,would have a more beneficial effect on the viability and differentiation of primary osteoblasts than either agent alone,and to analyze the dada for a possible synergistic,additive or antagonistic effect.The combinations of 10 μmol/L Zn,Ca and Mn with icariin and total flavonoids greatly improved the cell viability and meanwhile dramatically enhanced the alkaline phosphatase activity as compared to each agent alone.On the other hand,an increased cell growth inhibition was also observed by combining 0.1 μmol/L,1 μmol/L Zn with 10 μmol/L icariin,and 10 μmol/L Mn with 0.06 μg/mL total fla-vonoids.Meanwhile a decreased alkaline phosphatase activity was also found in several icariin-Zn/Mn and total flavonoids-Zn/Ca/Mn combinations.These results suggested that mineral elements(Zn,Ca,Mn)greatly enhanced the efficacy of icariin and total flavonoids from Herba epimedii on the viability and differentiation of primary osteoblasts by certain combinations. | ZHANG DaWei CHENG Yan ZHANG JinChao WANG XinLuan WANG NaiLi CHEN Yao YANG MengSu YAO XinSheng | 2008 | Chinese Science Bulletin2008,53,3: | 0 |
| 17 | Imipramine Inhibits Adipogenic Differentiation in Both 3T3-L1 Preadioocvtes and Mouse Marrow Stromal Cells显示文摘Imipramine(IM) has been widely used clinically for the treatment of mental disorders.Its actions on tissues or organs other than the nervous system also need to be understood for its proper clinical use.In this study,the effects of IM on adipogenic differentiation in both 3T3-L1 preadipocytes and mouse marrow stromal cells(MSCs) were investigated.The results showed that fewer adipocytic cells were developed from 3T3-L1 preadipocytes in the presence of 0.001 to 1μmol/L of IM as compared to control.Similar inhibitory effect was also observed in mouse MSCs.The decrease in the formation of adipocytes was accompanied with significant down-regulation at mRNA expression of the early adipogenic transcription factor,peroxisome proliferator-activated receptorγ2(PPARγ2).Western blot analysis further revealed that the protein expression of PPARγ2 was reduced markedly in cells treated with IM at concentrations of 0.01,0.1 and 1μmol/L,suggesting that the suppression on PPARγ2 was involved in IM's inhibition on MSCs adipogenesis.Moreover,IM at the above concentrations could stimulate the mRNA expression ofβ2-adrenergic receptor(AR) and P3-AR,which implicated that the effect of IM on adipogenic differentiation was partially mediated byβ-ARs.Our results demonstrated for the first time that the conventional antidepressive imipramine exerts accompanied inhibitory effect on adipocyte formation,which may have possible clinical implications. | Haifang Li Chi-chun Fong Yao Chen Guoping Cai Mengsu Yang | 2012 | Journal of Genetics and Genomics2012,39,4: | 0 |
| 18 | Value of contrast-enhanced ultrasound combined with percutaneous ultrasound-guided fine-needle aspiration in the diagnosis of solid pancreatic lesions显示文摘Background:Contrast-enhanced ultrasound(CEUS)can detect lesions hidden in inflammatory regions and find necrosis or areas of severe fibrosis within the lesion.This retrospective study aimed to compare the diagnostic accuracy of solid pancreatic lesions using percutaneous ultrasound(US)-guided fine-needle aspiration(FNA)with or without CEUS assessment.Methods:Clinical,imaging,and pathologic data of 181 patients from January 2014 to December 2018 in Pecking Union Medical College Hospital,with solid pancreatic masses who underwent percutaneous US-FNA and ThinPrep cytologic test were retrospectively evaluated.Patients were divided into CEUS and US groups according to whether CEUS was performed before the biopsy.According to FNA cytology diagnoses,we combined non-diagnostic,neoplastic,and negative cases into a negative category.The positive category included malignant,suspicious,and atypical cases.The final diagnosis was confirmed by pathology or clinical and radiological follow-up for at least 12 months.The sensitivity,specificity,positive predictive value(PPV),negative predictive value(NPV),and accuracy of US-FNA were evaluated between the two groups.Results:This study enrolled 107 male and 74 female patients(average age:60 years).There were 58 cases in the US group and 123 cases in the CEUS group.No statistically significant differences in age,gender,or lesion size were found between the two groups.The diagnostic accuracy of the CEUS group was 95.1%(117/123),which was higher than the 86.2%(50/58)observed in the US group(P=0.036).The sensitivity,specificity,PPV,and NPV of the CEUS group were increased by 7.5%,16.7%,3.4%,and 18.8%,respectively,compared with the US group.However,the differences of the two groups were not statistically significant.Conclusions:Compared with the conventional US,the use of CEUS could improve the biopsy accuracy and avoid the need for a repeat biopsy,especially for some complicated FNA cases. | Yang Gui Menghua Dai Zhilan Meng Xiaoyan Chang Li Tan Jing Zhang Xueqi Chen Tongtong Zhou Qing Zhang Mengsu Xiao Ke Lyu Yuxin Jiang | 2022 | Chinese Medical Journal2022,,4: | 0 |
