1ALM1, encoding a Fe-superoxide dismutase, is critical for rice chloroplast biogenesis and drought stress response显示文摘Chloroplasts are the center of plant life activities including photosynthesis,growth and development,and abiotic stress response.Chloroplast development and biogenesis in rice have been studied in detail,but how does abiotic stress affect chloroplasts is less studied.We obtained an albino mutant,alm1,whose chlorophyll content was greatly decreased.Transmission electron microscopy showed that chloroplast development in alm1 was blocked,especially in thylakoid-like structures,which could not form normally.The ALM1 gene encodes a chloroplast-localized superoxide dismutase.Full-length ALM1 successfully restored the non-albino phenotype,and in knockout lines,the albino phenotype reappeared.The ALM1gene is expressed mainly in young leaves.alm1 plants died as a consequence of excessive reactive oxygen accumulation after the third-leaf stage.A series of biochemical assays verified that ALM1 interacted with the OsTrxz protein,which is one of the components of plastid-encoded RNA polymerase (PEP) complexes.A western blot experiment indicated that ALM1 played an important role in stabilizing OsTrxz in rice.An overexpression test of ALM1 revealed that ALM1 can increase drought resistance by removing excess reactive oxygen in rice seedlings.This study suggests that ALM1 not only participates in rice chloroplast biogenesis,but also increases rice stress resistance by scavenging excess reactive oxygen.Yanwei Wang Chen Deng Pengfei Ai Xue’an Cui Zhiguo Zhang 2021The Crop Journal2021,9,5:4

4VSe_(2)/V_(2)C heterocatalyst with built-in electric field for efficient lithium-sulfur batteries:Remedies polysulfide shuttle and conversion kinetics显示文摘Lithium sulfur(Li-S)battery is a kind of burgeoning energy storage system with high energy density.However,the electrolyte-soluble intermediate lithium polysulfides(Li PSs)undergo notorious shuttle effect,which seriously hinders the commercialization of Li-S batteries.Herein,a unique VSe_(2)/V_(2)C heterostructure with local built-in electric field was rationally engineered from V_(2)C parent via a facile thermal selenization process.It exquisitely synergizes the strong affinity of V_(2)C with the effective electrocatalytic activity of VSe_(2).More importantly,the local built-in electric field at the heterointerface can sufficiently promote the electron/ion transport ability and eventually boost the conversion kinetics of sulfur species.The Li-S battery equipped with VSe_(2)/V_(2)C-CNTs-PP separator achieved an outstanding initial specific capacity of 1439.1 m A h g^(-1)with a high capacity retention of 73%after 100 cycles at0.1 C.More impressively,a wonderful capacity of 571.6 mA h g^(-1)was effectively maintained after 600cycles at 2 C with a capacity decay rate of 0.07%.Even under a sulfur loading of 4.8 mg cm^(-2),areal capacity still can be up to 5.6 m A h cm^(-2).In-situ Raman tests explicitly illustrate the effectiveness of VSe_(2)/V_(2)C-CNTs modifier in restricting Li PSs shuttle.Combined with density functional theory calculations,the underlying mechanism of VSe_(2)/V_(2)C heterostructure for remedying Li PSs shuttling and conversion kinetics was deciphered.The strategy of constructing VSe_(2)/V_(2)C heterocatalyst in this work proposes a universal protocol to design metal selenide-based separator modifier for Li-S battery.Besides,it opens an efficient avenue for the separator engineering of Li-S batteries.Yanwei Lv Lina Bai Qi Jin Siyu Deng Xinzhi Ma Fengfeng Han Juan Wang Lirong Zhang Lili Wu Xitian Zhang 2024Journal of Energy Chemistry2024,89,2:0
5A digital microfluidic system with 3D microstructures for single-cell culture显示文摘Despite the precise controllability of droplet samples in digital microfluidic(DMF)systems,their capability in isolating single cells for long-time culture is still limited:typically,only a few cells can be captured on an electrode.Although fabricating small-sized hydrophilic micropatches on an electrode aids single-cell capture,the actuation voltage for droplet transportation has to be significantly raised,resulting in a shorter lifetime for the DMF chip and a larger risk of damaging the cells.In this work,a DMF system with 3D microstructures engineered on-chip is proposed to form semi-closed micro-wells for efficient single-cell isolation and long-time culture.Our optimum results showed that approximately 20%of the micro-wells over a 30×30 array were occupied by isolated single cells.In addition,low-evaporation-temperature oil and surfactant aided the system in achieving a low droplet actuation voltage of 36V,which was 4 times lower than the typical 150 V,minimizing the potential damage to the cells in the droplets and to the DMF chip.To exemplify the technological advances,drug sensitivity tests were run in our DMF system to investigate the cell response of breast cancer cells(MDA-MB-231)and breast normal cells(MCF-10A)to a widely used chemotherapeutic drug,Cisplatin(Cis).The results on-chip were consistent with those screened in conventional 96-well plates.This novel,simple and robust single-cell trapping method has great potential in biological research at the single cell level.Jiao Zhai Haoran Li Ada Hang-Heng Wong Cheng Dong Shuhong Yi Yanwei Jia Pui-In Mak Chu-Xia Deng Rui PMartins 2020Microsystems & Nanoengineering2020,6,1:0
6The complete and fully-phased diploid genome of a male Han Chinese显示文摘Since the release of the complete human genome,the priority of human genomic study has now been shifting towards closing gaps in ethnic diversity.Here,we present a fully phased and well-annotated diploid human genome from a Han Chinese male individual(CN1),in which the assemblies of both haploids achieve the telomere-to-telomere(T2T)level.Comparison of this diploid genome with the CHM13 haploid T2T genome revealed significant variations in the centromere.Outside the centromere,we discovered 11,413 structural variations,including numerous novel ones.We also detected thousands of CN1 alleles that have accumulated high substitution rates and a few that have been under positive selection in the East Asian population.Further,we found that CN1 outperforms CHM13 as a reference genome in mapping and variant calling for the East Asian population owing to the distinct structural variants of the two references.Comparison of SNP calling for a large cohort of 8869 Chinese genomes using CN1 and CHM13 as reference respectively showed that the reference bias profoundly impacts rare SNP calling,with nearly 2 million rare SNPs miss-called with different reference genomes.Finally,applying the CN1 as a reference,we discovered 5.80 Mb and 4.21 Mb putative introgression sequences from Neanderthal and Denisovan,respectively,including many East Asian specific ones undetected using CHM13 as the reference.Our analyses reveal the advances of using CN1 as a reference for population genomic studies and paleo-genomic studies.This complete genome will serve as an alternative reference for future genomic studies on the East Asian population.Chentao Yang Yang Zhou Yanni Song Dongya Wu Yan Zeng Lei Nie Panhong Liu Shilong Zhang Guangji Chen Jinjin Xu Hongling Zhou Long Zhou Xiaobo Qian Chenlu Liu Shangjin Tan Chengran Zhou Wei Dai Mengyang Xu Yanwei Qi Xiaobo Wang Lidong Guo Guangyi Fan Aijun Wang Yuan Deng Yong Zhang Jiazheng Jin Yunqiu He Chunxue Guo Guoji Guo Qing Zhou Xun Xu Huanming Yang Jian Wang Shuhua Xu Yafei Mao Xin Jin Jue Ruan Guojie Zhang 2023Cell Research2023,33,10:0
