1Induced earthquakes in the development of unconventional energy resources显示文摘It has long been known that human activities such as waste fluid disposal and reservoir impoundment may cause earthquakes. Recently, anthropogenic activities to tackle the increasing energy demand and to address climate change issues are also reported to induce earthquakes. These activities have a common attribute in that fluids are injected and extracted underground and induce spatiotemporal changes of pore pressure and stress, which may cause slip on faults. Induced earthquakes not only pose significant impacts on seismic hazard assessment and preparation, but also raise the question to the society as how to balance the economic needs of resources development and the public's concerns about potential environmental impacts. Here we review the observations of fluid-injection/extraction induced earthquakes, ground deformation associated with these activities, and their physical mechanisms. 2Recombinant rabies virus expressing IL-15 enhances immunogenicity through promoting the activation of dendritic cells in mice显示文摘Rabies remains a public health threat that kills approximately 59,000 people worldwide each year,most of which are from the developing countries of Africa and Asia where dog rabies are endemic.Therefore, developing an affordable and efficacious vaccine is crucial for rabies control in these countries. Interleukin(IL)-15, an immunoregulatory cytokine, is a pluripotent molecule with therapeutic potential, which targets many cell types and links the innate and adaptive immune system. In this study, IL-15 gene was cloned and inserted into the genome of a recombinant rabies virus(RABV) strain LBNSE(designated as LBNSE-IL15), and the effect of over-expression of IL-15 on the immunogenicity of RABV was investigated. It was found that mice vaccinated with LBNSEIL15 could induce significantly higher level of virus-neutralizing antibody(VNA) than those immunized with LBNSE, resulting in the higher protection after challenge. Further investigation was performed to find out the possible role of IL-15 plays in the process of antibody induction, and it was found that LBNSE-IL15 could enhance the maturation of dendritic cells(DCs) in immunized mice. Furthermore, the mice immunized with LBNSE-IL15 could promote the T_(FH) cells differentiation and the generation of germinal center B cells and plasma cells. 3Nuclear Aurora kinase A triggers programmed death-ligand 1-mediated immune suppression by activating MYC transcription in triple-negative breast cancer显示文摘Background:Increasing studies have reported that oncogenes regulate components of the immune system,suggesting that this is a mechanism for tumorigenesis.Aurora kinase A(AURKA),a serine/threonine kinase,is involved in cell mitosis and is essential for tumor cell proliferation,metastasis,and drug resistance.However,the mechanism by which AURKA is involved in immune response regulation is unclear.Therefore,this study aimed to investigate the role of AURKA in immune regulation in triple-negative breast cancer(TNBC).Methods:Peripheral blood mononuclear cells(PBMCs)were co-cultured with TNBC cells.The xCELLigence Real-Time Cell Analyzer-MP system was used to detect the killing efficiency of immune cells on TNBC cells.The expression of immune effector molecules was tested by quantitative real-time polymerase chain reaction(qRT-PCR)to evaluate immune function.Furthermore,to validate AURKA-regulated immune response in vivo,4T1 murine breast cancer cell line with AURKA overexpression or downregulation was engrafted into BALB/c mice.The distribution and proportion of immune cells in tumors were further evaluated by immunohistochemistry and flow cytometry.Results:Downregulation of AURKA in TNBC cells increased immune response by activating CD8^(+)T cell proliferation and activity.Nuclear rather than cytoplasmic AURKA-derived programmed death-ligand 1(PD-L1)expression was independent of its kinase activity.Mechanistic investigations showed that nuclear AURKA increased PD-L1 expression via an MYC-dependent pathway.PD-L1 overexpression mostly reversed AURKA silencing-induced expression of immune effector molecules,including interleukin-(IL-2),interferon-γ(IFN-γ),and perforin.Moreover,AURKA expression was negatively correlated with the enrichment and activity of tumor-infiltrating CD8^(+)T cells in 4T1 engrafted BALB/c mouse model.Conclusions:Nuclear AURKA elevated PD-L1 expression via an MYCdependent pathway and contributed to immune evasion in TNBC.Therapies targeting nuclear AURKA may restore immune responses against tumors.Shulan Sun Wei Zhou Xiaoxi Li Fei Peng Min Yan Yajing Zhan Fan An Xiaoyan Li Yunyong Liu Quentin Liu Haozhe Piao 2021Cancer Communications2021,41,9:2

6Oxidative coupling of cinnamic acid derivatives and their radical-scavenging activities显示文摘4-Hydroxycinnamic acid derivatives,including ferulic acid (FA) (1),sinapic acid (SA) (2) and caffeic acid (CA) (3),are widely distributed in the plant kingdom,and undergo oxidative cross-coupling leading to the corresponding dehydrodimers,trimers and even higher oligomers in plants. In order to evaluate the antioxidative ability of these 4-hydroxycinnamic acid derivatives and their oligomers,we synthesized 8-8'-bis-lactone-dimers (8-8'-DiFA (4),8-8'-DiSA (5) and 8-8'-DiCA (6)),as well as a new trimer (7),by the reaction of the corresponding monomers (1-3) with Ag2O in methanol,and assayed their free radical-scavenging activity by the reaction with 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·). 7Deep white matter hyperintensity is spatially correlated to MRI-visible perivascular spaces in cerebral small vessel disease on 7 Tesla MRI显示文摘Background The association between perivascular space(PVS)and white matter hyperintensity(WMH)has been unclear.Normal-appearing white matter(NAWM)around WMH is also found correlated with the development of focal WMH.This study aims to investigate the topological connections among PVS,deep WMH(dWMH)and NAWM around WMH using 7 Tesla(7T)MRI.Methods Thirty-two patients with non-confluent WMHs and 16 subjects without WMHs were recruited from our department and clinic.We compared the PVS burden between patients with and without WMHs using a 5-point scale.Then,the dilatation and the number of PVS within a radius of 1 cm around each dWMH were compared with those of a reference site(without WMH)in the contralateral hemisphere.In this study,we define NAWM as an area within the radius of 1 cm around each dWMH.Furthermore,we assessed the spatial relationship between dWMH and PVS.Results Higher PVS scores in the centrum semiovale were found in patients with>5 dWMHs(median 3)than subjects without dWMH(median 2,p=0.014).We found there was a greater dilatation and a higher number of PVS in NAWM around dWMH than at the reference sites(p<0.001,p<0.001).In addition,79.59%of the dWMHs were spatially connected with PVS.Conclusion dWMH,NAWM surrounding WMH and MRI-visible PVS are spatially correlated in the early stage of cerebral small vessel disease.Future study of WMH and NAWM should not overlook MRI-visible PVS.Yajing Huo Yilin Wang Cen Guo Qianyun Liu Lili Shan Mingyuan Liu Haibo Wu Guanwu Li Huihui Lv Lingdan Lu Yintin Zhou Jianfeng Feng Yan Han 2023Stroke & Vascular Neurology2023,8,2:2

10Detection and Analysis of Pesticide Residues in Chinese Chives and Risk Assessment of Dietary Intake显示文摘[Objectives]This study was conducted to understand the status of pesticide residues and dietary intake risk of Chinese chives in Tangshan area. [Methods] Sixty eight kinds of pesticide residues in 415 Chinese chive samples collected from Tangshan area were qualitatively and quantitatively determined by high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) and gas chromatography(GC) in 2020. [Results] The results showed that 41 kinds of pesticide residues were detected in the 415 Chinese chive samples, and the detection rate was 69.4%(288/415), and there was a combination of pesticides in many samples. According to the National Food Safety Standard―Maximum Residue Limits of Pesticides in Food(GB 2763-2019), the residues of 12 pesticides exceeded the maximum residue limits(MRLs), and the unqualified rate was 38.07%(158/415). The highest detection rate of clothianidin was 41.20%(171/415), but there was no MRL in GB 2763-2019. The next was procymidone, the detection rate of which was 35.42%(147/415), and the over-standard rate was 30.12%(125/415). Forbidden and restricted pesticides were detected in some samples. According to the dietary exposure risk assessment, the NEDI/ADI values were all less than 1 and the intake risk was within acceptable range. In Tangshan area, the types of pesticides used in Chinese chive production are complex, and there are risks of multi-residue pollution and use of banned and restricted pesticides and unregistered pesticides. It is suggested that routine monitoring of pesticide residues and management of pesticide use should be strengthened to ensure the quality and safety of agricultural products. 13Multi-objective optimization of methane production system from biomass through anaerobic digestion显示文摘This work addressed the multi-objective optimization of a biogas production system considering both environmental and economic criteria. A mixed integer non-linear programming(MINLP) model was established and solved with non-dominated sorting genetic algorithm Ⅱ, from which the Pareto fronts, the optimal technology combinations and operation conditions were obtained and analyzed. It's found that the system is feasible in both environmental and economic considerations after optimization. The most expensive processing section is decarbonization; the most expensive equipment is anaerobic digester; the most power-consuming processing section is digestion, followed by decarbonization and waste management. The positive green degree value on the process is attributed to processing section of digestion and waste management. 3:1 chicken feces and corn straw, solar energy, pressure swing adsorption and 3:1 chicken feces and rice straw, solar energy, pressure swing adsorption are turned out to be two robust technology combinations under different prices of methane and electricity by sensitivity analysis. 14Green,Sustainable Architectural Bamboo with High Light Transmission and Excellent Electromagnetic Shielding as a Candidate for Energy-Saving Buildings显示文摘Currently,light-transmitting,energy-saving,and electromagnetic shielding materials are essential for reducing indoor energy consumption and improving the electromagnetic environment.Here,we developed a cellulose composite with excellent optical transmittance that retained the natural shape and fiber structure of bamboo.The modified whole bamboo possessed an impressive optical transmittance of approximately 60%at 6.23 mm,illuminance of 1000 luminance(lux),water absorption stability(mass change rate less than 4%),longitudinal tensile strength(46.40 MPa),and surface properties(80.2 HD).These were attributed to not only the retention of the natural circular hollow structure of the bamboo rod on the macro,but also the complete bamboo fiber skeleton template impregnated with UV resin on the micro.Moreover,a multilayered device consisting of translucent whole bamboo,transparent bamboo sheets,and electromagnetic shielding film exhibited remarkable heat insulation and heat preservation performance as well as an electromagnetic shielding performance of 46.3 dB.The impressive optical transmittance,mechanical properties,thermal performance,and electromagnetic shielding abilities combined with the renewable and sustainable nature,as well as the fast and efficient manufacturing process,make this bamboo composite material suitable for effective application in transparent,energy-saving,and electromagnetic shielding buildings.Jing Wang Xinyu Wu Yajing Wang Weiying Zhao Yue Zhao Ming Zhou Yan Wu Guangbin Ji 2023Nano-Micro Letters2023,15,1:1
15Allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci using Staphylococcus aureus Cas9(SaCas9)显示文摘Allele-specific DNA methylation is the most important imprinting marker localized to differentially methylated regions(DMRs),and aberrant genomic imprinted DNA methylation is associated with some human diseases,including Prader-Willi syndrome and cancer.Thus,the development of an effective strategy for the precise editing of allele-specific methylated genes is essential for the functional clarification of imprinting elements and the correction of imprinting disorders in human diseases.To discover a feasible allele-specific genome editing tool based on the CRISPR/Cas system,which is an efficient genetargeting technique in various organisms,we examined the targeting efficiency of Staphylococcus aureus Cas9(SaCas9)and Streptococcus pyogenes Cas9(SpCas9)in response to DNA methylation interference.We found that the targeting efficiency of SaCas9,but not SpCas9,was enhanced by targeted DNA demethylation using the d Cas9-Tet1 catalytic domain(CD)but suppressed by targeted DNA methylation using Dnmt3l-Dnmt3a-d Cas9.An in vitro cleavage assay further demonstrated that SaCas9 nuclease activity was inhibited by 5-methylcytosine(5mC)in a synthesized Cp G-containing context.Further analysis with Ch IP-Q-PCR demonstrated that the non-methylated sequence targeting of Sa Cas9 depends on the binding preference of SaCas9 to non-methylated sequences.Taking advantage of this feature of SaCas9,we have successfully obtained non-methylated allele-biased targeted embryos/mice for two imprinting genes,H19 and Snrpn,with relatively high efficiencies of 28.6%and 47.4%,respectively.These results indicate that the targeting efficiency of SaCas9 was strongly reduced by DNA methylation.By using SaCas9,we successfully achieved allele-specific genome editing of imprinting genes by preferentially targeting non-methylated loci.Yajing Liu Jianan Li Changyang Zhou Bin Meng Yu Wei Guang Yang Zongyang Lu Qingmei Shen Yu Zhang Hui Yang Yunbo Qiao 2019Science Bulletin2019,64,21:1
16The preparation process and feature of the topological insulator Bi_2Te_3显示文摘Topological insulators are insulating in the bulk but have metallic surface states. Its unique physicochemical properties can find numerous applications in electronics, spintronics, photonics, the energy sciences, and the signal control of transportation. We report an experimental approach to synthesize the high-quality single crystal of topological insulator Bi2Te3by using self-flux method. We obtained the optimal preparation conditions by adjusting the parameters of heat treatment, and successfully prepared the single-crystal Bi2Te3sample. The as-grown samples have a surface with bright metallic luster and are soft and fragile. Furthermore, Bi2Te3has the obvious layer structure from SEM results. The data of X-ray diffraction and scanning electron microscope show that Bi2Te3single crystal grows along the c-axis with the order of Te(1)–Bi–Te(2)–Bi–Te(1)and crystallizes in the hexagonal system with space group of R/3 m. 17Efficient gene transfection of suspension cells by highly branched poly(β-amino ester)显示文摘Suspension cells play a crucial role in many biological processes. However, compared to adherent cells, it is particularly challenging to introduce exogenous genes into suspension cells to regulate their biological functions with non-viral gene vectors, mainly due to the low cellular uptake and endosomal escape of polyplexes. Herein, to improve the interactions of polyplexes with cellular membranes, we design and synthesize highly branched poly(β-amino ester)(HPAE) via an “A2 + B4 + C2” Michael addition strategy.Results show that branching significantly increases DNA condensation of HPAE, cellular uptake and endosomal escape of HPAE/DNA polyplexes. In mast cells(MCs), HPAE exhibits up to 80-fold higher gene transfection efficiency compared to the corresponding linear poly(β-amino ester)(LPAE) and the leading commercial gene transfection reagents PEI25k, jetPEI, and Lipofectamine 3000, without causing obvious cytotoxicity. 20Evaluating damage in optical elements using an amplified spontaneous emission beam显示文摘A method to evaluate damage in optical elements with the near field of an amplified spontaneous emission(ASE) beam has been developed. Local peak intensities are generally distributed randomly in the near field of a laser beam. The partial coherence of the ASE source results in a very smooth beam profile. The coherence time of ASE is much less than the pulse width. Small-scale intensity modulations can be smoothed out rapidly within the time of a pulse width.In the experiments, ASE is generated from a multifunctional high-performance Nd:glass system, with a pulse duration of 3 ns, a spectral width(full width at half maximum, FWHM) of 1 nm and an adjustable energy range from 1 to 10 J.The damage thresholds of samples induced by ASE are two to three times higher than those induced by a laser with the same size of test spot.
