| 1 | PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. | 2 | Process mineralogy of Weishan rare earth ore by MLA显示文摘Mineral liberation analyser(MLA) was applied to quantitatively analyze the rare earth ore from Weishan in Shandong. Mineralogy parameters, such as mineral composition, occurrence states of rare earth elements(REEs) and valuable elements, mineral embedded grain size distribution, mineral association and liberation, are obtained. Results show that the contents of REEs and other valuable elements mainly contained in the ore were La 1.02 wt%, Ce 4.29 wt%, Pr 0.34 wt%, Nd 0.84 wt%, Sr 3.4 wt%and Ba 26.53 wt%, respectively. The REEs mainly occur in bastnaesite and carbocernaite in the form of independent mineral and the contents of bastnaesite and carbocernaite in the ore were 5.96 wt% and12.30 wt%, respectively. 67.34% of strontium occurs in carbocernaite and the rest occurs in celestobarite and strontianite mineral. 92.71% of barium occurs in barite. Liberation of main rare-earth minerals such as bastnaesite and carbocernaite is more than 80% when the grinding fineness is78.42% passing 74 μm. | 8 | MOF-derived zinc manganese oxide nanosheets with valence-controllable composition for high-performance Li storage显示文摘Zinc manganese oxide(ZMO)system represents a notable family of mixed transition metal oxides(MTMOs)because of their superiority of the high theoretical capacity,adequacy of natural content,and low cost.However,the methods to match both the reliable synthesis and the designable construction of large-sized two-dimensional(2D)ZMO nanosheets are still considered as grand challenges.Herein,we have successfully realized the preparation of 2D ZMO nanosheets with large lateral sizes up to~20 mm by simple pyrolysis of 2D metal–organic framework(MOF)nanosheets precursor.The growth mechanism of 2D MOF is proposed to be based on the lamellar micelles formed by polyvinyl pyrrolidone(PVP).The obtained 2D and porous ZMO nanosheets exhibit high specific capacity as well as good rate capability.More importantly,the as-prepared ZMO electrode shows a remarkable capacity increment upon cycling(from 832 mAh g^(-1) at the 2nd cycle to 1418 mAh g^(-1) at the 700th cycle,at 1 A g^(-1)).Through simple adjustment of the calcination temperature,the valence state of Mn species in the yielding ZMO samples can be fine-tuned.Through systematic investigation towards these ZMOs containing different Mn species,the extra specific capacity is revealed to be chiefly on account of the arising of the valence state of Mn upon the cycling process.Moreover,it is disclosed that the higher-valent Mn the pristine ZMO contains,the more additional capacity it gains upon cycling.We believe that this work will inspire more detailed analysis on the relationship between the valence state of Mn and extra capacity. | Yu Du Yihan Xu Weiwei Zhou Yaoyang Yu Xinzhou Ma Fei Liu Jinglong Xu Yongming Zhu | 2021 | Green Energy & Environment2021,6,5: | 1 |
