1Annual cost of illness of stomach and esophageal cancer patientsin urban and rural areas in China: A multi-center study显示文摘Objective: Stomach and esophageal cancer are imposing huge threats to the health of Chinese people whereas there were few studies on the financial burden of the two cancers.Methods: Costs per hospitalization of all patients with stomach or esophageal cancer discharged between September 2015 and August 2016 in seven cities/counties in China were collected, together with their demographic information and clinical details. Former patients in the same hospitals were sampled to collect information on annual direct non-medical cost, indirect costs and annual number of hospitalization. Annual direct medical cost was obtained by multiplying cost per hospitalization by annual number of hospitalization. Annual cost of illness(ACI)was obtained by adding the average value of annual direct medical cost, direct non-medical cost and indirect cost,stratified by sex, age, clinical stage, therapy and pathologic type in urban and rural areas. Costs per hospitalization were itemized into eight parts to calculate the proportion of each part. All costs were converted to 2016 US dollars(1 USD=6.6423 RMB).Results: Totally 19,986 cases were included, predominately male. Mean ages of stomach cancer and urban patients were lower than that of esophageal cancer and rural patients. ACI of stomach and esophageal cancer patients were $10,449 and $13,029 in urban areas, and $2,927 and $3,504 in rural areas, respectively. Greater ACI was associated with male, non-elderly patients as well as those who were in stage I and underwent surgeries.Western medicine fee took the largest proportion of cost per hospitalization.Conclusions: The ACI of stomach and esophageal cancer was tremendous and varied substantially among the population in China. 2Meso-damage cracking characteristics analysis for rock and soil aggregate with CT test显示文摘The aim of this paper is to investigate the damage cracking characteristics of rock and soil aggregate(RSA)by X-ray computed tomography(CT)under uniaxial compressive loading.The mean CT value for the region of interest(ROI)is used to analyze the cracking characteristics.Also,the mathematical morphology method based on the image threshold segmentation is used to obtain characteristic parameters of cracks to describe the cracking evolution of RSA.Results show that the elastic mismatch between rock blocks and soil matrix is the primary reason for RSA cracking.The mean CT value for the RSA specimen,rock block inclusions,and their adjacent soil regions decreases with the increasing stress level.However,it is more sensitive for block inclusions than soil regions.Using the image segmentation method,length,area and mean width of cracks obey to power function distribution.Crack statistical characteristics are closely related to the rock block’s distribution and morphology.These results may be useful to reveal the mesoscopic cracking mechanism,establish meso-damage evolution equation,and constitutive relation for RSA.WANG Yu LI Xiao ZHANG Bo WU YanFang 2014Science China(Technological Sciences)2014,57,7:21
3Isolation,characterization,and phylogenic analysis of three new severe fever with thrombocytopenia syndrome bunyavirus strains derived from Hubei Province,China显示文摘Hubei Province is a major epidemic area of severe fever with thrombocytopenia syndrome bunyavirus(SFTSV) in China. However, to date, a few SFTSV strains have been isolated from Hubei Province, preventing effective studies of epidemic outbreaks. Here, we report three confirmed patients(2015–2016) with typical symptoms of severe fever with thrombocytopenia syndrome disease(SFTS) who were farmers resident in different regions in Hubei Province. Three new SFTSV strains were isolated from the serum samples of each patient. Characterization of viral growth properties showed that there were no significant differences in virus production. All strains were completely sequenced, and phylogenetic analysis showed that unlike the other strains from Hubei province, which belonged to the SFTSV C3 genotype, one of the three strains belonged to the SFTSV C2 genotype. 4Analysis of the Fungal Community in Apple Replanted Soil Around Bohai Gulf显示文摘Apple replant disease(ARD) is a frequently occurring plant disease in replanted orchards around Bohai Gulf, which causes growth inhibition and even death of plants. The aim of this study was to investigate the etiology of ARD around Bohai Gulf. In this study, the primary growth inhibition of apple seedlings was evaluated in ten replanted soils, sampled around Bohai Gulf. A fungal clone library was used to identify changes in the structure and composition of the soil fungal community. The results revealed that the Simpson diversity indices of Laizhou and Pulandian orchards were higher than others, presenting severe ARD. Ascomycota dominated around Bohai Gulf at the phyla level. Fusarium and Saccharomyces were abundant in all replanted soils. In addition, correlations between the relative abundance of fungal genera in soils and the severity of ARD were analyzed. The results showed that Fusarium was correlated positively with the severity of ARD, but Mortierella was negatively correlated. Furthermore, the quantitative PCR of Fusarium oxysporum, which was regarded as a factor of ARD, was performed. 6The value of vegetation ecosystem services:a case of Qinling-Daba Mountains显示文摘According to differences in vegetation types and their coverage, combining the latest research, using theory and method on the value of vegetation ecosystem services, this paper not only calculated goods produced by different types of vegetation but also estimated the value of various vegetation ecosystem services and set up database, GIS and eco-account of vegetation ecosystem. The result was as follows: the value of vegetation’s primary productivity, soil and fertility conservation, water conservation, CO 2 fixation and O 2 release was 199.6 billion yuan/a, 22.64 billion yuan/a, 22.66 billion yuan/a, 352.24 billion yuan/a and 374.19 billion yuan/a, respectively. The total value of ecosystem services was 968.33 billion yuan/a. The temperate deciduous broad-leaved forest had the highest contribution rate, accounting for 16.42%. 7Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities.Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu 2011Cellular & Molecular Immunology2011,8,1:16
8Effect of pre-deformation on the stress corrosion cracking susceptibility of aluminum alloy 2519显示文摘The effects of pre-deformation and strain rate on the stress corrosion cracking (SCC) behavior of aluminum alloy 2519 in air and in 3.5% NaCl water solution were investigated by means of slow strain rate tension (SSRT), scanning elec- tron microscopy (SEM), and transmission electron microscopy (TEM). The results indicate that the alloy is susceptible to SCC in 3.5% NaCl water solution and not in air. At the same pre-deformation, the alloy is more susceptible to SCC at 1.33 × 10·5 s·1 than at 6.66 × 10·5 s·1. Moreover, it is more susceptible to SCC at free pre-deformation than at 10% pre-deformation at the same strain rate. 10M1 muscarinic acetylcholine receptor in Alzheimer's disease显示文摘The degeneration of cholinergic neurons and cholinergic hypofunction are pathologies associated with Alzheimer's disease(AD). Muscarinic acetylcholine receptors(mAChRs) mediate acetylcholine-induced neurotransmission and five mAChR subtypes(M1–M5) have been identified. Among them, M1 mAChR is widely expressed in the central nervous system and has been implicated in many physiological and pathological brain functions. In addition, M1 mAChR is postulated to be an important therapeutic target for AD and several other neurodegenerative diseases. 11Potential targets for protecting against hippocampal cell apoptosis after transient cerebral ischemiareperfusion injury in aged rats显示文摘Mitochondria play an important role in neuronal apoptosis caused by cerebral ischemia, and the role is mediated by the expression of mitochondrial proteins. This study investigated the involvement of mitochondrial proteins in hippocampal cell apoptosis after transient cerebral ischemia-reperfusion injury in aged rats using a comparative proteomics strategy. Our experimental results show that the aged rat brain is sensitive to ischemia-reperfusion injury and that transient ischemia led to cell apoptosis in the hippocampus and changes in memory and cognition of aged rats. Differential proteomics analysis suggested that this phenomenon may be mediated by mitochondrial proteins associated with energy metabolism and apoptosis in aged rats. 12Maize grain concentrations and above-ground shoot acquisition of micronutrients as affected by intercropping with turnip, faba bean, chickpea, and soybean显示文摘Most research on micronutrients in maize has focused on maize grown as a monocrop. The aim of this study was to determine the effects of intercropping on the concentrations of micronutrients in maize grain and their acquisition via the shoot. We conducted field experiments to investigate the effects of intercropping with turnip (Brassica campestris L.), faba bean (Vicia faba L.), chickpea (Cicer arietinum L.), and soybean (Glycine max L.) on the iron (Fe), manganese (Mn), copper (Cu) and zinc (Zn) concentrations in the grain and their acquisition via the above-ground shoots of maize (Zea mays L.). Compared with monocropped maize grain, the grain of maize intercropped with legumes showed lower concentrations of Fe, Mn, Cu, and Zn and lower values of their corresponding harvest indexes. The micronutrient concentrations and harvest indexes in grain of maize intercropped with turnip were the same as those in monocropped maize grain. Intercropping stimulated the above-ground maize shoot acquisition of Fe, Mn, Cu and Zn, when averaged over different phosphorus (P) application rates. To our knowledge, this is the first report on the effects of intercropping on micronutrient concentrations in maize grain and on micronutrients acquisition via maize shoots (straw+grain). The maize grain Fe and Cu concentrations, but not Mn and Zn concentrations, were negatively correlated with maize grain yields. The concentrations of Fe, Mn, Cu, and Zn in maize grain were positively correlated with their corresponding harvest indexes. The decreased Fe, Mn, Cu, and Zn concentrations in grain of maize intercropped with legumes were attributed to reduced translocation of Fe, Mn, Cu, and Zn from vegetative tissues to grains. This may also be related to the delayed senescence of maize plants intercropped with legumes. We conclude that turnip/maize intercropping is beneficial to obtain high maize grain yield without decreased concentrations of Fe, Mn, Cu, and Zn in the grain. 13How is mRNA expression predictive for protein expression? 14Abnormal activation of the occipital lobes during emotion picture processing in major depressive disorder patients显示文摘A large number of studies have demonstrated that depression patients have cognitive dysfunction. With recently developed brain functional imaging, studies have focused on changes in brain function to investigate cognitive changes. However, there is still controversy regarding abnormalities in brain functions or correlation between cognitive impairment and brain function changes. Thus, it is important to design an emotion-related task for research into brain function changes. We selected positive, neutral, and negative pictures from the International Affective Picture System. Patients with major depressive disorder were asked to judge emotion pictures. In addition, functional MRI was performed to synchronously record behavior data and imaging data. Results showed that the total correct rate for recognizing pictures was lower in patients compared with normal controls. Moreover, the consistency for recognizing pictures for depressed patients was worse than normal controls, and they frequently recognized positive pictures as negative pictures. The consistency for recognizing pictures was negatively correlated with the Hamilton Depression Rating Scale. Functional MRI suggested that the activation of some areas in the frontal lobe, temporal lobe, parietal lobe, limbic lobe, and cerebellum was enhanced, but that the activation of some areas in the frontal lobe, parietal lobe and occipital lobe was weakened while the patients were watching positive and neutral pictures compared with normal controls. The activation of some areas in the frontal lobe, temporal lobe, parietal lobe, and limbic lobe was enhanced, but the activation of some areas in the occipital lobe were weakened while the patients were watching the negative pictures compared with normal controls. These findings indicate that patients with major depressive disorder have negative cognitive disorder and extensive brain dysfunction. 15Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
16Antigenically dominant proteins within the human liver mitochondrial proteome identified by monoclonal antibodies显示文摘Analysis of the mitochondrial proteome would provide valuable insight into the function of this important organelle, which plays key roles in energy metabolism, apoptosis, free radical production, thermogenesis, and calcium signaling. It could also increase our understanding about the mechanisms that promote mitochondrial disease. To identify proteins that are antigenically dominant in human liver mitochondria, we generated >240 hybridoma cell lines from native mitochondrial proteins after cell fusion, screening, and cloning. Antibodies that recognized mitochondrial proteins were identified by screening human liver cDNA expression libraries. In this study, we identified 6 major antigens that were recognized by at least 2 different monoclonal antibodies (mAbs). The proteins that were antigenically dominant were: acetyl-Coenzyme A acyltransferase 2 (mitochondrial 3-oxoacyl-Coenzyme A thiolase), aldehyde dehydrogenase 1 family member A1, carbamoyl phosphate synthetase 1, dihydrolipoamide S-acetyltransferase (E2 component of pyruvate dehydrogenase complex), enoyl coenzyme A hydratase 1, and hydroxysteroid (11-beta) dehydrogenase 1. We also determined the subcellular localizations of these enzymes within the mitochondria using immunohistocytochemistry. We believe that these well-characterized antibodies will provide a valuable resource for the Human Liver Proteome Project (HLPP), and will make studies aimed at investigating liver mitochondrial function far easier to perform in future. 17A Method for Detection of Main Metabolites in Aromatic Rice Seeds显示文摘[Objective] A method was developed for the analysis of main metabolites in aromatic rice seeds by gas chromatography-mass spectrometry( GC-MS).[Method]The main metabolites in seeds of 9 different aromatic rice varieties were compared and analyzed with two methods. Method 1: the compound was extracted by the mixture of anhydrous ethanol and methylene chloride( V/V = 1∶ 1) at 60 ℃ for 3 h in water bath; the extract was isolated and detected by gas chromatography-mass spectrometry using HP-5 capillary column with scan monitoring mode. Method 2: the main metabolites in aromatic rice seeds were detected using headspace solid phase micro-extraction and gas chromatography mass spectrometry( GC-MS). [Result]Fifteen types of common metabolites were found in 9 different aromatic rice varieties. Method 1 was more effective than method 2 in detecting the main metabolites in aromatic rice seeds. 18Clinical spectrum and prognostic value of TP53 mutations in circulating tumor DNA from breast cancer patients in China显示文摘Background:TP53 mutations are common in breast cancer.There is currently no large-scale cohort study to investigate the TP53 landscape in breast cancer patients from China.The predictive value of TP53 mutations for the efficacy of human epidermal growth factor receptor 2(HER2)-targeted therapy in breast cancer remains controversial.In the present study,we aimed to analyze the clinical spectrum and prognostic value of TP53 mutations in circulating tumor DNA(ctDNA)from breast cancer patients in China.Methods:We retrospectively analyzed the clinical data and TP53 mutation features in ctDNA samples from 804 patients withmetastatic breast cancer.TP53 mutations were detected by target region capture-based next-generation sequencing.The relationship between TP53 mutation status and disease-free survival(DFS)was analyzed in 444 patientswithmetastatic breast cancer.Moreover,the relationship between TP53 mutation status and progression-free survival(PFS)was analyzed in 55 HER2-positive patients treated with first-line trastuzumab-based therapy.Kaplan-Meier analysis was performed to estimate the survival curves of the different subgroups,and the log-rank test was used to compare the curves.A Cox regression model was used to estimate multivariable-adjusted hazard ratios and their 95%confidence intervals(CIs)associated with the DFS and PFS.Results:Among the 804 investigated patients,431(53.6%)patients harbored TP53 mutations.TP53 mutations were differentially distributed among different molecular subtypes of breast cancer(P<0.05).Patients with TP53 mutations had a shorter DFS than those with wild-type TP53(hazard ratio=1.32,95%CI=1.09-1.61,P=0.005).TP53 mutations in exons 5-8 were associated with worse outcome(hazard ratio=1.50,95%CI=1.11-2.03,P=0.009).However,TP53 mutation status was not significantly associated with PFS in HER2-positive patients who received firstline trastuzumab-based therapy(P=0.966).Interestingly,in the taxane combination group,patients with TP53 mutations exhibited longer PFS than those without TP53 mutations(hazard ratio=0.08,95%CI=0.02-0.30,P<0.001).However,in the nontaxane combination group,patients with TP53 mutations displayed shorter PFS than those with wild-type TP53(hazard ratio=4.84,95%CI=1.60-14.66,P=0.005).Conclusions:TP53 mutations in exons 5-8 may be an independent prognostic marker for short DFS in patients with metastatic breast cancer.TP53 mutations had opposite effects on trastuzumab-treated patients treated with and without taxanes.Zongbi Yi Fei Ma Guohua Rong Yanfang Guan Chunxiao Li Binghe Xu 2020Cancer Communications2020,40,6:5

20Feasibility of Tea Saponin-Enhanced Soil Washing in a Soybean Oil-Water Solvent System to Extract PAHs/Cd/Ni Efficiently from a Coking Plant Site显示文摘Mixed contaminated brownfield sites have brought serious risks to human health and environmental safety. With the purpose of removing polycyclic aromatic hydrocarbons(PAHs) and heavy metals from a coking plant site, an innovative technology for ex-situ washing was developed in the present work. The combination of 15.0 mL L^(-1) soybean oil and 7.5 g L^(-1) tea saponin proved an effective method to extract co-pollutants from soil. After two consecutive washing cycles, the efficiency rates of removal for 3-, 4-,5(+6)-ring, and total PAHs, Cd, and Ni were approximately 98.2%, 96.4%, 92.3%, 96.3%, 94.1%, and 89.4%, respectively. Meanwhile,as evaluated by Tenax extraction method and metal stability indices, the residual PAHs and heavy metals after consecutive washing mainly existed in the form with extremely low bioaccessibility in the soil. Thus, in the soil after two washing cycles, there appeared limited environmental transfer risk of co-pollutants. Moreover, a subsequent precipitation method with alkaline solution and PAHdegrading strain Sphingobium sp. PHE9 inoculation effectively removed 84.6%–100% of Cd, 82.5%–91.7% of Ni, and 92.6%–98.4% of PAHs from the first and second washing solvents. The recovered solvents also exhibited a high recycling effectiveness.
