1Induced earthquakes in the development of unconventional energy resources显示文摘It has long been known that human activities such as waste fluid disposal and reservoir impoundment may cause earthquakes. Recently, anthropogenic activities to tackle the increasing energy demand and to address climate change issues are also reported to induce earthquakes. These activities have a common attribute in that fluids are injected and extracted underground and induce spatiotemporal changes of pore pressure and stress, which may cause slip on faults. Induced earthquakes not only pose significant impacts on seismic hazard assessment and preparation, but also raise the question to the society as how to balance the economic needs of resources development and the public's concerns about potential environmental impacts. Here we review the observations of fluid-injection/extraction induced earthquakes, ground deformation associated with these activities, and their physical mechanisms. 2Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells显示文摘RNase III DROSHA is upregulated in multiple cancers and contributes to tumor progression by hitherto unclear mechanisms.Here,we demonstrate that DROSHA interacts withβ-Catenin to transactivate STC1 in an RNA cleavage-independent manner,contributing to breast cancer stem-like cell(BCSC)properties.DROSHA mRNA stability is enhanced by N6-methyladenosine(m^(6)A)modification which is activated by AURKA in BCSCs.AURKA stabilizes METTL14 by inhibiting its ubiquitylation and degradation to promote DROSHA mRNA methylation.Moreover,binding of AURKA to DROSHA transcript further strengthens the binding of the m^(6)A reader IGF2BP2 to stabilize m^(6)A-modified DROSHA.In addition,wild-type DROSHA,but not an m^(6)A methylation-deficient mutant,enhances BCSC stemness maintenance,while inhibition of DROSHA m^(6)A modification attenuates BCSC traits.Our study unveils the AURKA-induced oncogenic m^(6)A modification as a key regulator of DROSHA in breast cancer and identifies a novel DROSHA transcriptional function in promoting the BCSC phenotype.Fei Peng Jie Xu Bai Cui Qilan Liang Sai Zeng Bin He Hong Zou Manman Li Huan Zhao Yuting Meng Jin Chen Bing Liu Shasha Lv Peng Chu Fan An Zifeng Wang Junxiu Huang Yajing Zhan Yuwei Liao Jinxin Lu Lingzhi Xu Jin Zhang Zhaolin Su Zhiguang Li Fangjun Wang Eric W-FLam Quentin Liu 2021Cell Research2021,31,3:9
3Allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci using Staphylococcus aureus Cas9(SaCas9)显示文摘Allele-specific DNA methylation is the most important imprinting marker localized to differentially methylated regions(DMRs),and aberrant genomic imprinted DNA methylation is associated with some human diseases,including Prader-Willi syndrome and cancer.Thus,the development of an effective strategy for the precise editing of allele-specific methylated genes is essential for the functional clarification of imprinting elements and the correction of imprinting disorders in human diseases.To discover a feasible allele-specific genome editing tool based on the CRISPR/Cas system,which is an efficient genetargeting technique in various organisms,we examined the targeting efficiency of Staphylococcus aureus Cas9(SaCas9)and Streptococcus pyogenes Cas9(SpCas9)in response to DNA methylation interference.We found that the targeting efficiency of SaCas9,but not SpCas9,was enhanced by targeted DNA demethylation using the d Cas9-Tet1 catalytic domain(CD)but suppressed by targeted DNA methylation using Dnmt3l-Dnmt3a-d Cas9.An in vitro cleavage assay further demonstrated that SaCas9 nuclease activity was inhibited by 5-methylcytosine(5mC)in a synthesized Cp G-containing context.Further analysis with Ch IP-Q-PCR demonstrated that the non-methylated sequence targeting of Sa Cas9 depends on the binding preference of SaCas9 to non-methylated sequences.Taking advantage of this feature of SaCas9,we have successfully obtained non-methylated allele-biased targeted embryos/mice for two imprinting genes,H19 and Snrpn,with relatively high efficiencies of 28.6%and 47.4%,respectively.These results indicate that the targeting efficiency of SaCas9 was strongly reduced by DNA methylation.By using SaCas9,we successfully achieved allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci.Yajing Liu Jianan Li Changyang Zhou Bin Meng Yu Wei Guang Yang Zongyang Lu Qingmei Shen Yu Zhang Hui Yang Yunbo Qiao 2019Science Bulletin2019,64,21:1
4COVID-19 and cardiovascular complications:updates of emergency medicine显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and SARS-CoV-2 variants,has become a global pandemic resulting in significant morbidity and mortality.Severe cases of COVID-19 are characterized by hypoxemia,hyperinflammation,cytokine storm in lung.Clinical studies have reported an association between COVID-19 and cardiovascular disease(CVD).Patients with CVD tend to develop severe symptoms and mortality if contracted COVID-19 with further elevations of cardiac injury biomarkers.Furthermore,COVID-19 itself can induce and promoted CVD development,including myocarditis,arrhythmia,acute coronary syndrome,cardiogenic shock,and venous thromboembolism.Although the direct etiology of SARS-CoV-2–induced cardiac injury remains unknown and underinvestigated,it is suspected that it is related to myocarditis,cytokine-mediated injury,microvascular injury,and stress-related cardiomyopathy.Despite vaccinations having provided the most effective approach to reducing mortality overall,an adapted treatment paradigm and regular monitoring of cardiac injury biomarkers is critical for improving outcomes in vulnerable populations at risk for severe COVID-19.In this review,we focus on the latest progress in clinic and research on the cardiovascular complications of COVID-19 and provide a perspective of treating cardiac complications deriving from COVID-19 in emergency medicine.Jianli Zhao Yaoli Xie Zhijun Meng Caihong Liu Yalin Wu Fuje Zhao Xinliang Ma Theodore A.Christopher Bernard J.Lopeza Yajing Wang 2023Emergency and Critical Care Medicine2023,3,3:0
5The repression of oncoprotein SET by the tumor suppressor p53 reveals a p53-SET-PP2A feedback loop for cancer therapy显示文摘The oncoprotein SET is frequently overexpressed in many types of tumors and contributes to malignant initiation and progression through multiple mechanisms,including the hijacking of the tumor suppressors p53 and PP2A.Targeting aberrant SET represents a promising strategy for cancer intervention.However,the mechanism by which endogenous SET is regulated in cancer cells remains largely unknown.Here,we identified the tumor suppressor p53 as a key regulator that transcriptionally repressed the expression of SET in both normal and cancer cells.In addition,p53 stimulated PP2A phosphatase activity via p53-mediated transcriptional repression of SET,whereby SET-mediated inhibition of PP2A was alleviated.Moreover,targeting the interaction between SET and PP2A catalytic subunit(PP2Ac)with FTY720 enhanced stress-induced p53 activation via PP2A-mediated dephosphorylation of p53 on threonine 55(Thr55).Therefore,our findings uncovered a previously unknown p53-SETPP2A regulatory feedback loop.To functionally potentiate this feedback loop,we designed a combined therapeutic strategy by simultaneously administrating a p53 activator and SET antagonist in cancer cells and observed a dramatic synergistic effect on tumor suppression.Our study reveals mechanistic insight into the regulation of the oncoprotein SET and raises a potential strategy for cancer therapy by stimulating the p53-SET-PP2A feedback loop.Han Yao Wenbin Xu Yajing Liu Zhijie Cao Jia Wen Mi Zhang Zhen Wu Xiaojun Yan Zishan Jiao Zijing Zhang Jianyuan Chen Meng Zhang Wei-Guo Zhu Donglai Wang 2023Science China(Life Sciences)2023,66,1:0
6Oxygen doping modulating thermal-activated charge transport of reduced graphene oxide for high performance temperature sensors显示文摘Graphene and its derivatives have sparked intense research interest in wearable temperature sensing due to their excellent electric properties,mechanical flexibility,and good biocompatibility.Despite these ad-vantages,the weak temperature dependence of charge transport makes them difficult to achieve a highly sensitive temperature response,which is one of the remaining bottlenecks in the progress towards practi-cal applications.Unfortunately,detailed knowledge about the key factors of the charge transport temper-ature dependence in this material that determines the critical performance of electrical sensors is very limited up to now.Here,we reveal that oxygen absorption on the ultrathin reduced graphene oxide(RGO)films(~3 nm)can significantly increase their conductance activation energy over 200%and thus greatly improve the temperature dependence of thermal-activated charge transport.Further investigations sug-gest that oxygen introduces the deep acceptor states,distributed at an energy level~0.175ev from the valence-band maximum,which allows a highly temperature-dependent impurity ionization process and the resulting vast holes release in a wide temperature range.Remarkably,our temperature sensors based on oxygen-doped ultrathin RGO films show a high sensitivity with temperature conductive coefficient of 14.58%K^(-1),which is one order of magnitude higher than the reported CNT or graphene-based devices.Moreover,the ultrathin thickness and high thermal conductivity of RGo film allow an ultrafast response time of~86ms,which represents the best level of temperature sensors based on soft materials.Profit-ing from these advantages,our sensors show good capacity to identify the slight temperature difference of human body,monitor respiratory rate,and detect the environmental temperature.This work not only represents substantial performance advances in temperature sensing,but also provides a new approach to modulate the charge transport temperature dependence,which could be benefited to both device design and fundamental research.Liqian Yuan Zhongwu Wang Yancheng Meng Shuguang Wang Yajing Sun Yinan Huang Liqiang Li Wenping Hu 2023Chinese Chemical Letters2023,34,12:0
7FOLR1-induced folate deficiency reduces viral replication via modulating APOBEC3 family expression显示文摘Folate receptor alpha(FOLR1)is vital for cells ingesting folate(FA).FA plays an indispensable role in cell pro-liferation and survival.However,it is not clear whether the axis of FOLR1/FA has a similar function in viral replication.In this study,we used vesicular stomatitis virus(VSV)to investigate the relationship between FOLR1-mediated FA deficiency and viral replication,as well as the underlying mechanisms.We discovered that FOLR1 upregulation led to the deficiency of FA in HeLa cells and mice.Meanwhile,VSV replication was notably sup-pressed by FOLR1 overexpression,and this antiviral activity was related to FA deficiency.Mechanistically,FA deficiency mainly upregulated apolipoprotein B mRNA editing enzyme catalytic subunit 3B(APOBEC3B)expression,which suppressed VSV replication in vitro and in vivo.In addition,methotrexate(MTX),an FA metabolism inhibitor,effectively inhibited VSV replication by enhancing the expression of APOBEC3B in vitro and in vivo.Overall,our present study provided a new perspective for the role of FA metabolism in viral infections and highlights the potential of MTX as a broad-spectrum antiviral agent against RNA viruses.Jing Wu Yajing Han Ruining Lyu Fang Zhang Na Jiang Hongji Tao Qiao You Rui Zhang Meng Yuan Waqas Nawaz Deyan Chen Zhiwei Wu 2023Virologica Sinica2023,38,3:0

9Continuity of the robustness of contextuality of empirical models显示文摘Recently, the robustness of contextuality(RoC) of an empirical model was discussed in [Sci. China-Phys. Mech. Astron. 59,640303(2016)], many important properties of the RoC have been proved except for its boundedness and continuity. The aim of this paper is to find an upper bound for the RoC over all of empirical models and prove that the RoC is a continuous function on the set of all empirical models. Lastly, a relationship between the RoC and the extent of violating the noncontextual inequalities is established for an n-cycle contextual box. 10Postpartum depression and major depressive disorder:the same or not?Evidence from resting-state functional MRI显示文摘Background Although postpartum depression(PPD)and non-peripartum major depressive disorder(MDD)occurring within and outside the postpartum period share many clinical characteristics,whether PPD and MDD are the same or not remains controversial.Methods The current study was devoted to identify the shared and different neural circuits between PPD and MDD by resting-state functionalmagnetic resonance imaging data from 77 participants(22 first-episodic drug-naleMDD,26 drug-nale PPD,and 29 healthy controls(HC)).Results Both the PPD andMDD groups exhibited higher fractional amplitude of low-frequency fluctuation(fALFF)in left temporal pole relative to the HC group;the MDD group showed specifically increased degree centrality in the right cerebellum while PPD showed specifically decreased fALFF in the left supplementary motor area and posterior middle temporal gyrus(pMTG_L),and specifically decreased functional connectivities between pMTG and precuneus and between left subgeneual anterior cingulate cortex(sgACC_L)and right sgACC.Moreover,sgACC and left thalamus showed abnormal regional homogeneity of functional activities between any pair of HC,MDD,and PPD.Conclusions These results provide initial evidence that PPD and MDD have common and distinct neural circuits,which may facilitate understanding the neurophysiological basis and precision treatment for PPD.Bochao Cheng Yi Guo Xijian Chen Bin Lv Yi Liao Haibo Qu Xiao Hu Haoxiang Yang Yajing Meng Wei Deng Jiaojian Wang 2022Psychoradiology2022,2,3:0
