| 3 | 扬州宋大城西门发掘报告显示文摘The excavation of the western gate of the Great City, Song period Yangzhou, discovered the stratigraphical superimposition of the western gates respectively belonging to the Five Dynasties period Zhou Dynasty Lesser City, the Northern Song period District City, the Southern Song period Great City and the Ming and Qing Yangzhou City,brought to light the formal evolution of this city-gate, and revealed the historical mystery that the Zhou Dynasty Lesser City was also the Song period Great City. At the end of the Northern Song period, the Song and Jin dynasties were confronting each other, and Yangzhou became an anti-Jin forward position. To meet military needs, a barbican was added outside the western city-gate. The gate itself was built of bricks and must have been vaulted judging from the structure of the two side gateways. The great importance of this discovery lies in the fact that the construction of this city-gate was more than 130 years earlier than that of the vaulted city-gate of Heyimen in the Yuan Dadu. The typological features of the western gate cum barbican of the Song period Great City distinctly reflect the defensive function of the three cities of Song period Yangzhou, and provide valuable material for studying ancient urban architecture, military defence and the character of Song period Yangzhou City. | 5 | Four kinds of ENU-induced white spot mice and chromosome locations of the mutant genes显示文摘Phenotype-driven is the name for an ap-proach used to study gene functions through mutagenesis, location and cloning of the mutant gene. In this study, 150 male C57BL/6J(B6) mice were treated with ENU and repro-duced a total offspring of 3860. Of these descendants, 210 exhibited mutation phenotypes by screening, and more than 10 of them are hereditable. Four kinds of mutant mice, named Wbct, W-1Bao, W-2Bao, and W-3Bao, showed dominant hereditary white spot mutation with partial albinism on their belly, distal limbs and tail terminal. To map these mutant genes, 39 microsatellites, equally distributed on the mouse genome and with difference between B6 and DBA/2J (D2), were selected to scan the genome after discrimination of the white spots in the F2 mice [(B6×D2)×D2]. It is found that, the log odds score (LODS) between W-1Bao and D5Mit168 is 0.56, and the LODS of W-1Bao and D5Mit352 is 4.47. With the gradual application of microsatellites D5Mit290, D5Mit312, D5Mit308 and D5Mit356 that are close to the mutant gene, and the number of F2 mice going up to 537, the mutant W-1Bao is located between D5Mit356 and D5Mit308 on chro-mosome 5, about 42.19 cM from the centromere. In the same way, W-2Bao and W-3Bao are mapped nearby W-1Bao, and Wbct is located on chromosome 1, about 41.6 cM from the centro-mere. | 8 | Evaluation of genetic diversity in Chinese indigenous chicken breeds using microsatellite markers显示文摘China is rich in chicken genetic resources, and many indigenous breeds can be found throughout the country. Due to poor productive ability, some of them are threatened by the commercial varieties from domestic and foreign breeding companies. In a large-scale investigation into the current status of Chinese poultry genetic resources, 78 indigenous chicken breeds were surveyed and their blood samples collected. The genomes of these chickens were screened using microsatellite analysis. A total of 2740 individuals were genotyped for 27 microsatellite markers on 13 chromosomes. The number of alleles of the 27 markers ranged from 6 to 51 per locus with a mean of 18.74. Heterozy-gosity (H) values of the 78 chicken breeds were all more than 0.5. The average H value (0.622) and polymorphism information content (PIC, 0.573) of these breeds suggested that the Chinese indige-nous chickens possessed more genetic diversity than that reported in many other countries. The fixa-tion coefficients of subpopulations within the total population (FST) for the 27 loci varied from 0.065 (LEI0166) to 0.209 (MCW0078), with a mean of 0.106. For all detected microsatellite loci, only one (LEI0194) deviated from Hardy-Weinberg equilibrium (HWE) across all the populations. As genetic drift or non-random mating can occur in small populations, breeds kept on conservation farms such as Langshan chicken generally had lower H values, while those kept on large populations within con-servation regions possessed higher polymorphisms. The high genetic diversity in Chinese indigenous breeds is in agreement with great phenotypic variation of these breeds. Using Nei’s genetic distance and the Neighbor-Joining method, the indigenous Chinese chickens were classified into six categories that were generally consistent with their geographic distributions. | 12 | Impact of living mulch on soil C:N:P stoichiometry in orchards across China:A meta-analysis examining climatic,edaphic,and biotic dependency显示文摘The use of living mulch in orchards is a widely accepted management strategy for improving soil quality and enhancing tree productivity.Although the effects of living mulch on soil organic carbon(C)and nutrients have been previously investigated,changes in the stoichiometric ratios of C,soil total nitrogen(N),and soil total phosphorous(P)under different climatic,edaphic,and biotic conditions are currently unknown.These factors are important indicators of elemental balance associated with ecological interactions.In order to examine the effects of living mulch in orchards on soil C:N:P stoichiometry under different conditions,a meta-analysis was undertaken.The results showed that in general,living mulch significantly(P<0.05)increased C:P and N:P ratio,while the impact on C:N ratio was not significant,a result that was related to the coupled increase of C and N.Phosphorous limitation occurred shortly after the addition of living mulch;after four years this effect receded.In contrast,an increase in C occurred simultaneously with N increase at all stages.Specifically,the treatment effect was context-dependent.The living mulch did not change soil stoichiometry in orchards with old trees(>10 years),an occurrence which may be related to changes in the amount of fungi.Grass life history also had a significant influence on the treatment effect on soil stoichiometry,while N-fixing characteristics did not.The treatment effect was significant in areas with moderate mean annual temperature and mean annual precipitation,which might be related to the litter ratio of grass and trees.Effects on stoichiometric ratios were significant in the top soil layer(0–20 cm),in contrast to the deep soil layers.Therefore,grass species and management practices,such as fertilization,should be selected according to the specific soil and climatic conditions of the management area. | Guo CHEN Shibin LIU Yangzhou XIANG Xiaolu TANG Haitao LIU Bin YAO Xuqiang LUO | 2020 | Pedosphere2020,30,2: | 7 |
| 13 | Dynamic airspace sectorization via improved genetic algorithm显示文摘This paper deals with dynamic airspace sectorization (DAS) problem by an improved genetic algorithm (iGA). A graph model is first constructed that represents the airspace static structure. Then the DAS problem is formulated as a graph-partitioning problem to balance the sector workload under the premise of ensuring safety. In the iGA, multiple populations and hybrid coding are applied to determine the optimal sector number and airspace sectorization. The sector constraints are well satisfied by the improved genetic operators and protect zones. This method is validated by being applied to the airspace of North China in terms of three indexes, which are sector balancing index, coordination workload index and sector average flight time index. | 14 | Expression and intercellular trafficking of the VP22 protein of CVI988/Rispens vaccine strain of Marek’s disease virus显示文摘The viral protein 22 (VP22) in the tegument of Marek’s disease virus serotype 1 (MDV-1) plays an im-portant role in cell-to-cell spread and viral propagation. Antiserum against the carboxyl terminus of VP22 was prepared by immunizing mice with recombinant VP22 expressed in E. coli, and used to in-vestigate its expression in chicken embryo fibroblast (CEF) cells infected with different MDV-1 strains. At an infection dose of PFU=50, intercellular trafficking of the VP22 into the nuclei of the surrounding receipt cells was detected as early as 3 hours post infection. By 6 hours after infection (before viral plague formation), the protein was detected in the whole nuclei of the recipient cells with no difference among MDV-1 strains CVI988/Rispens, GA and RB1B. Intra-nuclear accumulation of the VP22 protein was further increased when the viral plagues started to form. Increasing N application amount and/or the ratio of topdressing N after elongation stage could raise grain yield, protein content, wet gluten content, sedimentation, falling number, development time and stability time. To produce strong-gluten and medium-gluten wheat, the suitable ratio of basis : tillering: elongation: booting N fertilizer was 3: 1: 3 : 3 or 5: 1 : 2: 2with 180 - 240 kg N ha-1. | 18 | Source-sink Characteristics and the Translocation of Assi- milates in New Plant Type and Intersubspecific Hybrid Rice显示文摘Both new plant type (NPT) and intersubspecific hybrid rice (IHR) had large sink size (total number of spikelets per m2), however, poor grain filling limited their potential in the grain yield. Compared to the three-line indica hybrid of Shanyou 63 (CK), NPT and IHR showed higher photosynthetic potential,higher dry matter accumulation and higher ratio of dry weight to spikelets (total dry wt./total number of spikelets) from heading to harvest. But both exhibited a low export percentage and transfer ratio of assimilates, low partitioning of 14C to grains from labeled flag leaves, low harvest indices and low physiological activities (IAA content and activities of ATPase and starch synthase) of grains at early grainfilling stage. The physiological activities of grains at early filling stage were significantly correlated with the export percentage and transfer ratio of assimilates, ripened-grain percentage and grain plumpness (r = 0.85 *- 0.95* * ). The source-sink ratio (dry matter wt./spikelet and nonstructural carbohydrate/spikelet) at heading was positively correlated with physiological activities of grains (r = 0.84* - 0.97 * * ). | 20 | MDV-1 VP22 conjugated VP2 enhancing immune response against infectious bursal disease virus by DNA vaccination in mice显示文摘VP22 of Marek’s disease virus serotype 1 (MDV-1) could function in protein transduction. In this study, an infectious bursal disease virus VP2 gene was fused to the carboxyl termini of VP22. It showed that the fusion protein did not spread into the bystander cells from the cells transfected with pVP22-VP2, as the VP22 alone could. The VP22 proteins were found to be translocated into all the nuclei in the neighboring COS-1 cells, as analyzed by a fluorescence assay. Although mice were immunized with the recombinant DNAs mixed with polyethylenimine (PEI) at a dose of 1:2, it failed to enhance the antibody response against IBDV VP2, as measured by the indirect ELISA assay, yet the cell mediated immune response was significantly increased. The ratio of CD8+/CD4+ T cells was significantly increased in the immunized group with the fusion genes, compared with the group immunized with VP2 (P<0.05).
