Antibody-drug conjugates:Recent advances in linker chemistry显示文摘Antibody-drug conjugates(ADCs)are gradually revolutionizing clinical cancer therapy. The antibody-drug conjugate linker molecule determines both the efficacy and the adverse effects, and so has a major influence on the fate of ADCs.An ideal linker should be stable in the circulatory system and release the cytotoxic payload specifically in the tumor. However, existing linkers often release payloads nonspecifically and inevitably lead to off-target toxicity. This defect is becoming an increasingly important factor that restricts the development of ADCs.The pursuit of ADCs with optimal therapeutic windows has resulted in remarkable progress in the discovery and development of novel linkers. The present review summarizes the advance of the chemical trigger,linker-antibody attachment and linker-payload attachment over the last 5 years, and describes the ADMET properties of ADCs.This work also helps clarify future developmental directions for the linkers.Zheng Su Dian Xiao Fei Xie Lianqi Liu Yanming Wang Shiyong Fan Xinbo Zhou Song Li 2021Acta Pharmaceutica Sinica B2021,11,12:5
Multi-objective clustering analysis via combinatorial pigeon inspired optimization显示文摘Multi-objective data clustering is an important issue in data mining, and the realization of data clustering using the multiobjective optimization technique is a significant topic. A combinatorial multi-objective pigeon inspired optimization(CMOPIO)with ring topology is proposed to solve the clustering problem in this paper. In the CMOPIO, a delta-locus based coding approach is employed to encode the pigeons. Thus, the length of pigeon representation and the dimension of the search space are significantly reduced. Thereby, the computational load can be effectively depressed. In this way, the pigeon inspired optimization(PIO) algorithm can be discretized with an auxiliary vector to address data clustering. Moreover, an index-based ring topology with the ability of contributing to maintain flock diversity is adopted to improve the CMOPIO performance. Comparative simulation results demonstrate the feasibility and effectiveness of our proposed CMOPIO for solving data clustering problems.CHEN Lin DUAN HaiBin FAN YanMing WEI Chen 2020Science China(Technological Sciences)2020,63,7:4
Pigeon-Inspired Circular Formation Control for Multi-UAV System with Limited Target Information显示文摘The problem of cooperative circular formation with limited target information for multiple Unmanned Aerial Vehicle(UAV)system is addressed in this paper.A pigeon-inspired circular formation control method is proposed to form the desired circular distribution in a plane based on the intelligent pigeon behavior during hovering. To reach the goal of prescribed radius and angular distribution, the controller is designed consisting of a circular movement part and a formation distribution part.Therein, the circular movement part is designed to make each UAV rotate around the speci-ed circle at the same angular speed only using the relative position between the UAV and the target. The formation distribution part could adjust the angular distance between each UAV and its neighbors with the jointly connected network to reduce communication cost.To smooth the speed variation, nonlinear PID-type method is delivered throughout the evolution of the system. The convergence analysis of the proposed control protocol is presented using Lyapunov theory and graph tools. The e®ectiveness of the proposed control strategies is demonstrated through numerical simulations.Mengzhen Huo Haibin Duan Yanming Fan 2021Guidance, Navigation and Control2021,1,1:3
Bacteroides ovatus-mediated CD27^(−)MAIT cell activation is associated with obesity-related T2D progression显示文摘Type 2 diabetes(T2D)is highly associated with obesity. However, the factors that drive the transition from excessive weight gain to glucose metabolism disruption are still uncertain and seem to revolve around systemic immune disorder.Mucosal-associated invariant T(MAIT)cells, which are innate-like T cells that recognize bacterial metabolites, have been reported to be altered in obese people and to lead to metabolic dysfunction during obesity.By studying the immunophenotypes of blood MAIT cells from a cross-sectional cohort of obese participants with/without T2D,we found an elevation in CD27^(-)negative(CD27−)MAIT cells producing a high level of IL-17 under T2D obese conditions, which could be positively correlated with impaired glucose metabolism in obese people. We further explored microbial translocation caused by gut barrier dysfunction in obese people as a triggering factor of MAIT cell abnormalities.Specifically, accumulation of the bacterial strain Bacteroides ovatus in the peripheral blood drove IL-17^(-)producing CD27−MAIT cell expansion and could be associated with T2D risk in obese individuals.Overall, these results suggest that an aberrant gut microbiota–immune axis in obese people may drive or exacerbate T2D.Importantly,CD27−MAIT cell subsets and Bacteroides ovatus could represent targets for novel interventional strategies. Our findings extend current knowledge regarding the clinical relevance of body mass index(BMI)-associated variation in circulating MAIT cells to reveal the role of these cells in obesity-related T2D progression and the underlying cellular mechanisms.Yue Li Yi Yang Jin Wang Peihong Cai Mei Li Xixiang Tang Ying Tan Yuchan Wang Fan Zhang Xiaofeng Wen Qiaoxing Liang Yuanpeng Nie Tufeng Chen Xiang Peng Xuemin He Yanhua Zhu Guojun Shi Wai WCheung Lai Wei Yanming Chen Yan Lu 2022Cellular & Molecular Immunology2022,19,7:2

Promoting electrochemical reduction of CO_(2) to ethanol by B/N-doped sp^(3)/sp^(2) nanocarbon electrode显示文摘Electrochemical reduction of CO_(2) to value-added chemicals holds promise for carbon utilization and renewable electricity storage. However, selective CO_(2) reduction to multi-carbon fuels remains a significant challenge. Here,we report that B/N-doped sp^(3)/sp^(2) hybridized nanocarbon(BNHC),consisting of ultra-small nanoparticles with a sp^(3) carbon core covered by a sp^(2) carbon shell,is an efficient electrocatalyst for electrochemical reduction of CO_(2) to ethanol at relatively low overpotentials.CO_(2) reduction occurs with a Faradaic efficiency of 58.8%-69.1% for ethanol and acetate production at -0.5∼-0.6 V(vs.RHE),among which 51.6%-56.0% is for ethanol. The high selectivity for ethanol is due to the integrated effect of sp^(3)/sp^(2) carbon and B/N doping. Both sp^(3) carbon and B/N doping contribute to enhanced ethanol production with sp^(2) carbon reducing the overpotential for CO_(2) reduction to ethanol.Yanming Liu Haolei Yang Xinfei Fan Bing Shan Thomas J.Meyer 2022Chinese Chemical Letters2022,33,10:2

Density Functional Theory Study of Oxygen Atom Adsorption on Different Surfaces of Pyrite显示文摘We discussed the oxidation differential and mechanisms on different planes of pyrite. The experimental results show that the oxidation priority is:(222) plane>(200) plane>(200) plane, and there is no direct correlation between the crystal plane index, the atom numbers, and the oxidation priority. However, with more exchanged charge among atoms, the oxidation could be conducted more easily, and the distribution rule of the electric charge conforms with the variation trend of adsorption energy, which will provide more overall cognition on the oxidation mechanism of pyrite from the atomic scale.黄腾 雷绍民 JI Mengjiao LIU Yuanyuan FAN Yanming 2017Journal of Wuhan University of Technology(Materials Science)2017,32,6:1
Suppression of partially irreversible phase transition in O′3-Na_(3)Ni_(2)SbO_(6)cathode for sodium-ion batteries by interlayered structural modulation显示文摘As a promising cathode material for sodium ion batteries,honeycomb-ordered layered Na_(3)Ni_(2)Sb O_(6)still suffers from rapid capacity fading because of partially irreversible phase transition.Herein,a substitution of Na+by Rb+with a larger ionic radius in honeycomb layered Na_(3)-xRbxNi_(2)Sb O_(6)is proposed to modulate the interlayer structure. The results unveil that biphasic transition reversibility of the intermediate P′3phase is substantially enhanced, and the structure evolution behavior during the charge/discharge process changes due to the structural modulation, which contributes to a suppression of the unfavorable O_(1)phase and an alleviation of the lattice distortion. Moreover,Rb substituted samples exhibited an improved Na+(de)intercalation thermodynamics and kinetics.Attributed to the modifications, the sample with optimized Rb content delivers superior cycle stability and rate capacity, demonstrating a feasible strategy for suppressing irreversible phase transition and developing high-performance honeycomb layered materials for sodium ion batteries.Jingjie Feng Jiajie Li Ni Wen Siyuan Chen Jian Wu Qinghua Fan Youzhong Dong Quan Kuang Yanming Zhao 2022Journal of Energy Chemistry2022,31,10:0
Optimization of projective transformation matrix in image stitching based on chaotic genetic algorithm显示文摘Purpose–The purpose of this paper is to present weighted Euclidean distance for measuring whether the fitting of projective transformation matrix is more reliable in feature-based image stitching.Design/methodology/approach–The hybrid model of weighted Euclidean distance criterion and intelligent chaotic genetic algorithm(CGA)is established to achieve a more accurate matrix in image stitching.Feature-based image stitching is used in this paper for it can handle non-affine situations.Scale invariant feature transform is applied to extract the key points, and the false points are excluded using random sampling consistency(RANSAC)algorithm.Findings–This work improved GA by combination with chaos’s ergodicity,so that it can be applied to search a better solution on the basis of the matrix solved by Levenberg-Marquardt.The addition of an external loop in RANSAC can help obtain more accurate matrix with large probability.Series of experimental results are presented to demonstrate the feasibility and effectiveness of the proposed approaches.Practical implications–The modified feature-based method proposed in this paper can be easily applied to practice and can obtain a better image stitching performance with a good robustness.Originality/value–A hybrid model of weighted Euclidean distance criterion and CGA is proposed for optimization of projective transformation matrix in image stitching. The authors introduce chaos theory into GA to modify its search strategy.Yanming Fan Ming Li 2013International Journal of Intelligent Computing and Cybernetics2013,6,4:0
Cloning and Analysis of Genes SAMS From Glycine soja显示文摘S-adenosylmethionine (SAM) plays important role in trans-methyl reactions. Under the condition of drought (30% PEG), salinity (200 mmol · L-1 NaCl) and low temperature (4℃), total RNA was extracted from the leaf and the first strand of cDNA was synthesized with reverse transcription. S-adenosylmethionine synthetase gene (SAMS gene) was amplified by PCR with the first strand cDNA as template and a pair of primers which was based on constructed ESTs sequence. Full-length SAMS gene sequence was obtained by BLAST comparison. According to the analysis, completed sequence of SAMS gene was integrality. The sequence of the SAMS gene was 1 185 bp in length with an opening reading frame (ORF) encoding 394 amino acids. The cDNA sequence showed a significant homology to the SAM genes from Phaseolus lunatus (89%), Medicago sativa (85%). A prokaryotic expression vectors based on pET-32b had been constructed and prokaryotic expression was analyzed in order to lay a strong foundation for resist adversity function analysis through situation of genic expression analysis.FAN Jinping BAI Xi LI Yong JI Wei WANG Xi ZHU Yanming 2008Journal of Northeast Agricultural University(English Edition)2008,15,1:0
Seasonal variations in the composition and functional profiles of gut microbiota reflect dietary changes in plateau pikas显示文摘Seasonal variations in gut microbiota of small mammals and how they are influenced by environmental variables are relatively poorly understood. We sampled 162 wild plateau pikas(Ochotona curzoniae)in 4 seasons over 2 and a half years and recorded the air temperature, precipitation, and nutrient content in edible vegetation at the sampling site. After conducting 16S rRNA and shotgun metagenomic sequencing,we found that the highest alpha diversity, the relative abundance of Firmicutes, and the simplest co-occurrence network occurred in winter, whereas the highest relative abundance of Proteobacteria and the most complex network structure were observed in spring. The highest relative abundance of Verrucomicrobiota and Spirochaetota was seen in summer and autumn, respectively.Air temperature, precipitation, and the contents of crude protein, crude fiber, and polysaccharide in vegetation had significant effects on the seasonal changes in gut microbiota.Diet contributed more to microbial variation than climatic factors.Metagenomic analysis revealed that the amino acid metabolism pathway and axillary activity enzymes were most abundant in summer, while abundance of carbohydrate-binding modules and carbohydrate esterases was highest in spring. These microbial variations were related to the changes in dietary nutrition, indicating that gut microbiota of plateau pika contribute to the efficient use of food resources. This study provides new evidence of how external environmental factors affect the intestinal environment of small mammals.Chao FAN Liangzhi ZHANG Shangang JIA Xianjiang TANG Haibo FU Wenjing LI Chuanfa LIU He ZHANG Qi CHENG Yanming ZHANG 2022Integrative Zoology2022,17,3:0

Programmed self-assembly of DNA origami nanoblocks into anisotropic higher-order nanopatterns显示文摘Anisotropic nanopatterns have potentials in constructing novel plasmonic structures which have various applications in such as super-resolution microscopy, medicine, and sensors. However, it remains challenging to build big anisotropic nanopatterns that are suitable for big noble metal nanoparticles. Herein, we report a simple and reliable strategy for constructing DNA origami-based big anisotropic nanopatterns with controlled size and shape, nanoscale resolution, and fully addressability. Two kinds of basic DNA origami nanoblocks-cross-shaped and rectangular DNA origami units were used. We have demonstrated that by encoding nanoblocks' edges, anisotropic higher-order nanopatterns, such as dimer, trimer, tetramer and mini 'windmill' like pentamer nanopatterns could be constructed. To show the potential use as template to direct the assembly of anisotropic nanoparticles arrays, a proof of concept work was conducted by anchoring streptavidin nanoparticles on the 'windmill' template to form a chiral array. Significantly, these nanopatterns have the sizes of hundreds of nanometers, which are in principle also suitable for big noble metal nanoparticles arrays.FU YanMing CHAO Jie LIU HuaJie FAN ChunHai 2013Chinese Science Bulletin2013,58,21:0
From prodrug to pro-prodrug:hypoxia-sensitive antibody–drug conjugates显示文摘Dear Editor,Antibody‒drug conjugates(ADCs),famous as biological targeting prodrugs, are gradually revolutionizing clinical cancer therapy. However, less than 1%of the dosed ADCs accumulate in the tumors.1 Therefore, the nonspecific release of the highly toxic payload(MMAE et al.,10−12–10−10 M)is a real threat, which could induce severe off-target toxicity.2 This danger necessitates strict requirements for the design of the linker. To date, the mainstream enzyme cleavable linkers,includingβ-glucuronidase cleavable linkers,sulfatase-cleavable linkers, and the most popular cathepsin cleavable linkers(valine-citrulline linker),all face this nonspecific release problem,3 because their cleaving enzymes are widely distributed with no significant difference in their quantities between tumor tissues and normal tissues.Yanming Wang Dian Xiao Jiaguo Li Shiyong Fan Fei Xie Wu Zhong Xinbo Zhou Song Li 2022Signal Transduction and Targeted Therapy2022,7,2:0
Cloning and Functional Analysis of Porcine Cycling A显示文摘Cyclin A is a key regulator of the cell cycle. Its expression may become disrupted in virus-infected cells, leading to deregulation of the cell cycle and increased cell proliferation. Here,we cloned the porcine cyclin A gene and verified its functionality in swine umbilicus vein endothelial cells (SUVEC).The human cyclin A gene was used to probe databases to clone the pig cyclin A gene electronically. The identified porcine cDNA contained an open reading frame of 1,299 bp,encoding 432 amino acids, the same length as the human cyclin A protein. The porcine cyclin A gene comprises eight exons on chromosome 8.The sequence of the in silico clone and expression of this novel gene were confirmed in SUVEC by reverse transcription PCR.Western blotting of cell lysates from SUVEC transfected with a cyclin A enhanced green fluorescent protein (EGFP) fusion construct revealed a band at approximately 40 kDa.Confocal microscopy of CycA-EGFP-expressing cells showed that the fusion protein was expressed in the nucleus.Flow cytometry demonstrated that more stably expressing SUVEC-CycA-EGFP were in G1 phase (15% to 20% increase) and fewer were in S phase (18% decrease) compared with control cells.MTS assays showed that the proliferative activity of SUVEC-CycAG-EGFP was significantly higher than that of the control cells. In conclusion,we have cloned the pig cyclin A gene and demonstrated that its biological function is consistent with cyclin A in other mammalian species. This provides a foundation for future research on the impact of virus infection on cyclin A.Li Fan Qinghai Tang Yanming Zhang Wei Liu Gang Tong 2011Journal of Animal Science and Biotechnology2011,2,1:0
Lagging and Its Kinetic Mechanism of Hydrocarbon Re-generation from Organic Matters in Coal显示文摘Based on the composite analysis of the coal sample series with natural and artificial maturation, the lagging and its kinetic mechanism of the hydrocarbon re-generation from the organic matters in coals were studied using the Rock-Eval gas chromatogram (PyGC) method. The results show that the maturation at the hydrocarbon regeneration peak shifts regularly forward with increasing the starting maturation and the deadline of the hydrocarbon regeneration lies about at 4.0% R o. The difference value between the peak and starting maturation of the hydrocarbon re-generation develops in a parabolalike pattern with increasing the starting maturation, and the resolute and relative laggings evolve in stage, from which the lagging depth could be predicted. The peak halfwidth of the hydrocarbon re-generation curve develops as the starting maturation increases, which might indicate that the hydrocarbonderived rocks with the starting maturation lied about at oilgenerated peak might be relatively high in the hydrocarbonregenerated amount. In the meantime, the mean reactivated energy of the coal samples with starting maturation develops in four stages that are highly consistent with those of the hydrocarbonregenerated amount and lagging, which indicated that the hydrocarbon re-generation is strictly controlled by the geochemical mechanism of the reactive kinetics.Qin Yong Zhang Yousheng Zhu Yanming Fan Bingheng Jiang Bo Li Tianzhong College of Mineral Resources and Environment Science, China University of Mining and Technology, Xuzhou 221008 2000Journal of Earth Science2000,19,1:0
