| 2 | The Expression and Location of Midkine in Gastric Carcinomas of Chinese Patients显示文摘Midkine (MK), a heparin-binding growth factor, can regulate cell growth, survival and differentiation. MK is expressed at high levels in a variety of human carcinomas. Recently, the urine and serum MK concentration was analyzed in gastric cancer patient. However, the association of the cytokine mRNA expression with the categorical clinicopathological variables of the tumors and the location of its protein expression in the tumor tissues are still elusive. MK mRNA expression from the surgically resected specimens of healthy gastric tissues (9 cases), gastric cancer tissues and the matched non-cancerous tissues adjacent to the cancer (37 cases) were assessed by reverse transcriptase-polymerase chain reaction (RT-PCR) and real-time PCR. Immunohistochemical analysis was performed to locate MK in gastric cancer. The expression of MK mRNA in gastric cancer was much higher in tumor tissues than that in the non-cancerous tissues and control tissue samples. And its expression was significantly associated with the pTNM stage and distant metastasis, but not with the differentiation grade, tumor size and nodal involvement. MK protein was ubiquitous in the tumor, especially in the adenoid part of tumors. In addition, it was found in the cytoplasm of tumor cells and highly concentrated in nucleus and nucleolus. The expression level and location of MK in gastric tumor tissues of Chinese Patients may be related to the tumor genesis and progression. | 9 | H.sinensis mycelium inhibits epithelial–mesenchymal transition by inactivating the midkine pathway in pulmonary fibrosis显示文摘The medical fungus Hirsutella sinensis has been used as a Chinese folk health supplement because of its immunomodulatory properties.Our previous studies established the antifibrotic action of Hirsutella sinensis mycelium(HSM)in the lung.The epithelial–mesenchymal transition(EMT)is involved in the pathogenesis of idiopathic pulmonary fibrosis.The present study investigates the role of HSM in mediating EMT during the development of pulmonary fibrosis.HSM significantly inhibits bleomycin(BLM)-induced pulmonary fibrosis by blocking the EMT.In addition,the expression levels of midkine are increased in the lungs of the BLM-induced group.Further analysis of the results indicates that the mRNA level of midkine correlated positively with EMT.HSM markedly abrogates the transforming growth factorβ-induced EMT-like phenotype and behavior in vitro.The activation of midkine related signaling pathway is ameliorated following HSM treatment,whereas this extract also caused an effective attenuation of the induction of EMT(caused by midkine overexpression)in vitro.Results further confirm that oral medication of HSM disrupted the midkine pathway in vivo.Overall,findings suggest that the midkine pathway and the regulation of the EMT may be considered novel candidate therapeutic targets for the antifibrotic effects caused by HSM. | Li Lu Haiyan Zhu Hailin Wang Huaping Liang Yayi Hou Huan Dou | 2021 | Frontiers of Medicine2021,15,2: | 3 |

| 12 | Formalin-induced astrocyte glutamate-glutamine cycle involved in rat spinal cord central sensitization显示文摘BACKGROUND:Central sensitization,a state of increased excitability of nociceptive neurons in the spinal dorsal horn following peripheral tissue injury and/or inflammation,is an important mechanism underlying hyperalgesia and neuropathic pain.Participation of the glutamate-glutamine cycle in central sensitization of the spinal cord remains poorly understood. OBJECTIVE:To determine whether the astrocyte-neuronal glutamate-glutamine cycle is involved in formalin-induced central sensitization in the spinal cord. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Institute of Orthopedics,Second Hospital,Lanzhou University,China from September 2007 to August 2008. MATERIALS:Methionine sulfoximine(MSO,0.1 mmol/L),glutamine(0.25 mmol/L),and formalin were used for this study. METHODS:A total of 43 male,Sprague Dawley rats,aged 4 months,were randomly assigned to a sham operation group(n = 6) and a model group(n = 37).Rats in the model group received intrathecal infusion in the spinal cord.7 days later,37 model rats were randomly divided into PBS, MSO,glutamine,MSO + glutamine and formalin subcutaneous injection alone groups.The PBS, MSO,glutamine,MSO + glutamine groups were respectively intrathecally injected with PBS,MSO, glutamine,MSO + glutamine(50μL each),and then infused with 10μL of saline.Rats from the sham operation group were not subjected to intrathecal infusion in the spinal cord.At 15 minutes after intrathecal injection,a rat model of formalin-induced inflammatory pain was established by subcutaneous injection of 5%formalin(50μL) in the left hindpaw. MAIN OUTCOME MEASURES:Changes in spontaneous nociceptive behavior(licking/biting or flinching) were observed following formalin injection into the rat hindpaw. RESULTS:Compared with the PBS group,duration of licking/biting was significantly shortened,and flinching frequency was significantly diminished in the MSO group(P<0.05).Compared with the MSO group,duration of licking/biting was significantly prolonged,and flinching frequency was significantly increased in the MSO + glutamine group(P<0.05).There was no significant difference in inflammatory pain behaviors among the sham operation,PBS,glutamine,MSO + glutamine,and formalin subcutaneous injection alone groups(P>0.05). | 13 | Research progress in cold region wetlands, China显示文摘China has some of the most abundant wetland resources in the world. Cold region wetlands cover more than 60% of the total natural wetlands in China and play an indispensable role in global climate regulation, water holding, uptake and emission of greenhouse gases, and biodiversity conservation. Because cold region wetlands are sensitive to climatic and environmental changes, it is important for ecological conservation and environmental management to summarize and analyze current research progress on these specific ecosystems. This paper reviews the focus of present studies on the typical cold region wetlands in the northeast region and the Qinghai-Tibet Plateau of China from several aspects as follows: types and distribution, responses of permafrost to climatic changes, uptake and emission of greenhouse gas, eco-hydrological processes, and vegetation succession. Our conclusions are: global warming has a long-term and serious impact on cold region permafrost; emission of greenhouse gases has great temporal and spatial heterogeneity; and hygrophytes in the cold region wetlands have been generally replaced by xerophytes, although it is still unclear whether the vegetation diversity index has increased or decreased.
