1Rac1 Modulates Excitatory Synaptic Transmission in Mouse Retinal Ganglion Cells显示文摘Ras-related C3 botulinum toxin substrate 1(Racl),a member of the Rho GTPase family which plays important roles in dendritic spine morphology and plasticity,is a key regulator of cytoskeletal reorganization in dendrites and spines.Here,we investigated whether and how Racl modulates synaptic transmission in mouse retinal ganglion cells(RGCs)using selective conditional knockout of Racl(Racl-cKO).Racl-cKO significantly reduced the frequency of AMPA receptor-mediated miniature excitatory postsynaptic currents,while glycine/GABA_A receptor-mediated miniature inhibitory postsynaptic currents were not affected.Although the total GluA1 protein level was increased in Racl-cKO mice,its expression in the membrane component was unchanged.RaclcKO did not affect spine-like branch density in single dendrites,but significantly reduced the dendritic complexity,which resulted in a decrease in the total number of dendritic spine-like branches.These results suggest that Racl selectively affects excitatory synaptic transmission in RGCs by modulating dendritic complexity.Ling-Zhu Li Ning Yin Xue-Yan Li Yanying Miao Shuo Cheng Fang Li Guo-Li Zhao Shu-Min Zhong Xin Wang Xiong-Li Yang Zhongfeng Wang 2019Neuroscience Bulletin2019,35,4:2
2Dopamine D2 Receptor-Mediated Modulation of Rat Retinal Ganglion Cell Excitability显示文摘Ganglion cells(RGCs) are the sole output neurons of the retinal circuity. Here, we investigated whether and how dopamine D2 receptors modulate the excitability of dissociated rat RGCs. Application of the selective D2 receptor agonist quinpirole inhibited outward K^+ currents, which were mainly mediated by glybenclamide-and 4-aminopyridine-sensitive channels, but not the tetraethylammonium-sensitive channel. In addition,quinpirole selectively enhanced Nav1.6 voltage-gated Na^+ currents. The intracellular c AMP/protein kinase A,Ca^2+/calmodulin-dependent protein kinase Ⅱ, and mitogen-activated protein kinase/extracellular signal-regulated kinase signaling pathways were responsible for the effects of quinpirole on K^+ and Na^+ currents, while phospholipase C/protein kinase C signaling was not involved. Under current-clamp conditions, the number of action potentials evoked by positive current injection was increased by quinpirole. 4Simultaneous enhancement of strength and ductility in friction stir processed 2205 duplex stainless steel with a bimodal structure:experiments and crystal plasticity modeling显示文摘Achieving excellent strength-ductility synergy is a long-lasting research theme for structural materials.However,attempts to enhance strength usually induce a loss of ductility,i.e.,the strength-ductility trade-off.In the present study,the strength-ductility trade-off in duplex stainless steel(DSS)was overcome by developing a bimodal structure using friction stir processing(FSP).The ultimate tensile strength and elongation were improved by 140%and 109%,respectively,compared with those of the asreceived materials.Plastic deformation and concurrent dynamic recrystallization(DRX)during FSP were responsible for the formation of bimodal structure.Incompatible deformation resulted in the accumulation of dislocations at the phase boundaries,which triggered interpenetrating nucleation between the austenite and ferrite phases during DRX,leading to a bimodal structure.The in situ mechanical responses of the bimodal structure during tensile deformation were investigated by crystal plasticity finite element modeling(CPFEM).The stress field distribution obtained from CPFEM revealed that the simultaneous enhancement of strength and ductility in a bimodal structure could be attributed to the formation of a unique dispersion-strengthened system with the austenite and ferrite phases.It is indicated that the present design of alternating fine austenite and coarse ferrite layers is a promising strategy for optimizing the mechanical properties of DSSs.Yanying Hu Zexin Fang Yongbing Peng Weiqi Yang Xianfeng Ma Jinglong Li Yunqiang Zhao Biao Wang 2022Science China(Physics,Mechanics & Astronomy)2022,65,2:0
5Immunity-and-matrix-regulatory cells enhance cartilage regeneration for meniscus injuries: a phase I dose-escalation trial显示文摘Immunity-and-matrix-regulatory cells(IMRCs)derived from human embryonic stem cells have unique abilities in modulating immunity and regulating the extracellular matrix,which could be mass-produced with stable biological properties.Despite resemblance to mesenchymal stem cells(MSCs)in terms of self-renew and tri-lineage differentiation,the ability of IMRCs to repair the meniscus and the underlying mechanism remains undetermined.Here,we showed that IMRCs demonstrated stronger immunomodulatory and pro-regenerative potential than umbilical cord MSCs when stimulated by synovial fluid from patients with meniscus injury.Following injection into the knees of rabbits with meniscal injury,IMRCs enhanced endogenous fibrocartilage regeneration.In the dose-escalating phase I clinical trial(NCT03839238)with eighteen patients recruited,we found that intra-articular IMRCs injection in patients was safe over 12 months post-grafting.Furthermore,the effective results of magnetic resonance imaging(MRI)of meniscus repair and knee functional scores suggested that 5×107 cells are optimal for meniscus injury treatment.In summary,we present the first report of a phase I clinical trial using IMRCs to treat meniscus injury.Our results demonstrated that intra-articular injection of IMRCs is a safe and effective therapy by providing a permissive niche for cartilage regeneration.Liangjiang Huang Song Zhang Jun Wu Baojie Guo Tingting Gao Sayed Zulfiqar Ali Shah Bo Huang Yajie Li Bo Zhu Jiaqi Fan Liu Wang Yani Xiao Wenjing Liu Yao Tian Zhengyu Fang Yingying Lv Lingfeng Xie Sheng Yao Gaotan Ke Xiaolin Huang Ying Huang Yujuan Li Yi Jia Zhongwen Li Guihai Feng Yan Huo Wei Li Qi Zhou Jie Hao Baoyang Hu Hong Chen 2023Signal Transduction and Targeted Therapy2023,8,12:0
6Effects of genetic polymorphisms on methotrexate levels and toxicity in Chinese patients with acute lymphoblastic leukemia显示文摘Methotrexate(MTX)has an antitumor effect when used for the treatment of acute lymphoblastic leukemia(ALL).This study aims at evaluating the associations between 14 polymorphisms of six genes involved in MTX metabolism with serum MTX concentration and toxicity accompanying high-dose MTX.Polymorphisms in 183 Chinese patients with ALL were analyzed using TaqMan single nucleotide polymorphism genotyping assay.The serum MTX concentration was determined using homogeneous enzyme immunoassay.MTX-related toxicities were also evaluated.Renal toxicity was significantly associated with higher serum MTX concentrations at 24,48,and 72 hours,and MTX elimination delay(P=0.001,P<0.001,P<0.001,and P<0.001,respectively),whereas SLCO1B1 rs4149056 was associated with serum MTX concentrations at 48 and 72 hours,and MTX elimination delay in candidate polymorphisms(P=0.014,P=0.019,and P=0.007,respectively).SLC19A1 rs2838958 and rs3788200 were associated with serum MTX concentrations at 24 hours(P=0.016,P=0.043,respectively).MTRR rs1801394 was associated with serum MTX concentrations at 72 hours(P=0.045).Neutropenia was related to SLC19A1 rs4149056(odds ratio[OR]:3.172,95%confidence interval[CI]:1.310–7.681,P=0.011).Hepatotoxicity was associated with ABCC2 rs2273697(OR:3.494,95%CI:1.236–9.873,P=0.018)and MTRR rs1801394(OR:0.231,95%CI:0.084–0.632,P=0.004).Polymorphisms of SLCO1B1,SLC19A1,ABCC2,and MTRR genes help predict higher risk of increased MTX levels or MTX-related toxicities in adult ALL patients.Qishan Hao Yang Song Qiuyun Fang Yani Lin Long Chen Xiaodan Wang Ping Zhang Zhe Wang Xiaoyuan Gong Kaiqi Liu Qinghua Li Zheng Tian Min Wang Jianxiang Wang Yingchang Mi 2023Blood Science2023,5,1:0
7Toughening of vinyl ester resins by two-dimensional MXene nanosheets显示文摘Two-dimensional nanosheets are highly effective tougheners for vinyl ester resins.The toughening effect is related to the high specific surface area and unique two-dimensional planar structure of the nanosheets.In this study,a coupling agentγ-(2,3-epoxypropoxy)propytrimethoxysilane(Kh-560)was used to modify MXene nanosheets(M-MXene)for use in toughening vinyl ester resin.The mechanical properties,including the tensile strength,flexural strength,Young’s modulus and elongation,of neat vinyl ester resin and vinyl ester resin modified with MXene and M-MXene were investigated.The results showed that modification significantly improved the mechanical properties of the vinyl ester resin.The tensile and flexural strengths of the MXene-nanosheet-modified vinyl ester resin were 27.20%and 25.32%higher,respectively,than those of the neat vinyl ester resin.The coupling agent improved the interfacial compatibility between the MXene nanosheets and vinyl ester resin,which resulted in the tensile and flexural strengths of the M-MXene-nanosheet-modified vinyl ester resin being 52.57%and 54.60%higher,respectively,than those of the neat vinyl ester resin for a loading quantity of nanosheets of only 0.04 wt%,which is economically viable.The main mechanisms by which the nanosheets toughen the resin are crack deflection and crack pinning.Yurun Dai Heng Fang Zong Lu Zhuohong Yang Yanying Wei 2022Frontiers of Chemical Science and Engineering2022,16,11:0
8A Review of the Engineering Role of Burrowing Animals: Implication of Chinese Pangolin as an Ecosystem Engineer显示文摘Ecosystem engineers are organisms that alter the distribution of resources in the environment by creating,modifying,maintaining and/or destroying the habitat.They can affect the structure and function of the whole ecosystem furthermore.Burrowing engineers are an important group in ecosystem engineers as they play a critical role in soil translocation and habitat creation in various types of environment.However,few researchers have systematically summarized and analyzed the studies of burrowing engineers.We reviewing the existing ecological studies of burrowing engineer about their interaction with habitat through five directions:(1)soil turnover;(2)changing soil physicochemical properties;(3)changing plant community structure;(4)providing limited resources for commensal animals;and/or(5)affecting animal communities.The Chinese pangolin(Manis pentadactyla)is a typical example of burrowing mammals,in part(5),we focus on the interspecific relationships among burrow commensal species of Chinese pangolin.The engineering effects vary with environmental gradient,literature indicates that burrowing engineer play a stronger role in habitat transformation in the tropical and subtropical areas.The most common experiment method is comparative measurements(include different spatial and temporal scale),manipulative experiment is relatively few.We found that most of the engineering effects had positive feedback to the local ecosystem,increased plant abundance and resilience,increased biodiversity and consequently improved ecosystem functioning.With the global background of dramatic climate change and biodiversity loss in recent decades,we recommend future studies should improving knowledge of long-term engineering effects on population scale and landscape scale,exploring ecological cascades through trophic and engineering pathways,to better understand the attribute of the burrowing behavior of engineers to restore ecosystems and habitat creation.The review is presented as an aid to systematically expound the engineering effect of burrowing animals in the ecosystem,and provided new ideas and advice for planning and implementing conservation management.Song Sun Hongliang Dou Shichao Wei Yani Fang Zexu Long Jiao Wang Fuyu An Jinqian Xu Tingting Xue Huangjie Qiu Yan Hua Guangshun Jiang 2021Journal of Zoological Research2021,3,3:0
9P2X7/P2X4 Receptors Mediate Proliferation and Migration of Retinal Microglia in Experimental Glaucoma in Mice显示文摘Microglia are involved in the inflammatory response and retinal ganglion cell damage in glaucoma.Here,we investigated how microglia proliferate and migrate in a mouse model of chronic ocular hypertension(COH).In COH retinas,the microglial proliferation that occurred was inhibited by the P2X7 receptor(P2X7R)blocker BBG or P2X7R knockout,but not by the P2X4R blocker 5-BDBD.Treatment of primary cultured microglia with BzATP,a P2X7R agonist,mimicked the effects of cell proliferation and migration in COH retinas through the intracellular MEK/ERK signaling pathway.Transwell migration assays showed that the P2X4R agonist CTP induced microglial migration,which was completely blocked by 5-BDBD.In vivo and in vitro experiments demonstrated that ATP,released from activated Müller cells through connexin43 hemichannels,acted on P2X7R to induce microglial proliferation,and acted on P2X4R/P2X7R(mainly P2X4R)to induce microglial migration.Our results suggest that inhibiting the interaction of Müller cells and microglia may attenuate microglial proliferation and migration in glaucoma.Meng-Xi Xu Guo-Li Zhao Xin Hu Han Zhou Shu-Ying Li Fang Li Yanying Miao Bo Lei Zhongfeng Wang 2022Neuroscience Bulletin2022,38,8:0
10Histopathological Observation of Parvovirus Infection in Foxes显示文摘In this study,histopathological changes in the heart,liver,spleen,lung,kidney,intestine,brain and other organs of foxes died of parvovirus infection were observed. 11Antibiotic Sensitivity Test and Detection of Sulfonamide Resistance Gene Sul2 in Escherichia coli Isolates from Swine显示文摘[Objective] This study aimed to analyze antibiotic sensitivity and detect sulfonamide resistance gene Sul2 in 17 Escherichia coli isolates from swine.[Method]The antibiotic sensitivity of 17 E.coli isolates from swine was analyzed with Kirby-Bauer disk diffusion method.Sulfonamide resistance gene Sul2 was detected by PCR amplification with the extracted genomic DNA as a template.[Result] The 17 swine-derived E.coli isolates from different regions were resistant to various antibiotics and exhibited different multi-antibiotic resistance phenotypes,including nine isolates resistant to sulfamethoxazole,seven isolates resistant to oxacillin,ten isolates resistant to cefazolin and eight isolates resistant to erythromycin.Sulfonamide resistance gene Sul2 was successfully amplified from genomic DNA of E.coli isolates.The PCR results were basically consistent with antibiotic resistance phenotypes of these strains.[Conclusion] Sul2 gene was widespread in swine-derived E.coli,which was closely associated with sulfonamide resistance phenotypes of E.coli.Yanying ZHANG Fang XIANG Rui GUO Zhaoxing ZHANG Qiumei SHI Yuqin LIU Cairan YANG Zongze YANG Guangping GAO Guisheng GAO Zhi PENG Xun SUO 2016Agricultural Biotechnology2016,5,2:0
