15DFRXXL region of long myosin light chain kinase causes F-actin bundle formation显示文摘Long myosin light chain kinase (L-MLCK) contains five DFRXXL motifs with ability to bind F-actin. Binding stoichiometry data indicated that each DFRXXL motif might bind each G-actin, but its biological significance remained unknown. We hypothesized that L-MLCK might act as an F-actin bundle peptides by its multiple binding sites of 5DFRXXL motifs to actin. In order to characterize F-actin-bundle formation properties of 5DFRXXL region of long myosin light chain kinase, we expressed and purified 5DFRXXL peptides tagged with HA in vitro. The properties of 5DFRXXL peptides binding to myofilaments or F-actin were analyzed by binding stoichiometries assays. The results indicated that 5DFRXXL peptides bound to myofilaments or F-actin with high affinity. KD values of 5DFRXXL binding to myofilaments and F-actin were 0.45 and 0.41 μmol/L, re- spectively. Cross-linking assay demonstrated that 5DFRXXL peptides could bundle F-actin efficiently. Typical F-actin bundles were observed morphologically through determina- tion of confocal and electron microscopy after adding 5DFRXXL peptides. After transfection of pEGFP-5DFRXXL plasmid into eukaryocyte, spike structure was observed around cell membrane edge. We guess that such structure formation may be attributable to F-actin over-bundle forma- tion caused by 5DFRXXL peptides. 2Temporal and spatial changes in Bt toxin expression in Bt-transgenic poplar and insect resistance in field tests显示文摘Extensive planting of Bacillus thuringiensis(Bt)-transgenic plants economically benefits society; however, the potential risk they pose is receiving increasing attention. This study used enzyme-linked immunosorbent assay and fluorescence quantitative PCR(RT-PCR) to monitor the temporal and spatial dynamics of the expression of Bt toxic protein in a forest of 6- to 8-year-old trees of transgenic insect-resistant poplar 741 for three consecutive years. The enrichment, distribution, and degradation of Bt toxic protein and the influence of transgenic poplars on the targeted insect population, Hyphantria cunea, were investigated. The content of Cry1Ac toxic protein dynamically changed in transgenic poplar. During the annual growth cycle, the content initially increased, then decreased in the long and the short branches of the crown and in the root system, peaking in August. During the study, the protein did not accumulate overtime. The m RNA transcription of gene Cry1Ac was almost consistent with the level of the protein, but transcription peaked in July. In the transgenic and control forestland, microscale levels of the Cry1Ac toxic protein were detected from the soil, but increased accumulation was not observed with the planting year of transgenic poplar. Meanwhile, Bt was isolated and detected molecularly from the soil in the experimental forestland. A systematic investigation of the density of H.cunea in the experimental transgenic poplar forest indicated that transgenic Pb29 poplar could resist insects to a certain degree. 3Genomic Variations in the Tea Leafhopper Reveal the Basis of Its Adaptive Evolution显示文摘Tea green leafhopper(TGL),Empoasca onukii,is of biological and economic interest.Despite numerous studies,the mechanisms underlying its adaptation and evolution remain enigmatic.Here,we use previously untapped genome and population genetics approaches to examine how the pest adapted to different environmental variables and thus has expanded geographically.We complete a chromosome-level assembly and annotation of the E.onukii genome,showing notable expansions of gene families associated with adaptation to chemoreception and detoxification.Genomic signals indicating balancing selection highlight metabolic pathways involved in adaptation to a wide range of tea varieties grown across ecologically diverse regions.Patterns of genetic variations among 54 E.onukii samples unveil the population structure and evolutionary history across different tea-growing regions in China.Our results demonstrate that the genomic changes in key pathways,including those linked to metabolism,circadian rhythms,and immune system functions,may underlie the successful spread and adaptation of E.onukii.This work highlights the genetic and molecular basis underlying the evolutionary success of a species with broad economic impacts,and provides insights into insect adaptation to host plants,which will ultimately facilitate more sustainable pest management.Qian Zhao Longqing Shi Weiyi He Jinyu Li Shijun You Shuai Chen Jing Lin Yibin Wang Liwen Zhang Guang Yang Liette Vasseur Minsheng You 2022Genomics, Proteomics & Bioinformatics2022,20,6:1

7Integrative transcriptomic and metabolomic analyses reveal the flavonoid biosynthesis of Pyrus hopeiensis flowers under cold stress显示文摘Low temperature is among the most restrictive factors to limit the yield and distribution of pear. Pyrus hopeiensis is a valuable wild resource.PCA showed that P. hopeiensis had strong cold resistance. In this study, the mRNA and metabolome sequencing of P. hopeiensis flower organs exposed to different low temperatures were performed to identify changes of genes and metabolites in response to low-temperature stress. A total of 4 851 differentially expressed genes(DEGs) were identified. Trend analysis showed that these DEGs were significantly enriched in profiles 19, 18, 7, 14, 1, 4 and 11. And the KEGG enrichment analysis showed that the DEGs in profile 18 were significantly enriched in flavone and flavonol biosynthesis. Besides, the expressed trends as well as GO and KEGG functional enrichment analyses of DEGs under cold and freezing stress showed significantly difference. Analyses of flavonoid-related pathways indicated that flavonoid structural genes had undergone significant changes. Correlation analysis showed that b HLH and MYB TFs may affect flavonoid biosynthesis by regulating structural gene expression. And PhMYB308 and PhMYB330 were likely candidate repressors of flavonoid biosynthesis by binding to a specific site in bHLH proteins. In total, 92 differentially accumulated metabolites(DAMs) were identified in P. hopeiensis flowers including 12 flavonoids. WGCNA results showed that coral 1, pink and brown 4 modules were closely associated with flavonoids and 11 MYBs and 15 bHLHs among the three modules may activate or inhibit the expression of 23 structural genes of flavonoid biosynthesis. 15Effects of site conditions on growth and wood properties of Populus×euramericana cv.'74/76'显示文摘The growth and wood properties of 240 individual Populus×euramericana cv.’74/76’(hereafter poplar 107)trees planted in Hebei Plain,China was evaluated.Mean annual increments in height,breast height diameter and volume,as well as cellulose,hemicellulose and lignin contents,shrinkage,density,bending strength and modulus of elasticity in the heart wood and sap wood.Environmental factors influencing growth and wood properties were analyzed using correlation and stepwise regres sion.The results show that the coefficients of variation(CVs)of growth traits ranged from 10.6 to 22.4%.The CVs of the chemical properties of heartwood ranged from 4.3 to 30.2%,and for sap wood from 3.2 to 27.5%.The CVs of the physical and mechanical properties of heartwood ranged from 8.6 to 31.7%,and for sapwood from 6.4 to 29.9%.The results of one-way ANOVA showed that there were significant differences in growth traits and wood properties among sites.Soil pH,total and available phosphorus,total potassium,and soil organic matter were key soil factors affecting growth and wood properties of poplar 107,whereas mean annual ground temperatures and precipitation were the main climatic factors.To better cultivate poplar 107,area with less annual rainfall,slightly higher temperature and soil pH value close to neutral should be selected.Jianmin Fan Changjun Ding Shijie Wang Changming Ma Chao Zhang Yongtan Li Jinmao Wang Minsheng Yang Xiaohua Su 2023Journal of Forestry Research2023,34,2:0

17Intraspecific leaf morphological variation in Quercus dentata Thunb.:a comparison of traditional and geometric morphometric methods,a pilot study显示文摘To compare the application of traditional morphometric methods(TMMs)and geometric morphometric methods(GMMs)in the study of intraspecific leaf morphological characters of Quercus dentata,fifteen linear measurement indices and thirteen landmarks of leaves were used to study leaf morphology of three provenances(H1,H2,and H3).In TMMs,principal component analysis(PCA)showed that leaf size–related indices played an important role in population classification.Partial least square(PLS)analysis showed that the main morphological characters affecting leaf size were the average depth of the lobes and the length–width ratios.However,the important indices to distinguish the provenances were circularity,leaf width,and length–width ratio.The results of discriminant analysis(DA)showed that 74.0%of H1,68.0%of H2,and 74.0%of H3 were correctly classified.Cluster analysis showed that the Mahalanobis distances between H1 and H2,H1 and H3,and H2 and H3 were 4.3761,11.4629,and 10.2067,respectively.In GMMs,PCA based on symmetrical components showed that the difference in leaf morphology was mainly due to the changing trend of the leaf apex and base,petiole length,and degree of leaf cracking.PLS analysis showed that there was a significant covariation between the leaf symmetrical components and size:as the leaf enlarged,the widest part gradually moved up,and the shape changed from nearly oval to lanceolate.DA results showed that 86.0%and 78.0%of H1 and H2,70.0%and 80.0%of H1 and H3,and 82.0%and 76.0%of H2 and H3 were correctly classified.Canonical variate analysis results showed that the Mahalanobis distances between H1 and H2,H1 and H3,and H2 and H3 were 1.7238,1.5380,and 1.6329,respectively.Both GMMs and TMMs showed significant differences in morphology among the three Q.dentata provenances,but GMMs had higher classification accuracy and could provide more information about leaf shape,whereas TMMs could provide more information about leaf size.Based on our results,GMMs are promising in the study of leaf morphological variation within Q.dentata provenances.Kaiyu Yang Jianghao Wu Xinman Li Xinbo Pang Yangchen Yuan Guohui Qi Minsheng Yang 2022Journal of Forestry Research2022,33,6:0
18CRISPR/Cas9-based functional analysis of yellow gene in the diamondback moth,Plutella xylostella显示文摘The diamondback moth,Plutella xylostella(L.),is an economically important pest of cruciferous crops worldwide.This pest is notorious for rapid evolution of the resistance to diferent classes of insecticides,making it increasingly dificult to control.Genetics-based control approaches,through manipulation of target genes,have been reported as promising supplements or alternatives to traditional methods of pest management.Here we identified a gene of pigmentation(yellow)in P.xylostella,Pxyellow,which encodes 1674 bp complementary DNA sequence with four exons and three introns.Using the clustered regularly interspersed palindromic repeats(CRISPR)CRISPR-associated protein 9 system,we knocked out Pxyellow,targeting two sites in Exon III,to generate 272 chimeric mutants(57%of the CRISPR-treated individuals)with color-changed phenotypes of the Ist to 3rd instar larvae,pupae,and adults,indicating that Pxyellow plays an essential role in the body pigmentation of P xlostella.Fitness analysis revealed no significant difference in the oviposition of adults,the hatchability of eggs,and the weight of pupac between homozygous mutants and wildtypes,suggesting that Pxyellow is not directly involved in regulation of growth,development,or reproduction.This work advances our understanding of the genetic and insect science molecular basis for body pigmentation of P xylostella,and opens a wide avenue for development of the genctcally based pest control techniques using Pxyellow as a screening marker.Yajun Wang Yuping Huang Xuejiao Xu Zhaoxia Liu Jianyu Li Xue Zhan Guang Yang Minsheng You Shijun You 2021Insect Science2021,28,5:0
