| 1 | Effects of soil water and nitrogen availability on photosynthesis and water use efficiency of Robinia pseudoacacia seedlings显示文摘The efficient use of water and nitrogen (N) to promote growth and increase yield of fruit trees and crops is well studied.However,little is known about their effects on woody plants growing in arid and semiarid areas with limited water and N availability.To examine the effects of water and N supply on early growth and water use efficiency (WUE) of trees on dry soils,one-year-old seedlings of Robinia pseudoacacia were exposed to three soil water contents (non-limiting,medium drought,and severe drought) as well as to low and high N levels,for four months.Photosynthetic parameters,leaf instantaneous WUE (WUEi) and whole tree WUE (WUEb) were determined.Results showed that,independent of N levels,increasing soil water content enhanced the tree transpiration rate (Tr),stomatal conductance (Gs),intercellular CO2 concentration (Ci),maximum net assimilation rate (Amax),apparent quantum yield (AQY),the range of photosynthetically active radiation (PAR) due to both reduced light compensation point and enhanced light saturation point,and dark respiration rate (Rd),resulting in a higher net photosynthetic rate (Pn) and a significantly increased whole tree biomass.Consequently,WUEi and WUEb were reduced at low N,whereas WUE i was enhanced at high N levels.Irrespective of soil water availability,N supply enhanced Pn in association with an increase of Gs and Ci and a decrease of the stomatal limitation value (Ls),while Tr remained unchanged.Biomass and WUEi increased under non-limiting water conditions and medium drought,as well as WUEb under all water conditions;but under severe drought,WUEi and biomass were not affected by N application.In conclusion,increasing soil water availability improves photosynthetic capacity and biomass accumulation under low and high N levels,but its effects on WUE vary with soil N levels.N supply increased Pn and WUE,but under severe drought,N supply did not enhance WUEi and biomass. | Xiping Liu Yangyang Fan Junxia Long Ruifeng Wei Roger Kjelgren Chunmei Gong Jun Zhao | 2013 | Journal of Environmental Sciences2013,25,3: | 15 |
| 2 | Potassium Channels: A Potential Therapeutic Target for Parkinson's Disease显示文摘The pathogenesis of the second major neurodegenerative disorder, Parkinson's disease(PD), is closely associated with the dysfunction of potassium(K^+ ) channels. Therefore, PD is also considered to be an ion channel disease or neuronal channelopathy. Mounting evidence has shown that K^+ channels play crucial roles in the regulations of neurotransmitter release, neuronal excitability, and cell volume. Inhibition of K^+ channels enhances the spontaneous firing frequency of nigral dopamine(DA)neurons, induces a transition from tonic firing to burst discharge, and promotes the release of DA in the striatum.Recently, three K^+ channels have been identified to protect DA neurons and to improve the motor and non-motor symptoms in PD animal models: small conductance(SK)channels, A-type K^+ channels, and KV7/KCNQ channels.In this review, we summarize the physiological and pharmacological effects of the three K^+ channels. | 4 | Characteristics and dynamics analysis of Populus euphratica populations in the middle reaches of Tarim River显示文摘Populus euphratica Oliv. is widely distributed along the Tarim River. Maintaining stability of P. euphratica population is important to local development. This study explored the static life table, survivorship curves and four function curves (survival rate, cumulative mortality rate, mortality density, and hazard rate), and development index of P. euphratica population in the middle reaches of Tarim River. The results indicated that the age structure of P. euphratica population belonged to positive pyramidal type, which meant young age-class individuals occupied most populations. The number ofⅠ-Ⅱage classes accounted for 66.2% of whole population, and this indicated that there were abundant subsequent seedlings resources to support the growth of P. euphratica population in the middle reaches of Tarim River. The survivorship curve of P. euphratica belonged to the Deevey Ⅲ (concave-type) and the development index was 47.72%. Four function curves revealed that the individuals of P. euphratica sharply decreased at the initial stage and then leveled off at the late stage of survival curve. | 5 | Vertical distribution of aerosol optical properties based on aircraft measurements over the Loess Plateau in China显示文摘Vertical distributions of aerosol optical properties based on aircraft measurements over the Loess Plateau were measured for the first time during a summertime aircraft campaign, 2013 in Shanxi, China. Data from four flights were analyzed. The vertical distributions of aerosol optical properties including aerosol scattering coefficients(σsc), absorption coefficients(σab),Angstr?m exponent(α), single scattering albedo(ω), backscattering ratio(βsc), aerosol mass scattering proficiency(Q sc) and aerosol surface scattering proficiency(Q sc′) were obtained. The mean statistical values of σsc were 77.45 Mm-1(at 450 nm), 50.72 Mm-1(at 550 nm), and32.02 Mm-1(at 700 nm). The mean value of σab was 7.62 Mm-1(at 550 nm). The mean values ofα, βsc and ω were 1.93, 0.15, and 0.91, respectively. Aerosol concentration decreased with altitude. Most effective diameters(ED) of aerosols were less than 0.8 μm. The vertical profiles of σsc,, α, βsc, Q sc and Q sc′ showed that the aerosol scattering properties at lower levels contributed the most to the total aerosol radiative forcing. Both α and βsc had relatively large values, suggesting that most aerosols in the observational region were small particles. The mean values of σsc, α, βsc, Q sc, Q sc′, σab and ω at different height ranges showed that most of the parameters decreased with altitude. | 6 | Auranofin mitigates systemic iron overload and induces ferroptosis via distinct mechanisms显示文摘Iron homeostasis is essential for health;moreover,hepcidin-deficiency results in iron overload in both hereditary hemochromatosis and iron-loading anemia.Here,we identified iron modulators by functionally screening hepcidin agonists using a library of 640 FDA-approved drugs in human hepatic Huh7 cells.We validated the results in C57BL/6J mice and a mouse model of hemochromatosis(Hfe^(−/−)mice).Our screen revealed that the anti-rheumatoid arthritis drug auranofin(AUR)potently upregulates hepcidin expression.Interestingly,we found that canonical signaling pathways that regulate iron,including the Bmp/Smad and IL-6/Jak2/Stat3 pathways,play indispensable roles in mediating AUR’s effects.In addition,AUR induces IL-6 via the NF-κB pathway.In C57BL/6J mice,acute treatment with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling and decreased serum iron and transferrin saturation.Whereas chronically treating male Hfe^(−/−)mice with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling,decreasing systemic iron overload,but less effective in females.Further analyses revealed that estrogen reduced the ability of AUR to induce IL-6/hepcidin signaling in Huh7 cells,providing a mechanistic explanation for ineffectiveness of AUR in female Hfe^(−/−)mice.Notably,high-dose AUR(25 mg/kg)induces ferroptosis and causes lipid peroxidation through inhibition of thioredoxin reductase(TXNRD)activity.We demonstrate the ferroptosis inhibitor ferrostatin significantly protects liver toxicity induced by highdose AUR without comprising its beneficial effect on iron metabolism.In conclusion,our findings provide compelling evidence that TXNRD is a key regulator of ferroptosis,and AUR is a novel activator of hepcidin and ferroptosis via distinct mechanisms,suggesting a promising approach for treating hemochromatosis and hepcidin-deficiency related disorders. | Lei Yang Hao Wang Xiang Yang Qian Wu Peng An Xi Jin Weiwei Liu Xin Huang Yuzhu Li Shiyu Yan Shuying Shen Tingbo Liang Junxia Min Fudi Wang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 5 |
| 7 | Trends in Stroke Mortality Rate--China,2004-2019显示文摘Introduction:Stroke has been the leading cause of death in China for decades.This study described the trends in stroke mortality in China from 2004 to 2019.Methods:Data was obtained from the National Disease Surveillance Point(DSP)system.A descriptive analysis was conducted.The adjusted mortality rate(AMR)and age-standardized mortality rate(ASMR)of stroke were calculated.Results:From 2004 to 2019,the ASMR substantially decreased,with a reduction of 39.8%,but the AMR stayed relatively stable.The mortality rate of stroke in rural areas was consistently higher than in urban areas.A geographical gradient in mortality of stroke was also apparent,with an increased rate in the western part of China and a decreased rate in the eastern part of China.In central China,the rate remained relatively stable.Conclusions:Although the ASMR of stroke continued to decline in China,the stagnant crude mortality rates suggested that China had not achieved sufficient decline to offset the demographic forces of population growth and ageing.More vigorous and effective prevention and treatment strategies are urgently needed to mitigate the disease burden of stroke in China,especially in areas with high stroke burden and limited resources. | Junxia Cheng Wei Wang Jianwei Xu Ling Yin Yunning Liu Jing Wu | 2022 | China CDC weekly2022,4,24: | 3 |
| 8 | The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. | 9 | Temporal and spatial changes in Bt toxin expression in Bt-transgenic poplar and insect resistance in field tests显示文摘Extensive planting of Bacillus thuringiensis(Bt)-transgenic plants economically benefits society; however, the potential risk they pose is receiving increasing attention. This study used enzyme-linked immunosorbent assay and fluorescence quantitative PCR(RT-PCR) to monitor the temporal and spatial dynamics of the expression of Bt toxic protein in a forest of 6- to 8-year-old trees of transgenic insect-resistant poplar 741 for three consecutive years. The enrichment, distribution, and degradation of Bt toxic protein and the influence of transgenic poplars on the targeted insect population, Hyphantria cunea, were investigated. The content of Cry1Ac toxic protein dynamically changed in transgenic poplar. During the annual growth cycle, the content initially increased, then decreased in the long and the short branches of the crown and in the root system, peaking in August. During the study, the protein did not accumulate overtime. The m RNA transcription of gene Cry1Ac was almost consistent with the level of the protein, but transcription peaked in July. In the transgenic and control forestland, microscale levels of the Cry1Ac toxic protein were detected from the soil, but increased accumulation was not observed with the planting year of transgenic poplar. Meanwhile, Bt was isolated and detected molecularly from the soil in the experimental forestland. A systematic investigation of the density of H.cunea in the experimental transgenic poplar forest indicated that transgenic Pb29 poplar could resist insects to a certain degree. | 11 | Synthesis and gelation behaviors of five new dimeric cholesteryl derivatives显示文摘Five new diacid amides of di-cholesteryl L-glycinates were designed and prepared.The compounds with linkers containing 0,1, 2,3,or 4 methylene units are denoted as 1,2,3,4,and 5,respectively.Their gelation behaviors in 25 solvents were tested as novel low-molecular-mass organic gelators(LMOGs).It was shown that the length of the linker connecting the two-cholesteryl residues in a gelator plays a crucial role in the gelation behavior of the compound.1 gels 11 of the 25 solvents tested at a concentration lower than 1.0%,while 2 gels 17 of the solvents tested.4 and 5,however,gel only 2 and 4 of them,respectively. SEM observation reveals that the lengths of the linkers and the identity of the solvents are the main factors affecting the structures of the aggregates in the gels.Experimentally,a clear linker effect on the microstructures of the gels was observed.As example,the aggregates of 1,2 and 3 in benzene or 1-heptanol adopt structures of thin fibers,rods or lamellas,respectively. | 12 | Characterization of coal burning-derived individual particles emitted from an experimental domestic stove显示文摘Coal combustion in the domestic stoves, which is common in most parts of the Chinese countryside, can release harmful substances into the air and cause health issues. In this study, particles emitted from laboratory stove combustion of the raw powder coals were analyzed for morphologies and chemical compositions by using transmission electron microscopy(TEM) coupled with energy-dispersive X-ray spectrometry(EDX). The coal burning-derived individual particles were classified into two groups: carbonaceous particles(including soot aggregates and organic particles) and non-carbonaceous particles(including sulfate, mineral and metal particles). The non-carbonaceous particles, which constituted a majority of the coal burning-derived emissions, were subdivided into Si-rich, S-rich, K-rich,Ca-rich, and Fe-rich particles according to the elemental compositions. The Si-rich, S-rich and K-rich particles are commonly observed in the coal burning emission. The proportions for particles of different types exhibit obvious coal-issue dependence. Burning of coal with high ash yield could emit more non-carbonaceous particles, and burning of coal with high sulfur content can emit more S-rich particles. | 14 | SIZ1 negatively regulates aluminum resistance by mediating the STOP1–ALMT1 pathway in Arabidopsis显示文摘Sensitive to proton rhizotoxicity 1(STOP1) functions as a crucial regulator of root growth during aluminum(Al) stress. However, how this transcription factor is regulated by Al stress to affect downstream genes expression is not well understood. To explore the underlying mechanisms of the function and regulation of STOP1, we employed a yeast two hybrid screen to identify STOP1-interacting proteins. The SUMO E3 ligase SIZ1, was found to interact with STOP1 and mainly facilitate its SUMO modification at K40 and K212 residues. Simultaneous introduction of K40 R and K212 R substitutions in STOP1 enhances its transactivation activity to upregulate the expression of aluminum-activated malate transporter 1(ALMT1)via increasing the association with mediator 16(MED16) transcriptional co-activator. Loss of function of SIZ1 causes highly increased expression of ALMT1, thus enhancing Al-induced malate exudation and Al tolerance. Also, we found that the protein level of SIZ1 is reduced in response to Al stress. Genetic evidence demonstrates that STOP1/ALMT1 is epistatic to SIZ1 in regulating root growth response to Al stress. | 15 | Dynamic Microwave-Assisted Extraction of Total Ginsenosides from Ginseng Fibrous Roots显示文摘A dynamic microwave-assisted extraction(DMAE) method is established for the extraction of total ginsenosides from ginseng fibrous roots. The extraction process has been simulated and its main affection factors(liquid/solid ratio K of solvent to ginseng powders(V/m), irradiation time, irradiation temperature, extracting solution concentration, flow rate of solvent and microwave power) have been optimized by response surface methodology(RSM). The optimum conditions of extraction are the liquid/solid ratio of 270 m L/g, the extraction temperature of 75 ℃, the extraction time of 35 min, ethanol concentration of 70%(V/V), the solvent flow rate of 1.3 m L/min, and the microwave power of 500 W. The yield of ginsenosides obtained by the proposed DAME method is(15.0±0.7) %, which is well agreement with the yield predicted by the model. | 20 | NAC1 regulates root ground tissue maturation by coordinating with the SCR/SHR-CYCD6;1 module in Arabidopsis显示文摘Precise spatiotemporal control of the timing and extent of asymmetric cell divisions(AcDs)is essential for plant development.In the Arabidopsis root,ground tissue maturation involves an additional AcD of the endo-dermis that maintains the inner cell layer as the endodermis and generates the middle cortex to the outside.Through regulation of the cell cycle regulator CYCLIND6;1(CYCD6;1),the transcription factors SCARECROW(SCR)and SHORT-ROOT(SHR)play critical roles in this process.In the present study,we found that loss of function of NAC1,a NAC transcription factor family gene,causes markedly increased periclinal cell divisions in the root endodermis.Importantly,NAC1 directly represses the transcription of CYCD6;1 by recruiting the co-repressor TOPLESS(TPL),creating a fine-tuned mechanism to maintain proper root ground tissue patterning by limiting production of middle cortex cells.Biochemical and genetic analyses further showed that NAC1 physically interacts with SCR and SHR to restrict excessive periclinal cell divisions in the endo-dermis during root middle cortex formation.Although NAC1-TPL is recruited to the CYCD6;1 promoter and represses its transcription in an SCR-dependent manner,NAC1 and SHR antagonize each other to regu-late the expression of CYCD6;1.Collectively,our study provides mechanistic insights into how the NAC1-TPL module integrates with the master transcriptional regulators SCR and SHR to control root ground tissue patterning by fine-tuning spatiotemporal expression of CYCD6;1 in Arabidopsis. | Chuantian Xie Cuiling Li Fengxia Wang Feng Zhang Jiajia Liu Junxia Wang Xiansheng Zhang Xiangpei Kong Zhaojun Ding | 2023 | Molecular Plant2023,16,4: | 1 |
