1Immobilization of functional nano-objects in living engineered bacterial biofilms for catalytic applications显示文摘Nanoscale objects feature very large surface-area-to-volume ratios and are now understood as powerful tools for catalysis,but their nature as nanomaterials brings challenges including toxicity and nanomaterial pollution.Immobilization is considered a feasible strategy for addressing these limitations.Here,as a proof-of-concept for the immobilization of nanoscale catalysts in the extracellular matrix of bacterial biofilms,we genetically engineered amyloid monomers of the Escherichia coli curli nanofiber system that are secreted and can self-assemble and anchor nano-objects in a spatially precise manner.We demonstrated three scalable,tunable and reusable catalysis systems:biofilm-anchored gold nanoparticles to reduce nitro aromatic compounds such as the pollutant p-nitrophenol,biofilm-anchored hybrid Cd0.9Zn0.1S quantum dots and gold nanoparticles to degrade organic dyes and biofilm-anchored CdSeS@ZnS quantum dots in a semi-artificial photosynthesis system for hydrogen production.Our work demonstrates how the ability of biofilms to grow in scalable and complex spatial arrangements can be exploited for catalytic applications and clearly illustrates the design utility of segregating high-energy nano-objects from injury-prone cellular components by engineering anchoring points in an extracellular matrix.Xinyu Wang Jiahua Pu Yi Liu Fang Ba Mengkui Cui Ke Li Yu Xie Yan Nie Qixi Mi Tao Li Lingli Liu Manzhou Zhu Chao Zhong 2019National Science Review2019,6,5:9
2Folate-chitosan-gemcitabine core-shell nanoparticles targeted to pancreatic cancer显示文摘Objective:Human pancreatic cancer is one of the most common clinical malignancies.The effect of comprehensive treatment based on surgery is general.The effects of chemotherapy were not obvious mainly because of lack of targeting and chemoresistance in pancreatic cancer.This study aimed to investigate the effects of folate receptor(FR)-mediated gemcitabine FA-Chi-Gem nanoparticles with a core-shell structure by electrostatic spray on pancreatic cancer.Methods:In this study,the levels of expression of FR in six human pancreatic cancer cell lines were studied by immunohistochemical analysis.The uptake rate of isothiocyanate-labeled FA-Chi nanoparticles in FR high expression cell line COLO357 was assessed by fluorescence microscope and the inhibition rate of FAChi-Gem nanoparticles on COLO357 cells was evaluated by MTT assay.Moreover,the biodistribution of PEG-FA-ICGDER02-Chi in the orthotopic pancreatic tumor model was observed using near-infrared imaging and the human pancreatic cancer orthotopic xenografts were treated with different nanoparticles and normal saline control.Results:The expression of FR in COLO357 was the highest among the six pancreatic cancer cell lines.The FR mainly distributed on cell membrane and fewer in the cytoplasm in pancreatic cancer.Moreover,the absorption rate of the FA-Chi-Gem nanoparticles was more than the Chi nanoparticles without FA modified.The proliferation of COLO357 was significantly inhibited by FA-Chi-Gem nanoparticles.The PEG-FAICGDER02-Chi nanoparticles were enriched in tumor tissue in human pancreatic cancer xenografts,while non-targeted nanoparticles were mainly in normal liver tissue.PEG-FA-Gem-Chi significantly inhibited the growth of human pancreatic cancer xenografts(PEG-FA-Gem-Chi vs.Gem,t=22.950,P=0.000).Conclusions:PEG-FA-FITC-Chi nanoparticles might be an effective targeted drug for treating human FR-positive pancreatic cancer.Jiahua Zhou Junying Wang Qian Xu Shi Xu Jin Wen Zeqian Yu Detong Yang 2013Chinese Journal of Cancer Research2013,25,5:5
3Retrospective analysis of interventional treatment of hepatic metastasis from gastroenteropancreatic neuroendocrine tumors显示文摘Objective: To analyze the angiography appearance of liver metastases from gastroenteropancreatic neuroendocrine tumors(GEP-NETs),and evaluate the clinical efficacy and prognostic factors of interventional treatment for hepatic metastases.Methods: Fifty GEP-NETs patients with hepatic metastases were treated from January 2012 to December 2016,and received transarterial embolization(TAE) in the hepatic tumor or hepatic arterial infusion chemotherapy(HAIC).All patients received 179 times of the intervention therapy in total.Results: Blood supplies were identified in the 50 cases with angiography,which showed that 35 cases had abundant vessels,while 15 cases had poor blood supply.Twenty-two cases were found either collateral blood supply,or portal vein invasion or arterial-portal vein fistula.The best curative efficacy was complete remission(CR)in 4 cases,partial remission(PR) in 28 cases and stable disease(SD) in 18 cases during the process of treatment.The angiography(P=0.047) and the frequency of intervention(P=0.037) showed significantly statistical difference with Kaplan-Meier analysis.The Cox analysis showed that more than 3 times of interventional therapy was an independent prognostic factor.Conclusions: Interventional treatment is safe and effective for GEP-NETs,and is beneficial to patients with main hepatic metastases after endocrine therapy.Peng Liu Xu Zhu Jie Li Ming Lu Jiahua Leng Ying Li Jiangyuan Yu 2017Chinese Journal of Cancer Research2017,29,6:4
4On the Influences of Urbanization on the Extreme Rainfall over Zhengzhou on 20 July 2021: A Convection-Permitting Ensemble Modeling Study显示文摘This study investigates the influences of urban land cover on the extreme rainfall event over the Zhengzhou city in central China on 20 July 2021 using the Weather Research and Forecasting model at a convection-permitting scale[1-km resolution in the innermost domain(d3)].Two ensembles of simulation(CTRL,NURB),each consisting of 11 members with a multi-layer urban canopy model and various combinations of physics schemes,were conducted using different land cover scenarios:(i)the real urban land cover,(ii)all cities in d3 being replaced with natural land cover.The results suggest that CTRL reasonably reproduces the spatiotemporal evolution of rainstorms and the 24-h rainfall accumulation over the key region,although the maximum hourly rainfall is underestimated and displaced to the west or southwest by most members.The ensemble mean 24-h rainfall accumulation over the key region of heavy rainfall is reduced by 13%,and the maximum hourly rainfall simulated by each member is reduced by 15–70 mm in CTRL relative to NURB.The reduction in the simulated rainfall by urbanization is closely associated with numerous cities/towns to the south,southeast,and east of Zhengzhou.Their heating effects jointly lead to formation of anomalous upward motions in and above the planetary boundary layer(PBL),which exaggerates the PBL drying effect due to reduced evapotranspiration and also enhances the wind stilling effect due to increased surface friction in urban areas.As a result,the lateral inflows of moisture and high-θe(equivalent potential temperature)air from south and east to Zhengzhou are reduced.Yali LUO Jiahua ZHANG Miao YU Xudong LIANG Rudi XIA Yanyu GAO Xiaoyu GAO Jinfang YIN 2023Advances in Atmospheric Sciences2023,40,3:4
5Thymosin Alpha-1 Inhibits Complete Freund’s Adjuvant-Induced Pain and Production of Microglia-Mediated Pro-inflammatory Cytokines in Spinal Cord显示文摘Activation of inflammatory responses regulates the transmission of pain pathways through an integrated network in the peripheral and central nervous systems.The immunopotentiator thymosin alpha-1(Tal)has recently been reported to have anti-inflammatory and neuroprotective functions in rodents.However,how Tα1 affects inflammatory pain remains unclear.In the present study,intraperitoneal injection of Tal attenuated complete Freund's adjuvant(CFA)-induced pain hypersensitivity,and decreased the up-regulation of pro-inflammatory cytokines(TNF-α,IL-1β,and IL-6)in inflamed skin and the spinal cord.We found that CFA-induced peripheral inflammation evoked strong microglial activation,but the effect was reversed by Tα1.Notably,Tα1 reversed the CFA-induced up-regulation of vesicular glutamate transporter(VGLUT)and down-regulated the vesicular γ-aminobutyric acid transporter(VGAT)in the spinal cord.Taken together,these results suggest that Tα1 plays a therapeutic role in inflammatory pain and in the modulation of microgliainduced pro-inflammatory cytokine production in addition to mediation of VGLUT and VGAT expression in the spinal cord.Yunlong Xu Yanjun Jiang Lin Wang Jiahua Huang Junmao Wen Hang Lv Xiaoli Wu Chaofan Wan Chuanxin Yu Wenjie Zhang Jiaying Zhao Yinqi Zhou Yongjun Chen 2019Neuroscience Bulletin2019,35,4:4
6Effect of particle motion on the hydraulic collection of coarse spherical particles显示文摘Deep-sea mining,through which shattered coarse solid mineral resources are hydraulically collected and transported,is a promising solution for problems associated with resource exhaustion.This study reports some interesting phenomena observed in a series of upward pumping experiments conducted on a coarse sphere with a 2-cm radius.The sphere was hydraulically lifted using a vertical pipe with a 5-cm radius suspended above the sphere.Remarkably,the tangential motion benefited the collection.The discrete element method-computational fluid dynamics was used to investigate the collection mechanism;this method was used to simulate the collecting processes of the sphere under different initial motion conditions.The simulated results agreed well with the experimental observations.The vortices over the sphere induced by its motion coupled with the main stream mainly provide sufficient lift force to raise the sphere.Yuxiang Chen Hong Xiong Hui Cheng Chunliang Yu Jiahua Xie 2020Acta Mechanica Sinica2020,36,1:4
7Controlling the Balance of Photoluminescence and Photothermal Effect in Cyanostilbene-Based Luminescent Liquid Crystals显示文摘Molecular motions of the luminescent liquid crystals(LLCs)show a significant effect on fluorescent emission and heat generation.In this article,a series of cyanostilbene-based LLCs(CSs:CS1-6,CS1-12,CS2-6 and CS2-12)are synthesized to investigate how the pho-toluminescence and photothermal effect balanced.Among these materials,the mesogens peripheried by single alkyl chains formed enantiotropic nematic(CS1-6)or smectic C(CS1-12)phase with different alkyl tail lengths.When the single aliphatic chain is re-placed by mini-dendrons,room temperature(RT)monotropic hexagonal columnar phase(CS2-12)or molecular liquid(CS2-6)is formed.The results revealed that all these materials exhibited high efficiency emission with the highest quantum yield reaching 59.0%.The photoluminescence and photothermal effect can be effectively tuned by dispersing CSs into a commercially available RT liquid crystal matrix 8CB,which output significantly improved photothermal conversion efficiency of 63.2%.Furthermore,the pho-tothermal can rapidly trigger the Smectic A-Nematic-Isotropic sequence transitions of 8CB doped with CSs.This work paves a way of adjusting the balance of photoluminescence and photothermal properties of the LLC materials.Xiang-Jian Cao Wei Li Jiahua Li Lin Zou Xing-Wang Liu Xiang-Kui Ren Zhen-Qiang Yu 2022Chinese Journal of Chemistry2022,40,8:3
8Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR.Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang 2023Cell Research2023,33,5:2
9An Efficient Priority-Driven Congestion Control Algorithm for Data Center Networks显示文摘With the emerging diverse applications in data centers,the demands on quality of service in data centers also become diverse,such as high throughput of elephant flows and low latency of deadline-sensitive flows.However,traditional TCPs are ill-suited to such situations and always result in the inefficiency(e.g.missing the flow deadline,inevitable throughput collapse)of data transfers.This further degrades the user-perceived quality of service(QoS)in data centers.To reduce the flow completion time of mice and deadline-sensitive flows along with promoting the throughput of elephant flows,an efficient and deadline-aware priority-driven congestion control(PCC)protocol,which grants mice and deadline-sensitive flows the highest priority,is proposed in this paper.Specifically,PCC computes the priority of different flows according to the size of transmitted data,the remaining data volume,and the flows’deadline.Then PCC adjusts the congestion window according to the flow priority and the degree of network congestion.Furthermore,switches in data centers control the input/output of packets based on the flow priority and the queue length.Different from existing TCPs,to speed up the data transfers of mice and deadline-sensitive flows,PCC provides an effective method to compute and encode the flow priority explicitly.According to the flow priority,switches can manage packets efficiently and ensure the data transfers of high priority flows through a weighted priority scheduling with minor modification.The experimental results prove that PCC can improve the data transfer performance of mice and deadline-sensitive flows while guaranting the throughput of elephant flows.Jiahua Zhu Xianliang Jiang Yan Yu Guang Jin Haiming Chen Xiaohui Li Long Qu 2020China Communications2020,17,6:2
10A thermal insulation method for a piezoelectric transducer显示文摘This study deals with the sources of signal distortion of a piezoelectric transducer heated by measured gas flow. These signal distortions originate from both unloading of preload on a piezocrystal because of expansion of a diaphragm in the test apparatus and the pyroelectric effect of a heated piezoelectric crystal. A plastic film on the diaphragm of the transducer can effectively insulate the diaphragm and the piezocrystal within transducer from heating by gas flow, eliminating the sources of distortion.
