2Umbilical cord mesenchymal stem cell transplantation for the treatment of Duchenne muscular dystrophy显示文摘Due to their relative abundance,stable biological properties and excellent reproductive activity,umbilical cord mesenchymal stem cells have previously been utilized for the treatment of Duchenne muscular dystrophy,which is a muscular atrophy disease.Three patients who were clinically and pathologically diagnosed with Duchenne muscular dystrophy were transplanted with umbilical cord mesenchymal stem cells by intravenous infusion,in combination with multi-point intramuscular injection.They were followed up for 12 months after cell transplantation.Results showed that clinical symptoms significantly improved,daily living activity and muscle strength were enhanced,the sero-enzyme,electromyogram,and MRI scans showed improvement,and dystrophin was expressed in the muscle cell membrane.Hematoxylin-eosin staining of a muscle biopsy revealed that muscle fibers were well arranged,fibrous degeneration was alleviated,and fat infiltration was improved.These pieces of evidence suggest that umbilical cord mesenchymal stem cell transplantation can be considered as a new regimen for Duchenne muscular dystrophy.Xiaofeng Yang Yifeng Xu Naiwu Lu Yibin Zhang Hongmei Wang Xin Lu Jiping Cui JinxuZhou Hong Shan Yanxiang Wu Xinping Liu 2011Neural Regeneration Research2011,6,10:4

4RNA干扰技术裸鼠体内沉默缺氧诱导因子1α的表达对宫颈癌的抑瘤效应(英文)显示文摘Objective: The aim of our study was to observe the anti-tumor effect of silencing the expression of HIF-1α on cervical cancer in nude mice and to explore its mechanism of action. Methods: Human cervical cancer cell line Siha cells were divided into 3 groups: mock control group, control group transfected with scrambled sequence plasmid, and experimental group transfected with pU-HIF-1a-shRNA eukaryotic expression plasmid. Cultured cells of the three groups were inoculated in nude mice to establish cervical cancer-bearing nude mice. HIF-1α RNAi assay was performed to evaluate the tumor-suppressive effect of HIF-1α silencing on cervical cancer-bearing nude mice. Immunohistochemistry and Western blot were used to observe the distribution and protein expression of HIF-1α and GLUT1, while RT-PCR was adopted to detect the gene expression of HIF-1α, GLUTl and HK II. The product of glycolysis (lactic acid) and apoptosis in tumor cells were detected by colorimetry and semi-quantitative TUNEL staining, respectively. Results: The tumor growth in experimental group was significantly slower than that in the two control groups (P < 0.05). In the 50th day after transplantation, the tumor weight in the experimental group was (1.90 ± 0.28) g, significantly lower than (2.95 ± 0.77) g in the control group and (2.54 ± 0.56) g in the mock group (P < 0.01). In the experimental group, the gene and protein levels of HIF-1α were 0.45 ± 0.04 and 1.25 ± 0.92, and the levels of GLUT1 were 0.32 ± 0.02 and 1.25 ± 0.48, respectively. Both indicators in HIF-1α and GLUT1 were lower than that in the two control groups (P < 0.05). The expression levels of HK II gene and lactic acid in the experimental group were lower than that in the two control groups (P < 0.05), but the apoptotic cells were much more numerous in the experimental group than that in matched control groups (P < 0.01). Conclusion: The gene therapy by siRNA targeted silencing of HIF-1α may down-regulate its downstream genes GLUT1 and HK II expression, therefore, to reduce the tumor glycolysis activity and promote tumor cell apoptosis, and exert a tumor-suppressing effect in vivo.Yanxiang Cheng Wen Fan Bingshu Li Qin Wang Min Hu Li Hong 2010The Chinese-German Journal of Clinical Oncology2010,9,7:1
5Ion-exchange polymers modified bacterial cellulose electrodes for the selective removal of nitrite ions from tail water of dyeing wastewater显示文摘Ion-exchange polymer and modified carbonization bacterial cellulose(CBC)electrodes were fabricated using varying amounts of cation-exchange polymers(glutaric acid(GA)and sulfosuccinic acid(SSA))and assembled within an asymmetric capacitive deionization unit(p-CDI).The performance of selective NO2-electro-adsorption was studied.The AC/CBCSSA group showed a better salt adsorption capacity(14.56 mg/g)and nitrite removal efficiency(71.01%)than the AC/CBC-GA(10.72 mg/g,47.83%)and AC/AC(4.81 mg/g,12.74%)groups.It was confirmed that the CBC-SSA/GA electrodes enhanced nitrite selectivity and increased the adsorption capacity,and the total amounts of adsorbed anions increased when the applied voltage was increased from 0.8 to 1.2 V,while the molar fraction of nitrate decreased.The competitive and preferential adsorption of anions was further investigated using different binary solutions of anions and occurred in the following sequence:NO2->SO42->NO3->F-≈Cl-.Furthermore,the p-CDI units were applied to remove nitrite in real wastewater samples,and the results showed that they had excellent reusability and application for use in dyeing wastewater treatment.Danping Li Xun-an Ning Yiqian Yuan Yanxiang Hong Jianpei Zhang 2020Journal of Environmental Sciences2020,32,5:1
6A controllable fabrication improved silicon nanowire array sensor on(111)SOI for accurate bio-analysis application显示文摘Silicon nanowire field-effect transistor(SiNW-FET)sensors possess the ability of rapid response,real-time,and label-free detection with high sensitivity and selectivity in the analysis of charged molecules.Their nano-scale size makes them well suited for ultralow detection of charged molecules,but also brings the uniformity fabrication challenging,thus limiting their large-scale application.By a horizontal control approach,highly controllable silicon nanowires arrays at the top of the silicon-on-insulator(SOI)wafer(T-SiNW)were developed in our previous work.To further improve the device uniformity,here a novel SiNW fabricated approach was carefully designed by the combination of horizontal and vertical control.The new silicon nanowires appeared at the bottom of the top silicon layer(B-SiNW).The B-SiNW has a relatively low requirement on the fabrication process and better device uniformity compared to T-SiNW.These improvements resulted in the B-SiNW device with a lower current fluctuation(4.1 nA with 5.1%variations)in the flowing liquid,compared to the T-SiNW device(4.4 nA with 11%variations).Further,in quantitative detection of 40 ng/mL MMP-9,the B-SiNW sensors provided larger signals and lower fluctuation(normalized average response value:0.57 with 4.2%variations),compared to the T-SiNW sensors(0.41 with 12.1%variations),thus indicating a more accurate bio-analysis application of the B-SiNW sensor.This work advances the nanowire sensor technology a step closer toward large-scale application to create stable sensing platforms in disease diagnosis and monitoring.Zicheng Lu Hong Zhou Yi Wang Yanxiang Liu Tie Li 2022Nano Research2022,15,8:0

8RNA干扰HIF-1α的表达能抑制宫颈癌血管生成和糖代谢(英文)显示文摘Objective:Cervical cancer has become a major public health problem.The development of effective,systemic therapies for cervical cancer is highly desired.We show here that hypoxia inducible factor-1α(HIF-1α) was indicated as an attractive therapeutic molecular target for cervical cancer.Methods:Firstly,we observed the expressional level of HIF-1α in cervical cancer and Hela and Siha cell lines.Secondly,by constructuring HIF-1α shRNA targeting human HIF-1α mRNA common sequence and transfecting it with plasmid to cervical cell,we detected the changes of HIF-1α and its downstream genes levels VEGF.Then we injected selected stably transfected cell line into athymic nude mice to estimate its' antitumor effects.Results:We observed that HIF-1α inhibition was related to down-regulated VEGF resulting in prevention of angiogenesis,then leading to slower-growing tumors.Conclusion:The underlying concept of transfecting a HIF-1α shRNA expression vector to block the HIF-1α holds promise as the clinical potential of gene therapy for cervical cancer.Yanxiang Cheng Li Hong Xuexian Xu Wen Fan 2010The Chinese-German Journal of Clinical Oncology2010,9,5:0

11Morphological and Molecular Identification of Alternaria alternata Causing Leaf Spot Disease on Huangdi Banana in China显示文摘[Objectives]The paper was to identify Alternaria alternata causing leaf spot disease on Huangdi banana in China.[Methods]Fungal isolates were isolated and identified by morphological features,molecular identification and pathogenicity test.[Results]There were light to dark brown,tiny oval spots on leaves.The causal agent isolated from affected leaves was identified as A.alternata based on the morphological properties,coupled with sequence analyses of the internal transcribed spacer(ITS)region,large subunit ribosomal DNA(LSU rDNA)and the translation elongation factor 1-alpha(TEF-1α)gene.Koch s postulates were fulfilled by successful re-isolation of pathogen from the artificial inoculated leaves.[Conclusions]To our knowledge,this is the first report of leaf spot caused by A.alternata on Huangdi banana in China.The identification of A.alternata as the causal agent of the observed leaf spot disease on Huangdi banana is critical to the prevention and control of this disease in the future.Xin ZHANG Hong ZHAO Yanfei OUYANG Yixian XIE Jun PENG Fanyun ZENG Yanxiang QI 2023Plant Diseases and Pests2023,14,3:0
12The impact of nanoporous SiN_x interlayer growth position on high-quality GaN epitaxial films显示文摘The impact of nanoporous SiN x interlayer growth position on high-quality GaN epitaxial film was elucidated from the behavior of dislocations. The best quality GaN film was achieved when a nanoporous SiN x interlayer was grown on a rough layer, with the high-resolution X-ray diffraction rocking curve full width at half maximum for ( 1102 ) reflection decreasing to 223 arcs, and the total dislocation density reduced to less than 1.0×10 8 cm 2 . GaN films were grown on sapphire substrates by metal organic chemical vapor deposition. The quality of these films was investigated with high-resolution X-ray diffraction, atomic force microscopy, and cross-sectional transmission electron microscopy. A preference for the formation of half-loops to reduce threading dislocations was observed when an SiN x interlayer was grown on a rough layer.
