| 1 | Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control. | Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu | 2021 | Science China(Life Sciences)2021,64,5: | 46 |
| 2 | Modelling urban expansion using a multi agentbased model in the city of Changsha显示文摘Although traditional urban expansion simulation models can simulate dynamic features, these models fail to address complex changes produced by different agents' behaviors. The paper has built up a set of spatial-temporal land resource allocation rules and developed a dynamic urban expansion model based on a multi-agent system, which can simulate the interaction among different agents, such as residents, peasants, and governments. This model is applied to simulate urban expansion process taking Changsha City, in China as a study area. The results show that this model can not only reflect basic characteristics of urban expansion, but also help explain the reasons for urban expansion process and understand the effect of agents' behavior on the expansion process, and provide insights into the causing factors behind the expansion. In addition, in contrast to simulation results with land use classification map from remote sensing images, the precision of the simulation reached over 68% with higher precision than cellular automata model according to the cell-by-cell comparison. | 3 | Temperature pattern dynamics in shocked porous materials显示文摘The physical fields in porous materials under strong shock wave reaction are very complicated. We simulate such systems using the grain contact material point method. The complex temperature fields in the material are treated with the morphological characterization. To compare the structures and evolution of characteristic regimes under various temperature thresholds, we introduce two concepts, structure similarity and process similarity. It is found that the temperature pattern dynamics may show high similarity under various conditions. Within the same material, the structures and evolution of high-temperature regimes may show high similarity if the shock strength and temperature threshold are chosen appropriately. For process similarity in materials with high porosity, the required temperature threshold increases parabolically with the impact velocity. When the porosity becomes lower, the increasing rate becomes higher. | 4 | A conservative local discontinuous Galerkin method for the solution of nonlinear Schrdinger equation in two dimensions显示文摘In this study, we present a conservative local discontinuous Galerkin(LDG) method for numerically solving the two-dimensional nonlinear Schrdinger(NLS) equation. The NLS equation is rewritten as a firstorder system and then we construct the LDG formulation with appropriate numerical flux. The mass and energy conserving laws for the semi-discrete formulation can be proved based on different choices of numerical fluxes such as the central, alternative and upwind-based flux. We will propose two kinds of time discretization methods for the semi-discrete formulation. One is based on Crank-Nicolson method and can be proved to preserve the discrete mass and energy conservation. The other one is Krylov implicit integration factor(IIF) method which demands much less computational effort. | 5 | Microbial community functional structure in response to micro-aerobic conditions in sulfate-reducing sulfur-producing bioreactor显示文摘Limited oxygen supply to anaerobic wastewater treatment systems had been demonstrated as an effective strategy to improve elemental sulfur(S0) recovery, coupling sulfate reduction and sulfide oxidation. However, little is known about the impact of dissolved oxygen(DO) on the microbial functional structures in these systems. We used a high throughput tool(GeoChip) to evaluate the microbial community structures in a biological desulfurization reactor under micro-aerobic conditions(DO: 0.02–0.33 mg/L). The results indicated that the microbial community functional compositions and structures were dramatically altered with elevated DO levels. The abundances of dsrA/B genes involved in sulfate reduction processes significantly decreased(p < 0.05, LSD test) at relatively high DO concentration(DO: 0.33 mg/L). The abundances of sox and fccA/B genes involved in sulfur/sulfide oxidation processes significantly increased(p < 0.05, LSD test) in low DO concentration conditions(DO: 0.09 mg/L) and then gradually decreased with continuously elevated DO levels. Their abundances coincided with the change of sulfate removal efficiencies and elemental sulfur(S0) conversion efficiencies in the bioreactor. In addition, the abundance of carbon degradation genes increased with the raising of DO levels, showing that the heterotrophic microorganisms(e.g., fermentative microorganisms) were thriving under micro-aerobic condition. | 6 | Sulfonic acid-based metal organic framework functionalized magnetic nanocomposite combined with gas chromatography-electron capture detector for extraction and determination of organochlorine显示文摘The metal organic framework functionalized with sulfonic acid was combined with magnetic nanoparticles to fabricate a new nanocomposite(denoted as Fe3O4@PDA@Zr-SO3H).By combining with gas chromatography-electron capture detector,the resulting Fe3O4@PDA@Zr-SO3H nanocomposite was successfully used as a high-efficiency adsorbent for pre-concentrating eight organochlorine pesticides from water sample in environment.Apart from the ability of fast separation,the as-prepared Fe3O4@PDA@Zr-SO3H nanocomposite also exhibited high adsorption capacity for organochlorine pesticides.With the use of optimal experimental conditions,the linear relationship can be obtained in the range of 0.05~300μg/L,the correlation coefficient was over 0.9978,and the relative standard deviation was located in 2.5%-7.7%.Moreover,the limit of detection and quantification was between0.005-0.016μg/L and 0.017~0.050μg/L.Finally,the nanocomposite was used for the determination of organochlorine pesticides from environmental water samples,and displayed the recovery of 82%-118%. | Ying Wang Qing Ye Menghuan Yu Xijun Zhang Chunhui Deng | 2020 | Chinese Chemical Letters2020,31,7: | 3 |
| 7 | H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus. | Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen | 2022 | National Science Review2022,9,1: | 3 |
| 8 | High-throughput metabolomics reveals the perturbed metabolic pathways and biomarkers of Yang Huang syndrome as potential targets for evaluating the therapeutic effects and mechanism of geniposide显示文摘High-throughput metabolomics can clarify the underlying molecular mechanism of diseases via the qualitative and quantitative analysis of metabolites.This study used the established Yang Huang syndrome(YHS)mouse model to evaluate the efficacy of geniposide(GEN).Urine metabolic data were quantified by ultraperformance liquid chromatography-tandem mass spectrometry.The non-target screening of the massive biological information dataset was performed,and a total of 33 metabolites,including tyramine glucuronide,aurine,and L-cysteine,were identified relating to YHS.These differential metabolites directly participated in the disturbance of phase I reaction and hydrophilic transformation of bilirubin.Interestingly,they were completely reversed by GEN.While,as the auxiliary technical means,we also focused on the molecular prediction and docking results in network pharmacological and integrated analysis part.We used integrated analysis to communicate the multiple results of metabolomics and network pharmacology.This study is the first to report that GEN indirectly regulates the metabolite“tyramine glucuronide”through its direct effect on the target heme oxygenase 1 in vivo.Meanwhile,heme oxygenase-1,a prediction of network pharmacology,was the confirmed metabolic enzyme of phase I reaction in hepatocytes.Our study indicated that the combination of high-throughput metabolomics and network pharmacology is a robust combination for deciphering the pathogenesis of the traditional Chinese medicine(TCM)syndrome. | Heng Fang Aihua Zhang Xiaohang Zhou Jingbo Yu Qi Song Xijun Wang | 2020 | Frontiers of Medicine2020,14,5: | 3 |
