| 1 | Overexpression of Dickkopf-3 induces apoptosis through mitochondrial pathway in human colon cancer显示文摘AIM:To investigate the mechanisms of the biological roles of Dickkopf-3(Dkk-3) in cell invasion,survival and apoptosis in colon cancer cells.METHODS:Three human colon cancer cell lines,i.e.,HT-29,LoVo and SW480,were used.Overexpression of Dkk-3 induced by pEGFP-N1-Dkk-3-GFP plasmid in LoVo cells was performed using Lipofectamine 2000 reagent.Reverse transcription polymerase chain reaction and Western blotting were performed to determine the mRNA and protein expression levels of Dkk-3,respectively.Cell proliferation assay,cell cycle analysis,hoechst 33258 assay and Matrigel invasion assay were performed on Dkk-3 overexpressing transfectants.RESULTS:The mRNA and protein expressions of Dkk-3 in HT-29(mRNA:0.06 ± 0.02,protein:0.06 ± 0.01) and LoVo(mRNA:0.07 ± 0.02,protein:0.07 ± 0.02) cells were significantly lower than that in SW480 cells(mRNA:0.92 ± 0.04,protein:0.69 ± 0.13;all P < 0.05),and the greatest levels of invasiveness wasin LoVo cells.Dkk-3 overexpression inhibited the proliferation and invasion of LoVo cells and induced cell cycle arrest at G0/G1 phase and subsequent apoptosis,as indicated by increased chromatin condensation and fragments,upregulated Bax and cytochrome c protein,downregulated survivin and Bcl-2 protein,and the activation of caspase-3 and caspase-9.Furthermore,Dkk-3 overexpression reduced the accumulation of cytosolic fraction of β-catenin.CONCLUSION:Dkk-3 overexpression induced apoptosis in human colon cancer possibly through the mitochondrial pathway.Dkk-3 may be involved in the Wnt/β-catenin signaling pathways in colon cancer. | Zi-Rong Yang Wei-Guo Dong Xiao-Fei Lei Meng Liu QiSheng Liu | 2012 | World Journal of Gastroenterology2012,18,14: | 7 |
| 2 | Observation and research of deep underground multi-physical fields—Huainan–848 m deep experiment显示文摘Compared with the surface,the deep environment has the advantages of allowing“super-quiet and ultra-clean”-geophysical field observation with low vibration noise and little electromagnetic interference,which are conducive to the realization of long-term and high-precision observation of multi-physical fields,thus enabling the solution of a series of geoscience problems.In the Panyidong Coal Mine,where there are extensive underground tunnels at the depth of 848 m below sea level,we carried out the first deep-underground geophysical observations,including radioactivity,gravity,magnetic,magnetotelluric,background vibration and six-component seismic observations.We concluded from these measurements that(1)the background of deep subsurface gravity noise in the long-period frequency band less than 2 Hz is nearly two orders of magnitude weaker than that in the surface observation environment;(2)the underground electric field is obviously weaker than the surface electric field,and the relatively high frequency of the underground field,greater than 1 Hz,is more than two orders of magnitude weaker than that of the surface electric field;the east-west magnetic field underground is approximately the same as that at the surface;the relatively high-frequency north-south magnetic field underground,below 10 Hz,is at least one order of magnitude lower than that at the surface,showing that the underground has a clean electromagnetic environment;(3)in addition to the highfrequency and single-frequency noises introduced by underground human activities,the deep underground space has a significantly lower background vibration noise than the surface,which is very beneficial to the detection of weak earthquake and gravity signals;and(4)the underground roadway support system built with ferromagnetic material interferes the geomagnetic field.We also found that for deep observation in the“ultra-quiet and ultra-clean”environment,the existing geophysical equipment and observation technology have problems of poor adaptability and insufficient precision as well as data cleaning problems,such as the effective separation of the signal and noise of deep observation data.It is also urgent to interpret and comprehensively utilize these high-precision multi-physics observation data. | Yun WANG Yaxin YANG Heping SUN Chengliang XIE Qisheng ZHANG Xiaoming CUI Chang CHEN Yongsheng HE Qiangqiang MIAO Chaomin MU Lianghui GUO Jiwen TENG | 2023 | Science China Earth Sciences2023,66,1: | 3 |
| 3 | Spatial distribution and reconstruction potential of Japanese anchovy(Engraulis japonicus) based on scale deposition records in recent anaerobic sediment of the Yellow Sea and East China Sea显示文摘Many studies have revealed that anchovy has exhibited large variability in population size on decadal timescales. However, such works concerning anchovy population are mainly based on short historical catch records. In order to understand the causes of variability in fish stocks(natural and/or anthropogenic) and calibrate the error between catches and standing stocks, it is essential to develop long-term time series of fish stocks from the time when human impacts are minimal or negligible. Well preserved fish scales from sediment record are regarded as useful indicators revealing the history of fish population dynamics over the last centuries. Anchovy scales was first analyzed over the Yellow Sea and East China Sea and the largest abundance was found in the central South Yellow Sea where is regarded as the largest overwintering ground for Japanese anchovy(Engraulis japonicas). Thus in the central South Yellow Sea, two cores covering the last 150 years were collected for estimating fish scale flux. The scale deposition rate(SDR) records show that the decadal scale SDRs were obviously coherent between cores with independent chronologies. The calibration of downcore SDRs to the standing stocks of anchovy further validated that SDR is a reliable proxy to reconstruct the long-term anchovy population dynamic in the central South Yellow Sea where anoxic conditions prevail in the sediment. | 4 | Effects of fish meal replacement with Chlorella meal on growth performance,pigmentation,and liver health of largemouth bass(Micropterus salmoides)显示文摘Chlorella meal is a potential protein source for aquafeeds.However,the physiological response of carnivorous fish fed Chlorella meal remains elusive.This study evaluated the effects of replacing dietary fish meal with Chlorella meal on growth performance,pigmentation,and liver health in largemouth bass.Five diets were formulated to replace dietary fish meal of 0%(C0,control),25%(C25),50%(C50),75%(C75),and 100%(C100)with Chlorella meal,respectively.Total 300 fish(17.6±0.03 g)were randomly assigned to 15 tanks(3 tanks/group).Fish were fed the experimental diet twice daily for 8 weeks.The increased dietary Chlorella meal quadratically influenced the final body weight(FBW),weight gain rate(WGR),specific growth rate(SGR),and feed intake(FI),which were significantly lower in the C100 group than in the other groups(P<0.05).The feed conversion ratio(FCR)increased linearly or quadratically with dietary Chlorella meal.Dietary Chlorella meal linearly or quadratically increased the lutein content of plasma,liver,and dorsal muscle of largemouth bass(P<0.05).Compared to the C0 group,all supplemented Chlorella meal groups significantly improved the yellowness(b*)of the dorsal body(1.5 to 2.0 fold),abdominal body(1.5 to 1.8 fold),and dorsal muscle(3.8 to 5.4 fold)of largemouth bass(P<0.05).In addition,compared to the C0 group,the liver vacuolation area of fish was significantly increased in the C75 and C100 groups(P<0.05).Transcriptional levels of apoptosis-related genes of b-cell lymphoma-2(bcl2),caspase-9-like(casp9),and caspase-3a(casp3)were markedly upregulated(0.9 to 1.6 fold)in the C100 group compared to the C0 group(P<0.05).Based on the quadratic regression analysis between FBW,WGR,or SGR and dietary Chlorella meal level,largemouth bass had the best growth when replacing 31.7%to 32.6%of fish meal with 15.03%to 15.43%dietary Chlorella meal.The present results indicated that dietary supplementation with Chlorella meal(11.85%to 47.45%)significantly enhanced the pigmentation;however,total replacement of fish meal(40%)with Chlorella meal(47.45%)caused growth retardation,apoptosis,and liver damage in largemouth bass. | Longwei Xi Qisheng Lu Yulong Liu Jingzhi Su Wen Chen Yulong Gong Dong Han Yunxia Yang Zhimin Zhang Junyan Jin Haokun Liu Xiaoming Zhu Shouqi Xie | 2022 | Animal Nutrition2022,,3: | 2 |

| 7 | Characterizing the spatiotemporal variations of evapotranspiration and aridity index in mid-western China from 2001 to 2016显示文摘Mid-western China is one of the most sensitive and fragile areas on the Earth.Evapotranspiration(ET)is a key part of hydrological cycle in these areas and is affected by both global climate change and human activities.The dynamic changes in ET and potential evapotranspiration(PET),which can reflect water consumption and demand,are still unclear,and there is a lack of predictive capacity on drought severity.In this study,we used global MODIS(moderate-resolution imaging spectroradiometer)terrestrial ET(MOD16)products,Morlet wavelet analysis,and simple linear regression to investigate the spatiotemporal variations of ET,PET,reference ET(ET0),and aridity index(AI)in mid-western pastoral regions of China(including Gansu Province,Qinghai Province,Ningxia Hui Autonomous Region,and part of Inner Mongolia Autonomous Region)from 2001 to 2016.The results showed that the overall ET gradually increased from east to southwest in the study area.Actual ET showed an increasing trend,whereas PET tended to decrease from 2001 to 2016.The change in ET was affected by vegetation types.During the study period,the average annual ET0 and AI tended to decrease.At the monthly scale within a year,AI value decreased from January to July and then increased.The interannual variations of ET0 and AI showed periodicity with a main period of 14 a,and two other periodicities of 11 and 5 a.This study showed that in recent years,drought in these pastoral regions of mid-western China has been alleviated.Therefore,it is foreseeable that the demand for irrigation water for agricultural production in these regions will decrease. | MU Le LU Yixiao LIU Minguo YANG Huimin FENG Qisheng | 2021 | Journal of Arid Land2021,13,12: | 1 |
