| 2 | miR-124 suppresses multiple steps of breast cancer metastasis by targeting a cohort of pro-metastatic genes in vitro显示文摘Metastasis is a multistep process involving modification of morphology to suit migration, reduction of tumor cell adhesion to the extracellular matrix, increase of cell mobility, tumor cell resistance to anoikis, and other steps. MicroRNAs are well-suited to regulate tumor metastasis due to their capacity to repress numerous target genes in a coordinated manner, thereby enabling their intervention at multiple steps of the invasion-metastasis cascade. In this study, we identified a microRNA exemplifying these attributes, miR-124, whose expression was reduced in aggressive MDA-MB-231 and SK-3rd breast cancer cells. Down-regulation of miR-124 expression in highly aggressive breast cancer cells contributed in part to DNA hypermethylation around the promoters of the three genes encoding miR-124. Ectopic expression of miR-124 in MDA-MB-231 cells suppressed metastasis-related traits including formation of spindle-like morphology, migratory capacity, adhesion to fibronectin, and anoikis. | 3 | TCP7 functions redundantly with several Class Ⅰ TCPs and regulates endoreplication in Arabidopsis显示文摘TCP(TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR) proteins, a family of plantspecific transcription factors, play important roles in many developmental processes. However, genetic and functional redundancy among class I TCP limits the analysis of their biological roles. Here, we identified a dominantnegative mutant of Arabidopsis thaliana TCP7 named leaf curling-upward(lcu), which exhibits smaller leaf cells and shorter hypocotyls than the wild type, due to defective endoreplication. A septuple loss-of-function mutant of TCP7, TCP8, TCP14, TCP15, TCP21, TCP22, and TCP23 displayed similar developmental defects to those of lcu.Genome-wide RNA-sequencing showed that lcu and the septuple mutant share many misexpressed genes.Intriguingly, TCP7 directly targets the CYCLIN D1;1 (CYCD1;1) locus and activates its transcription. We determined that the C-terminus of TCP7 accounts for its transcriptional activation activity. Furthermore, the mutant protein LCU exhibited reduced transcriptional activation activity due to the introduction of an EAR-like repressive domain at its C-terminus. | 5 | Bi-level Coordinated Planning of Active Distribution Network Considering Demand Response Resources and Severely Restricted Scenarios显示文摘Due to the uncertainty and fluctuation of distributed generation(DG)and load,the operation of active distribution network(ADN)is affected by multi-dimension factors which are described by massive operation scenarios.Efficient and accurate screening of severely restricted scenarios(SRSs)has become a new challenge in ADN planning.In this paper,a novel bi-level coordinated planning model which combines the short-time-scale operation problem with the long-time-scale planning problem is proposed.At the upper level,the demand response(DR)resource,an effective non-component planning resource characterized by low capacity price,high energy price,and short contract term,is co-optimized with the configuration of lines and energy storage systems(ESSs)to achieve the economic trade-off between the investment cost and the operation cost under SRSs.At the lower level,with the planning scheme obtained from the upper level,massive operation problems are optimized to minimize the daily operation cost;and the SRSs are provided to the upper level through a shadow-price-based scenario screening method,which simulates the planning information(i.e.,the restricted degrees of operation scenarios)feedback process from ADN operators to ADN planners.Case studies on a 62-node distribution system in Jianshan New District,Zhejiang Province,China,illustrate the effectiveness of the proposed bi-level coordinated planning model considering DR resources and SRSs. | Huang Yixin Lin Zhenzhi Liu Xinyi Yang Li Dan Yangqing Zhu Yanwei Ding Yi Wang Qin | 2021 | Journal of Modern Power Systems and Clean Energy2021,9,5: | 3 |

| 9 | A study on melanoma treatment using dendritic cells loaded with antigens purified from melanoma cell lines显示文摘Objective The aim of this study was to purify effective tumor peptide complexes from human melanoma cell lines to enhance the treatment effects on melanoma.Methods We purified heat shock protein 70(HSP70)-peptide complexes(PCs)from human melanoma cell lines A375,A875,M21,M14,WM-35,and SK-HEL-1.We named the purified product as M-HSP70-PCs and determined its immunological activities.Autologous HSP70-PCs purified from primary tumor cells of melanoma patients(9 cases)were used as controls.These two tumor antigenic complexes were loaded into dendritic cells(DCs)and used to stimulate an antitumor response against tumor cells in the corresponding patients.Results Mature DCs pulsed with M-HSP70-PCs stimulated autologous T cells to secrete the same levels of type I cytokines as the autologous HSP70-PCs.Moreover,DCs pulsed with M-HSP70-PCs endued CIK cells with an equal ability as autologous HSP70-PCs to kill melanoma cells in the patients.Conclusion M-HSP70-PCs may be used as an efficient and generalized tumor antigen in the treatment of DC-based malignant melanoma. | Yanwei Gao Xia Chen Weishi Gao Xiangji Lu Lin Peng | 2020 | Oncology and Translational Medicine2020,6,1: | 2 |
