Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies. However, few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases. In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms. Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes, like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors, including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes. In summary, our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications. | Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu | 2021 | Acta Pharmaceutica Sinica B2021,11,11: | 15
Ce and Er Co-doped TiO2 for rapid bacteria- killing using visible light显示文摘Bacterial infection and related diseases are threatening the health of human beings.Photocatalytic disinfection as a simple and low-cost disinfection strategy is attracting more and more attention. In this work,TiO2 nanoparticles(NPs)were modified by co-doping of Ce and Er using the sol-gel method, which endowed TiO2 NPs with enhanced visible light photocatalytic performance but not pure ultraviolet photocatalytic properties compared the untreated TiO2.Our results disclosed that as the doping content of Er increased, the photocatalytic activity of modified TiO2 NPs initially increased and subsequently decreased. The same trend occurred for Ce doping. When the doping dose of Er and Ce is 0.5 mol%and 0.2 mol%,the 0.5Ce0.2Ti-O calcined at 800℃ presented the best antibacterial properties, with the antibacterial efficiency of 91.23%and 92.8%for Staphylococcus aureus and Escherichia coli, respectively. The existence of Er ions is thought to successfully turn the near-infrared radiation into visible region, which is easier to be absorbed by TiO2 NPs.Meanwhile, the addition of Ce ions can effectively extend spectral response range and inhibit the recombination of electrons and holes, enhancing the photocatalytic disinfection activity of co-doped TiO2. | Yawei Ren Yajing Han Zhaoyang Li Xiangmei Liu Shengli Zhu Yanqin Liang Kelvin Wai Kwok Yeung Shuilin Wu | 2020 | Bioactive Materials2020,5,2: | 7
An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. Combined analyses of laboratory biomarkers with their clinical manifestations before and after salvage treatment showed that the persistent immunosurveillance mediated by CAR-T-19 cells would inevitably potentiate the leukemia-killing effectiveness of subsequent chemotherapy in patients who showed relapse after CAR-T-19-induced remission. | Yajing Zhang Wenying Zhang Hanren Dai Yao Wang Fengxia Shi Chunmeng Wang Yelei Guo Yang Liu Meixia Chen Kaichao Feng Yan Zhang Chuanjie Liu Qingming Yang Suxia Li Weidong Han | 2016 | Science China(Life Sciences)2016,59,4: | 6
Formation of Bulk Intermetallic Compound AgaSn in Slowly-Cooled Lead-Free Sn-4.0 wt pct Ag Solders显示文摘Sn-Ag alloy system has been regarded as one of the most promising lead-free solder to substitute conventional Sn-Pb eutectic solder. But the formation of bulk Ag3Sn intermetallic compounds (IMCs) during reflow and post heattreatment significantly influences the performance of the solder joints. With an effort to clarify its microstructuralevolution as a function of slow cooling rates, the fraction of bulk IMCs within the slowly solidified Sn-4.0 wt pctAg solder was investigated by standard metallographic and compared with that detected by thermal analysis. It wasfound that the bulk IMCs fraction determined by thermal analysis corresponds quite well with the microstructureobservation results. In accordance with the conventional solidification theory, the lower the applied cooling rate, thefewer the amount of bulk Ag3Sn IMCs formed in Sn-4.0 wt pct Ag alloy. In addition, Vickers hardness measurementresults indicated that the relative coarse eutectic Ag3Sn IMCs distributing in the lamellar eutectic structure favoredthe improvement of the mechanical performance. | Jun SHEN+, Yongchang LIU, Yajing HAN, Peizhen ZHANG and Houxiu GAO College of Materials Science & Engineering, Tianjin University, Tianjin 300072, China | 2005 | Journal of Materials Science & Technology2005,21,6: | 4
Carbonaceous and nitrogenous disinfection byproduct precursor variation during the reversed anaerobic–anoxic–oxic process of a sewage treatment plant显示文摘Disinfection byproduct(DBP)precursors in wastewater during the reversed anaerobic–anoxic–oxic(A^2/O)process,as well as their molecular weight(MW)and polarity-based fractions, were characterized with UV scanning, fluorescence excitation emission matrix,Fourier transform infrared and nuclear magnetic resonance spectroscopy.Their DBP formation potentials(DBPFPs)after chlorination were further tested. Results indicated that the reversed A^2/O process could not only effectively remove the dissolved organic carbon(DOC)and dissolved total nitrogen in the wastewater, but also affect the MW distribution and hydrophilic–hydrophobic properties of dissolved organic matter(DOM).The accumulation of low MW and hydrophobic(HPO)DOM was possibly due to the formation of soluble microbial product-like(SMP-like)matters in the reversed A^2/O treatment, especially in the anoxic and aerobic processes. Moreover,DOM in the wastewater displayed a high carbonaceous disinfection byproduct formation potential(C-DBPFP)in the fractions of MW>100 k Da and MW<5 k Da,and revealed an increasing tendency of nitrogenous disinfection byproduct formation potential(N-DBPFP)with decrease of MW.For polarity-based fractions, the HPO fraction of wastewater showed significantly higher C-DBPFP and N-DBPFP than hydrophilic and transphilic fractions. Therefore, although the reversed A^2/O process could remove most DBP precursors by DOC reduction,it led to the enhancement of DBPFP with the formation and accumulation of low MW and HPO DOM.In addition, strong correlations between C-DBPFPs and SUVA,and between N-DBPFPs and DON/DOC,were observed in the wastewater, which might be helpful for DBPFP prediction in wastewater and reclaimed water chlorination. | Huihui Han Hengfeng Miao Yajing Zhang Minfeng Lu Zhenxing Huang Wenquan Ruan | 2018 | Journal of Environmental Sciences2018,30,3: | 3
2022 Chinese expert consensus and guidelines on clinical management of toxicity in anti-CD19 chimeric antigen receptor T-cell therapy for B-cell non-Hodgkin lymphoma显示文摘Adoptive cellular immunotherapy with chimeric antigen receptor(CAR)T cells has emerged as a novel modality for treating relapsed and/or refractory B-cell non-Hodgkin lymphoma(B-NHL).With increasing approval of CAR T-cell products and advances in CAR T cell therapy,CAR T cells are expected to be used in a growing number of cases. However,CAR T-cell-associated toxicities can be severe or even fatal, thus compromising the survival benefit from this therapy.Standardizing and studying the clinical management of these toxicities are imperative. In contrast to other hematological malignancies, such as acute lymphoblastic leukemia and multiple myeloma,anti-CD19 CAR T-cell-associated toxicities in B-NHL have several distinctive features, most notably local cytokine-release syndrome(CRS).However, previously published guidelines have provided few specific recommendations for the grading and management of toxicities associated with CAR T-cell treatment for B-NHL.Consequently,we developed this consensus for the prevention, recognition, and management of these toxicities,on the basis of published literature regarding the management of anti-CD19 CAR T-cell-associated toxicities and the clinical experience of multiple Chinese institutions. This consensus refines a grading system and classification of CRS in B-NHL and corresponding measures for CRS management, and delineates comprehensive principles and exploratory recommendations for managing anti-CD19 CAR T-cell-associated toxicities in addition to CRS. | Ping Li Yang Liu Yun Liang Jian Bo Sujun Gao Yongxian Hu Yu Hu He Huang Xiaojun Huang Hongmei Jing Xiaoyan Ke Jianyong Li Yuhua Li Qifa Liu Peihua Lu Heng Mei Ting Niu Yongping Song Yuqin Song Liping Su Sanfang Tu Jianxiang Wang Depei Wu Zhao Wang Kailin Xu Zhitao Ying Qingming Yang Yajing Zhang Fengxia Shi Bin Zhang Huilai Zhang Xi Zhang Mingfeng Zhao Weili Zhao Xiangyu Zhao Liang Huang Jun Zhu Wenbin Qian Weidong Han Aibin Liang | 2023 | Cancer Biology & Medicine2023,20,2: | 3
The model of cytokine release syndrome in CAR T-cell treatment for B-cell non-Hodgkin lymphoma显示文摘Chimeric antigen receptor T(CAR T)cell therapy has demonstrated efficacy in the treatment of haematologic malignancies. However, the accompanying adverse events, the most common of which is cytokine release syndrome(CRS),substantially limit its wide application.Due to its unique physiological characteristics,CRS in CAR T-cell treatment for B-cell non-Hodgkin lymphoma(BNHL)may exhibit some special features.Although existing guidelines had greatly promoted the recognition and management of CRS,many recommendations are not fully applicable to B-NHL.Therefore,it is imperative to identify responses that are specific to CRS observed following CAR T treatment for B-NHL.Based on underlying biological processes and known pathophysiological mechanisms,we tentatively propose a new model to illustrate the occurrence and evolution of CAR T-cell-therapy-related CRS in BNHL.In this model, tumour burden and bone marrow suppression are considered determinants of CRS.Novel phenomena after CAR T-cell infusion(such as local inflammatory response)are further identified. The proposed model will help us better understand the basic biology of CRS and recognize and manage it more rationally. | Jianshu Wei Yang Liu Chunmeng Wang Yajing Zhang Chuan Tong Guanghai Dai Wei Wang John E.J.Rasko J.Joseph Melenhorst Wenbin Qian Aibin Liang Weidong Han | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 2

Biocompatible chitosan/polyethylene glycol/multi-walled carbon nanotube composite scaffolds for neural tissue engineering显示文摘Carbon nanotube(CNT)composite materials are very attractive for use in neural tissue engineering and biosensor coatings.CNT scaffolds are excellent mimics of extracellular matrix due to their hydrophilicity, viscosity, and biocompatibility.CNTs can also impart conductivity to other insulating materials improve mechanical stability guide neuronal cell behavior and trigger axon regeneration. The performance of chitosan(CS)/polyethylene glycol(PEG)composite scaffolds could be optimized by introducing multi-walled CNTs(MWCNTs).CS/PEG/CNT composite scaffolds with CNT content of 1%,3%,and 5%(1%=0.01 g/mL)were prepared by freeze-drying.Their physical and chemical properties and biocompatibility were evaluated.Scanning electron microscopy(SEM)showed that the composite scaffolds had a highly connected porous structure.Transmission electron microscope(TEM)and Raman spectroscopy proved that the CNTs were well dispersed in the CS/PEG matrix and combined with the CS/PEG nanofiber bundles.MWCNTs enhanced the elastic modulus of the scaffold. The porosity of the scaffolds ranged from 83%to 96%.They reached a stable water swelling state within 24 h,and swelling decreased with increasing MWCNT concentration. The electrical conductivity and cell adhesion rate of the scaffolds increased with increasing MWCNT content.Immunofluorescence showed that rat pheochromocytoma(PC12)cells grown in the scaffolds had characteristics similar to nerve cells. We measured changes in the expression of nerve cell markers by quantitative real-time polymerase chain reaction(qRT-PCR),and found that PC12 cells cultured in the scaffolds expressed growth-associated protein 43(GAP43),nerve growth factor receptor(NGFR),and class IIIβ-tubulin(TUBB3)proteins.Preliminary research showed that the prepared CS/PEG/CNT scaffold has good biocompatibility and can be further applied to neural tissue engineering research. | Shengbo SANG Rong CHENG Yanyan CAO Yayun YAN Zhizhong SHEN Yajing ZHAO Yanqing HAN | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,1: | 2
Deep white matter hyperintensity is spatially correlated to MRI-visible perivascular spaces in cerebral small vessel disease on 7 Tesla MRI显示文摘Background The association between perivascular space(PVS)and white matter hyperintensity(WMH)has been unclear.Normal-appearing white matter(NAWM)around WMH is also found correlated with the development of focal WMH.This study aims to investigate the topological connections among PVS,deep WMH(dWMH)and NAWM around WMH using 7 Tesla(7T)MRI.Methods Thirty-two patients with non-confluent WMHs and 16 subjects without WMHs were recruited from our department and clinic. We compared the PVS burden between patients with and without WMHs using a 5-point scale. Then, the dilatation and the number of PVS within a radius of 1 cm around each dWMH were compared with those of a reference site(without WMH)in the contralateral hemisphere. In this study,we define NAWM as an area within the radius of 1 cm around each dWMH.Furthermore,we assessed the spatial relationship between dWMH and PVS.Results Higher PVS scores in the centrum semiovale were found in patients with>5 dWMHs(median 3)than subjects without dWMH(median 2,p=0.014).We found there was a greater dilatation and a higher number of PVS in NAWM around dWMH than at the reference sites(p<0.001,p<0.001).In addition,79.59%of the dWMHs were spatially connected with PVS.Conclusion dWMH,NAWM surrounding WMH and MRI-visible PVS are spatially correlated in the early stage of cerebral small vessel disease.Future study of WMH and NAWM should not overlook MRI-visible PVS. | Yajing Huo Yilin Wang Cen Guo Qianyun Liu Lili Shan Mingyuan Liu Haibo Wu Guanwu Li Huihui Lv Lingdan Lu Yintin Zhou Jianfeng Feng Yan Han | 2023 | Stroke & Vascular Neurology2023,8,2: | 2

Effects of rapid solidification on the microstructure and microhardness of a lead-free Sn-3.5Ag solder显示文摘A lead-free Sn-3.5Ag solder was prepared by rapid solidification technology. The high solidification rate, ob-tained by rapid cooling, promotes nucleation, and suppresses the growth of Ag3Sn intermetallic compounds (IMCs) in Ag-rich zone, yielding fine Ag3Sn nanoparticulates with spherical morphology in the matrix of the solder. The large amount of tough homogeneously-dispersed IMCs helps to improve the surface area per unit volume and obstructs the dislocation lines passing through the solder, which fits with the dispersion-strengthening theory. Hence, the rapidly-solidified Sn-3.5Ag solder exhibits a higher microhardness when compared with a slowly-solidified Sn-3.5Ag solder. | SHEN Jun LIU Yongchang Han Yajing GAO Houxiu | 2006 | Rare Metals2006,25,4: | 1

Colony-stimulating factor 3 receptor (CSF3R) M696T mutation does not impact on clinical outcomes of a Ph+ acute lymphoblastic leukemia patient显示文摘Colony-stimulating factor 3 receptor(CSF3R)mutations have been identified in a variety of myeloid disorders.Although CSF3R point mutations(eg,T618I)are emerging as key players in chronic neutrophilic leukemia/atypical chronic myelogenous leukemia, the significance of rarer CSF3R mutations is unknown. Here,we report a 32-year-old female who was diagnosed as Philadelphia chromosome-positive acute lymphoblastic leukemia(Ph^(+)ALL)with the CSF3R M696T mutation and was undergone unrelated donor hematopoietic stem cell transplantation. The patient achieved complete remission with chemotherapy in combination with tyrosine kinase inhibitor(TKI)and long-term survival by unrelated donor transplantation.Meanwhile,we performed a series of experiments using murine interleukin 3(IL-3)-dependent Ba/F3 cell line to evaluate the transforming capacity of the CSF3R M696T mutation. We confirmed the presence of a CSF3R M696T germline mutation in this patient which was inherited from her mother. The in vitro experiment results showed that the CSF3R M696T mutation contributes marginally to the tumor transformation of Ba/F3 cells, indicating that CSF3R M696T mutation was neutral in tumor transformation ability. We concluded that TKI is effective in patients with the CSF3R M696T mutation in Ph+ALL and donors with CSF3R M696T mutation might still be selected as the candidate for transplantation. | Xin Chen Bichen Wang Aiming Pang Weiping Yuan Erlie Jiang Yajing Chu Sizhou Feng Mingzhe Han | 2021 | Blood Science2021,3,3: | 0
The repression of oncoprotein SET by the tumor suppressor p53 reveals a p53-SET-PP2A feedback loop for cancer therapy显示文摘The oncoprotein SET is frequently overexpressed in many types of tumors and contributes to malignant initiation and progression through multiple mechanisms, including the hijacking of the tumor suppressors p53 and PP2A.Targeting aberrant SET represents a promising strategy for cancer intervention. However, the mechanism by which endogenous SET is regulated in cancer cells remains largely unknown. Here,we identified the tumor suppressor p53 as a key regulator that transcriptionally repressed the expression of SET in both normal and cancer cells. In addition,p53 stimulated PP2A phosphatase activity via p53-mediated transcriptional repression of SET,whereby SET-mediated inhibition of PP2A was alleviated. Moreover, targeting the interaction between SET and PP2A catalytic subunit(PP2Ac)with FTY720 enhanced stress-induced p53 activation via PP2A-mediated dephosphorylation of p53 on threonine 55(Thr55).Therefore,our findings uncovered a previously unknown p53-SETPP2A regulatory feedback loop.To functionally potentiate this feedback loop,we designed a combined therapeutic strategy by simultaneously administrating a p53 activator and SET antagonist in cancer cells and observed a dramatic synergistic effect on tumor suppression. Our study reveals mechanistic insight into the regulation of the oncoprotein SET and raises a potential strategy for cancer therapy by stimulating the p53-SET-PP2A feedback loop. | Han Yao Wenbin Xu Yajing Liu Zhijie Cao Jia Wen Mi Zhang Zhen Wu Xiaojun Yan Zishan Jiao Zijing Zhang Jianyuan Chen Meng Zhang Wei-Guo Zhu Donglai Wang | 2023 | Science China(Life Sciences)2023,66,1: | 0

FOLR1-induced folate deficiency reduces viral replication via modulating APOBEC3 family expression显示文摘Folate receptor alpha(FOLR1)is vital for cells ingesting folate(FA).FA plays an indispensable role in cell pro-liferation and survival. However,it is not clear whether the axis of FOLR1/FA has a similar function in viral replication. In this study,we used vesicular stomatitis virus(VSV)to investigate the relationship between FOLR1-mediated FA deficiency and viral replication,as well as the underlying mechanisms. We discovered that FOLR1 upregulation led to the deficiency of FA in HeLa cells and mice.Meanwhile,VSV replication was notably sup-pressed by FOLR1 overexpression, and this antiviral activity was related to FA deficiency.Mechanistically,FA deficiency mainly upregulated apolipoprotein B mRNA editing enzyme catalytic subunit 3B(APOBEC3B)expression,which suppressed VSV replication in vitro and in vivo.In addition, methotrexate(MTX),an FA metabolism inhibitor, effectively inhibited VSV replication by enhancing the expression of APOBEC3B in vitro and in vivo.Overall, our present study provided a new perspective for the role of FA metabolism in viral infections and highlights the potential of MTX as a broad-spectrum antiviral agent against RNA viruses. | Jing Wu Yajing Han Ruining Lyu Fang Zhang Na Jiang Hongji Tao Qiao You Rui Zhang Meng Yuan Waqas Nawaz Deyan Chen Zhiwei Wu | 2023 | Virologica Sinica2023,38,3: | 0
